Selected hybrid natural products as tubulin modulators.

Modulators of microtubule dynamics have received increasing attention because of their potential to stop cancer growth. Although it belongs to the category of complex protein-protein interactions (PPIs), which are generally considered difficult to modulate through small molecules, the use of microtubule is considered a well-validated target. There are a number of bioactive natural products and related compounds that are currently in use as drugs or in clinical trials as next generation anti-cancer agents. The present review article is focused on two such bioactive natural products, epothilone

Diazepam induces mitotic defects and cytotoxicity through modulation of tubulin and Eg5.

Diazepam (DZP) is a widely prescribed drug for central nervous system disorders. However, it also exhibits a significant inhibitory effect on tubulin and the mitotic kinesin Eg5. These novel mitotic mechanisms explain its cytotoxicity and provide valuable insights for designing safer, more targeted antimitotic agents. Studies in mammalian cancer and noncancerous cell lines, including HeLa, MCF-7, A549, and L929, demonstrated that DZP inhibits cell proliferation in a concentration-dependent manner, with IC50 values ranging from 42 μM to 76 μM. Fluorescence spectroscopy confirmed direct binding

Tubulin deacetylase NDST3 modulates lysosomal acidification: Implications in neurological diseases.

Neurological diseases (NDs), featured by progressive dysfunctions of the nervous system, have become a growing burden for the aging populations. N-Deacetylase and N-sulfotransferase 3 (NDST3) is known to catalyze deacetylation and N-sulfation on disaccharide substrates. Recently, NDST3 is identified as a novel deacetylase for tubulin, and its newly recognized role in modulating microtubule acetylation and lysosomal acidification provides fresh insights into ND therapeutic approaches using NDST3 as a target. Microtubule acetylation and lysosomal acidification have been reported to be critical f

HDAC6 deacetylates alpha tubulin in sperm and modulates sperm motility in Holtzman rat.

Histone deacetylase 6 (HDAC6) is an alpha (α)-tubulin deacetylase and its over-expression has been demonstrated to promote chemotactic cell movement. Motility in sperm is driven by the flagella, the cytoskeletal structure comprising the microtubules, which are heterodimers of α- and β-tubulins. We have hypothesized that HDAC6, by virtue of being an α-tubulin deacetylase, might modulate sperm motility. However, the presence of HDAC6 on sperm has hitherto not been reported. In this study, we have demonstrated, for the first time, the presence of HDAC6 transcript and protein in the testicular and

5-fluorouracil derivatives induce differentiation mediated by tubulin and HLA class I modulation.

Neoplastic cells exhibit defects in their ability to differentiate; therefore, differentiation therapy represents a viable option to control cancer growth and progression. Rhabdomyosarcomas (RMS), a malignant tumor of skeletal muscle, is the most common soft tissue sarcoma in children and is characterized by its poor response to cytotoxic treatment and significant morbidity. Since modulation of alpha-tubulin and human leukocyte antigen (HLA) class I expression has been detected during malignant transformation, we analyzed in this study the expression pattern of both kinds of proteins after the

ClC-3 promotes paclitaxel resistance via modulating tubulins polymerization in ovarian cancer cells.

Epithelial ovarian cancers (EOC) present as malignant tumors with high mortality in the female reproductive system diseases. Acquired resistance to paclitaxel (PTX), one of the first-line treatment of EOC, remains a therapeutic challenge. ClC-3, a member of the voltage-gated Cl- channels, plays an essential role in a variety of cellular activities, including chemotherapeutic resistance. Here, we demonstrated that the protein expression and channel function of ClC-3 was upregulated in PTX resistance A2780/PTX cells compared with its parental A2780 cells. The silence of ClC-3 expression by siRNA

Tubulin Post-Translational Modifications in Cardiovascular Diseases: Emerging Mechanisms and Therapeutic Modulation by Traditional Chinese Medicine.

Cardiovascular diseases (CVDs) severely threaten human health, and microtubule dysfunction is one of the major contributing factors. Tubulin post-translational modifications (PTMs), such as detyrosination, acetylation, and polyglutamylation, regulate microtubule homeostasis, among which the upregulation of detyrosination is a key feature of cardiovascular diseases. Traditional Chinese Medicine (TCM) can alleviate cardiac pathologies by targeting these modifications. This review discusses the driving mechanisms of PTMs in relation to the pathological changes that occur in CVDs and summarizes th

Pan-cancer Myc modulator that targets Myc-α-tubulin interaction to drive selective mitotic catastrophe.

MYC overexpression is a well-established cancer vulnerability, yet direct therapeutic targeting of Myc remains a challenge. Here, we identify DL78 as a potent antimitotic agent with selective anticancer activity through its regulation of Myc. DL78 demonstrated broad efficacy by inhibiting growth across nine cancer types and significantly reducing tumor burden in an in vivo model of platinum-resistant high-grade serous ovarian cancer, with no overt toxicity. DL78 preferentially targets chromosomally unstable, MYC-overexpressing cancer cells, a hallmark of high-grade serous ovarian cancer. Mecha

Modulating microtubule stability via α-tubulin acetylation partially restores Golgi fragmentation in spinal muscular atrophy.

