Phosphoglucomutase and trehalase isoenzymes of Venezuelan simulium vectors of Onchocerca volvulus.

Phosphoglucomutase (PGM) and trehalase (Tre) isoenzymes of five species of Simulium blackflies (Diptera: Simuliidae), vectors of onchocerciasis in Venezuela, were investigated by means of a portable electrophoresis field kit. Tre differed between S. incrustatum and S. oyapockense s.l. Electrophoretic variation of Tre in other members of the S. amazonicum and S. incrustatum groups merit further investigation. PGM appears to be more useful for separating populations within species complexes. Multiple populations and/or seasonal changes in population structure of S. guianense s.l., S. exiguum s.l

Elevation of urinary trehalase activity in patients of itai-itai disease.

The elevation of urinary trehalase activity in patients of itai-itai disease was examined. Urinary trehalase was correlated with tubular reabsorption of phosphorus (%TRP): the lower the trehalase activity, the worse was %TRP. Furthermore, this activity was inversely correlated with urinary glucose and urinary total protein. In itai-itai disease, the excretion of beta 2-microglobulin seems to be maximal, and urinary trehalase activity was low in the latter stages of the disease. It is inferred that itai-itai disease produces extremely severe tubular damage as well as glomerular dysfunction.

Control of yeast neutral trehalase by distinct polyphosphates and ribonucleic acid.

The activity of yeast trehalase when assayed at pH 7 in a crude extract was found to increase 2- to 3-fold upon incubation with 0.1% (v/v) polyethyleneimine or other polycations such as polylysine (0.075-mMol) and calf thymus histones (0.08 mMol). Incubation with 3 mM-Mn2+ and 5 mM-Ca2+ also led to 3- and 1.6-fold increases in trehalase activity, respectively. The activities of 11 other enzymes assayed in the crude yeast extract did not increase after addition of polyethylene imine. At concentrations of polyethyleneimine that maximally stimulated trehalase activity, 97% of the total RNA presen

Prenatal diagnosis of cystic fibrosis by trehalase enzyme assay in amniotic fluid.

Amniocentesis and amniotic fluid trehalase enzyme assay were offered to 14 pregnant women at a 1 in 4 risk for a child with cystic fibrosis. Twelve of these pregnancies were screened at the 18th week of gestation; ten proceeded to term, seven following the finding of a normal trehalase activity and three despite the low enzyme level in amniotic fluid. In all ten cases prenatal diagnosis proved to be correct. In two cases with low enzyme activity parents opted for termination at the 19th week, and with PAS-Alcian Blue staining some slight histochemical lesions characteristic of cystic fibrosis

Trehalase-trehalose axis in the human brain: A potential modulator of neuroprotection and neurodegeneration.

Trehalase, the primary enzyme responsible for the degradation of gastrointestinal trehalose ("mushroom sugar"), is well-characterised in the human gut, but has not been conclusively identified in the human brain. Trehalose itself has shown promise in neuroprotection through diverse molecular mechanisms, including the autophagy-driven clearance of cellular debris and neurotoxic aggregates. However, the mechanisms activating trehalose and its integration into human central nervous system processes remain elusive. To investigate the modulatory role of trehalase in the trehalose-mediated neuroprot

Low trehalase activity is associated with abdominal symptoms caused by edible mushrooms.

BACKGROUND: The purpose of the study was to evaluate whether maldigestion of trehalose causes abdominal symptoms and which available diagnostic method best distinguishes intolerant from tolerant subjects. METHODS: A 25-g oral trehalose load test was performed in 64 subjects. The 19 experiencing clear symptoms constituted the trehalose-intolerant subjects. Changes from base-line levels of blood glucose, breath hydrogen, and methane and symptoms were recorded after the test. Trehalase activity was determined in serum and on a duodenal biopsy specimen obtained by endoscopy. RESULTS: Intolerant

The involvement of the Candida glabrata trehalase enzymes in stress resistance and gut colonization.

Candida glabrata is an opportunistic human fungal pathogen and is frequently present in the human microbiome. It has a high relative resistance to environmental stresses and several antifungal drugs. An important component involved in microbial stress tolerance is trehalose. In this work, we characterized the three C. glabrata trehalase enzymes Ath1, Nth1 and Nth2. Single, double and triple deletion strains were constructed and characterized both in vitro and in vivo to determine the role of these enzymes in virulence. Ath1 was found to be located in the periplasm and was essential for growth

Comparative transcriptomic and metabolomics analysis of ovary in Nilaparvata lugens after trehalase inhibition.

The fecundity of Nilaparvata lugens (brown planthopper) is influenced by trehalase (TRE). To investigate the mechanism by which trehalose affects the reproduction of N. lugens, we conducted a comparative transcriptomic and metabolomic analysis of the ovaries of N. lugens following injection with dsTREs and validamycin (a TRE inhibitor). The results revealed that 844 differentially expressed genes (DEGs) were identified between the dsGFP and dsTREs injection groups, with 317 up-regulated genes and 527 down-regulated genes. Additionally, 1451 DEGs were identified between the water and validamyci

Fungal pathogen promotes caterpillar feeding and weight gain using a host-like trehalase.

Parasite-mediated extended phenotypes in hosts are of particular interest in biology. However, few parasite genes have been characterized for their selfish role in altering host behaviors to benefit parasite transmission or reproduction. The entomopathogenic fungus Cordyceps militaris infects caterpillar larvae without killing them until after pupation. Here, we report that fungal infection of silkworm larvae induces increased feeding and weight gain, which is manifested by starvation-like responses, including the constant upregulation of the orexigenic peptide HemaP and a sharp reduction in h

[Genetically determined trehalase deficiency in various population groups of Russia and neighboring countries].

