Bisacridine derivatives as effective eukaryotic topoisomerase II inhibitors.

Fungal infections, particularly those caused by Candida species, pose a growing clinical challenge due to the increasing resistance to conventional antifungal agents and, in general, the limited arsenal of such drugs. In this study, we synthesized and tested a series of novel derivatives of bisacridines as potential antifungal compounds targeting fungal topoisomerase II. These compounds were evaluated for their ability to inhibit yeast topoisomerase II, their antifungal activity, and their selectivity over the human enzyme. Enzymatic assays confirmed that among them, compound IKE16 exhibited a

Leukemias related to treatment with DNA topoisomerase II inhibitors.

The epipodophyllotoxins etoposide and teniposide and other DNA topoisomerase II inhibitors including anthracyclines and dactinomycin are highly efficacious anticancer drugs. All are associated with a distinct form of leukemia characterized by chromosomal translocations as a treatment complication. Most of the translocations disrupt a breakpoint cluster region (bcr) of the MLL gene at chromosome band 11q23. Other characteristic translocations also may occur. The normal function of the nuclear enzyme DNA topoisomerase II is to catalyze changes in DNA topology between relaxed and supercoiled stat

Cadmium is a catalytic inhibitor of DNA topoisomerase II.

Cadmium (Cd(2+)) is a highly toxic and carcinogenic metal that is an environmental and occupational hazard. DNA topoisomerase II is an essential nuclear enzyme and its inhibition can result in the formation of genotoxic and recombinogenic DNA double strand breaks. In this study we showed that cadmium chloride strongly inhibited the DNA decatenation activity of human topoisomerase IIα in the low micromolar concentration range and that its inhibitory effects were reduced by glutathione. Because the activity of topoisomerase II is strongly inhibited by thiol-reactive compounds this result suggest

Antiparasitic effects of topoisomerase II inhibitors on Giardia intestinalis trophozoites.

Giardiasis is a common intestinal infection caused by the parasitic protozoan Giardia intestinalis. Current treatment relies on drugs such as metronidazole, which are associated with therapeutic failure and potential toxicity, highlighting the need for new treatment strategies. Topoisomerases, enzymes involved in regulating DNA topology, represent promising targets for drug development, including treatment of protozoan infections. The present study investigated the effects of type II topoisomerase inhibitors on G. intestinalis. Trophozoites were exposed to 1, 5, 10, or 20 μM nalidixic acid, en

Inhibitors of mTOR overcome drug resistance from topoisomerase II inhibitors in solid tumors.

The present study was performed to investigate the possible role of mTOR inhibitors in restoring chemosensitivity to adriamycin/cisplatin and elucidate the underlying mechanism. Combining adriamycin/cisplatin with torisel synergistically inhibited the cell proliferation in human oropharyngeal carcinoma cell line KB and its multidrug-resistant subclone KB/7D. Combining adriamycin and torisel inhibited the phosphorylation of 4EBP-1 and p70S6K, the proteins involved in mTOR pathway, increased expression of γH2AX indicative of DNA damage, triggered cell cycle arrest at G2/M and apoptosis. Topoisomerase I/II inhibitors: from established drugs to next-generation therapeutics.

Topoisomerases are crucial enzymes that control the DNA topology in replication, transcription, recombination, and chromosomal segregation, and therefore are validated as the targets of anticancer therapy. Topoisomerase-targeting agents are mechanistically divided into poison-type and catalytic-type inhibitors. Poison-type inhibitors stabilize covalent enzyme-DNA cleavage complexes, whereas catalytic inhibitors prevent enzymatic turnover without stabilizing cleavage complexes. Both classes may selectively target either topoisomerase I or topoisomerase II isoforms. Established and clinically us

Antiviral activity of topoisomerase II catalytic inhibitors against Epstein-Barr virus.

Herpesviruses require several cellular proteins for their lytic DNA replication including topoisomerase II (Topo II). Thus, Topo II could be an effective drug target against herpesviral infection. In this study, we examined several Topo II catalytic inhibitors for their potentials in blocking EBV replication and becoming efficacious antiviral agents. Topo II catalytic inhibitors in general exhibited marked inhibition of EBV lytic replication and minimal cytotoxicity. In particular, (+)-rutamarin, with the best selectivity index (SI>63) among the inhibitors tested in this study, is effective in

Antibacterial activity of novel dual bacterial DNA type II topoisomerase inhibitors.

In this study, a drug discovery programme that sought to identify novel dual bacterial topoisomerase II inhibitors (NBTIs) led to the selection of six optimized compounds. In enzymatic assays, the molecules showed equivalent dual-targeting activity against the DNA gyrase and topoisomerase IV enzymes of Staphylococcus aureus and Escherichia coli. Consistently, the compounds demonstrated potent activity in susceptibility tests against various Gram-positive and Gram-negative reference species, including ciprofloxacin-resistant strains. The activity of the compounds against clinical multidrug-resi

G2 arrest in response to topoisomerase II inhibitors: the role of p53.

We have previously found that the overexpression of p53 causes G(2) arrest and represses the synthesis of cyclin-dependent kinase 1 and cyclin B1, two proteins required for cells to traverse from G(2) into M. G(2) arrest occurs in response to DNA damage caused by a variety of agents and treatments. Here, we investigate the role of p53 in the G(2) arrest that occurs in response to the topoisomerase inhibitors etoposide and merbarone. In HT1080 cells expressing a dominant-negative form of p53, treatment with etoposide still caused G(2) arrest, but the arrest could be overcome by additional treat

Recent advancements in the medicinal chemistry of bacterial type II topoisomerase inhibitors.

