Evaluation of tumour promoting potency of fish borne toxaphene residues, as compared to technical toxaphene and UV-irradiated toxaphene.

In this study the potential impact of food chain-based biotransformation and physico-chemical weathering of toxaphene on its tumour promoting potential was investigated in vitro and in vivo. Human exposure to toxaphene is mainly through consumption of contaminated fish, therefore fish-borne residues of toxaphene (cod liver extract, CLE) were prepared by exposing cod to technical toxaphene (TT) for 63 days. UV-irradiated toxaphene (uvT) was included to represent a physico-chemical weathered toxaphene mixture. In vitro, TT, uvT and CLE all showed a dose- and time-dependent inhibition of gap junc

Bioassay of toxaphene for possible carcinogenicity.

A bioassay of technical-grade toxaphene for possible carcinogenicity was conducted by administering the test chemical in feed to Osborne-Mendel rats and B6C3F1 mice. Groups of 50 rats of each sex were administered toxaphene at one of two doses for 80 weeks, then observed for 28 or 30 weeks. Time-weighted average doses for males were 556 or 1,112 ppm; for females they were 540 or 1,080 ppm. Matched controls consisted of groups of 10 untreated rats of each sex; pooled controls consisted of the matched-control groups for toxaphene combined with 45 untreated male and 45 untreated female rats from

Studies on the tissue disposition and fate of [14C]toxaphene in Japanese quail.

The disposition and the fate of a [14C]toxaphene preparation was studied in adult and juvenile female Japanese quail. The distribution of [14C]toxaphene in the body is dominated by high concentrations of radioactivity in the adipose tissue and the egg yolk, and, especially in juvenile birds, in the bone marrow. The [14C]toxaphene compounds present in the fat tend to be less polar than the parent [14C]toxaphene. More than half of the radioactive dose is excreted via the droppings, eggs, and preen gland within a few days, with biliary excretion playing a major role. The main part of the radioact

Thyroid function and thyroid tumors in toxaphene-treated rats.

Historically, a direct and irreversible genotoxic reaction of a xenobiotic with DNA has been considered to be a universal and obligatory initiating event in the etiology of neoplasia, and it was assumed therefore that (1) there was no threshold other than zero exposure for cancer initiation, and (2) like radiation, exposure was additive over a lifetime. Human exposure to xenobiotics causing neoplasia in laboratory rodents has been regulated in many countries on that basis. In the last decade evidence has accumulated indicating that some neoplasia in laboratory rodents may not be caused by a di

Induction of rat hepatic microsomal enzymes by toxaphene pretreatment.

The effects of pretreatment of rats with toxaphene on hepatic drug metabolizing enzymes and several other parameters of the mixed function oxidase system were investigated. Adult male Sprague-Dawley rats were fed diets containing 0, 50, 100, 150 and 200 ppm of toxaphene for 14 days. The body weight gain was unaltered as well as the food consumption in all the toxaphene fed groups. There was no change in the weights of brain, kidney, heart, and testes but the liver weight was significantly increased. The thymus weight in all the toxaphene fed grups was decreased. Hydroxylation of pentobarbital

Toxaphene is antiestrogenic in a human breast-cancer cell assay.

Toxaphene is a complex mixture of chlorinated bornanes, bornenes, and bornadienes and was a heavily used insecticide in the United States until its use was restricted in 1982. There are conflicting reports regarding the potential for toxaphene to induce estrogenic responses in human and nonhuman animals. Due to the public concern over environmental estrogens, the estrogenicity of toxaphene was examined in a human breast-cancer cell assay, the MCF-7 focus assay, which is based on in vitro postconfluent cell proliferation and tissue restructuring. In this assay, 0.1-1 nM 17beta-estradiol (E2) pr

Model predictions of toxaphene degradation in the atmosphere over North America.

Technical toxaphene, a broad-spectrum pesticide mixture, degrades in the environment, resulting in potential changes in toxicity. The present study uses a multimedia model that the authors developed to estimate toxaphene degradation in the atmosphere over North America. The predicted degradation has strong spatial and temporal variability determined by processes such as emission and transport of technical toxaphene, as well as the complex interactions among many species (e.g., toxaphene, hydroxyl [OH] radicals, and ozone). More toxaphene is degraded in warmer months due to higher concentration

Developmental effects of embryonic exposure to toxaphene in the American alligator (Alligator mississippiensis).

A variety of organochlorine pesticides have been shown to adversely affect embryonic development. A number of abnormalities have been documented in alligators (Alligator mississippiensis) from highly-contaminated Lake Apopka, FL, USA that are similar to the results of experimental studies exposing embryos to pesticides. In the current study, we exposed developing alligator embryos to varying concentrations of toxaphene, a broad-spectrum pesticide found in relatively high concentration in Lake Apopka alligator egg yolk. The toxaphene, dissolved in 50 microl of ethanol, was applied topically to

Synthesis of low and high chlorinated toxaphene and comparison of their toxicity by zebrafish (Danio rerio) embryo test.

Toxaphene, also known as camphechlor, is a persistent organochlorine pesticide of complex composition. It is technically produced by photochlorination of camphene with elemental chlorine gas under ultraviolet irradiation. In the present work, a novel, laboratory-scale synthesis using sulfuryl chloride as a chlorinating reagent is described. This approach allowed the degree of chlorination of the resulting mixtures to be arbitrarily adjusted by varying the reaction conditions. Both the compositions and the chlorine contents of the low- and high-chlorinated mixtures acquired using this method we

Up-regulation of the alligator CYP3A77 gene by toxaphene and dexamethasone and its short term effect on plasma testosterone concentrations.

