Light-induced stimulation of carbonic anhydrase activity in pea thylakoids.

Stimulation of the bicarbonate dehydration reaction in thylakoid suspension under conditions of saturating light at pH 7.6-8.0 was discovered. This effect was inhibited by nigericin or the lipophilic carbonic anhydrase (CA) inhibitor ethoxyzolamide (EZ), but not by the hydrophilic CA inhibitor, acetazolamide. It was shown that the action of EZ is not caused by an uncoupling effect. It was concluded that thylakoid CA is the enzyme utilizing the light-generated proton gradient across the thylakoid membrane thus facilitating the production of CO(2) from HCO(3)(-) and that this enzyme is covered f

Thylakoids: A Novel Food-Derived Supplement for Obesity - A Mini-Review.

Nowadays, overweight and obesity are major epidemic health problems that can bring about some other health issues such as cardiovascular disease which is the first cause of mortality worldwide. Thylakoids are disc-like membranes responsible for photosynthetic light reactions in chloroplasts of green plants. Although only a few animal and human studies have been conducted regarding the impact of thylakoids on overweight- and obesity-related factors, all of them have resulted in positive outcomes. These outcomes are as follows: increment of satiety response; suppression of hunger sensations, par

Chlorophyll-protein and polypeptide composition of Mn-deficient sugar beet thylakoids.

The chlorophyll-protein and polypeptide composition of manganese deficient and control sugar beet thylakoids was examined using three different detergent-electrophoresis systems. On a per chlorophyll basis, manganese deficiency reduced the amounts of CPa complex (separated by sodium dodecylsulfate (SDS)-polyacrylamide gel electrophoresis), and CP 47 and CP 43 complexes (separated by octylglucoside/SDS-polyacrylamide gel electrophoresis) without decreasing the amounts of light harvesting complexes. Lithium dodecylsulfate/Triton X-100 polyacrylamide gel electrophoresis showed that manganese defi

Lumen alkalization under elevated bicarbonate concentration outside isolated thylakoids of higher plants.

Illumination causes alkalization of chloroplast stroma, which is accompanied with a substantial increase in stromal bicarbonate concentration there. Whether bicarbonate itself contributes to regulation of thylakoid lumen pH has remained unclear. Here we show that, increasing bicarbonate concentration in the suspension of the isolated pea thylakoids increases luminal pH and stimulates electron transport through the photosynthetic electron transport chain. Addition of bicarbonate stimulated the electron transport in the functionally isolated photosystem I in the case when a pH sensitive electron

Protein identification of purified particles isolated from spinach thylakoids by deoxycholate electrophoresis.

Three thylakoid complexes were isolated by deoxycholate preparative electrophoresis. The protein composition of each fraction was analyzed by SDS analytical electrophoresis. No protein of the PS 1 enriched fraction (fraction 1) was found in the PS 2 enriched fraction (fraction 2) and inversely. The antenna complex (fraction 3) did not have any contamination by proteins of fraction 1 or fraction 2. Fraction 1 was mainly composed of the CP1, the reaction center complex of the PS1, and by low molecular weight proteins, previously found in other PS 1 preparations. Tentative assignments of these pr

Effect of phosphorylation on the thermal and light stability of the thylakoid membranes.

Higher plant thylakoid membranes contain a protein kinase that phosphorylates certain threonine residues of light-harvesting complex II (LHCII), the main light-harvesting antenna complexes of photosystem II (PSII) and some other phosphoproteins (Allen, Biochim Biophys Acta 1098:275, 1992). While it has been established that phosphorylation induces a conformational change of LHCII and also brings about changes in the lateral organization of the thylakoid membrane, it is not clear how phosphorylation affects the dynamic architecture of the thylakoid membranes. In order to contribute to the eluci

Heterogeneous origin of carbonic anhydrase activity of thylakoid membranes.

Carbonic anhydrase activities of pea thylakoids as well as thylakoid fragments enriched either in Photosystem 1 (PS1-membranes) or Photosystem 2 (PS2-membranes) were studied. The activity of PS1-membranes if calculated on chlorophyll basis was much higher than the activity of PS2-membranes. Acetazolamide, a non-permeable inhibitor of carbonic anhydrases, increased carbonic anhydrase activity of PS2-membranes at concentrations lower than 10(-6) M and suppressed this activity only at higher concentrations. A lipophilic inhibitor of carbonic anhydrases, ethoxyzolamide, effectively suppressed the

The effect of metal chelators on the production of hydroxyl radicals in thylakoids.

The effect of metal chelators (EDTA, DTPA and Desferal) on the metal-catalyzed decomposition of hydrogen peroxide was studied using EPR spin-trapping spectroscopy. The formation of hydroxyl radicals (OH*) in both chemical (Fenton reaction) and biological (thylakoids) systems was stimulated by EDTA. DTPA promoted the generation of OH* in the presence of strong reducing agents, whereas in their absence it acted as an antioxidant. Desferal suppressed OH* production even in the presence of reductants. In our study, we have shown that metal chelators can both stimulate and suppress the formation of

Correlation between membrane-localized protons and flash-driven ATP formation in chloroplast thylakoids.

Flash-driven ATP formation by spinach chloroplast thylakoids, using the luciferin luminescence assay to detect ATP formed in single turnover flashes, was studied under conditions where a membrane protein amine buffering pool was either protonated or deprotonated before the beginning of the flash trains. The flash number for the onset of ATP formation was delayed by about 10 flashes (from 15 to about 25) when the amine pool was deprotonated as compared to the protonated state. The delay was substantially reversed again by reprotonating the pool upon application of 20-30 single-turnover flashes

How Did Thylakoids Emerge in Cyanobacteria, and How Were the Primary Chloroplast and Chromatophore Acquired?

