Tropical theileriosis is a significant tick-borne disease affecting cattle. For decades an empirical live attenuated vaccine has been the primary method of controlling disease. The vaccine is produced through prolonged culture of Theileria annulata schizont-transformed macrophages, but how loss of virulence occurs remains unclear. Notably attenuated (vaccine) macrophages display dampened dissemination potential compared with their original, virulent counterparts. In addition, parasite schizonts in attenuated macrophages have significantly lost their ability to differentiate into merozoites. Th

Bovine tropical theileriosis, caused by Theileria annulata, poses a significant threat to livestock health and productivity, particularly in endemic regions. Early and accurate diagnosis is crucial for effective disease management and control. Molecular techniques, such as conventional PCR (cPCR), reverse line blot (RLB) and real-time PCR (qPCR), offer high sensitivity and specificity but are limited by cost, technical expertise, and the requirement for specialized laboratory infrastructure. Multiplex PCR (mPCR), a variant of standard cPCR, offers the advantage of simultaneously detecting two

During 1973-1990 cattle immunization, in Iran, was induced by two strains within an interval of one month: first the milder and then the mild strain. Although this method of vaccination rendered satisfactory results in the field, yet production, maintenance in deep-freezers and transportation in liquid nitrogen particularly to remote areas of the country proved to be uneconomical and time consuming. Therefore, in order to reduce cost and save time, a new method involving only one local and live attenuated vaccine strain was sought. Reports received from different ecological areas of the countr

Western blot analysis of Theileria annulata antigens was carried out using sera collected from cattle which had been immunised and challenged with either T. annulata sporozoites or schizont-infected cells. Three antigens between 71 and 73 kDa proved to be common to the three stages of parasite studied: sporozoites, schizonts and piroplasms. An antigen was found at 32 kDa which was specific to T. annulata piroplasms. Results were reproducible using sera from Morocco and the UK. At least one of the proteins at 71-73 kDa, but not that at 32 kDa were also recognised by sera from animals infected w

Theileriosis is a disease of domesticated animals in tropical and subtropical countries and causes significant reductions in livestock productivity. The arid region of Punjab in Pakistan is notorious for the presence of the vector tick (Acari: Ixodidae) and tick-borne diseases, such as theileriosis and babesiosis. The distribution of Theileria annulata and T. orientalis in the Chakwal district of Punjab was determined by developing a multiplex recombinase polymerase amplification (RPA) assay as a scientific basis for formulating control strategies for bovine theileriosis. Specificity was evalu

Tropical theileriosis is a significant health risk for cattle especially in tropical and subtropical regions where it contributes to high mortality rates and economic losses. Tick-borne protozoan parasites belonging to Theileria genera are responsible for this infection. Theileriosis has been associated with abortion and stillbirth in cattle, primarily as an indirect consequence of maternal illness; however, this association has not been investigated in Pakistan. Therefore, the present study aimed to determine the molecular prevalence and distribution of Theileria species in cattle with a rece

Theileria annulata and Theileria parva macroschizont-infected bovine cells formed tumours at the inoculation site when injected subcutaneously into C.B.-17 scid mice. T. annulata tumours showed more vigorous growth than T. parva tumours. The tumours did not regress and infected cells spread to other tissues. Intraperitoneal injection of high doses of T. annulata-infected cells resulted in the development of ascites: the infected cells colonized abdominal organs, in particular mesenteric tissue. Low doses of cells did not establish when administered by this route. Evidence for a role for macrop

Theileria annulata, an apicomplexan parasite, causes lymphoproliferative disease in cattle and threatens the bovine industry in tropical and subtropical regions. To clarify life-cycle mechanisms and parasite-host interactions, we conducted high-resolution Orbitrap proteomic and phosphoproteomic analyses of the schizont and piroplasm stages. In total, we confidently identified and quantified 2,093 proteins; among these, 459 proteins were observed in a phosphorylated state, comprising 5,069 distinct phosphorylation sites. Functional enrichment analysis of differentially abundant proteins indicat

The growing emergence of resistance to current anti-theilerial agents necessitates the exploration of alternative approaches to drug discovery. This study evaluated the antiparasitic efficacy of 148 compounds derived from an epigenetic inhibitor library against the schizont stage of a Theileria annulata-infected cell line. Initial screening at a concentration of 10 µM identified 27 compounds exhibiting promising anti-theilerial activity. Further investigation, including determination of the 50% inhibitory concentration (IC50) and host cell cytotoxicity assay, highlighted seven highly effective

Theileriosis caused by Theileria lestoquardi (malignant ovine theileriosis) in sheep and Theileria annulata (topical theileriosis) in cattle, is a tick-borne hemoprotozoal disease that causes major economic losses in animal production caused by a tick-transmitted haemoprotozoan Theileria species, infecting various domestic and wild animals. It is a threat to the buffalo population in Pakistan due to high mortality rates and economic losses. Extensive mixed farming in Pakistan poses threats due to mixed infections and increased mortality rates. The current case report describes the mixed infect