In this study we describe an alligator hepatic CYP3A gene, CYP3A77, which is inducible by dexamethasone and toxaphene. CYP3A plays a broad role in biotransforming both exogenous compounds and endogenous hormones such as testosterone and estradiol. Alligators collected from sites in Florida that are contaminated with organochlorine compounds exhibit differences in sex steroid concentrations. Many organochlorine compounds induce CYP3A expression in other vertebrates; hence, CYP3A induction by organochlorine contaminants could increase biotransformation and clearance of sex steroids by CYP3A and

Development of a reference dose for the persistent congeners of weathered toxaphene based on in vivo and in vitro effects related to tumor promotion.

Toxaphene is a mixture of chlorinated camphenes and bornanes that was produced and used in the United States until 1982. 1.3 million tons of toxaphene have been released worldwide. "Technical" toxaphene (TT) consists of a mixture of up to 800 different chemicals, known as congeners. TT weathers in the environment by both biotic and abiotic processes. The human body burden of toxaphene consists of only five persistent congeners that are not metabolized; three of these occur in considerably greater amounts than the other two. Because of the rapid metabolism and excretion of the non-persistent co

Growth, nutritional composition, and hematology of Arctic charr (Salvelinus alpinus) exposed to toxaphene and tapeworm (Diphyllobothrium dendriticum) larvae.

Toxaphene, an organochlorine pesticide, is the major contaminant of Arctic charr (Salvelinus alpinus) in the Canadian Arctic. The objective of this study was to investigate the combined effects of toxaphene exposure and infection by the larval stage of the cestode Diphyllobothrium dendriticum on fish growth, nutritional composition, and hematology. Hatchery-reared Arctic charr were subjected to one of four treatments: (1) oral administration of corn oil (control); (2) single oral dose of 10 microg/g wet wt toxaphene dissolved in corn oil; (3) exposure to 15 larval D. dendriticum; and (4) expos

The organochlorine pesticide toxaphene reduces non-mitochondrial respiration and induces heat shock protein 70 expression in early-staged zebrafish (Danio rerio).

Toxaphene is a restricted-use pesticide produced by reacting chlorine gas with camphene. It was heavily used as a pesticide for agricultural purposes in the 1960-1970s, but despite being banned >30 years ago, it can remain elevated in the soil due to its resistance to metabolic degradation; this has led to longstanding concerns about elevated levels of toxaphene and other organochlorine pesticides (OCPs) in the environment. The objective of this study were to determine the effects of waterborne exposure to toxaphene on early life stages of zebrafish. Based on the LC50, zebrafish embryos were e

Geographic variation of chlorinated pesticides, toxaphenes and PCBs in human milk from sub-arctic and arctic locations in Russia.

The concentrations of HCB, alpha-, beta- and gamma-HCH, 3 chlordanes (CHLs), p,p'-DDE, p,p'-DDD, p,p'-DDT, and 30 PCBs (polychlorinated biphenyls) were determined in 140 human milk samples from Kargopol (n=19), Severodvinsk (n=50), Arkhangelsk (n=51) and Naryan-Mar (n=20). Pooled samples were used for determination of three toxaphenes (chlorobornanes, CHBs). The concentrations of HCB, beta-HCH and p,p'-DDE in Russian human milk were 2, 10 and 3 times higher than corresponding levels in Norway, respectively, while concentrations of sum-PCBs and sum-TEQs (toxic equivalent quantities) of the mono

Very high concentrations of DDE and toxaphene residues in crocodiles from the Ord River, Western Australia: an investigation into possible endocrine disruption.

Organochlorine pesticide concentrations, particularly those of the DDT family and of toxaphene, were measured by gas chromatography in samples of liver and body fat taken from Australian freshwater crocodiles Crocodylus johnstoni at three locations along the Ord River in Western Australia. The three sampling sites were the irrigation area, downstream of the irrigation area, and well upstream of the irrigation area; the last site serving as the control. DDT and toxaphene were applied in large and known quantities to cotton grown in the Ord Irrigation Area from 1964 to 1974. Thus the residues in

Investigation of hepatic cytochrome P-450 enzyme induction and DNA adduct formation in male CD/1 mice following oral administration of toxaphene.

Exposure of experimental animals to toxaphene induces hepatic cytochrome P-450 (CYP). Although chronic administration of toxaphene to mice was found to cause an increased incidence of liver tumors, a mechanism for its carcinogenicity has yet to be elucidated. We investigated two potential mechanisms of toxaphene-induced carcinogenicity: peroxisomal proliferation and DNA binding. Peroxisomal proliferation was evaluated by measuring the level of immunodetectable CYP 4A1, an isozyme of CYP that is specifically induced by peroxisomal proliferators, in hepatic microsomes from CD1 mice that were tre

Chlorinated insecticides (toxaphene and endrin) affect oxytocin, testosterone, oestradiol and prostaglandin secretion from ovarian and uterine cells as well as myometrial contractions in cow in vitro.

We examined the direct effects of toxaphene and endrin, chlorinated insecticides that are widespread in the environment, on myometrial contractions and on the secretion of hormones involved in regulating these contractions. Granulosa, luteal, endometrial and myometrial cells, and myometrial strips from non-pregnant cows were incubated with both insecticides at environmentally relevant doses. Toxaphene inhibited and endrin stimulated the secretion of testosterone and oestradiol from granulosa cells.
