3,5-diiodo-L-thyronine: A Possible Pharmacological Agent?

Overweight and obesity related metabolic disorders, commonly sharing a pathogenic excess of body adiposity, are world-wide epidemic leading to increasing morbidity and mortality. The related conditions include, among the others, liver steatosis, insulin resistance, and cardiovascular risk. Effective and safe anti-obesity drugs are still needed. Likely without undesirable side effects, an ideal treatment should be able to counteract the numerous causes associated with excess of body adiposity putatively modulating the delicate balance between feeding and energy expenditure, untimely controlling

Comparative effects of 3,5-diiodo-L-thyronine and 3,5,3'-triiodo-L-thyronine on mitochondrial damage and cGAS/STING-driven inflammation in liver of hypothyroid rats.

Maintaining a well-functioning mitochondrial network through the mitochondria quality control (MQC) mechanisms, including biogenesis, dynamics and mitophagy, is crucial for overall health. Mitochondrial dysfunction caused by oxidative stress and further exacerbated by impaired quality control can trigger inflammation through the release of the damage-associated molecular patterns (mtDAMPs). mtDAMPs act by stimulating the cyclic GMP-AMP synthase (cGAS) stimulator of interferon genes (STING) pathway. Recently, aberrant signalling of the cGAS-STING axis has been recognised to be closely associate

A radioimmunoassay for measurement of thyronine and its acetic acid analog in urine.

A sensitive and specific RIA has been developed to measure thyronine (To) in urine. The RIA used an anti-To antibody obtained from a rabbit immunized with a L-To-human serum albumin conjugate and [3H]To as the radioligand. The acetic acid analog of To (ToAc), that is the diphenyl structure with an acetic acid side-chain, cross-reacted strongly with the antibody. Relative to To, it cross-reacted 160% in phosphate-buffered saline, pH 7.4, and 100% in 0.075 mol/L barbital buffer, pH 8.6, containing sodium salicylate (final concentration, 8 mg/mL). The latter conditions were employed for the RIA,

Thyronines and thyronamines: dual modulators of synaptic plasticity, cognitive decline, and geriatric depression in the aging brain.

The aging brain induces cognitive deterioration and geriatric depression via synaptic dysfunction, neuroinflammation, and bioenergetic failure. Recent evidence identifies thyroid hormones (thyronines: T3, T4) and their decarboxylated derivatives (thyronamines, especially 3-iodothyronamine, T1AM) as essential endogenous regulators of these processes. Thyronines influence both genomic and non-genomic pathways in glutamatergic, GABAergic, cholinergic, and monoaminergic signaling, thereby modulating learning and memory. With aging, diminished expression of thyroid hormone transporters (MCT8, OATP1

Effect of 3,5-di-iodo-L-thyronine on the mitochondrial energy-transduction apparatus.

We examined the effect of a single injection of 3,5-di-iodo-L-thyronine (3,5-T2) (150 microg/100 g body weight) on the rat liver mitochondrial energy-transduction apparatus. We applied 'top-down' elasticity analysis, which allows identification of the site of action of an effector within a metabolic pathway. This kinetic approach considers oxidative phosphorylation as two blocks of reactions: those generating the mitochondrial inner-membrane potential (DeltaPsi; 'substrate oxidation') and those 'consuming' it ('proton leak' and 'phosphorylating system'). The results show that 1 h after the inj

Effects of Thyroxine (T4), 3,5,3'-triiodo-L-thyronine (T3) and their Metabolites on Osteoblast Differentiation.

Studies involving human genetic mutations and mutant mouse models have provided irrevocable evidence for a key role for thyroid hormones (THs) in the regulation of skeletal growth. While T3 binds to TH receptors with higher affinity than T4, T4 occupied TH receptors have also been reported in the nucleus under euthyroid conditions raising the possibility that T4 bound nuclear receptors may be biologically relevant in thyroid syndromes with elevated free T4 and reduced T3 levels. We, therefore, evaluated the direct effects of T4, T3, and their metabolites (rT3 and T2) in stimulating osteoblast

Preferential binding of tri-substituted thyronine analogs by bullfrog tadpole tail fin cytosol.

The relative strength of binding of several triiodothyronine (T3) analogs by cytosol prepared from the tail fin of the bullfrog tadpole has been tested and compared with their thyromimetic activity in tail tissue. In competitive binding experiments, four tri-substituted analogs were bound much more strongly than two tetra-derivatives. Correlation between binding and relative thyromimetic activity was observed only for three triiodo-substituted compounds, triiodothyronine, its methylene bridge analog and the acetic acid analog. The affinity for T3 was 250 times that of T4, making it unlikely th

[A 125I-thyronine derivative of high specific activity and radiochemical stability (pseudo-T4) (author's transl)].