Due to the low specificity and sensitivity of non-invasive clinical tests trehalose malabsorption remained out of sight of gastroenterologists. Therefore, the specialists regard this disorder as rare. Trehalose became widely used in the food industry as a harmless sucrose substitute, sweetener and stabilizer. After the discovery of the trehalase gene (rs2276064 TREH), it was found that the A*TREH allele is the determinant of the disaccharide absorption disorders, and the allele's carriership may be high in some groups. There is not enough information on the A*TREH frequency in the population o

Prevalence of genetically determined trehalase deficiency in populations of Siberia and Russian Far East.

In order to be digested, the disaccharide trehalose needs to be cleaved by the trehalase enzyme. There were reports suggesting that trehalase deficiency was more common in high-latitude than in the temperate climate populations. New horizons were opened for the epidemiologic research of trehalase enzymopathy when it became clear that reduced trehalase activity is determined by the A allele of tTREH gene (rs2276064). The aim of this study was to analyze the frequencies of the trehalase gene alleles and genotypes among the indigenous peoples of Siberia and the Russian Far East. We genotyped 567

The secreted acid trehalase encoded by the CgATH1 gene is involved in Candida glabrata virulence.

BACKGROUND: Candida glabrata yeast is the second cause of candidiasis worldwide. Differs from other yeasts since assimilates only glucose and trehalose (a characteristic used in rapid identification tests for this pathogen) by secreting into the medium a highly active acid trehalase encoded by the CgATH1 gene. OBJECTIVE: This study aimed to characterise the function of the acid trehalase in the physiopathology of C. glabrata. METHODS: Gene deletion was performed to obtain a mutant ath1Δ strain, and the ability of the ath1Δ strain to grow in trehalase, or the presence of trehalase activity in

Intestinal trehalase activity in a UK population: establishing a normal range and the effect of disease.

Trehalose is a disaccharide, the main dietary source being mushrooms. It has been approved as an additive in the preparation of dried food. Isolated intestinal trehalase deficiency is found in 8% of Greenlanders, but is rare elsewhere. The normal range of trehalase activity and the incidence of isolated trehalase deficiency in the UK have not been reported. Patients (n 400) were investigated for suspected malabsorption. Endoscopic distal duodenal biopsies were taken for histological assessment and maltase, sucrase, lactase and trehalase estimation. Disaccharidase activities were determined by

Urinary trehalase as an early indicator of cadmium-induced renal tubular damage in rabbit.

The significance of urinary trehalase as a possible early indicator of renal disorder was examined using Cd-treated rabbits, which received 1 mg/kg Cd thrice weekly for 3 months subcutaneously. The results showed that urinary trehalase increased significantly from 1 week after treatment, earlier than LAP, ALP, proteinuria and glucosuria, with no changes in plasma trehalase level. A marked decrease in trehalase activity in renal brush border membranes prepared from Cd-treated rabbits was observed. It was also confirmed by immunohistological techniques that Cd treatment resulted in a marked decr

The role of urinary trehalase levels in the early diagnosis of acute kidney injury in dehydrated children.

BACKGROUND: Trehalase is a brush-border glycoprotein enzyme found in the small intestine and in renal proximal tubules, where it breaks down trehalose into glucose. We examined whether a non-invasive urine measurement of trehalase could help with earlier recognition of acute kidney injury (AKI) in children presenting with acute dehydration. METHODS: We included 80 children who were evaluated in the Pediatric Emergency Department for acute dehydration and 40 healthy controls from the general pediatric outpatient clinic. Baseline laboratory data were extracted from the hospital information syst

Identification of natural inhibitors targeting trehalase of Anopheles funestus in the management of malaria: A Biocomputational assessment.

BACKGROUND OBJECTIVES: Anopheles funestus is playing an increasingly important role in malaria transmission in sub-Saharan Africa. Trehalase, an enzyme required for trehalose breakdown, is important for mosquito flight and stress adaptation. Hence, its inhibition has emerged as a promising malaria management strategy. METHODS: A collection of 1900 natural compounds from the ZINC database were screened against the 3D modeled structure of An. funestus trehalase protein using in silico tools. ADMET-AI, a web-based platform, was used to predict the absorption, distribution, metabolism, excretion,

Trehalase plays a role in macrophage colonization and virulence of Burkholderia pseudomallei in insect and mammalian hosts.

Trehalose is a disaccharide formed from two glucose molecules. This sugar molecule can be isolated from a range of organisms including bacteria, fungi, plants and invertebrates. Trehalose has a variety of functions including a role as an energy storage molecule, a structural component of glycolipids and plays a role in the virulence of some microorganisms. There are many metabolic pathways that control the biosynthesis and degradation of trehalose in different organisms. The enzyme trehalase forms part of a pathway that converts trehalose into glucose. In this study we set out to investigate w

A Ca2⁺-responsive GH37 neutral trehalase from Lentinula edodes: Cloning, heterologous expression and biochemical characterization.

Trehalases (EC 3.2.1.28) hydrolyze trehalose into two glucose molecules and play important roles in fungal trehalose metabolism. Although GH37 neutral trehalases have been characterized in yeasts and filamentous fungi, their biochemical properties in basidiomycetes remain poorly understood. In this study, we cloned LeNTH1, a putative GH37 trehalase gene from Lentinula edodes, expressed it in Pichia pastoris, and characterized the purified recombinant enzyme. The deduced protein comprised 745 amino acid residues and contained a predicted N-terminal signal peptide. The purified recombinant form
