Known for its valuable agarwood, Aquilaria malaccensis Lam. is an evergreen tropical forest tree species endemic to the Indo-malesian region. Indiscriminate damaging and harvesting of the trees in the wild have resulted in it being listed in the CITES Appendix II for controlled trade and in the IUCN Red List as 'Vulnerable (VU)'. In this study, the complete chloroplast genome of A. malaccensis was assembled using data from high-throughput Illumina sequencing. The chloroplast genome was 174,832 bp in size, which included two inverted repeat regions of 42,091 bp each, separated by a large single

The species of Aquilaria and Gyrinops are renowned for being the primary sources of Agarwood, a valued resin, produced by these trees in response to fungal infections or physical damage. The major derivatives of Agarwood are wood chips and oil which are used in perfumes, incenses, cosmetics, and medicines. Due to high international trade and exploitation in the wild, the members of both genera are listed under Appendix II of the Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES). Aquilaria khasiana, endemic to the Indian state of Meghalaya, is a less-studie

Despite the success of combination antiretroviral therapy (cART), HIV persists in low- and middle-income countries (LMIC) due to emerging drug resistance and insufficient drug accessibility. Furthermore, cART does not target latently-infected CD4+ T cells, which represent a major barrier to HIV eradication. The “shock and kill” therapeutic approach aims to reactivate provirus expression in latently-infected cells in the presence of cART and target virus-expressing cells for elimination. An attractive therapeutic prototype in LMICs would therefore be capable of simultaneously inhibiting viral r

Lignans are phenylpropanoid dimers in which the monomers are linked by the central carbon atoms of their propyl side chains. In addition to the biosynthesis of biologically active lignans, the unique stereochemical mechanisms of biosynthetic reactions have been of long-standing interest. The enantiomeric composition of pinoresinol and lariciresinol-the two lignans farthest upstream in the lignan biosynthetic pathway-varies greatly among the plant species; these compounds are not enantiomerically pure and are mixtures of both enantiomers. The enantiomeric composition of the next intermediate, s

Eight novel organic sulfur compounds featuring an unprecedented structure among natural organosulfur molecules are reported. The identified compounds, named dirchromones (1-8), were isolated from a dichloromethane extract of the roots, bark, and wood of Dirca palustris, an endemic shrub of eastern North America. Their identification was based on thorough NMR, IR, and HRMS spectroscopic data interpretation. These compounds showed cytotoxic and mild Gram-positive antibacterial activities in vitro, while being inactive against Gram-negative Escherichia coli and the yeast Candida albicans. This is

The mountains of Southwest China comprise a significant large mountain range and biodiversity hotspot imperiled by global climate change. The high species diversity in this mountain system has long been attributed to a complex set of factors, and recent large-scale macroevolutionary investigations have placed a broad timeline on plant diversification that stretches from 10 million years ago (Mya) to the present. Despite our increasing understanding of the temporal mode of speciation, finer-scale population-level investigations are lacking to better refine these temporal trends and illuminate t

Agarwood is a type of resinous wood found in the trunks of Aquilaria, Gonystylus, and Gyrinops species [1]. High-quality agarwood is extraordinarily expensive and therefore its source plant species have become depleted due to exploitation. In 2005, these species were added to Appendix II of the Convention on International Trade in Endangered Species of Wild Fauna and Flora [1]. Because these wild agarwood resources have become depleted, commercial production of agarwood has long been a desirable goal. In addition, inauthentic agarwood is sometimes produced from non-agarwood species. Few report

Agarwood is the resinous part of the injured tree of the Aquilaria species of the Thymelaeceae family, Qi-Nan is a type of agarwood that forms easily and benefits from a shortened inducing time, noted for features like its substantial resin, (2-phenylethyl) chromone and 2-[2-(4-methoxyphenyl) ethyl] chromone content. Although the content of agarotetrol in Qi-Nan is very low, it is still an important indicator for the agarwood quality. A validated method was developed to precisely quantify both agarotetrol and two aforementioned chromones, as well as ethanal extracts, within Qi-Nan to explore h

