Species belonging to the filamentous cyanobacterial genus Trichodesmium are responsible for a significant fraction of oceanic nitrogen fixation. The availability of phosphorus (P) likely constrains the growth of Trichodesmium in certain regions of the ocean. Moreover, Trichodesmium species have recently been shown to play a role in an emerging oceanic phosphorus redox cycle, further highlighting the key role these microbes play in many biogeochemical processes in the contemporary ocean. Here, we show that Trichodesmium erythraeum IMS101 can grow on the reduced inorganic compound phosphite as i

The factors that control the growth and nitrogen fixation rates of marine diazotrophs such as Trichodesmium have been intensively studied because of the role that these processes have in the global cycling of carbon and nitrogen, and in the sequestration of carbon to the deep sea. Because the phosphate concentrations of many ocean gyres are low, the bioavailability of the larger, chemically heterogeneous pool of dissolved organic phosphorus could markedly influence Trichodesmium physiology. Here we describe the induction, by phosphorus stress, of genes from the Trichodesmium erythraeum IMS101

Mixotrophy, the combination of heterotrophic and autotrophic nutrition modes, is emerging as the rule rather than the exception in marine photosynthetic plankton. Trichodesmium, a prominent diazotroph ubiquitous in the (sub)tropical oceans, is generally considered to obtain energy via autotrophy. While the ability of Trichodesmium to use dissolved organic phosphorus when deprived of inorganic phosphorus sources is well known, the extent to which this important cyanobacterium may benefit from other dissolved organic matter (DOM) resources is unknown. Here we provide evidence of carbon-, nitroge

Nitrogen-fixing (N2) cyanobacteria provide bioavailable nitrogen to vast ocean regions but are in turn limited by iron (Fe) and/or phosphorus (P), which may force them to employ alternative nitrogen acquisition strategies. The adaptive responses of nitrogen fixers to global-change drivers under nutrient-limited conditions could profoundly alter the current ocean nitrogen and carbon cycles. Here, we show that the globally important N2 fixer Trichodesmium fundamentally shifts nitrogen metabolism toward organic-nitrogen scavenging following long-term high-CO2 adaptation under iron and/or phosphor

The nonheterocystous cyanobacterium Trichodesmium fixes C and N concurrently during the light period in tropical and subtropical oceans. Synchrotron mapping of Fe, P and S in trichomes of Trichodesmium erythraeum Erhenberg IMS 101 (CCMP 1985) collected during exponential and senescent growth revealed that 16% of trichomes contained sections of up to 25 cells with ca. 2-fold elevated Fe and S but ca. 2-fold lowered P in comparison to neighboring trichome sections. The correlation between Fe and S in these trichomes was moderate to strongly positive (R > 0.35), while the correlation between Fe a

Size is an important trait among marine phytoplankton as it influences a vast range of physiological, ecological, and evolutionary processes. For Trichodesmium, a cyanobacterial diazotroph important for the global nitrogen cycle, size is a flexible trait that can change because of its ability to form colonies. Trichodesmium colonies can persist from 10 µm to greater than 1 mm in the natural environment. Despite this known ability, we still do not know whether a maximum size limit exists for Trichodesmium colonies and, more importantly, whether any mechanisms regulate this limit. In this paper,

Trichodesmium is a suspected toxin-producing nonheterocystous cyanobacteria ubiquitous in tropical, subtropical, and temperate seas. The genus is known for its ability to fix nitrogen and form massive blooms. In oligotrophic seas, it can dominate the biomass and be a major component of oceanic primary production and global nitrogen cycling. Numerous reports suggest Trichodesmium-derived toxins are a cause of death of fish, crabs, and bivalves. Laboratory studies have demonstrated neurotoxic effects in T. thiebautii cell extracts and field reports suggest respiratory distress and contact dermat

Trichodesmium spp. are diazotrophic cyanobacteria that exist as single filaments (trichomes) and as macroscopic colonies of varying shapes formed by aggregating trichomes. The causes and dynamics of colony formation and disassociation are not yet elucidated. we demonstrate that limited availability of dissolved phosphorus (P) or iron (Fe) stimulated trichome mobility and induced colony formation in Trichodesmium erythraeum IMS101 cultures. The specific nutrient limitation differentially affected the rate of colony formation and morphology of the colonies. Fe starvation promoted rapid colony fo

The nitrogen (N)-fixing cyanobacterium Trichodesmium is globally distributed in warm, oligotrophic oceans, where it contributes a substantial proportion of new N and fuels primary production. These photoautotrophs form macroscopic colonies that serve as relatively nutrient-rich substrates that are colonized by many other organisms. The nature of these associations may modulate ocean N and carbon (C) cycling, and can offer insights into marine co-evolutionary mechanisms. Here we integrate multiple omics-based and experimental approaches to investigate Trichodesmium-associated bacterial consorti

Marine pelagic cyanobacteria Trichodesmium are widespread in the New Caledonia lagoon. Blooms of these Oscillatoriales are suspected to be a potential source of toxins in the ciguatera food chain and were previously reported to contain certain types of paralysing toxins. In the present study, toxicity experiments were conducted on lipid- and water-soluble extracts of freeze-dried samples of these cyanobacteria. Lipid-soluble fractions revealed a ciguatoxin-like activity in both in vivo (mouse bioassay) and in vitro (mouse neuroblastoma cells assay and receptor binding assay using tritiated bre

