The serologic specificity of tropocollagen telopeptides.

Tropocollagen preparations from carp, buffalo fish, rats, calves, sheep, and humans have been studied by electron microscopy and serologic methods. Tropocollagens from each species appeared identical by electron microscopy but they were readily distinguished (except between sheep and calves) by C'-fixation tests with rabbit antisera against the various tropocollagens. Tests with calf tropocollagen antiserum showed no distinction between tropocollagen isolated from different tissues nor between individuals of the same or different strains. The major immunogenic sites in native tropocollagen are

Effect of changes in tropocollagen residue sequence and hydroxyapatite mineral texture on the strength of ideal nanoscale tropocollagen-hydroxyapatite biomaterials.

Changes in mineral texture (e.g. hydroxyapatite (HAP) or aragonite) and polypeptide (e.g. tropocollagen (TC)) residue sequence are characteristic features of a disease known as osteogenesis imperfecta (OI). In OI, different possibilities of changes in polypeptide residue sequence as well as changes in polypeptide helix replacement (e.g. 3 alpha1 chains instead of 2 alpha1 and 1 alpha2 chain in OI murine) exist. The cross section of the HAP crystals could be needle like or plate like. Such texture and residue sequence related changes can significantly affect the material strength at the nanosca

Tropocollagen springs allow collagen fibrils to stretch elastically.

The mechanical properties of connective tissues are tailored to their specific function, and changes can lead to dysfunction and pathology. In most mammalian tissues the mechanical environment is governed by the micro- and nano-scale structure of collagen and its interaction with other tissue components, however these hierarchical properties remain poorly understood. In this study we use the human cornea as a model system to characterise and quantify the dominant deformation mechanisms of connective tissue in response to cyclic loads of physiological magnitude. Synchronised biomechanical testi

Single molecule effects of osteogenesis imperfecta mutations in tropocollagen protein domains.

Osteogenesis imperfecta (OI) is a genetic disease characterized by fragile bones, skeletal deformities and, in severe cases, prenatal death that affects more than 1 in 10,000 individuals. Here we show by full atomistic simulation in explicit solvent that OI mutations have a significant influence on the mechanical properties of single tropocollagen molecules, and that the severity of different forms of OI is directly correlated with the reduction of the mechanical stiffness of individual tropocollagen molecules. The reduction of molecular stiffness provides insight into the molecular-scale mech

Alport syndrome mutations in type IV tropocollagen alter molecular structure and nanomechanical properties.

Alport Syndrome is a genetic disease characterized by breakdown of the glomerular basement membrane (GBM) around blood vessels in the kidney, leading to kidney failure in most patients. It is the second most inherited kidney disease in the US, and many other symptoms are associated with the disease, including hearing loss and ocular lesions. Here we probe the molecular level structure-property relationships of this disease using a bottom-up computational materiomics approach implemented through large-scale molecular dynamics simulation. Since the GBM is under constant mechanical loading due to

Effects of Hydroxylation and Packing Geometry on Tropocollagen Stability: Insights from Molecular Dynamics Simulations.

Collagen is the most prevalent protein in living organisms, playing diverse roles across multiple tissues. Its hierarchical structure relies on the assembly of tropocollagen, the fundamental building block of collagen fibrils. This assembly occurs in a predominantly random manner, allowing for variations in packing. This randomness can lead to regions of both tight and nontight packing within the fibrils. The mechanisms by which these regions influence collagen's packing configurations, structural organization, and functional properties remain poorly understood. This study provides a focused i

Fabrication and Characterization of Fish Tropocollagen Sponge Enriched with Nanodiamonds for Potential Wound Dressing Applications.

The development of collagen-based composite materials offers new opportunities for designing bioactive porous structures with tunable properties. This study focuses on sponges or scaffolds fabricated from fish skin-derived tropocollagen combined with detonation nanodiamonds (NDs), aiming to explore how incorporation of NDs and application of radiation, as a potential sterilization method, influence structural and functional characteristics of the material. Freeze-dry methods of sponge fabrication resulted in a bilayered structure of open porosity, with microporosity at the top and a microchann

Exploration of tropocollagen unwinding by cathepsin K dimer with chondroitin 4-sulphate through microsecond timescale molecular dynamics.

Cathepsin K (CatK), a promising therapeutic target for bone-related diseases, uniquely unfolds triple-helical collagen and digests tropocollagen into soluble peptides in the presence of chondroitin 4-sulphate (C4-S). However, the molecular mechanism of CatK-mediated collagenolysis remains poorly understood, hindering the rational design of selective inhibitors. In this study, we performed microsecond-scale molecular dynamics simulations of a fully solvated ternary complex comprising the CatK dimer, tropocollagen segment, and C4-S, to indicate the structural and dynamical basis of tropocollagen

Pilot study on the effect of tropocollagen injections on re-epithelialisation rate of hard-to-heal venous leg ulcers.

