The pharmacological responses produced by alpha chaconine and tomatine on guinea pig ileum, on the isolated electrically stimulated frog ventricle, and recordings of EEG, ECG, respiration and blood pressure in the rabbit showed no essential differences from those produced by alpha solanine. The LD50 values of chaconine and solanine in the mouse and rabbit are also similar and suggest that compounds other than these are probably responsible for the predominant toxic effects of certain hybrid potatoes in man and animals. The failure of the three glycoalkaloids to produce a significant teratologi

The potent adaptive immune response of mRNA vaccines requires that sufficient antigens translated from mRNA cargos and highly efficient activation of innate immunity. However, it remains unclear if simple introduction of an adjuvant can help mRNA vaccines achieve these functions. In this study, a nature product, tomatine, was experimentally incorporated into the widely used nanocarriers, serving a multi-functional adjuvant to improve mRNA expression efficiency and activate innate immunity. Tomatine based liposome (T-LPX) exhibited elevated uptake in dendritic cells (DCs) via caveolin-mediated

There is a growing interest in the cytotoxic effects of bioactive glycoalkaloids, such as α-tomatine on tumor cells. Here, for the first time, we determine the antitumor potential of tomatine, a mixture of α-tomatine and dehydrotomatine, in metastatic melanoma (MM) cell lines harboring different BRAF and MC1R variants. We performed cytotoxicity experiments and annexin-V/propidium iodide staining to assess the apoptotic/necrotic status of the cells. ER stress and autophagy markers were revealed by Western Blot, whereas antiangiogenic and vascular-disrupting effects were evaluated through a capi

Head and neck squamous cell carcinoma (HNSCC) is frequently associated with cisplatin resistance, which limits the therapeutic efficacy of conventional chemotherapy. In this study, we investigated whether α-tomatine could enhance cisplatin sensitivity and augment its antitumor efficacy in HNSCC cells. Treatment with 20 μM cisplatin alone induced relatively low cytotoxicity in FaDu and YD38 cells (18.45 ± 2.59% and 9.40 ± 2.33%, respectively). In contrast, co-treatment of FaDu and YD38 cells with cisplatin (20 μM) and a non-cytotoxic concentration of α-tomatine (2 μM) significantly increased ce

The present study investigated the inhibitory effects of the commercial tetrasaccharide tomato glycoalkaloid tomatine and the aglycone tomatidine on three mucosal pathogenic protozoa that are reported to infect humans, cattle, and cats, respectively: Trichomonas vaginalis strain G3, Tritrichomonas foetus strain D1, and Tritrichomonas foetus strain C1. A preliminary screen showed that tomatine at 100 μM concentration completely inhibited the growth of all three trichomonads. In contrast, the inhibition of all three pathogens by tomatidine was much lower, suggesting the involvement of the lycote

Oral cancer still poses a serious threat to world health and has limited feasible remedies. Recently, Alpha-tomatine (AT), a naturally occurring glycoalkaloid, has attracted attention due to its possible anticancer effects. The study investigates its cytotoxic and apoptotic impact of AT against KB human oral carcinoma cells and examine its role in regulating molecular mechanism, with an emphasis on oxidative stress and DNA damage-mediated apoptosis. The MTT assay for cytotoxicity of AT against KB cells showed an IC50 of 11.8 µM. The DCFH-DA staining technique was used to measure ROS formation,

First-generation vaccine candidates against leishmaniasis, based on chemically or physically inactivated whole parasite, have attracted considerable attention owing to their feasibility, scalability of production, and comparatively low manufacturing costs. Vaccine formulations typically require immunostimulatory compounds to elicit robust cell-mediated immunity. Traditionally, adjuvants have been incorporated into vaccine formulation to augment their immunogenic potential. Recent advances over the past decade have yielded novel adjuvant system capable of stimulating both antibody-mediated and

Tuta absoluta is a worldwide invasive pest that causes severe losses in tomato industry in China, while its resistance mechanism to tomato's protective alkaloid of α-tomatine remains unclear. This study explores the role of its gut microbiota, and results showed that the abundance of the gut symbiont Enterococcus mundtii was significantly higher in T. absoluta feeding on wild-type tomatoes than that in those feeding on α-tomatine-deficient GAME2 mutant tomatoes. Isolated E. mundtii strain E14 degraded 72.2 ± 1.3% of α-tomatine in vitro; reinoculating it into antibiotic-treated T. absoluta rest

INTRODUCTION: The objective of this study is to investigate the shear bonding strength of a glycoalkaloid, also a novel matrix metalloproteinase enzyme known as α-tomatine, on two different surfaces of dentin (sound & caries-affected) and its efficacy against cariogenic microorganisms using in vitro and in silico methods.
METHODS: The effect of a-tomatine at different concentrations (0.75 / 1 / 1.5 µM) on shear bonding strength in caries-affected and sound dentin was also investigated (n = 10; each per subgroup). The analysis of shear bonding and failure tests was conducted after a 24-hour st

α-Tomatine is a steroidal glycoalkaloid found in immature tomatoes that has been shown to have multiple beneficial effects on health. However, its anti-inflammatory properties have been little investigated thus far. The aim of this study was to evaluate its potential anti-inflammatory properties and underlying molecular mechanisms in rat splenocytes in vitro and ex vivo. We measured the lipopolysaccharide (LPS)-stimulated secretion of inflammatory molecules by splenocytes to assess the anti-inflammatory effects of α-tomatine. The underlying mechanism was investigated via western blotting. Next

