Several TRP channels exhibit highly temperature-dependent gating properties, which leads to steep changes in depolarising current upon either cooling or heating. Based on this characteristic feature, these so-called "thermoTRPs" have been widely studied with the aim to elucidate their potential key role as thermosensors in the somatosensory system and to understand the basis of their high thermal sensitivity. In this chapter, I provide a brief critical overview of current knowledge on the role of TRP channels in thermosensing and on the thermodynamic and molecular basis of their steep temperat

Somatosensory neurons can sense external temperature by converting sensation of temperature information to neural activity via afferent input to the central nervous system. Various populations of somatosensory neurons have specialized gene expression, including expression of thermosensitive transient receptor potential (TRP) ion channels. Thermosensitive TRP channels are responsible for thermal transduction at the peripheral ends of somatosensory neurons and can sense a wide range of temperatures. Here we focus on several thermosensitive TRP channels including TRPV1, TRPV4, TRPM2, TRPM3, TRPM8

The thermal sense of triatomine bugs, vectors of Chagas disease, is unique among insects. Not only do these bugs exhibit the highest sensitivity to heat known in any animal up to date, but they can also perceive the infrared radiation emitted by the body of their warm-blooded hosts. The sensory basis of this capacity has just started to be unravelled. To shed additional light on our understanding of thermosensation, we initiated an analysis of the genetic basis of the thermal sense in Rhodnius prolixus. We tested the hypothesis that a TRPV (transient receptor potential vanilloid) channel recep

Objectives: Sweating, hot flushes, and blushing are symptoms frequently reported by individuals with anxiety disorders. They represent important reinforcers of anxiogenic cognitions and behaviours. One system that may be involved in the manifestation of these symptoms is the thermosensory/thermoregulatory system. The aim of the present study was to investigate to what extent individuals with anxiety disorders are characterised by alterations in this system. Methods: PubMed and PsycINFO were systematically searched. Studies were eligible if they (i) assessed individuals with anxiety disorders,

A patient recently presented with long-standing severe itching originating from lichen sclerosus et atrophicus at the vulva area. We successively treated her using a topical formulation of TRPM8 agonist which produces a cooling sensation. The cooling sensation, an afferent sensory perception in various skin neuronal pathways, could be a useful mechanism to relieve an itchy sensation in various skin disorders. Mechanoreceptors are related to touch vibration and pressure sensations and have a special morphology where the nerve endings are optimized to receive sensory inputs. However, unmyelinate

Opportunistic pathogens switch from a commensal to pathogenic state by sensing and responding to a variety of environmental cues, including temperature fluctuations. Minor temperature oscillations can alert the pathogen to a changing niche ecosystem, necessitating efficient sensing and rapid integration to trigger behavioral change. This is typically achieved through master regulators, dictating pleiotropic phenotypes. Here, we uncover a pivotal role of minor temperature shifts in transition of Streptococcus pneumoniae (SPN) from commensal to virulent lifestyles, mediated via an RNA thermosens

Skin-penetrating parasitic nematodes infect approximately one billion people worldwide and are a major source of neglected tropical disease [1-6]. Their life cycle includes an infective third-larval (iL3) stage that searches for hosts to infect in a poorly understood process that involves both thermal and olfactory cues. Here, we investigate the temperature-driven behaviors of skin-penetrating iL3s, including the human-parasitic threadworm Strongyloides stercoralis and the human-parasitic hookworm Ancylostoma ceylanicum. We show that human-parasitic iL3s respond robustly to thermal gradients.

Temperature is a biologically active physical variable in skeletal tissues, but apparently contradictory findings often arise because thermal magnitude is reported without the accompanying time, spatial, mechanical, and pathological context. This critical review synthesizes evidence on temperature-dependent regulation of bone and cartilage using a thermal-dose framework that distinguishes local mild hypothermia, chronic systemic cold exposure, adaptive mild heating, and hyperthermic injury. Representative studies indicate that no universal boundary temperature exists: transient exposures of ap

Plant thermomorphogenesis represents the adjustment of growth to warm ambient temperatures. The mechanism has long been organised around the transcription factor PHYTOCHROME-INTERACTING FACTOR 4 (PIF4) and the thermosensors that act upon it. Internal circadian control and external light intensity and quality have been the primary contexts that gate thermosensor-PIF signal transduction. Here, we argue that cellular oxygen and the reduction/oxidation (redox) state of the cell form additional layers that gate or calibrate thermosensing signal transduction. Current evidence points to discrete mech

