Serum triiodothyronine and reverse triiodothyronine concentrations after surgical operation.

Serum-triiodothyronine (T3) concentrations fell rapidly after surgery in six out of seven initially euthyroid patients. Simultaneous increases in reverse triiodothyronine (rT3) concentrations suggested that the peripheral monodeiodination of thyroxine (T4) proceeds by an alternative pathway in the postoperative period.

Reverse triiodothyronine nuclear binding in rat brain.

Reverse triiodothyronine (rT3) had been believed biologically inactive, but recently we demonstrated nuclear binding sites for rT3 in human placenta. In this study we examined rT3 binding sites in rat brain. Male Wistar normal rats aged 2, 4 and 9 weeks were killed and the brain was removed for rT3 binding assay. In another series, male Wistar rats weighing about 40g were divided into two groups: group 1 (rickets group), kept on our original rachitogenic diet and group 2, kept on standard diet. After 28 days on either diet they were killed and the brain was removed for assay. Cerebral nuclear

Amniotic fluid reverse triiodothyronine in complicated pregnancy.

The concentrations of reverse triiodothyronine (3,3',5'-T3 or rT3) in amniotic fluid (AF) were measured by radioimmunoassay in 81 patients with various complications of pregnancy and in 39 normal pregnant patients at equivalent gestational age. In normal pregnancy, AFrT3 concentrations decreased with advancing gestational age. At 21-25, 26-30, 31-35, and 36-40 weeks of normal pregnancy, AFrT3 concentrations (mean +/- SE) were 353 +/- 62 (n = 6), 131 +/- 49 (n = 7), 94 +/- 25 (n = 14), and 93 +/- 5 (n = 20) ng/dl, respectively (ranges: 200-600, 57-350, 66-135, and 50-135). Measurement of Reverse Triiodothyronine Level and the Triiodothyronine to Reverse Triiodothyronine Ratio in Dried Blood Spot Samples at Birth May Facilitate Early Detection of Monocarboxylate Transporter 8 Deficiency.

Background: Monocarboxylate transporter 8 (MCT8) deficiency is an X-chromosome-linked neurodevelopmental disorder resulting from impaired thyroid hormone transport across the cell membrane. The diagnosis of MCT8 deficiency is typically delayed owing to the late appearance of signs and symptoms as well as the inability of standard biomarkers of neonatal screening to provide early detection. In this study, we report, for the first time, the ability to detect MCT8 deficiency at birth using dried blood spot (DBS) samples. Methods: We retrospectively measured triiodothyronine (T3), thyroxine (T4),

Low serum triiodothyronine and high serum reverse triiodothyronine in old age: an effect of disease not age.

Serum concentrations of T4, T3, T3, free T4, free T3, and TSH were determined in four groups of adult subjects: group A, 27 healthy young volunteers (aged 18-29 yr); group B, 24 carefully selected healthy elderly subjects (aged 70-90 yr); group C, 41 subjects living at a municipal nursing home for the elderly (aged 70-90 yr); and group D, 35 hospitalized patients (aged 70-90 yr). Identical levels of iodothyronines in serum were found in the young and in the elderly healthy subjects. Moderate and severe disease induced alterations in iodothyronine concentrations in serum comparable to those rep

Clinical and laboratory aspects of 3,3',5'-triiodothyronine (reverse T3).

Reverse T3 (3,3',5'-triiodothyronine or rT3) is the third most abundant iodothyronine circulating in human blood and is produced by the inner ring deiodination of the pro-hormone thyroxine (T4). Unlike the more abundant and active metabolite T3, the measurement of serum rT3 is yet to find a routine clinical application. As rT3 binds weakly to the T3 thyroid nuclear hormone receptors, it is thought to represent an inactive end-product of thyroid hormone metabolism, diverting T4 away from T3 production. The analysis of serum rT3 has, up until recently, been measured by competitive radioimmunoass

Dissociated thyroxine, triiodothyronine and reverse triiodothyronine levels in patients with familial goitre due to iodide organification defects.

The thyroid function of patients with three different types of organification defect was studied. All patients were characterized by a high thyroidal 131I uptake and a positive perchlorate discharge. Patients with Pendred's syndrome who had goitre and congenital nerve deafness were mostly euthyroid with normal circulating thyroid hormone levels. Only two of them had compensated euthyroidism with elevated total T3, high basal TSH and delayed return to basal value with TRH. The patients who were euthyroid with large goitres and normal hearing had elevated total T3 and an exaggerated TSH response

Reverse triiodothyronine does not alter pituitary-thyroid function in normal subjects.

Serum rT3 concentrations are often increased in patients with nonthyroid illness. Such elevations could be responsible for some of the alterations in pituitary-thyroid function that occur in such patients, particularly since rT3 is a potent inhibitor of extrathyroidal T3 production in vitro. To evaluate the role of serum rT3 elevations in the regulation of the hypothalamic-pituitary-thyroid axis, 10 normal subjects were given 3 mg rT3, orally, in divided doses for 4 days. Serum rT3 concentrations were elevated at least 10-fold by the end of the first day of treatment. Mean serum T4 and T3 conc

Serum reverse triiodothyronine in dogs with hypothyroidism, nonthyroidal illness syndrome and healthy dogs.

