Cladistic analysis of Tospovirus using molecular characters.

The genus Tospovirus was thought to be composed only of the tomato spotted wilt virus (TSWV), but now at least four Tospovirus species have been proposed based on serological and molecular data. A classification of tospoviruses has been proposed taking into account global similarities of the N gene and N protein sequences of 7 isolates of Tospovirus. Because phylogenetic analyses based on global similarities can lead to classifications which do not mirror the genealogy of the group, we have employed a cladistic analysis using parsimony of this genus with RNA sequences of 450 nucleotides of the

Nucleocapsid of Tomato spotted wilt tospovirus forms mobile particles that traffic on an actin/endoplasmic reticulum network driven by myosin XI-K.

A number of viral proteins from plant viruses, other than movement proteins, have been shown to traffic intracellularly along actin filaments and to be involved in viral infection. However, there has been no report that a viral capsid protein may traffic within a cell by utilizing the actin/endoplasmic reticulum (ER) network. We used Tomato spotted wilt tospovirus (TSWV) as a model virus to study the cell biological properties of a nucleocapsid (N) protein. We found that TSWV N protein was capable of forming highly motile cytoplasmic inclusions that moved along the ER and actin network. The di

Berberine from Corydalis saxicola Bunting reveals anti-tomato spotted wilt orthotospovirus activity with a novel mechanism.

Tomato spotted wilt orthotospovirus (TSWV) is a serious plant pathogen of crops that causes serious damage in the agriculture industry. The damage caused by TSWV has made the development of new and effective botanical antiviral agents against the disease increasingly urgent. Many kinds of compounds have been found to control disease caused by TSWV. However, novel alkaloid for the effectively control of this disease remain rarely. In this study, a series of compounds of novel alkaloid were extracted from Corydalis saxicola Bunting and their structure was elucidated by nuclear magnetic resonance

HMG-like DSP1 is a damage signal to mediate the western flower thrips, Frankliniella occidentalis, immune responses to tomato spotted wilt virus infection.

Tomato spotted wilt virus (TSWV) causes a serious plant disease and is transmitted by specific thrips including the western flower thrips, Frankliniella occidentalis. The persistent and circulative virus transmission suggests an induction of immune defenses in the thrips. We investigated the immune responses of F. occidentalis to TSWV infection. Immunofluorescence assay demonstrated viral infection in the larval midguts at early stage and subsequent propagation to the salivary gland in adults. In the larval midgut, TSWV infection led to the release of DSP1, a damage-associated molecular patter

Evaluation of Soybean for Resistance to Neohyadatothrips variabilis (Thysanoptera: Thripidae) Noninfected and Infected With Soybean Vein Necrosis Virus.

Soybean vein necrosis virus (SVNV) was first identified in Arkansas and Tennessee in 2008 and is now known to be widespread in the United States and Canada. Multiple species of thrips transmit this and other tospoviruses with Neohydatothrips variabilis (Beach) (soybean thrips) cited as the most efficient vector for SVNV. In this study, 18 soybean, Glycine max (L.) Merr., genotypes were evaluated in four experiments by infesting plants with noninfected and SVNV-infected thrips using choice and no-choice assays. In both choice experiments with noninfected and SVNV-infected thrips, the lowest num

Salicylic Acid Is Involved in the Basal Resistance of Tomato Plants to Citrus Exocortis Viroid and Tomato Spotted Wilt Virus.

Tomato plants expressing the NahG transgene, which prevents accumulation of endogenous salicylic acid (SA), were used to study the importance of the SA signalling pathway in basal defence against Citrus Exocortis Viroid (CEVd) or Tomato Spotted Wilt Virus (TSWV). The lack of SA accumulation in the CEVd- or TSWV-infected NahG tomato plants led to an early and dramatic disease phenotype, as compared to that observed in the corresponding parental Money Maker. Addition of acibenzolar-S-methyl, a benzothiadiazole (BTH), which activates the systemic acquired resistance pathway downstream of SA signa

Nucleotide sequence and coding strategy of the Uukuniemi virus L RNA segment.