Under the described reaction conditions for the synthesis of 125I-labelled thyroid hormones by the chloramine-T method, electrophilic substitution of the phenolic ring at positions 3' and 5' is accompanied by an exchange reaction. The proportions of mono- and diiodo-thyronine derivatives formed in this reaction depend on the specific activity of the 125I starting material. Doubly labelled molecules are far less stable than mono products, because the radioactive decay of one iodine atom is followed by destruction of the molecule itself, and the remaining iodine atom is released as I-. Transplacental stimulation of lung development in the fetal rabbit by 3,5-dimethyl-3'-isopropyl-L-thyronine.

The effect of thyroid hormone on maturation of fetal rabbit lung was studied with maternal treatment using 3,5-dimethyl-3'-isopropyl-L-thyronine (DIMIT), a synthetic analogue of triiodothyronine. To investigate the in vivo kinetics and distribution of DIMIT, we prepared [3H]DIMIT and injected both pregnant rats (18-21 d gestation) and rabbits (25 d gestation). In the rat, maximal concentrations of radioactivity in maternal plasma, fetal plasma, and amniotic fluid occurred within 10 min, 1-2 h, and 4-6 h, respectively, after intramuscular injection. After 7 h the concentration of radioactivity

Comparison of high-performance liquid chromatographic detection methods for thyronine and tyrosine residues in toxicological studies of the thyroid.

Four high-performance liquid chromatographic methods for the detection of thyroid hormones (iodinated thyronines) and precursors (iodinated tyrosines) have been developed and evaluated. Two methods consist of direct determination of the parent compounds with detection at ultraviolet wavelength (230 nm) and with electrochemical detection. The two other methods consist of a pre-column derivatization (with fluorenylmethyl chloroformate and dabsyl chloride) prior to high-performance liquid chromatographic analysis. The various methods were evaluated based on their practical use and sensitivity. Th

Potential contribution of translational factors to triiodo-L-thyronine-induced insulin synthesis by pancreatic beta cells.

BACKGROUND: Thyroid hormones (THs) are known to regulate protein synthesis by acting at the transcriptional level and inducing the expression of many genes. However, little is known about their role in protein expression at the post-transcriptional level, even though studies have shown enhancement of protein synthesis associated with mTOR/p70S6K activation after triiodo-L-thyronine (T3) administration. On the other hand, the effects of TH on translation initiation and polypeptidic chain elongation factors, being essential for activating protein synthesis, have been poorly explored. Therefore,

Iodine and tri-iodo-thyronine reduce the incidence of type 1 diabetes mellitus in the autoimmune prone BB rats.

Thyroid hormones modulate the immune system and metabolism, influence insulin secretion, and cause decreased glucose tolerance. Thyroid hormones have been described to change the incidence of spontaneous autoimmune thyroiditis in Bio-Breeding/Worcester (BB) rats but it is unknown how these hormones affect the development of type 1 diabetes mellitus (T1DM). The aim was to investigate the influence of changes in thyroid function during postnatal development on the prevalence of T1DM in BB rats and the influence of T3 on the beta cell mass in non-diabetic Wistar rats. BB rats were treated with so

3,3',5-Triiodo-L-thyronine inhibits ductal pancreatic adenocarcinoma proliferation improving the cytotoxic effect of chemotherapy.

The pancreatic adenocarcinoma is an aggressive and devastating disease, which is characterized by invasiveness, rapid progression, and profound resistance to actual treatments, including chemotherapy and radiotherapy. At the moment, surgical resection provides the best possibility for long-term survival, but is feasible only in the minority of patients, when advanced disease chemotherapy is considered, although the effects are modest. Several studies have shown that thyroid hormone, 3,3',5-triiodo-l-thyronine (T(3)) is able to promote or inhibit cell proliferation in a cell type-dependent mann

Induction of cytosolic triiodo-L-thyronine (T3) binding protein (CTBP) by T3 in primary cultured rat hepatocytes.

Cytosolic 3, 5, 3'-triiodo-L-thyronine (T3)-binding protein (CTBP) plays an important role in the regulation of intracellular T3 translocation from cytoplasm to the nuclear T3 receptor. We examined whether the CTBP activity could be induced by T3 or not in cultured hepatocytes prepared from thyroidectomized rats. CTBP activity was not detected in primary cultured hepatocytes from thyroidectomized rats. However, the protein was induced by the addition of T3 to the culture medium. The increase in the activity of CTBP was time dependent and the maximal level was obtained by 48 h in the presence o

Hippocampal gene expression of deiodinases 2 and 3 and effects of 3,5-diiodo-L-thyronine T2 in mouse depression paradigms.

Central thyroid hormone signaling is important in brain function/dysfunction, including affective disorders and depression. In contrast to 3,3',5-triiodo-L-thyronine (T3), the role of 3,5-diiodo-L-thyronine (T2), which until recently was considered an inactive metabolite of T3, has not been studied in these pathologies. However, both T3 and T2 stimulate mitochondrial respiration, a factor counteracting the pathogenesis of depressive disorder, but the cellular origins in the CNS, mechanisms, and kinetics of the cellular action for these two hormones are distinct and independent of each other.