Daphne retusa belongs to the Daphne genus of Thymelaeaceae, which is widespread in northwest China, and its roots and stem bark are commonly used in Chinese folk medicine to treat rheumatism and congestion. In this study, four new (1-4) and eleven known (5-15) flavones were isolated from the branches and leaves of Daphne retusa, including six monomeric flavones (2-7) and nine dimeric flavones (1, 8-15). On the basis of chemical evidence and spectral data analysis (UV, ECD, NMR, and HR-ESI-MS), the structures of compounds 1-4 was elucidated. Furthermore, all compounds have been tested for their

Dried flowers of Daphne genkwa Sieb. et Zucc. (Thymelaeaceae) are a Chinese herbal medicine used as an abortifacient with purgative, diuretic and anti-inflammatory activities. However, the activity of this medicine against enteroviral infections has not been investigated. The water-extract of dried buds of D. genkwa Sieb. et Zucc. (DGFW) was examined against various strains of enterovirus 71 (EV71) by neutralization assay, and its initial mode of action was characterized by time-of-addition assay followed by attachment and penetration assays. Pretreatment of DGFW with virus abolished viral rep

The endangered tree species of the Aquilaria genus produce agarwood, a high value material produced only after wounding; however, conservation of Aquilaria seeds is difficult. The B3 transcription factor family has diverse important functions in plant development, especially in seed development, although their functions in other areas, such as stress responses, remain to be revealed. Here germination tests proved that the seeds of A. sinensis were recalcitrant seeds. To provide insights into the B3 superfamily, the members were identified and characterized by bioinformatic approaches and class

PURPOSE OF REVIEW: Combination antiretroviral therapy (cART) does not act on latent HIV reservoirs, and no latency-reversing agent (LRA) to date consistently reduces viral reservoirs in humans. In Sub-Saharan Africa and elsewhere, complementary and alternative medicines (CAM) are traditionally used to manage HIV/AIDS, including a subset with LRA properties.
RECENT FINDINGS: Several plants from the Euphorbiaceae and Thymelaeaceae families have been recently documented for traditional HIV/AIDS management and contain LRAs that function through protein kinase C activation. These CAM are increasin

Since plants are an important source of anticancer drugs, we have carried out a random screening for selective anticancer activity of 57 extracts from 45 plants collected in Grazalema Natural Park, an area in the South of Spain of high plant diversity and endemism. Using lung cancer cells (A549) and lung non-malignant cells (MRC-5), we found that several extracts were more cytotoxic and selective against the cancer cells than the standard anticancer agent cisplatin. Five active extracts were further tested in cancer and normal cell lines from other tissues, including three skin cell lines with

Agarwood is a highly valued aromatic resinous wood formed in Aquilaria species following stress or infection, yet the putative microbial drivers of its quality remain poorly understood, particularly outside its native range. In this study, we investigated the bacterial and fungal communities associated with agarwood produced from Aquilaria crassna Pierre ex Lecomte planted in French Guiana and examined their relationships with volatile chemical compounds relevant to agarwood fragrance. Using high‑throughput sequencing and comprehensive chemical profiling, we characterized microbial community c

Three highly modified sesquiterpenoids, stelleroids A-C (1-3, respectively), were isolated from the roots of Stellera chamaejasme. Biosynthetically, these molecules are derived from the 5/7-fused bicyclic guaiane, involving a series of skeletal rearrangements, leading to the formation of an adamantane-like 12-oxa-tetracyclo[6.2.1.13,10.03,7]dodecane scaffold for 1, a tricyclo[6.2.1.02,6]undecane backbone for 2, and a tetracyclo[6.2.2.01,7.05,11]dodecane backbone for 3. The structures of 1-3 were elucidated through a combination of HR-ESI-MS, NMR, quantum chemical calculations, and X-ray diffra