Insulin resistance (IR) disrupts hepatic glucose metabolism and mitochondrial function, which contributes to metabolic disorders. The present study examined the effects of tomatine on glucose metabolism in high-glucose-induced IR hepatocytes and explored its underlying mechanisms using AML12 and HepG2 cell models. The results showed that tomatine did not exhibit cytotoxic effects. Under IR conditions, tomatine dose-dependently improved glucose metabolism by enhancing glucose consumption and restoring the mRNA expression of the glucose transporter Glut2 and gluconeogenesis-related genes (Pepck

Currently, hormonal therapy is the main treatment option for advanced prostate cancer; however, a certain number of cases progress to metastatic, castration-resistant prostate cancer. Therefore, we designed in vitro and in vivo studies of a new molecular targeted therapy using alpha-tomatine (α-tomatine), a glycoalkaloid extracted from tomatoes, for the growth inhibition of both castration-sensitive human LNCaP and castration-resistant mouse TRAMP-C2 and metastatic human C4-2B prostate cancer cell lines. In vitro, α-tomatine supplementation showed a dose-dependent decrease in the proliferation

α-Tomatine and chlorophyll (a and b) decreased, and β-carotene and lycopene increased with ripening of tomatoes. α-Tomatine was localised in peel of immature green tomatoes. The dose-response curve of α-tomatine determined by WST-1 (water soluble tetrazolium) assay was the same as that by LDH (lactate dehydrogenase) assay, suggesting that the cytotoxicity of α-tomatine depends on the destruction of plasma membrane. Immature green tomatoes had little cytotoxic effect after one month-incubation with 25% ethanol or 4.5% acetate at 7°C, and α-tomatine was decomposed by crude enzymes extracted from

Esculeoside A and tomatine are two major steroidal alkaloids in tomato fruit (Solanum lycopersicum) that exhibit anti-inflammatory, anticancer, and anti-hyperlipidemia activities. Tomatine contained in immature tomato fruit is converted to esculeoside A as the fruit matures. To develop new tomato varieties based on the content analysis of functional secondary metabolites, 184 mutant lines were generated from the original cultivar (S. lycopersicum cv. Micro-Tom) by radiation breeding. Ultra-performance liquid chromatography coupled with evaporative light scattering detector was used to identify

The actinomycete Streptomyces scabies 87-22 is the causal agent of common scab, an economically important disease of potato and taproot crops. Sequencing of the S. scabies 87-22 genome revealed the presence of a gene with high homology to the gene encoding the alpha-tomatine-detoxifying enzyme tomatinase found in fungal tomato pathogens. The tomA gene from S. scabies 87-22 was cotranscribed with a putative family 1 glycosyl hydrolase gene, and purified TomA protein was active only on alpha-tomatine and not potato glycoalkaloids or xylans. Tomatinase-null mutants were more sensitive to alpha-to

During their interactions with plants, fungal pathogens employ large numbers of pathogenesis-associated molecules including secreted effectors and enzymes that can degrade various defence compounds. However, in many cases, in planta targets of pathogen-produced enzymes remain unknown. We identified a gene in the wheat pathogen Fusarium graminearum, encoding a putative enzyme that shows 84% sequence identity to FoTom1, a tomatinase produced by the tomato pathogen Fusarium oxysporum f. sp. lycopersici. In F. oxysporum f. sp. lycopersici, FoTom1 is a virulence factor involved in the degradation o

BACKGROUND: We previously reported that the tomato glycoalkaloid tomatine inhibited the growth of Trichomonas vaginalis strain G3, Tritrichomonas foetus strain D1, and Tritrichomonas foetus-like strain C1 that cause disease in humans and farm and domesticated animals. The increasing prevalence of antibiotic resistance requires development of new tools to enhance or replace medicinal antibiotics.
METHODS: Wild tomato plants were harvested and divided into leaves, stems, and fruit of different colors: green, yellow, and red. Samples were freeze dried and ground with a handheld mill. The resulti

ABCB6 is a member of the adenosine triphosphate (ATP)-binding cassette family of transporter proteins that is increasingly recognized as a relevant physiological and therapeutic target. Evaluation of modulators of ABCB6 activity would pave the way toward a more complete understanding of the significance of this transport process in tumor cell growth, proliferation and therapy-related drug resistance. In addition, this effort would improve our understanding of the function of ABCB6 in normal physiology with respect to heme biosynthesis, and cellular adaptation to metabolic demand and stress res

Leishmaniasis, caused by Leishmania spp., is a major public health concern in tropical and subtropical regions and is classified as a neglected tropical disease (NTD). Visceral leishmaniasis (VL), the most severe form, affects the reticuloendothelial system and is often fatal if untreated. Current treatment relies on a limited number of chemotherapeutic agents, and no licensed vaccine is available to prevent infection. This highlights the urgent need for a prophylactic vaccine to protect against the infection and its potentially fatal consequences. Therefore, in the present study, we evaluated

Botrytis cinerea is a widely studied model organism for plant-fungal interactions. This chapter provides simple and easy protocols for live-cell imaging techniques and subcellular protein localization studies in B. cinerea in vitro. Using a simple microscopy setup, we describe methods for imaging spore germlings and hyphae and for studying protein dynamics in response to the antifungal compound α-tomatine. The protocols highlight the application of fluorescence microscopy to visualize sterol distribution and protein localization patterns, enabling deeper insights into fungal cell biology.