The complete nucleotide sequence of the L RNA segment of Uukuniemi virus has been determined from cloned cDNA. The L RNA is 6423 nucleotides in length, and is of negative polarity. The viral-complementary RNA contains a single large open reading frame of 2104 codons which corresponds to the L protein (M(r) 241039). Comparison with the L protein sequences of other members of the Bunyaviridae showed homology with the Rift Valley fever phlebovirus L protein (38% amino acid identity), but no detectable similarity with bunyavirus, hantavirus or tospovirus L proteins. These data lend further support

Induction of innate immunity and plant growth promotion in tomato unveils the antiviral nature of bacterial endophytes against groundnut bud necrosis virus.

Tomato is an important crop worldwide, but groundnut bud necrosis virus (GBNV) often hampers its growth. This study investigates the antiviral potential of bacterial endophytes, including Brucella melitensis CNEB54, Bacillus licheniformis CNEB4, Bacillus velezensis CNEB26, and Bacillus vallismortis BAVE5 against GBNV, as well as their ability to enhance immunity and growth in tomato. All four bacterial isolates demonstrated a significant delay in GBNV symptom development 10 days post-inoculation, with disease incidence ranging from 18% to 36% compared to 84% in control. Generation of a Bivalent Recombinant Vaccine Conferring Dual Protection Against Potyvirus and Orthotospovirus in Cucurbits.

Climate warming has facilitated the expansion of insect vectors and plant viral pathogens, leading to increased incidence of viral diseases in crops. Cucurbit crops, including cucumber (Cucumis sativus), melon (Cucumis melo), squash (Cucurbita pepo), and watermelon (Citrullus lanatus), are of major economic importance worldwide, but their production is severely threatened by viral infections. Among the most damaging viruses are zucchini yellow mosaic virus (ZYMV; genus Potyvirus), transmitted by aphids, and melon yellow spot virus (MYSV; genus Orthotospovirus), transmitted by thrips, both of w

Rift Valley fever virus.

Rift Valley fever is considered to be one of the most important viral zoonoses in Africa. In 2000, the Rift valley fever virus spread to the Arabian Peninsula and caused two simultaneous outbreaks in Yemen and Saudi Arabia. It is transmitted to ruminants and to humans by mosquitoes. The viral agent is an arbovirus, which belongs to the Phlebovirus genus in the Bunyaviridae family. This family of viruses comprises more than 300 members grouped into five genera: Orthobunyavirus, Phlebovirus, Hantavirus, Nairovirus, and Tospovirus. Several members of the Bunyaviridae family are responsible for fa

Preparation and application of a polyclonal antibody against the N structural protein of chilli yellow ringspot orthotospovirus.

Chilli yellow ringspot orthotospovirus (CYRSV), which is a newly discovered virus in Yunnan Province and is a member of the genus Orthotospovirus, family Tospoviridae, order Bunyavirales, causes severe disease in vegetable crops. The nucleocapsid (N) protein is a structural protein that is crucial for viral replication and virulence. To understand the exact role of N from CYRSV in viral replication and the host immune response, a qualified antibody against this protein is needed. In this study, the coding region of the CYRSV N gene was cloned into a bacterial expression vector (pTOPO-D1), and

Linking vector biology, transmission, and genome architecture in southeastern orthotospoviruses.

Orthotospoviruses remain major threats to specialty and row crops in the southeastern United States through thrips-borne, persistent-propagative transmission. We synthesize the emergence and biology of tomato spotted wilt virus (TSWV) and soybean vein necrosis virus (SVNV), with context from impatiens necrotic spot virus, groundnut ringspot virus, and tomato chlorotic spot virus (TCSV). TSWV has a broad host range and global importance, whereas SVNV has a narrower known host range but continues to expand across soybean-producing regions. Comparative genomic analyses suggest distinct evolutiona

Orthotospovirus tomatomaculae (Tomato spotted wilt virus [TSWV]) infects ovules and pollen to achieve vertical transmission in Capsicum annuum.

Orthotospovirus tomatomaculae (Tomato spotted wilt virus, TSWV) is an economically important plant pathogen reported to be transmissible by seeds. However, the pathways by which the virus enters the seed remain poorly understood. Here, we characterized the distribution and vertical transmission pathways of TSWV within reproductive organs of Capsicum annuum. Transmission electron microscopy (TEM) confirmed systemic distribution of TSWV throughout floral tissues, with virions detected in anthers, pollen grains, ovaries, ovules, and styles. Immunofluorescence histochemistry further revealed accum

Pathological Features and Genetic Polymorphism Analysis of Tomato Spotted Wilt Virus in Infected Tomato Fruit.

