Hybrid reference genome assembly for the archetypal Leishmania (Viannia) guyanensis bearing the dsRNA totivirus LRV1.

We report a high-quality hybrid genome assembly and annotation for a Leishmania (Viannia) guyanensis line (M4147) expressing a firefly luciferase reporter gene and bearing the totivirus LRV1 strongly implicated in parasite hypervirulence. This reference genome should facilitate the study of infectivity and pathogenesis of cutaneous and severe leishmaniasis.

Tracking Mycoviruses in Public RNAseq Datasets of Malassezia: Three Original Totiviruses Revealed.

Mycoviruses are viruses that selectively infect and multiply in fungal cells. Malassezia is the most abundant fungus on human skin and is associated with a variety of conditions, including atopic eczema, atopic dermatitis, dandruff, folliculitis, pityriasis versicolor, and seborrheic dermatitis. Here, we conducted mycovirome studies on 194 public transcriptomes of Malassezia (2,568,212,042 paired-end reads) screened against all available viral proteins. Transcriptomic data were assembled de novo resulting in 1,170,715 contigs and 2,995,306 open reading frames (ORFs) that were subsequently trac

Comparative metagenomic profiling of microbial pathogen diversity in Haemaphysalis longicornis and Hyalomma dromedarii ticks.

Ticks are globally important vectors of human and animal pathogens. This study characterized microbial communities in Haemaphysalis longicornis (from humid Zhangjiachuan County) and Hyalomma dromedarii (from arid Minqin County, Gansu Province, China) using metagenomic sequencing. We identified diverse bacteria and viruses including known pathogens (Anaplasma phagocytophilum, Totivirus spp., Escherichia spp.) and potentially novel agents (Totivirus-like and Trachysalambria curvirostris-associated sequences). These results highlight the role of these tick species in pathogen transmission across

Discovery of two novel totiviruses from Culex tritaeniorhynchus classifiable in a distinct clade with arthropod-infecting viruses within the family Totiviridae.

Two double-stranded RNA viruses, named Culex tritaeniorhynchus totivirus NJ2 (CTV_NJ2) and NJ3 (CTV_NJ3), were discovered from wild-captured Culex tritaeniorhynchus mosquitoes. The complete genomes (7,624 and 7,612 bp in length) were obtained using RNA sequencing. Both CTV_NJ2 and CTV_NJ3 encode a putative capsid protein and an RNA-dependent RNA polymerase. The most similar strain to CTV_NJ2/3 is Omono River virus strain AK4 (ORV-AK4). The CP and RdRp identities of AK4 are different to CTV_NJ2 (84% and 87%) and CTV_NJ3 (47% and 62%). Phylogenetic analysis showed that taxonomically speaking CTV

The L-A dsRNA Virus and Its Satellites: Totiviruses and Killers in Saccharomyces cerevisiae.

A secreted protein toxin encoded by a satellite dsRNA-enabled dissection of the genetic control of replication and expression of a single-segment dsRNA virus, the first Totivirus, L-A. Among the then-novel findings were i. "head-full replication", ii. a supposedly forbidden "T = 2" capsid symmetry based on an asymmetric dimer, iii. a host N-acetyltransferase whose modification of the coat protein is necessary for packaging, iv. Kex1 and Kex2 pro-toxin peptidases leading to discovery of the pre-pro-insulin processing enzymes, and v. specific viral (+) strand sites/structures needed for RNA pack

Capsid Structure of Leishmania RNA Virus 1.

Leishmania parasites cause a variety of symptoms, including mucocutaneous leishmaniasis, which results in the destruction of the mucous membranes of the nose, mouth, and throat. The species of Leishmania carrying Leishmania RNA virus 1 (LRV1), from the family Totiviridae, are more likely to cause severe disease and are less sensitive to treatment than those that do not contain the virus. Although the importance of LRV1 for the severity of leishmaniasis was discovered a long time ago, the structure of the virus remained unknown. Here, we present a cryo-electron microscopy reconstruction of the

A Novel Mycovirus Evokes Transcriptional Rewiring in the Fungus Malassezia and Stimulates Beta Interferon Production in Macrophages.