Replication proteins are sought as a potential targets for antimicrobial agents. Despite their promising target characteristics, only topoisomerase II inhibitors targeting DNA gyrase and/or topoisomerase IV have reached clinical use. Topoisomerases are the enzymes that are essential for cellular functions and various biological activities. A wide range of natural and synthetic compounds have been identified as potential topoisomerase inhibitors but the resistance is most commonly found in these drugs. The emergence of FQ resistance has increased the need for the development of novel topoisomer

Tilmicosin derivatives as topoisomerase I/II inhibitors: Rational design, synthesis, and antibacterial evaluation.

The antibacterial mechanisms of macrolides primarily focus on ribosome inhibition, while their potential interactions with topoisomerases (Topo) remain to be thoroughly explored. In this study, novel tilmicosin (TIM) derivatives were designed and screened with strong binding affinities to Topo I/II by using molecular docking. Then, four TIM derivatives (T-1, T-2, T-16, and T-17) were selected as potential candidates for subsequent synthesis. In vitro antibacterial activities were evaluated, among them, T-1 and T-16 exhibited superior antibacterial effects against most bacteria, with a particul

The DNA topoisomerase II catalytic inhibitor merbarone is genotoxic and induces endoreduplication.

In the last years a number of reports have shown that the so-called topoisomerase II (topo II) catalytic inhibitors are able to induce DNA and chromosome damage, an unexpected result taking into account that they do not stabilize topo II-DNA cleavable complexes, a feature of topo II poisons such as etoposide and amsacrine. Merbarone inhibits the catalytic activity of topo II by blocking DNA cleavage by the enzyme. While it was first reported that merbarone does not induce genotoxic effects in mammalian cells, this has been challenged by reports showing that the topo II inhibitor induces effici

Amonafide, a topoisomerase II inhibitor, is unaffected by P-glycoprotein-mediated efflux.

Over-expression of P-glycoprotein (Pgp+) has been related to resistance to classical Topo II inhibitors used in the treatment of AML and is common in patients with poor-prognosis, such as those with secondary AML (sAML). Since clinical trials with amonafide, a unique ATP-independent Topo II inhibitor, in combination with cytarabine, have shown significant efficacy for remission induction in patients with sAML, we compared the cytotoxic effect of amonafide (amonafide l-malate, Xanafide) to the classical Topo II inhibitors (daunorubicin, doxorubicin, idarubicin, etoposide, and mitoxantrone) in K

Phase II study of XR 5000, an inhibitor of topoisomerases I and II, in advanced colorectal cancer.

XR 5000 is one of a series of tricyclic carboxamide-based cytotoxic agents. It binds to DNA by intercalation and stimulates DNA cleavage by inhibition of both topoisomerase I and II, thus possibly overcoming the resistance resulting from downregulation of either enzyme. Twenty patients with advanced or metastatic colorectal cancer, unpretreated for metastatic disease, received XR 5000 at the dose of 3010 mg/m(2) in a 120-h central intravenous (i.v.) infusion every 3 weeks. Response was evaluated every two cycles. No complete (CR) or partial responses (PR) were observed in eligible patients (re

Flavonoids as Dual Topoisomerase I and II Inhibitors: Mechanistic Insights and Emerging Anticancer Strategies.

Flavonoids, a diverse class of plant-derived polyphenolic compounds found in fruits, vegetables, and medicinal herbs, have gained considerable attention for their chemopreventive and anticancer properties. A key mechanism underlying their therapeutic potential is the modulation of DNA topoisomerases, enzymes essential for DNA replication, transcription, and recombination. Inhibition of topoisomerase I and II induces DNA damage, replication stress, and apoptosis in rapidly proliferating cancer cells. Flavonoids such as quercetin, kaempferol, myricetin, and epigallocatechin-3-gallate (EGCG) act

Biological evaluation of benzothiazole ethyl urea inhibitors of bacterial type II topoisomerases.

The type II topoisomerases DNA gyrase (GyrA/GyrB) and topoisomerase IV (ParC/ParE) are well-validated targets for antibacterial drug discovery. Because of their structural and functional homology, these enzymes are amenable to dual targeting by a single ligand. In this study, two novel benzothiazole ethyl urea-based small molecules, designated compound A and compound B, were evaluated for their biochemical, antibacterial, and pharmacokinetic properties. The two compounds inhibited the ATPase activity of GyrB and ParE with 50% inhibitory concentrations of <0.1 μg/ml. Prevention of DNA supercoil

Activities of the Novel Bacterial Topoisomerase Inhibitor OSUAB-0284 against the Biothreat Pathogen Bacillus anthracis and Its Type II Topoisomerases.

Bacillus anthracis is the etiological agent of anthrax and is classified as a Tier 1 biothreat pathogen. The fluoroquinolone ciprofloxacin is a preferred prophylactic drug for potential anthrax infections and acts by stabilizing DNA strand breaks formed by the bacterial type II topoisomerases, gyrase and topoisomerase IV. Unfortunately, widespread fluoroquinolone usage has increased levels of resistance in common bacterial pathogens, raising concern that resistant B. anthracis strains could be misused. Therefore, there is great interest in developing new classes of antibacterials that are effi