Diazotrophic cyanobacteria such as Trichodesmium play a crucial role in the nitrogen budget of the oceans due to their capability to bind atmospheric nitrogen. Little is known about their interoceanic transport pathways and their distribution in upwelling regions. Trichodesmium has been detected using a Video Plankton Recorder (VPR) mounted on a remotely operated towed vehicle (TRIAXUS) in the southern and northern Benguela Upwelling System (BUS) in austral autumn, Feb/Mar 2019. The TRIAXUS, equipped with a CTD as well as fluorescence and nitrogen sensors, was towed at a speed of 8 kn on two o

As diazotrophic cyanobacteria of tremendous biomass, Trichodesmium continuously provide a nitrogen source for carbon-fixing cyanobacteria and drive the generation of primary productivity in marine environments. However, ocean iron deficiencies limit growth and metabolism of Trichodesmium. Recent studies have shown the co-occurrence of Trichodesmium and siderophore-producing Synechococcus in iron-deficient oceans, but whether siderophores secreted by Synechococcus can be used by Trichodesmium to adapt to iron deficiency is not clear. We constructed a mutant Synechococcus strain unable to produc

To protect the O2-sensitive nitrogenase enzyme, diazotrophic cyanobacteria separate N2 fixation from photosynthesis in space or time. However, in the filamentous Trichodesmium, technical limitations and conflicting results have so far prevented a consensus on the separation mechanism. Here, we specifically adapted single-cell methods for simultaneously capturing both photosynthesis and N2 fixation at the levels of gene expression, fixation and transport of carbon and nitrogen. Testing expression of nifH (nitrogenase) and psbA (photosystem II), we found that about half of the cells exclusively

The Trichodesmium genus comprises some of the most abundant N2-fixing organisms in oligotrophic marine ecosystems. Since nitrogenase, the key enzyme for N2 fixation, is irreversibly inhibited upon O2 exposure, these organisms have to coordinate their N2-fixing ability with simultaneous photosynthetic O2 production. Although being the principal object of many laboratory and field studies, the overall process of how Trichodesmium reconciles these two mutually exclusive processes remains unresolved. This is in part due to contradictory results that fuel the Trichodesmium enigma. In this review, w

The diazotrophic cyanobacterium, Trichodesmium, is an integral component of the marine nitrogen cycle and contributes significant amounts of new nitrogen to oligotrophic, tropical/subtropical ocean surface waters. Trichodesmium forms macroscopic, fusiform (tufts), spherical (puffs) and raft-like colonies that provide a pseudobenthic habitat for a host of other organisms including marine invertebrates, microeukaryotes and numerous other microbes. The diversity and activity of denitrifying bacteria found in association with the colonies was interrogated using a series of molecular-based methodol

An assessment of the major pigments and neurotoxins and a description of the phytoplankton community were carried out within the coastal region of Rio de Janeiro State (Brazil), during winter and the following spring of 2018. Overall, six stations were investigated for oceanographic conditions (with CTD casts). Filtered water samples were used to estimate the chlorophyll a (CHL-a), carotenoids (CAR), and phycobiliproteins (PHY) using UV-Vis spectrophotometry, as well as the quantification of saxitoxins (STX) and domoic acid (DA), through High Performance Liquid Chromatography (HPLC). Palytoxin (PLTX), one the most potent marine toxins, and/or its analogs, have been identified in different marine organisms, such as Palythoa soft corals, Ostreopsis dinoflagellates, and Trichodesmium cyanobacteria. Although the main concern for human health is PLTXs entrance in the human food chain, there is growing evidence of adverse effects associated with inhalational, cutaneous, and/or ocular exposure to aquarium soft corals contaminated by PLTXs or aquaria waters. Indeed, the number of case reports describing human poisonings after handling these cnidarians is continuously increasing. I

Methylphosphonate is an organic phosphorus compound used by microorganisms when phosphate, a key nutrient limiting growth in most marine surface waters, becomes unavailable. Microbial methylphosphonate use can result in the formation of methane, a potent greenhouse gas, in oxic waters where methane production is traditionally unexpected. The extent and controlling factors of such aerobic methane formation remain underexplored. Here, we show high potential net rates of methylphosphonate-driven methane formation (median 0.4 nmol methane L-1 d-1) in the upper water column of the western tropical

Diazotrophy is the most important nitrogen source in the oligotrophic surface ocean, but the organisms involved and their contributions are incompletely understood due to limited observations. Only diazotrophic organisms possess the nifH gene to reduce dinitrogen to ammonium, but their distribution and activity can only be quantified through sampling and experiments during research cruises. Some recent studies document small diatoms with symbionts able to fix nitrogen, a new source of biologically available nitrogen in addition to the well-known cyanobacterial species such as Trichodesmium or

Palytoxin (PLTX) is a complex marine toxin produced by Zoanthids (Palyhtoa), dinoflagellates (Ostreopsis), and cyanobacteria (Trichodesmium). Contact with PLTX-like compounds present in aerosols or marine organisms has been associated with adverse effects on humans. The worldwide distribution of producer species and seafood contaminated with PLTX-like molecules illustrates the global threat to human health. The identification of species capable of palytoxin production is critical for human safety. We studied the presence of PLTX analogues in Palythoa canariensis, a coral species collected in t
