A female heterozygous patient with Fabry's disease with renal accumulation of trihexosylceramide detected with a monoclonal antibody.

We describe a female patient with heterozygous Fabry's disease. The patient had persistent proteinuria and microhematuria but lacked any other diagnostic signs such as corneal and cutaneous involvement. Kidney tissue obtained at biopsy showed the segmentally distributed enlarged glomerular epithelia. These cells were filled with vacuolated foamy cytoplasm, which had lamellar and myelinoid structures under electronmicroscopic observation. Accumulation of trihexosylceramide (CTH) in these foamy epithelial cells was confirmed with immunohistochemical staining with the use of anti-CTH monoclonal a

Metabolic differences in female adolescents with obsessive-compulsive disorder and trichotillomania: insights from plasma untargeted metabolomic analysis.

BACKGROUND: Obsessive-Compulsive Disorder (OCD) and Trichotillomania (TTM) are classified together as Obsessive-Compulsive and Related Disorders, yet their distinct neurobiological underpinnings remain poorly understood. This study represents the first untargeted metabolomic analysis comparing adolescents with OCD and TTM to identify differential metabolic signatures. METHOD: This cross-sectional study included 62 female adolescents aged 10-18 years: 20 with OCD, 22 with TTM, and 20 healthy controls. Plasma samples were analyzed using liquid chromatography-quadrupole time-of-flight mass spect

In-vitro biosynthesis of sulphatoglycosphingolipids by rat submandibular salivary glands.

A sulphotransferase activity, concentrated mainly in the microsomal fraction, which catalyses the transfer of sulphate group from 3'-phosphoadenosine-5'-phosphosulphate to galactosylceramide and lactosylceramide was demonstrated in rat submandibular and sublingual glands. However, the sulphotransferase activity of this fraction in submandibular glands was about ten times higher than in sublingual glands. Optimum enzyme activity was obtained using the detergent Triton X-100, F-, and Mg2+ at a pH of 6.8. The enzyme did not catalyse the transfer of sulphate to glucosylceramide, trihexosylceramide

Lipid composition of microdomains is altered in a cell model of Gaucher disease.

The formation of cholesterol and sphingolipids into specialized liquid-ordered membrane microdomains (rafts) has been proposed to function in the intracellular sorting and transport of proteins and lipids. Defined by biochemical criteria, rafts resist solubilization in nonionic detergents, enabling them to be isolated as detergent-resistant membranes (DRM). In this study, we characterized the lipid composition of DRM from a cell model of the sphingolipid storage disorder, Gaucher disease, in which the catabolism of the sphingolipid glucosylceramide (GC) is impaired. In this cell model, we show

Isolation and partial characterization of fucose- and N-acetylglucosamine-containing neutral glycosphingolipids from human senile cataracts.

Five neutral glycosphingolipids were isolated from human cataracts using silicic acid column chromatography and preparative thin-layer chromatography. Three of these glycolipids were partially identified by gas-liquid chromatography as glucosylceramide, lactosylceramide, and trihexosylceramide. Two glucosamine-containing glycosphingolipids (one of which contained fucose) were also detected. One of these two lipids contained galactose, glucose, N-acetylglucosamine in the molar ratio of 2:1:1, while the other contained fucose, galactose, glucose, and N-acetylglucosamine with the molar ratio of 1

Human Gb3/CD77 synthase: a glycosyltransferase at the crossroads of immunohematology, toxicology, and cancer research.

Human Gb3/CD77 synthase (α1,4-galactosyltransferase, P1/Pk synthase, UDP-galactose: β-D-galactosyl-β1-R 4-α-D-galactosyltransferase, EC 2.4.1.228) forms Galα1 → 4Gal structures on glycosphingolipids and glycoproteins. These glycans are recognized by bacterial adhesins and toxins. Globotriaosylceramide (Gb3), the major product of Gb3/CD77 synthase, is a glycosphingolipid located predominantly in plasma membrane lipid rafts, where it serves as a main receptor for Shiga toxins released by enterohemorrhagic Escherichia coli and Shigella dysenteriae of serotype 1. On the other hand, accumulation of

The glycosphingolipids of rat sublingual and submaxillary glands.

1. Glycosphingolipids have been isolated from rat sublingual and submaxillary glands by the procedure involving lipid extraction, column fractionation and thin-layer chromatography. 2. The major neutral glycosphingolipids in rat sublingual and submaxillary glands were monohexosylceramide, dihexosylceramide, tetrahexosylceramide and pentahexosylceramide. Both types of glands exhibited a low content of trihexosylceramide. The fucose-containing glycosphingolipids were not found. 3. The acidic glycosphingolipids in rat sublingual and submaxillary glands were composed of monohexose sulfatide, dihex

Glucosylceramide accumulation is not confined to the lysosome in fibroblasts from patients with Gaucher disease.