OBJECTIVE: Collagen is a key component of the extracellular matrix, providing structural integrity and supporting tissue remodelling. Its biodegradability, biocompatibility and resistance to degradation make it ideal for biomedical applications, such as wound healing, tissue engineering and surgical adhesives. Collagen also plays a vital role in wound healing by supporting clot formation, attracting growth factors and promoting tissue regeneration. The aim of this study was to use the technique of tropocollagen injections and evaluate its effect on wound healing. METHOD: In this pilot study,

Obesity-induced leptin resistance is directly involved in skin fragility by decreasing type I collagen synthesis.

The relationship between obesity-induced leptin resistance and skin function remains unclear. In this study, we examined the effects of leptin signaling on collagen production in mouse skin. Mice were divided into three groups: control diet, high-fat diet, and switching diet (from the high-fat diet to the control diet). We measured the expression of leptin signaling-related genes and type I tropocollagen levels in the skin. In an additional experiment, leptin was administered to leptin-deficient ob/ob mice to examine the direct effect of leptin signaling on type I collagen synthesis in the ski

Importance of amino acid composition to improve skin collagen protein synthesis rates in UV-irradiated mice.

Skin collagen metabolism abnormalities induced by ultraviolet (UV) radiation are the major causes of skin photoaging. It has been shown that the one-time exposure of UV irradiation decreases procollagen mRNA expression in dermis and that chronic UV irradiation decreases collagen amounts and induces wrinkle formation. Amino acids are generally known to regulate protein metabolism. Therefore, we investigated the effects of UV irradiation and various orally administered amino acids on skin collagen synthesis rates. Groups of 4-5 male, 8-week-old HR-1 hairless mice were irradiated with UVB (66 mJ/

Unidirectional binding of clostridial collagenase to triple helical substrates.

Histotoxic clostridia produce collagenases responsible for extensive tissue destruction in gas gangrene. The C-terminal collagen-binding domain (CBD) of these enzymes is the minimal segment required to bind to collagen fibril. Collagen binding efficiency of CBD is more pronounced in the presence of Ca(2+). We have shown that CBD can be functional to anchor growth factors in local tissue. A (1)H-(15)N HSQC NMR titration study with three different tropocollagen analogues ((POG)(10))(3), ((GPOG)(7)PRG)(3), and (GPRG(POG)(7)C-carbamidomethyl)(3), mapped a saddle-like binding cleft on CBD. Strain-induced collagen denaturation is rate dependent in failure of cerebral arteries.

While soft tissues are commonly damaged by mechanical loading, the manifestation of this damage at the microstructural level is not fully understood. Specifically, while rate-induced stiffening has been previously observed in cerebral arteries, associated changes in microstructural damage patterns following high-rate loading are largely undefined. In this study, we stretched porcine middle cerebral arteries to failure at 0.01 and >150 s-1, both axially and circumferentially, followed by probing for denatured tropocollagen using collagen hybridizing peptide (CHP). We found that collagen fibrils

Healing Process, Pain, and Health-Related Quality of Life in Patients with Venous Leg Ulcers Treated with Fish Collagen Gel: A 12-Week Randomized Single-Center Study.

The aim of the study was to assess the effectiveness of fish skin collagen and its impact on healing, pain intensity, and quality of life in patients with venous leg ulcers (VLUs). This study included 100 adults with VLUs. Eligible patients were randomized to either tropocollagen gel treatment (group A, n = 47) or placebo alone (group B, n = 45). We applied the gel to the periwound skin for 12 weeks. All groups received standard wound care, including class 2 compression therapy and wound hygiene procedures. We assessed the healing rate (cm2/week) and quality of life (QoL) using the Skindex-29

Collagen Fibril Intermolecular Spacing Changes with 2-Propanol: A Mechanism for Tissue Stiffness.

Materials composed primarily of collagen are important as surgical scaffolds and other medical devices and require flexibility. However, the factors that control the suppleness and flexibility of these materials are not well understood. Acellular dermal matrix materials in aqueous mixtures of 2-propanol were studied. Synchrotron-based small-angle X-ray scattering was used to characterize the collagen structure and structural arrangement. Stiffness was measured by bend tests. Bend modulus increased logarithmically with 2-propanol concentration from 0.5 kPa in water to 103 kPa in pure 2-propanol

Collagen pre-strain discontinuity at the bone-Cartilage interface.

The bone-cartilage unit (BCU) is a universal feature in diarthrodial joints, which is mechanically-graded and subjected to shear and compressive strains. Changes in the BCU have been linked to osteoarthritis (OA) progression. Here we report existence of a physiological internal strain gradient (pre-strain) across the BCU at the ultrastructural scale of the extracellular matrix (ECM) constituents, specifically the collagen fibril. We use X-ray scattering that probes changes in the axial periodicity of fibril-level D-stagger of tropocollagen molecules in the matrix fibrils, as a measure of micro

Ordered-Disorder Transition Induced Stretchable Collagen Ionic Gels Toward Flexible Sensors.

Natural structural proteins such as collagen offer exceptional biocompatibility and mechanical versatility, making them promising candidates for wearable and implantable sensing applications. However, achieving integrated high stretchability, stable electrical performance, and long-term durability in protein-based sensing materials remains a critical challenge. Conventional modification strategies are largely confined to molecular-scale adjustments, which often fail to simultaneously address these functional demands. This study introduces a fundamentally different approach by leveraging Hofmei
