Nicotinamide mononucleotide maintains cytoskeletal stability and fortifies mitochondrial function to mitigate oocyte damage induced by Triocresyl phosphate.

Triocresyl phosphate (TOCP) was commonly used as flame retardant, plasticizer, lubricant, and jet fuel additive. Studies have shown adverse effects of TOCP on the reproductive system. However, the potential harm brought by TOCP, especially to mammalian female reproductive cells, remains a mystery. In this study, we employed an in vitro model for the first time to investigate the effects of TOCP on the maturation process of mouse oocytes. TOCP exposure hampered the meiotic division process, as evidenced by a reduction in the extrusion of the first polar body from oocytes. Subsequent research re

Tricresyl phosphate inhibits fertilization in Japanese medaka (Oryzias latipes): Emphasizing metabolic toxicity.

As tricresyl phosphate (TCrP) is commonly found in global water sources, its potential reproductive toxicity to fish is of increasing concern. Japanese medaka larvae were exposed to TCrP at 657.9, 1,511, and 4042 ng/L for 100 days. We identified significant fertilization inhibition (6.9%-12.8%) in all exposure groups. Intersex was significantly induced at 4042 ng/L, with an incidence of 22.0%. TCrP exposure also caused dilation of the efferent duct in the testes with maximum duct widths of 83.3, 93.2, and 149.7 μm in the 657.9, 1,511, and 4042 ng/L exposure groups, respectively. These widths w

Health risk assessment of exposure to TriCresyl Phosphates (TCPs) in aircraft: a commentary.

Possible exposure to TriCresyl Phosphates (TCPs) has led to concerns among airline crew members. One isomer, Tri-ortho-Cresyl Phosphate (ToCP) is known to be neurotoxic and exposure to ToCP via contaminated cabin air has been suggested to be associated with the alleged Aerotoxic syndrome. The symptoms associated with Aerotoxic syndrome are diverse, including headaches, loss of balance, numbness and neurobehavioral abnormalities such as emotional instability, depression and cognitive dysfunction. Other ortho-isomers are toxic as well, but the non-ortho isomers are regarded as less toxic. In a c

Evaluation of the hazards of industrial exposure to tricresyl phosphate: a review and interpretation of the literature.

Commercial tricresyl phosphate (TCP) is a heterogeneous mixture of isomers and aryl phosphate congeners, known for many years to induce delayed neurotoxicity (OPIDN) in humans and experimental animals. In the past the isomer tri-o-cresyl phosphate (TOCP) was thought to be the component primarily responsible for OPIDN. It is now clear that other constituents, particularly the mono-o-esters, are not only neurotoxic but also may be more potent than pure TOCP. As a generality, the toxicity potential of a particular brand of TCP is related to its content of o-phenolic residues, whereby the maximal

An acute toxic neuropathy caused by organo-phosphate poisoning in hens.

OBJECTIVE: Organophosphorus compounds can induce an acute toxic peripheral neuropathy. In hens, the acute peripheral neuropathy was induced by poisoning with organo-phosphorus compound (OPC)--tri-ortho-cresyl phosphate (TOCP). METHODS: In the course of an acute TOCP-induced toxic neuropathy in hens the activity of following enzymes was analysed: asparaginase, glutaminase, glutamat-dehydrogenase, AMP and adenosine deaminases and 5'nucleotidase; ALT (SGPT), AST (SGOT) and proteins levels were estimated. RESULTS: A decrease in activity of all analysed enzymes was observed; the amount of protein

Acute tricresylphosphate intoxication in childhood.

Tricresylphosphates (TCP) have been responsible for a large number of epidemic and individual intoxications since the recognition, in 1899, that phospho-creosote was the cause of paralysis in patients being treated for tuberculosis. Although children are mentioned as being among the victims of epidemic intoxication, no well documented reports of acute tricresylphosphate ingestion in childhood are available in the literature. We report a case of severe intoxication in a 4 1/2-year-old child following ingestion of a lubricant containing TCP. Clinical findings in this child were typical of previo

Allergic contact dermatitis to adhesive bandages.

More than two billion Band-Aid Brand Sheer Strips are used in the United States yearly, yet allergic contact dermatitis resulting from their use is nearly nonexistent. We report four patients with allergic reactions to these strips. One patient reacted to tricresyl phosphate, the plasticizer in the vinyl backing; another patient was allergic to 2,5-di(tertiary-amyl)hydroquinone, the antioxidant in the adhesive. In the other two patients, the allergic contact dermatitis remains unexplained.

Neuroprotective effect of copper on neurotoxicity of TOCP in vivo.

A single or repeated dose of tri-ortho-cresyl phosphate (TOCP) induces in humans and animals a known neuropathy as organophosphorus-induced delayed polyneuropathy (OPIDP). This syndrome is related to the inhibition of neuropathy target esterase (NTE), causing signs and symptoms in the nervous system, such as ataxia and paralysis. Currently, there is no antidotal treatment for OPIDP. In the present study, the neuroprotective effect of Cu(II) on the acute in vivo neurotoxicity of TOCP is characterized. Adult hens (27 and 65 weeks old) were administered a single dose of 380, 500, 750, or 1000 mg/

Subchronic Exposure of Tri-Ortho-Cresyl Phosphate Reduces T-Cell Population in Mice.