Gaucher disease (GD) is an inborn error of glycosphingolipid metabolism resulting from a deficiency of the lysosomal enzyme beta-glucosidase leading to the accumulation of glucosylceramide (GC) in lysosomes of affected cells. In order to determine the effect of GC accumulation on intracellular lipid content in fibroblasts from patients with GD, we measured individual species of ceramide, di- and trihexosylceramide, sphingomyelin, phosphatidylcholine, phosphatidylinositol and phosphatidylglycerol using electrospray ionisation-tandem mass spectrometry. The different subspecies of each lipid clas

[Laboratory diagnosis of Fabry disease: historical perspectives and recent breakthroughs].

Due to its high sensitivity and specificity, determination of the c-galactosidase A activity in leukocytes is the gold standard to confirm the diagnosis of Fabry disease in hemizygous moles. In contrast, heterozygous females cannot be dependably diagnosed by this method and genotyping should always be carried out. Two recent breakthroughs have contributed to the rise of screening programs for Fabry disease: first, the use of filter papers to collect blood or urine spots from individuals in at-risk populations for Fabry disease, second, the development of moss spectrometry Substrate accumulatio

Identification of Four New Mutations in the GLA Gene Associated with Anderson-Fabry Disease.

Anderson-Fabry disease is a hereditary, progressive, multisystemic lysosomal storage disorder caused by a functional deficiency of the enzyme α-galactosidase A (α-GalA). This defect is due to mutations in the GLA gene, located in the long arm of the X chromosome (Xq21-22). Functional deficiency of the α-GalA enzyme leads to reduced degradation and accumulation of its substrates, predominantly globotriaosylceramide (Gb3), which accumulate in the lysosomes of numerous cell types, giving rise to the symptomatology. Clinical diagnosis can still be difficult today due to the peculiarities of the di

An antibody binding-based fluorescent assay for the rapid quantification of globotriaosylceramide levels in human Fabry cells.

Fabry disease is caused by reduced α-GAL A activity and accumulation of globotriaosylceramide (Gb3). Here, we describe a microplate Gb3 assay using fluorophore-tagged antibody and crude cellular lipid extracts. The assay is able to detect higher Gb3 concentrations in human Fabry cells compared to non-diseased cells. This result was verified by immunofluorescence staining that revealed large amounts of Gb3 deposits in Fabry cell lines, demonstrating the accuracy of this method. This assay may provide the basis for detecting Fabry disease by quantifying Gb3 deposits from human biological samples

Is it Fabry disease?

Fabry disease is caused by mutations in the GLA gene that lower α-galactosidase A activity to less than 25-30% of the mean normal level. Several GLA variants have been identified that are associated with relatively elevated residual α-galactosidase A. The challenge is to determine which GLA variants can cause clinical manifestations related to Fabry disease. Here, we review the various types of GLA variants and recommend that pathogenicity be considered only when associated with elevated globotriaosylceramide in disease-relevant organs and tissues as analyzed by mass spectrometry. This criteri

D313Y Variant in Fabry Disease: A Systematic Review and Meta-analysis.

BACKGROUND AND OBJECTIVES: There is accumulating evidence in the literature indicating a strong correlation between Fabry disease (FD) phenotypes and specific sequence variations in the Galactosidase Alpha (GLA) gene. Among them, the potential pathogenicity and clinical relevance of D313Y variation in patients with FD remain debated. METHODS: We performed a systematic review and meta-analysis of studies reporting D313Y as single occurring variant in the GLA gene and sought to evaluate (1) the prevalence of D313Y variation in different populations with or without clinical manifestations of FD,

Characterization of a novel bacterial adhesion specificity of Streptococcus suis recognizing blood group P receptor oligosaccharides.

Streptococcus suis causes sepsis, meningitis, and other serious infections in piglets, and meningitis in humans. Hemagglutination inhibition experiments with mono- and oligosaccharides and glycoproteins indicated that galactose-binding strains of S. suis recognized the Gal alpha 1-4Gal sequence present in the P1 and Pk blood group antigen structures. In thin-layer chromatography overlay assays the bacteria bound to trihexosylceramide (GbO3) but not to globoside (GbO4) or Forssman glycolipid (GbO5), in contrast to P-fimbriated Escherichia coli, which bound only to the latter two. The S. suis ad

Plasma lipidomic analysis of sphingolipids in patients with large artery atherosclerosis cerebrovascular disease and cerebral small vessel disease.

BACKGROUND: Sphingolipids mainly consist of ceramides (Cer), sphingomyelins (SM) and glycosphingolipids. Sphingolipids are related with coronary heart disease and metabolic disease, but there're few studies about cerebrovascular disease. The purpose was to detect sphingolipids in plasma of patients with large artery atherosclerosis (LAA) cerebrovascular disease and cerebral small vessel disease (CSVD) to explore the similarities and differences of pathogenesis of the two subtypes. METHODS: 20 patients with LAA cerebrovascular disease, 20 patients with age-related CSVD, 10 patients with Fabry