BACKGROUND: Diagnosing canine hypothyroidism remains challenging because decreased total thyroxine (tT4) concentrations are not specific for hypothyroidism and are frequently observed in dogs with non-thyroidal illness syndrome (NTIS). Although additional thyrotropin (TSH) and free thyroxine (fT4) measurements may improve diagnostic accuracy, differentiation between hypothyroidism and NTIS remains difficult in clinical practice. In human medicine, reverse triiodothyronine (rT3), an inactive metabolite of thyroxine, has been proposed as a useful marker to distinguish hypothyroidism from NTIS. H

Association between reverse triiodothyronine and cardiac complications in patients with uncontrolled Graves' disease.

BACKGROUND: Cardiac complications associated with poorly controlled Graves' disease present a significant challenge in managing hyperthyroid patients. The relationship between thyrotoxicosis severity and cardiac complications remains to be confirmed. This study investigates the association between reverse triiodothyronine (rT3) and cardiac complications in hospitalized patients with uncontrolled Graves' disease. METHODS: This cross-sectional study evaluated 178 hospitalized patients with uncontrolled Graves' disease from 2016 to 2020. After excluding cardiac diseases unrelated to Graves' dise

Triiodothyronine (T3) antagonizes adverse effects of high circulating reverse-T3 (rT3) during hemorrhagic shock.

To examine whether triiodothyronine (T3) could counteract the lethal effect of exogenous reverse T3 (rT3) in hemorrhagic shock, 21 anesthetized, heparinized mongrel dogs were given 15 micrograms/kg of rT3 IV. Thirty minutes later, the dogs were bled rapidly into a reservoir to achieve and maintain a mean arterial pressure of 40 mm Hg. After 60 minutes at 40 mm Hg (compensated shock), the reservoir line was clamped for 30 minutes (uncompensated shock). The shed blood was then reinfused over 30 minutes, and the dogs were monitored for an additional 60 minutes. At the start of uncompensated shock

[The conversion of thyroxine to triiodothyronine (T3) or to reverse T3 in patients with thyroid dysfunction].

In order to clarify the conversion of thyroxine (T4) to triiodothyronine (T3) or to reverse T3 (rT3), serum concentrations of T4, T3, rT3, thyrotropin (TSH), thyroxine-binding globulin (TBG) and values of T3 uptake (T3 U) were measured in 61 hyperthyroid and 31 hypothyroid patients, 8 patients with subacute thyroiditis, and 40 normal subjects. Then, free T4 index (FT4I), T3/T4, rT3/T4, and rT3/T3 ratio were calculated. The rT3/T3 ratio was high in the hyperthyroid patients and low in the hypothyroid patients compared with that in the normal subjects. The ratio was positively related to serum T

Triiodothyronine administration reverses vitamin A deficiency-related hypo-expression of retinoic acid and triiodothyronine nuclear receptors and of neurogranin in rat brain.

Recent studies have revealed that retinoids play an important role in the adult central nervous system and cognitive functions. Previous investigations in mice have shown that vitamin A deficiency (VAD) generates a hypo-expression of retinoic acid (RA, the active metabolite of vitamin A) receptors and of neurogranin (RC3, a neuronal protein involved in synaptic plasticity) and a concomitant selective behavioural impairment. Knowing that RC3 is both a triiodothyronine (T3) and a RA target gene, and in consideration of the relationships between the signalling pathways of retinoids and thyroid ho

Increased reverse triiodothyronine is associated with shorter survival in independently-living elderly: the Alsanut study.

OBJECTIVE: Increased reverse tritiodothyronine (T(3)) used to be described as a part of euthyroid sick syndrome (ESS). It was demonstrated to be associated with increased mortality in acutely ill patients. It can also be found with low or normal T(3) in non-severely ill subjects but its significance remains unclear. PATIENTS AND DESIGN: The Alsanut study included a representative sample of 440 independently-living subjects aged 65 or over constituted between January 1988 and September 1989. Past and current medical history and nutritional data were collected at inclusion. Baseline thyroid hor

Thyroid hormone status and nutrient intake in the free-living elderly. Interest of reverse triiodothyronine assessment.

Fasting or hypercaloric diets are established methods of inducing low triiodothyronine (T3) levels that resemble the sick euthyroid syndrome in adults, but little is known on the mechanisms of this syndrome in the elderly. Decreasing T3 does not seem to be an unavoidable consequence of ageing, but the role of illness or other factors in this decline remains unclear. The aim of this work was to study the influence of nutritional factors on thyroid hormone levels in free-living elderly subjects. A 3-day dietary survey was conducted in 440 randomized subjects aged between 65 and 96. Cholesterol,

Effects of thyroid hormones and reverse-triiodothyronine (rT3) pretreatment on beta-adrenoreceptors in the rat heart.

Effects of thyroxine (T4), triiodothyronine (T3) or reverse-triiodothyronine (rT3) in vivo pretreatment or the effect of hypothyroidism on properties of beta-adrenoreceptors in the rat heart were investigated. beta-adrenergic antagonist hydroxybenzylpindolol (HYP125I) was used to estimate the maximal binding capacity (MBC) and the affinity (KD) of beta-adrenoreceptors. Both T4 and T3 in dose and time dependent manner increased MBC value but only slightly and insignificantly affected affinity of the beta-adrenoreceptor. The effect of T3 on MBC was higher than that of an equal dose of T4, but th
