Nanopatterns of molecular spoked wheels as giant homologues of benzene tricarboxylic acids.

Molecular spoked wheels with D 3h and C s symmetry are synthesized by Vollhardt trimerization of C 2v-symmetric dumbbell structures with central acetylene units and subsequent intramolecular ring closure. Scanning tunneling microscopy of the D 3h-symmetric species at the solid/liquid interface on graphite reveals triporous chiral honeycomb nanopatterns in which the alkoxy side chains dominate the packing over the carboxylic acid groups, which remain unpaired. In contrast, C s-symmetric isomers partially allow for pairing of the carboxylic acids, which therefore act as a probe for the reduced a

Inhibition of rat liver rhodanese by di-, tricarboxylic, and alpha-keto acids.

Rat liver rhodanese [EC 2.8.1.1] purified by ammonium sulfate fractionation, CM-cellulose and Sephadex G-200 chromatography yielded two active fractions (I & II). Their molecular weights were estimated to be 1.75 X 10(4) (I) and 1.26 X 10(4) (II) by the gel filtration method. Kinetic studies revealed that Fraction I rat liver rhodanese catalyzes thiocyanate formation from thiosulfate and cyanide by a double displacement mechanism. Carboxylic acids such as DL-isocitric, citric malic, pyruvic, and oxaloacetic acid were competitive inhibitors with respect to thiosulfate, whereas fumaric, succinic

Paper chromatographic separation of phosphate esters, tricarboxylic cycle acids and amino acids in extracts from malaria parasites.

A paper chromatographic method for the separation and identification of complex mixtures of phosphate esters, tricarboxylic cycle acids and amino acids in biological extracts is described. The method has been applied to the investigation of carbohydrate metabolism pathways in the intraerythrocytic, simian malaria parasite Plasmodium knowlesi; on treatment of parasitized erythrocytes with [U-C14]glucose, a perchloric acid extract was prepared and separated by chromatography on Whatman 31 ET filter-paper. A simple procedure for the measurement of the specific activities of radioactive compounds

Tricarboxylic acid cycle enzymes following thiamine deficiency.

Thiamine (Vitamin B1) deficiency (TD) leads to memory deficits and neurological disease in animals and humans. The thiamine-dependent enzymes of the tricarboxylic acid (TCA) cycle are reduced following TD and in the brains of patients that died from multiple neurodegenerative diseases. Whether reductions in thiamine or thiamine-dependent enzymes leads to changes in all TCA cycle enzymes has never been tested. In the current studies, the pyruvate dehydrogenase complex (PDHC) and all of enzymes of the TCA cycle were measured in the brains of TD mice. Non-thiamine-dependent enzymes such as succin

Metabolic Flexibility and Essentiality of the Tricarboxylic Acid Cycle in Plasmodium.

The complete tricarboxylic acid (TCA) cycle, comprising a series of 8 oxidative reactions, occurs in most eukaryotes in the mitochondria and in many prokaryotes. The net outcome of these 8 chemical reactions is the release of the reduced electron carriers NADH and FADH2, water, and carbon dioxide. The parasites of the Plasmodium spp., belonging to the phylum Apicomplexa, have all the genes for a complete TCA cycle. The parasite completes its life cycle across two hosts, the insect vector mosquito and a range of vertebrate hosts including humans. As the niches that the parasite invades and occu

The Tricarboxylic Acid Cycle Metabolites for Cancer: Friend or Enemy.

BACKGROUND/AIMS: Changes in the skin conditions after exposure to low humidity have been generally experienced in everyday life, but there have been few reports to approach it-especially in healthy skin. We have examined the effect of low humidity on healthy human skin by using noninvasive measurement devices. METHODS: Skin conditions on the ventral forearm and the cheek before and after 3 or 6 h exposure to low humidity were evaluated by measuring skin surface conductance, skin surface capacitance and transepidermal water loss. Skin surface replicas were also taken before and after exposure

Lysosomes contribute to the synthesis of tricarboxylic acid-related metabolites in the hippocampus.