BACKGROUND/AIM: Spinal muscular atrophy (SMA) is a neurodegenerative disease caused by the loss of survival of motor neuron (SMN) protein. SMN deficiency leads to perturbations of the cytoskeleton, including microtubules, which are mainly involved in motility-related cellular processes. However, the molecular mechanisms of microtubule dysregulation in SMA remain elusive. Alpha (α)-tubulin is a structural component of microtubules, and its posttranslational modifications affect microtubule dynamics. Here, we aimed to investigate α-tubulin acetylation and related molecular mechanisms in SMA. MA

Chalcogen bond-assisted syn-locked scaffolds: DFT analysis and biological implications of novel tubulin inhibitors.

A series of new tubulin inhibitors containing chalcogen bonds have been discovered. Density functional theory (DFT) analysis of the O-C-C-S torsion profile shows a preference of 0.8 kcal/mol for the syn-conformer over the anti-conformer. Besides, the O-S natural bond orbital (NBO) analysis reveals that the OLP ∼ C-SBD∗ energy potential is 0.62 kcal/mol. Further pharmacochemical screening of several series of (4-arylthiophen-2-yl)(3,4,5-trimethoxyphenyl)methanones identified IPO-10 as a highly effective tubulin inhibitor with an IC50 of 23 nm for MCF-7.

Insight into the Tubulin-Targeted Anticancer Potential of Noscapine and its Structural Analogs.

Cancer is known as a notorious disease responsible for threatening millions of lives every year. Natural products which act by disrupting the microtubule assembly and dynamics have proven to be highly successful as anticancer agents but their high toxicity owing to lower selectivity has limited their usage. Recently, Noscapine (NOS), a known anti-tussive, has come out to be an effective anti-tubulin candidate with far lesser toxicity. Since its first report as an anti-mitotic agent in 1998, NOS has been extensively studied and modified by various groups of researchers to optimize its anti-tubu

Novel Beta-Tubulin-Immobilized Nanoparticles Affinity Material for Screening β-Tubulin Inhibitors from a Complex Mixture.

In order to efficiently screen and isolate β-tubulin inhibitors, β-tubulin was immobilized on core-shell PMMA/CS (poly(methyl methacrylate)/Chitosan) nanoparticles to produce a new type of immobilized affinity material named β-tubulin-immobilized nanoparticles (β-TIN). The selectivity and adsorption performance of β-TIN were characterized using various control drugs. The β-TIN, the paclitaxel molecularly imprinted ploymers (MIP), and the C18 adsorbing material were compared for selectivity and enrichment ratio. Microtubule-targeting antitumor compounds were screened and isolated from a typical

Verubulin and its derivatives: Progress and promise in tubulin-targeted cancer therapy.

Verubulin (MPC-6827) is a microtubule-destabilizing agent that binds to the colchicine site of tubulin and has been primarily investigated for the treatment of glioblastoma and other malignancies. Its ability to interfere with microtubule dynamics, induce mitotic arrest, and retain efficacy in multidrug-resistant tumour cells has positioned it as a valuable scaffold in the development of next-generation anticancer agents. Over the past decade, extensive efforts have been dedicated to optimizing verubulin and its derivatives through modifications that enhance potency and selectivity. Additional

An overview on anti-tubulin agents for the treatment of lymphoma patients.

Anti-tubulin agents constitute a large class of compounds with broad activity both in solid tumors and hematologic malignancies, due to the interference with microtubule dynamics. Since microtubules play crucial roles in the regulation of the mitotic spindles, the interference with their function usually leads to a block in cell division with arrest at the metaphase/anaphase junction of mitosis, followed to apoptosis. This explains the reason why tubulin-binding agents (TBAs) proved to be extremely active in patients with cancer. Several anti-tubulin agents are indicated in the treatment of pa

Computational Based Identified EAAT2 Transporter Activator Attenuates Amyloid-β Induced Excitotoxicity in Primary Neuronal-Astroglial Mixed Culture by Ameliorating Glutamate Clearance.

Astrocytes play a central role in maintaining synaptic homeostasis, regulating neurotransmitter clearance, and supporting neuronal function, all of which are integral to cognition. In Alzheimer's brain, astrocytic dysfunction contributes significantly to the rapid progression of the disease pathology. Aβ deposition induces oxidative alterations in the astrocytic glutamate transporter EAAT2, leading to impaired glutamate uptake and elevated extracellular glutamate levels, which drive excitotoxicity. This disruption adversely impacts synaptic plasticity, neuronal survival, and cognitive integrit

Exploring the potential of Ziziphus nummularia and luteolin-7-O-glucoside as tubulin inhibitors in cancer therapy and survival.

Cancer is responsible for approximately 10 million deaths worldwide, with 70% of the deaths occurring in low- and middle-income countries; as such safer and more effective anti-cancer drugs are required. Therefore, the potential benefits of Ziziphus nummularia and Ziziphus spina-christi as sources of anti-cancer agents were investigated. Z. nummularia and Z. spina-christi extracts were prepared using chloroform, ethanol, ethyl acetate, and water. The extracts' anti-cancer properties were determined using the MTT Cell Viability Assay in four cancer cell lines: breast (KAIMRC2 and MDA-MB-231), c