Tri-ortho-cresyl phosphate (TOCP) is a widely used organophosphorus compound in industry and agriculture, but its immunotoxicity has not been thoroughly investigated. Therefore, this study aimed to assess the immunotoxicity of TOCP in mice by examining changes in immune cell populations and histopathological alterations in the spleen. Male ICR mice were administered TOCP via daily oral gavage at doses of 200 and 400 mg/kg body weight (BW)/d for 28 days to evaluate its effects on immune cell populations in blood and spleen. TOCP treatment at 400 mg/kg BW/d reduced white blood cell counts by 18%

Triaryl phosphate poisoning in cattle.

Clinical signs, pathologic changes and biochemical changes occurred in cattle with natural and experimental triaryl phosphate poisoning. Natural poisoning was caused by triaryl phosphates escaping from a gas pipeline compressor station. The clinical signs were posterior motor paralysis, dyspnea, diarrhea and agalactia. Experimental doses of 1/2-1 gm/kg body weight of these organophosphate compounds caused depression of cholinesterase and axonal degeneration in the spinal cord.

Thermal oxidative degradation studies of phosphate esters.

Five phosphate esters - tri-p-tolyl phosphate, tributyl phosphate, tris(1,3-dichloro-2-propyl)phosphate, tris(2,3-dibromopropyl)phosphate, and tris(2-chloroethyl)phosphate- were subjected to thermal oxidative degradation in air at 370 degrees C. Degradation mechanisms were postulated and the toxic hazards assessed based on the volatiles produced. Tri-p-tolyl phosphate was found to undergo only minimal degradation; the other compounds were decomposed extensively. Butene was the main product formed on tributyl phosphate decomposition; hydrogen halides and halogenated C2- and C3- species were the

Quantitation of ortho-cresyl phosphate adducts to butyrylcholinesterase in human serum by immunomagnetic-UHPLC-MS/MS.

Tri-ortho-cresyl phosphate (ToCP) is an anti-wear, flame retardant additive used in industrial lubricants, hydraulic fluids and gasoline. The neurotoxic effects of ToCP arise from the liver-activated metabolite 2-(o-cresyl)-4H-1,3,2-benzodioxaphosphoran-2-one (cresyl saligenin phosphate or CBDP), which inhibits esterase enzymes including butyrylcholinesterase (BChE). Following BChE adduction, CBDP undergoes hydrolysis to form the aged adduct ortho-cresyl phosphoserine (oCP-BChE), thus providing a biomarker of CBDP exposure. Previous studies have identified ToCP in aircraft cabin and cockpit ai

October 1942: a strange epidemic paralysis in Saval, Verona, Italy. Revision and diagnosis 50 years later of tri-ortho-cresyl phosphate poisoning.

In the autumn of 1942 a strange epidemic paralysis started in Saval, at that time a country area but now part of the city of Verona. The epidemic went on for several months and affected 41 people, all working as owners or labourers on the same farm. Some of the farm animals (chickens, horses, cattle, pigs) also became ill. About 20 patients were admitted to the nearby city hospital. The outbreak was diagnosed as polyneuritis with a probable viral cause. Fifty years later, seven people with sequelae of the disease were examined. The most severe cases present a spastic paraplegia and lower leg m

Involvement of neuropathy target esterase in tri-ortho-cresyl phosphate-induced testicular spermatogenesis failure and growth inhibition of spermatogonial stem cells in mice.

Tri-ortho-cresyl phosphate (TOCP) has been widely used in industry and reported to induce delayed neurotoxicity in humans and animals. In addition, it is known to have a deleterious effect on the male reproductive system in animals, but the precise mechanism is yet to be elucidated. The present study shows that TOCP could disrupt the seminiferous epithelium in the mouse testis and decrease the sperm density in the epididymis in a dose-dependent manner. Neuropathy target esterase (NTE) was shown to exist in mouse spermatogenic cells, including spermatogonial stem cells and to be significantly i

Delayed neurotoxic effects of tri-o-tolyl phosphate in the European ferret.

The development of organophosphorus-induced delayed neurotoxicity (OPIDN) was studied in the European ferret (Mustela putorius furo). A single oral or dermal dose of 250, 500, or 1000 mg tri-o-tolyl phosphate (TOTP)/kg body weight was administered to adult male ferrets. Corn oil served as the vehicle in the oral test and 95% ethanol was the vehicle in the dermal test. At 48 h posttreatment, half the animals in each group were killed by cervical dislocation for assessment of whole-brain neuropathy target esterase (NTE) activity. The remaining 5 animals per group were observed and examined neuro

Subchronic exposure to tri-ortho-cresyl phosphate induces nephrotoxicity through induction of oxidative stress with neuropathy target esterase inhibition.

Tri-ortho-cresyl phosphate (TOCP), an organophosphate compound widely used in industry as a lubricant, plasticizer, and hydraulic fluid, could cause organophosphate-induced delayed neuropathy (OPIDN) which is a phospholipase and involved in phospholipid metabolism. However, whether TOCP could induce nephrotoxicity is not clear. In this study, we used a mouse model with subchronic exposure to TOCP, and a human renal proximal tubular epithelial cell line HK-2 cells as the in vitro model to study the TOCP-induced nephrotoxicity. We found that TOCP-exposed mice developed severe renal dysfunctions,

Clinical and electrophysiological study of neuropathy after organophosphorus compounds poisoning.

Clinical and electrophysiological examinations were performed on 12 patients with toxic neuropathy following accidental ingestion of alcohol polluted by triorthocresyl phosphate (TOCP). Concurrent PNS and CNS lesions were found in all patients. Two to three months after ingestion, five of them showed prevalent signs of mixed, sensorimotor polyneuropathy, especially motor and distal, and the electrophysiological data pointed to the mixed process of axonal degeneration and secondary demyelination. In two of these five patients in whom examinations were repeated 13 years after TOCP ingestion, the