The central nervous system is highly sensitive to energy supply, and the hippocampus operates under sustained metabolic load due to continuous synaptic activity and information processing. Lysosomes couple nutrient status to cellular energetics through the mechanistic target of rapamycin complex 1 (mTORC1) and the autophagy-lysosome pathway, yet their -subcellular contribution to neuronal metabolic profiles remains unclear. To address this, we established an in vivo AAV-LysoTag/Lyso-IP workflow combined with metabolomics to quantify metabolites within mouse hippocampal lysosomes. An in vitro L

Kinetics overcome thermodynamics in primitive analogs of the reverse tricarboxylic acid cycle.

The reverse/reductive tricarboxylic acid (rTCA) cycle is a metabolic pathway that facilitates CO2 fixation in certain anaerobic bacteria and archaea. Its presence in phylogenetically ancient organisms has led to hypotheses about its role in the early evolution of CO2 fixation pathways. While the thermodynamics of the pathway is well studied, the kinetic feasibility of uncatalyzed rTCA cycle reactions remains uncertain. In this study, we report a systematic, mechanistic, and kinetic characterization of the uncatalyzed rTCA cycle and its side reactions. A primitive, uncatalyzed rTCA reaction net

Enhanced selective copper precipitation by mechanochemically activated benzene tricarboxylic acid.

Toward the treatment of waste solution containing heavy metals, direct precipitation of the metal ions from an acidic solution without alkaline neutralisation is still the greatest challenge. Based on the ligand properties of benzene tricarboxylic acid (BTC) to copper ions, a simple ball milling with 90 min at 400 rpm was used to activate BTC to enhance its capacity for copper removal from the pH of the original solution around 3-4. A set of analytical methods were used to characterise the activated BTC sample and BTC-Cu precipitate before and after copper precipitation. Compared with the raw

Genetic investigation of tricarboxylic acid metabolism during the Plasmodium falciparum life cycle.

New antimalarial drugs are urgently needed to control drug-resistant forms of the malaria parasite Plasmodium falciparum. Mitochondrial electron transport is the target of both existing and new antimalarials. Herein, we describe 11 genetic knockout (KO) lines that delete six of the eight mitochondrial tricarboxylic acid (TCA) cycle enzymes. Although all TCA KOs grew normally in asexual blood stages, these metabolic deficiencies halted life-cycle progression in later stages. Specifically, aconitase KO parasites arrested as late gametocytes, whereas α-ketoglutarate-dehydrogenase-deficient parasi

Abnormalities in the tricarboxylic Acid cycle in Huntington disease and in a Huntington disease mouse model.

Glucose metabolism is reduced in the brains of patients with Huntington disease (HD). The mechanisms underlying this deficit, its link to the pathology of the disease, and the vulnerability of the striatum in HD remain unknown. Abnormalities in some of the key mitochondrial enzymes involved in glucose metabolism, including the pyruvate dehydrogenase complex (PDHC) and the tricarboxylic acid (TCA) cycle, may contribute to these deficits. Here, activities for these enzymes and select protein levels were measured in human postmortem cortex and in striatum and cortex of an HD mouse model (Q175); m

Tricarboxylic acid cycle related-metabolites and risk of atrial fibrillation and heart failure.

BACKGROUND: Tricarboxylic acid (TCA) cycle deregulation may predispose to cardiovascular diseases, but the role of TCA cycle-related metabolites in the development of atrial fibrillation (AF) and heart failure (HF) remains unexplored. This study sought to investigate the association of TCA cycle-related metabolites with risk of AF and HF. METHODS: We used two nested case-control studies within the PREDIMED study. During a mean follow-up for about 10 years, 512 AF and 334 HF incident cases matched by age (±5 years), sex and recruitment center to 616 controls and 433 controls, respectively, wer

Deficits in a tricarboxylic acid cycle enzyme in brains from patients with Parkinson's disease.

Parkinson's disease (PD) is associated with mitochondrial dysfunction, specifically a deficiency of complex I of the electron transport chain. Most, although not all, studies indicate that this deficiency is limited to brain regions with neurodegeneration. The current studies tested for deficiencies in other mitochondrial components in PD brain in a neuropathologically unaffected region where the abnormality cannot be attributed to secondary effects of neurodegeneration. The activity of a key (and arguably rate-limiting) tricarboxylic acid cycle enzyme, the alpha-ketoglutarate dehydrogenase co
