Current Pharmacological Approaches to Reduce Chorea in Huntington's Disease.

There are currently no effective pharmacological agents available to stop or prevent the progression of Huntington's disease (HD), a rare hereditary neurodegenerative disorder. In addition to psychiatric symptoms and cognitive impairments, HD causes progressive motor disturbances, in particular choreiform movements, which are characterized by unwanted contractions of the facial muscles, trunk and extremities. Management of choreiform movements is usually advised if chorea interferes with daily functioning, causes social isolation, gait instability, falls, or physical injury. Although drugs to

Efficacy of tiapride in the treatment of psychiatric disorders: A systematic review.

BACKGROUND: Tiapride is an atypical antipsychotic used to treat alcohol withdrawal, aggressiveness and agitation, headache, dyskinesias, tic and Tourette's disorder. More recently, it has been proposed for the treatment of delirium and agitation in hospitalised patients with COVID-19. Although its safety profile makes it suitable for use in vulnerable populations, the use of tiapride for psychiatric disorders is limited. This work aims to systematically review the available evidence on the efficacy and tolerability of tiapride in individuals with a psychiatric disorder. METHODS: We searched P

[Tiapride therapy of centrally produced dyskinesia. Examination of the serum levels of protein, LH and FSH (author's transl)].

The effect of N-(2-diethylaminoethyl)-2-methoxy-5-(methylsulfonyl)-benzamide hydrochloride (tiapride, Tiapridex), a dopamine antagonist, on the serum levels of prolactin, luteinising hormone (LH) and follicle-stimulating hormone (FSH) was studied on 20 healthy individuals and 10 patients with dyskinesia resulting from extrapyramidal disorders. Daily doses of 300 mg in healthy subjects and 300-800 mg in patients with dyskinesia resulted from CNS disorders, were found to increase serum prolactin levels without causing amenorrhoea or galactorrhoea. The drug seemed to have no effect on LH and FSH

The Efficacy of Tiapride and Carbamazepine Combination Therapy in Reducing Alcohol Withdrawal Symptoms: A Systematic Review and Meta-Analysis.

The combination of tiapride (TIA) and carbamazepine (CBZ) as an alternative treatment option to benzodiazepines and clomethiazole has been investigated by several investigations. We performed a systematic review and meta-analysis to further explore the efficacy of this combination in order to render more definite answers whether this combination can be recommendable in the clinical practice. We systematically searched electronic databases including PubMed (MEDLINE), EMBASE, OVID, Cochrane, Google Scholar, and Scopus for human studies. Statistical homogeneity was checked by χ2 test and I2 using

Diagnosis and treatment of acute alcohol intoxication and alcohol withdrawal syndrome: position paper of the Italian Society on Alcohol.

The chronic use of alcohol can lead to the onset of an alcohol use disorder (AUD). About 50% of subjects with an AUD may develop alcohol withdrawal syndrome (AWS) when they reduce or discontinue their alcohol consumption and, in 3-5% of them, convulsions and delirium tremens (DTs), representing life-threatening complications, may occur. Unfortunately, few physicians are adequately trained in identifying and treating AWS. The Italian Society on Alcohol has, therefore, implemented a task force of specialists to draw up recommendations for the treatment of AWS with the following main results: (1)

Pharmacological treatment of tic disorders and Tourette Syndrome.

The present review gives an overview of current pharmacological treatment options of tic disorders and Tourette Syndrome (TS). After a short summary on phenomenology, clinical course and comorbid conditions we review indications for pharmacological treatment in detail. Unfortunately, standardized and large enough drug trials in TS patients fulfilling evidence based medicine standards are still scarce. Treatment decisions are often guided by individual needs and personal experience of treating clinicians. The present recommendations for pharmacological tic treatment are therefore based on both

Treatment-resistant diabetic chorea manifesting with psychiatric symptoms: a case report.

We report a case of a 69-year-old man with treatment-resistant diabetic chorea presenting psychiatric symptoms. The right chorea lasted for 3 months and was refractory to control of diabetes mellitus or administration of haloperidol and benzodiazepines. Only administration of tiapride was efficacious. Magnetic resonance spectrometry and dopamine transporter-single photon emission computed tomography suggested that sustained ischemia at the striatum may lead to impaired expression of dopamine transporters, thereby resulting in deterioration in the indirect pathway. Tiapride inhibited dopamine D

[Facial chorea and hemichorea due to cardiogenic cerebral embolism in the cortex and subcortical white matter].

A 62-year-old man was admitted to our hospital because of suddenly developed bilateral facial chorea and left-sided hemichorea. He had neither hemiparesis nor sensory disturbance. Diffusion-weighted magnetic resonance images of the brain showed acute cortical and subcortical infarctions at the right insula, frontal, temporal and parietal lobes. Tiapride hydrochloride was administered for his chorea. The chorea disappeared on the next day. We diagnosed him as cardiogenic cerebral embolism because he had a paroxysmal atrial fibrillation. We supposed that his chorea was induced by interruption of

Unsuccessful Suicide Attempt With Tiapride.

To the best of our knowledge, no case has been published in the literature that reports an overdose of tiapride, either alone or in combination with other drugs. We report a self-poisoning case in an 18-year-old girl, with approximately 10 times the usual daily dose (ie, 2.5 g). Although the blood concentration was 20/30-fold higher than usually observed after therapeutic drug intakes (17,300 mcg/L), the patient remained almost asymptomatic.

Comparison of tetrabenazine, tiapride and olanzapine in Huntington's disease: a one-year French randomized multicenter study (Neuro-HD).

INTRODUCTION: Chorea and behavioral symptoms in Huntington's disease (HD) have been treated with neuroleptics and related drugs for over 50 years, yet direct randomized comparisons are scarce. This study assessed the efficacy and safety of olanzapine, tetrabenazine, and tiapride in HD, with the Independence Scale as the primary outcome and components of the Unified Huntington's Disease Rating Scale as secondary outcomes. METHODS: We conducted a pragmatic, randomized, open-label trial across 11 centers of the French-Speaking Huntington's Network, enrolling 179 patients with Huntington's diseas

Long-Term Use of Tiapride and Tetrabenazine in Huntington's Disease: A 25-Year Retrospective Single-Center Analysis.

PURPOSE: Chorea is a key motor feature of Huntington's disease (HD). In Germany, Austria, and Switzerland, tiapride and tetrabenazine are established and formally approved for symptomatic management of chorea in HD. However, long-term real-world comparative data are lacking, and early documentation is often incomplete. The purpose of this study was to analyze prescription patterns, tolerability, and reasons for discontinuation of both medications in a retrospective real-world HD cohort and to examine outcomes of reinitiation and combination therapy. METHODS: We retrospectively reviewed medica