The oral cavity is exposed to a remarkable range of noxious and innocuous conditions, including temperature fluctuations, mechanical forces, inflammation, and environmental and endogenous chemicals. How such changes in the oral environment are sensed is not completely understood. Transient receptor potential (TRP) ion channels are a diverse family of molecular receptors that are activated by chemicals, temperature changes, and tissue damage. In non-neuronal cells, TRP channels play roles in inflammation, tissue development, and maintenance. In somatosensory neurons, TRP channels mediate nocice

Axons are the initial sites of thermal signal detection and transduction, but their localized response to heat remains poorly characterized. Here, we present a photothermal nerve-on-a-chip platform that reconstructs in vitro the key elements of axonal thermosensation. By integrating graphene-based photothermal heaters with microelectrode arrays inside axon-guiding microchannels, the platform enables localized, millisecond-scale thermal stimulation and simultaneous extracellular recording of action potentials. Using this platform, we uncovered rapid and reproducible heat-evoked responses in rat

While thermosensation from external environment has been extensively studied, physiological responses to temperature changes inside the body and the underlying regulatory mechanisms are less understood. As a critical link between body and brain that relays visceral organ information and regulates numerous physiological functions, the vagus nerve has been proposed to mediate diverse visceral thermal reflexes and indirectly regulate body temperature. However, the precise role of the vagus nerve in body thermal responses or visceral organ-related thermoregulation is still under debate due to exte

Female yellow-fever mosquitoes, Aedes aegypti, are obligate blood-feeders and vectors of the pathogens that cause dengue fever, yellow fever and Chikungunya. This feeding behavior concludes a series of multisensory events guiding the mosquito to its host from a distance. The antennae and maxillary palps play a major role in host detection and other sensory-mediated behaviors. Compared to the antennae, the maxillary palps are a relatively simple organ and thus an attractive model for exploration of the neuromolecular networks underlying chemo- and mechanosensation. In this study, we surveyed th

TRP cation channels transduce mechanical, thermal, and pain-related inflammatory signals. In this issue of Cell, it is reported that TRPA1 has a central role in the pain response to endogenous inflammatory mediators and to a diverse array of volatile irritants, including those found in tear gas and garlic. In contrast, mechano- and thermosensation are normal in TRPA1-deficient mice.

Ophthalmic, maxillary, and mandibular branches of the trigeminal nerve provide sensory innervation to orofacial tissues. Trigeminal sensory neurons respond to a diverse array of sensory stimuli to generate distinct sensations, including thermosensation, mechanosensation, itching, and pain. These sensory neurons also detect the distinct sharpness or pungency of many foods and beverages. This SnapShot highlights the transduction ion channels critical to orofacial sensation.

OBJECTIVES: A no-flap design based on actual tongue anatomy has been reported to achieve precise reconstruction, but large carcinologic tongue defects remain extremely challenging to treat. The aim of this pilot study was to explore an anatomy-based flap design for precision subtotal tongue reconstruction.
MATERIAL AND METHODS: Thirty-five patients were randomly divided into two groups, a "five-points eight-line-segments" (FIPELS) technique flap design group (18 patients) and a conventional surgery group (17 patients), to undergo subtotal tongue reconstruction. All patients received an antero

The transient receptor potential (TRP) family of cation channels are evolutionarily conserved proteins with critical roles in sensory physiology. Despite extensive studies in model species, knowledge of TRP channel functional diversity and physiological impact remains limited in many non-model insect species. To assess the TRP channel repertoire in a non-model agriculture pest species (Lygus hesperus), publicly available transcriptomic datasets were mined for potential homologs. Among the transcripts identified, 30 are predicted to encompass complete open reading frames that encode proteins re

Plants coordinate their growth and developmental programs through dynamic communication in the core of the nucleus, enabling them to adapt to persistently high-temperature environments. Upon heat stress, plants primarily perceive and transduce thermal signals between the nucleus and other organelles, a process defined as nuclear-organellar communication. The published research on plants in response to heat environments mainly focuses on sensing heat physical signals from the plasma membrane (PM)/cytosol to the nucleus, eliciting nuclear transcriptional reprogramming and decoding organelle-deri

Three different sensory loss tests, for anaesthesia to light touch, for diminished pain sensation and for loss of thermosensation, were compared with histopathological examination results in the diagnosis of suspected tuberculoid leprosy in 120 individuals with 126 lesions. Though none of the 3 tests used in this study was found to be strikingly superior to any of the others, the results indicate potentially important differences in their usefulness in different subgroups of suspected patients. The methodological problems inherent in such studies are discussed.