The emergence of thylakoid membranes in cyanobacteria is a key event in the evolution of all oxygenic photosynthetic cells, from prokaryotes to eukaryotes. Recent analyses show that they could originate from a unique lipid phase transition rather than from a supposed vesicular budding mechanism. Emergence of thylakoids coincided with the great oxygenation event, more than two billion years ago. The acquisition of semi-autonomous organelles, such as the mitochondrion, the chloroplast, and, more recently, the chromatophore, is a critical step in the evolution of eukaryotes. They resulted from pr

Cardiomyocyte Membrane-Camouflaged Spinach Nano-Thylakoids Enhance Myocardial Energy Metabolism and Ameliorate Heart Failure.

BACKGROUND: Energy deficiency and mitochondrial dysfunction are key features of heart failure (HF), while current therapies only indirectly improve cardiac metabolism. We developed a biomimetic photoactivatable nanoplatform, cardiomyocyte membrane-camouflaged photosynthetic nano-thylakoids (MM-NTU), to enhance myocardial bioenergetics. METHODS AND RESULTS: Nano-thylakoids were isolated from spinach chloroplasts, downsized, and coated with cardiomyocyte membranes to improve cardiac association and retention. In transverse aortic constriction (TAC) rats, intramyocardial MM-NTU administration co

Multiple sources of carbonic anhydrase activity in pea thylakoids: soluble and membrane-bound forms.

Carbonic anhydrase (CA) activity of pea thylakoids, thylakoid membranes enriched with photosystem I (PSI-membranes), or photosystem II (PSII-membranes) as well as both supernatant and pellet after precipitation of thylakoids treated with detergent Triton X-100 were studied. CA activity of thylakoids in the presence of varying concentrations of Triton X-100 had two maxima, at Triton/chlorophyll (triton/Chl) ratios of 0.3 and 1.0. CA activities of PSI-membranes and PSII-membranes had only one maximum each, at Triton/Chl ratio 0.3 or 1.0, respectively. Two CAs with characteristics of the membrane

Microdomains heterogeneity in the thylakoid membrane proteins visualized by super-resolution microscopy.

The investigation of spatial heterogeneity within the thylakoid membrane (TM) proteins has gained increasing attention in photosynthetic research. The recent advances in live-cell imaging have allowed the identification of heterogeneous organisation of photosystems in small cyanobacterial cells. These sub-micrometre TM regions, termed microdomains in cyanobacteria, exhibit functional similarities with granal (Photosystem II dominant) and stromal (Photosystem I dominant) regions observed in TM of higher plants. This study delves into microdomain heterogeneity using super-resolution Airyscan-bas

VIA1 is a conserved regulator of thylakoid membrane integrity that acts through VIPP1.

Thylakoid membranes are indispensable for oxygenic photosynthesis, yet the mechanisms that protect these membranes from photooxidative damage remain poorly understood. By screening poorly characterized proteins induced during the chloroplast unfolded protein response, we identify VIA1 as an essential factor for preserving thylakoid integrity under high light in the model green alga Chlamydomonas reinhardtii. Loss of VIA1 causes hypersensitivity to photo-oxidative stress and rapid thylakoid swelling. VIA1 localizes to thylakoid membranes and directly binds Vesicle-Inducing Protein in Plastids 1

Lipid Phase Behaviour of the Curvature Region of Thylakoid Membranes of Spinacia oleracea.

Thylakoid membranes (TMs) of oxygenic photosynthetic organisms are flat membrane vesicles, which form highly organised, interconnected membrane networks. In vascular plants, they are differentiated into stacked and unstacked regions, the grana and stroma lamellae, respectively; they are densely packed with protein complexes performing the light reactions of photosynthesis and generating a proton motive force (pmf). The maintenance of pmf and its utilisation for ATP synthesis requires sealing the TMs at their highly curved regions (CRs). These regions are devoid of chlorophyll-containing protei

Dual-Mediator Redox Cascade Enhances Photoelectron Harvesting from Thylakoid Membranes.

Natural light reactions carried out in the thylakoid membrane (TM) generate abundant photoelectrons from water and sunlight, thereby providing a green platform for solar energy conversion. However, creating an efficient pathway for targeted photoelectron transfer at the TM-electrode interface remains challenging. Most studies have focused on extending the electrochemical active area through electrode modification to increase the photocurrent. Here, we establish a redox cascade between TM and the electrode using two newly identified mediators. One mediator is NADP+, the terminal electron accept

Thylakoid membrane transporter SecE1 is required for chloroplast development and photosynthetic function in Arabidopsis.

Chloroplast biogenesis depends on protein transport, pigment biosynthesis, and thylakoid membrane assembly, which are coordinated through anterograde (nucleus-to-plastid) and retrograde (plastid-to-nucleus) signaling. The chloroplast Sec translocon mediates the import of nuclear-encoded precursor proteins across the thylakoid membrane, yet the regulatory role of its core subunit SecE1 in chloroplast development and photosynthetic performance remains incompletely understood. Here, we characterized a chlorotic Arabidopsis mutant with partial loss of SecE1 function. Compared with the wild type an

Enhanced thermal stability of the thylakoid membranes from spruce. A comparison with selected angiosperms.

Recently, we have found that thermal stability of photosystem II (PSII) photochemistry in spruce needles is higher than in other plants (barley, beech) cultivated under the same temperatures. In this work, temperature dependences of various characteristics of PSII organization were studied in order to obtain complex information on the thermal stability of PSII function and organization in spruce. Temperature dependency of circular dichroism spectra revealed by about 6 °C higher thermal stability of macrodomain organization in spruce thylakoid membranes in comparison with Arabidopsis and barley