Theileria annulata is a haemoprotozoan parasite that causes a cancer-like illness known as tropical theileriosis in cattle. In the course of analyzing the genetic diversity of T. annulata in Sri Lanka, we observed that merozoite-piroplasm surface antigen (tams1) and surface protein (tasp)-like gene sequences obtained from bovine blood DNA samples, which were PCR-positive for T. annulata, were conserved but shared low identity with T. annulata GenBank sequences. Moreover, the 18S rRNA sequences from the Sri Lankan samples contained ten unique single-nucleotide polymorphisms compared with all kn

Theileria annulata is an obligate intracellular parasite that induces bovine leukocyte transformation leading to uncontrolled proliferation and heightened dissemination of infected leukocytes. Early passage T. annulata-transformed macrophages are virulent, but with long-term culture, they become attenuated for dissemination. To investigate this phenotype from an epigenetic perspective, we focused on the important repressive histone marks, tri-methylated lysine 27 of histone H3 (H3K27me3) catalyzed by the Polycomb Repressive Complex 2. ChIP-seq revealed that the genomic distribution of H3K27me3

Theileriosis caused by Theileria annulata can be effectively prevented by vaccination with attenuated, cultured schizonts. Although these attenuated vaccines have been applied for a long time, not much is known about the fate of the vaccine strain in the field. Here, two experimental Spanish vaccine strains originating in Cádiz and Cáceres, and one Sudanese strain are studied to address the development of a carrier status and the infectivity for Hyalomma ticks. Moreover, the heterogeneity of the merozoite surface protein, Tams1, was analyzed in search for an attenuation marker. Using the sensi

We report the detection of Theileria annulata, the causative agent of tropical theileriosis, by PCR in blood samples obtained from carrier cattle. The assay employs primers specific for the gene encoding the 30-kDa major merozoite surface antigen of T. annulata. A 721-bp fragment was amplified from blood samples taken monthly from calves experimentally infected with one of four different stocks of T. annulata originating in either Mauritania, Portugal, Spain, or Turkey. At the end of the experiment, five animals carried the infection for 12 months and two animals remained infected for 15 month

The protozoan pathogen, Theileria annulata, is transmitted by ticks which is very detrimental to various breeds of cattle, leading to the development of theileriosis. Early diagnosis and control of the disease is a crucial aspect of preventing and treating any infection. In order to detect T. annulata on-site, we selected sporozoite surface antigen gene (SPAG) as the target gene to design Recombinase Polymerase Amplification (RPA) primers and crRNA to build a one-pot SHERLOCK method. The results showed the assay could specifically detect T. annulata infection, but have no-cross reaction with t

Bovine tropical theileriosis (Theileria annulata infection) is of great economic importance to several countries of Asia and Africa where millions of cattle are at risk of exposure to this disease. The authors describe symptoms, sequence of appearance of the parasite and pathology of the infection by three virulent strains of the parasite in fully susceptible calves. Symptoms observed were: enlargement of local lymph gland, fever, haemorrhages on visible mucous membranes and sometime on skin, anaemia and jaundice particularly in subacute and chronic forms of the disease. The parasite "schizont

Babesia spp., Theileria spp. and Anaplasma spp. are significant tick-borne pathogens of livestock globally. In this study, we investigated the presence and distribution of Babesia bigemina, Theileria annulata, Theileria orientalis and Anaplasma marginale in cattle from 6 provinces of Turkey using species-specific PCR assays. The PCR were conducted using the primers based on the B. bigemina rhoptry-associated protein 1a (BbiRAP-1a), T. annulata merozoite surface antigen-1 (Tams-1), T. orientalis major piroplasm surface protein (ToMPSP) and A. marginale major surface protein 4 (AmMSP4) genes, re

In a series of experiments, sporozoite stabilates of a Theileria lestoquardi (Lahr) and a T. annulata (Ankara) stock prepared from Hyalomma anatolicum anatolicum ticks, were used to examine the infectivity of both parasite species for sheep and cattle and to study the development of cross-immunity between these parasite species. In the first experiment sheep and cattle were inoculated with T. lestoquardi sporozoites. Surviving animals and naive sheep and cattle were, in the second experiment, inoculated with T. annulata. In the third experiment, naive sheep and sheep previously infected with T

Theileria annulata, the causative agent of tropical theileriosis, poses a significant threat to cattle industries, a challenge further intensified by increasing drug resistance. In this study, the food-grade phenolic compound octyl gallate (OG) was evaluated for its antitheilerial activity and identified as a potent and selective inhibitor of T. annulata. Comparative screening of gallic acid (GA) derivatives revealed that OG exhibited the strongest activity against T. annulata-transformed cell lines, with IC50 values of 374.0 nM in TaNM cells and 448.7 nM in the buparvaquone-resistant TaXJS ce