An in-house tomato inbred line, YNAU335, was planted in a greenhouse in spring from 2014 to 2017, and showed immunity to tomato spotted wilt virus (TSWV). YNAU335 was infected with TSWV in the spring from 2018 to 2020, and disease was observed on the leaves, sepals, and fruits. In 2021 and 2022, YNAU335 was planted in spring in the same greenhouse, which was suspected of being infected with TSWV, and visible disease symptoms were observed on the fruits. Transmission electron microscopy, deep sequencing of small RNAs, and molecular mutation diagnosis were used to analyze the pathological featur

Evaluation of Alternatives to an Organophosphate Insecticide with Selected Cultural Practices: Effects on Thrips, Frankliniella fusca, and Incidence of Spotted Wilt in Peanut Farmscapes.

Peanut growers use a combination of tactics to manage spotted wilt disease caused by thrips-transmitted Tomato spotted wilt virus (TSWV). They include planting TSWV-resistant cultivars, application of insecticides, and various cultural practices. Two commonly used insecticides against thrips are aldicarb and phorate. Both insecticides exhibit broad-spectrum toxicity. Recent research has led to the identification of potential alternatives to aldicarb and phorate. In this study, along with reduced-risk, alternative insecticides, we evaluated the effect of conventional versus strip tillage; singl

Tomato Spotted Wilt Virus Reprogrammes Host Glycolysis to Facilitate Proliferation by a Phase-Separated Co-Aggregate of Nucleocapsid Protein and Phosphoglycerate Kinase.

Efficient viral proliferation within the host is a critical step in pathogenicity and requires adenosine triphosphate (ATP). The replication, movement and immune evasion of many plant viruses within their hosts are associated with phase separation (PS)-derived aggregates formed by viral components. However, the host factors that drive the formation of these condensates remain largely unknown. This study provides evidence that the nucleocapsid protein (N) of tomato spotted wilt virus (TSWV) recruits the host factor phosphoglycerate kinase (NbPGK) from Nicotiana benthamiana to form phase-separat

Evolutionary origin and demographic trajectory of tomato spotted wilt virus (Orthotospovirus tomatomaculae).

Tomato spotted wilt virus (TSWV) is a harmful pathogen that causes severe disease in tomato, pepper, and other horticultural and agronomic crops. Its genome comprises three linear single-stranded RNA molecules (segments L, M, and S), which are unequally packed in nucleocapsids. Although its genome structure and molecular mechanism of infection are well understood, the evolutionary dynamics of this virus require further analysis to determine the most probable viral ancestor, the date of divergence, the ancestral geographical source of dispersal, and the demographic history of TSWV. For this, we

Emergence and molecular characterization of a resistance-breaking tomato spotted wilt virus isolate on bell pepper plants harboring the Tsw resistance gene in Japan.

Tomato spotted wilt virus (TSWV) is distributed all over Japan and globally, seriously damaging infected plants. A resistance-conferring gene, Tsw, that controls the yellow spotted wilt disease in bell pepper (Capsicum annuum) plants caused by TSWV infection in Japan, is now commercially available. In this study, we isolated for the first time in Japan, a resistance-breaking isolate, TSWV-JRB, from bell pepper plants harboring Tsw. The virus overcame the resistance conferred by Tsw in its heterozygous and homozygous configurations. The host range or virulence of TSWV-JRB and one of the non-res

Resistance breaking: integrated genomic and phenotypic insights into Orthotospovirus tomatomaculae strains overcoming Sw-5b and Tsw resistance.

BACKGROUND: Tomato spotted wilt virus (TSWV) is a globally significant plant pathogen infecting more than 1,000 species and causing billion-dollar crop losses annually. The emergence of resistance-breaking (RB) strains has undermined single-gene resistance conferred by Sw-5b in tomato and Tsw in pepper, threatening durable resistance strategies. However, genomic and phenotypic characterization of novel RB strains remains limited. RESULTS: We characterized six novel TSWV strains—four tomato RB strains (Tom-BL1, Tom-BL2, Tom-CA, Tom-MX) and two pepper strains (RB Pep-BL and non-RB Pep-Non-RB)—us