Mycoviruses infect fungi, and while most persist asymptomatically, there are examples of mycoviruses having both beneficial and detrimental effects on their host. Virus-infected Saccharomyces and Ustilago strains exhibit a killer phenotype conferring a growth advantage over uninfected strains and other competing yeast species, whereas hypovirus-infected Cryphonectria parasitica displays defects in growth, sporulation, and virulence. In this study, we identify a double-stranded RNA (dsRNA) mycovirus in five Malassezia species. Sequence analysis reveals it to be a totivirus with two dsRNA segmen

Investigation on Mosquito-Borne Viruses at Lancang River and Nu River Watersheds in Southwestern China.

During 2007 and 2010, an extensive entomological survey was performed to assess the distribution of mosquitoes and mosquito-borne arboviruses at Lancang River and Nu River watersheds in southwestern China. A total of 20,450 mosquitoes consisting 20 species was trapped and submitted 261 pools according to species and location. Culex tritaeniorhynchus and Anopheles sinensis were the most abundant species. Eighty-seven isolates representing 11 virus species in 8 genera were obtained from 6 mosquito species. The new isolates were identified as Getah virus (GETV), Japanese encephalitis virus (JEV),

Metagenomic sequencing characterizes a wide diversity of viruses in field mosquito samples in Nigeria.

Mosquito vectors are a tremendous public health threat. One in six diseases worldwide is vector-borne transmitted mainly by mosquitoes. In the last couple of years, there have been active Yellow fever virus (YFV) outbreaks in many settings in Nigeria, and nationwide, entomological surveillance has been a significant effort geared towards understanding these outbreaks. In this study, we used a metagenomic sequencing approach to characterize viruses present in vector samples collected during various outbreaks of Yellow fever (YF) in Nigeria between 2017 and 2020. Mosquito samples were grouped in

Viral discovery and diversity in trypanosomatid protozoa with a focus on relatives of the human parasite Leishmania.

Knowledge of viral diversity is expanding greatly, but many lineages remain underexplored. We surveyed RNA viruses in 52 cultured monoxenous relatives of the human parasite Leishmania (Crithidia and Leptomonas), as well as plant-infecting PhytomonasLeptomonas pyrrhocoris was a hotbed for viral discovery, carrying a virus (Leptomonas pyrrhocoris ostravirus 1) with a highly divergent RNA-dependent RNA polymerase missed by conventional BLAST searches, an emergent clade of tombus-like viruses, and an example of viral endogenization. A deep-branching clade of trypanosomatid narnaviruses was found,

A mycovirus from entomogenous fungi Beauveria acts as a mobile effector of phytopathogen antagonism through cross-taxonomic transmission.

Fungal antagonism shapes microbial community structure and provides an ecological basis for biological control. Although mycoviruses have been implicated in fungal antagonism, their effects are generally attributed to indirect modulation of resident host physiology. Here, we show that a mycovirus can function as a mobile effector of antagonism through cross-taxonomic transmission, representing the first documentation of viral transfer from an entomopathogenic/endophytic fungus (Beauveria) to a phytopathogenic fungus (Sclerotinia) via a plant‑mediated route. We characterized Beauveria brongniar

Discovery of a novel orthototivirus-like virus in patients with vulvovaginal candidiasis.

INTRODUCTION: Vulvovaginal candidiasis (VVC) is a common fungal infection affecting women worldwide. Although the vaginal microbiome has been extensively studied, the diversity of viruses present in the vaginal microenvironment remains poorly characterized. METHODS: Vaginal swab samples from patients diagnosed with VVC were subjected to viral metagenomic sequencing using an Illumina NovaSeq platform. Viral contigs were assembled, annotated, and screened against public databases. Genome organization, pairwise sequence identity, and phylogenetic relationships were analyzed to determine the evol

Exploration of viral diversity in Aedes mosquitoes employing different shotgun metagenomic data analysis pipelines.

Metagenomics or metaviromics is emerging as a powerful technology for pathogen surveillance and pandemic preparedness. Mosquitoes are important vectors for transmission of many emerging viruses responsible for numerous outbreaks. Monitoring mosquitoes becomes essential to investigate its virome which leads to understanding of disease dynamics and allow preventive actions. In this study, shotgun metagenomic methodology using Ion GeneStudio S5 System was optimized for exploration of viral diversity. A total of 1913 Aedes larvae were collected from Central India during post monsoon season of 2024
