RGS1 targeted by miR-191-3p inhibited the stemness properties of esophageal cancer cells by suppressing CXCR4/PI3K/AKT signaling.

BACKGROUND: Esophageal cancer is one of the most common malignant tumors in the world. It is urgent to prevent the development and progression of esophageal cancer. Cancer stem cells (CSCs) were reported to have the ability to initiate tumorigenesis, and reducing the stem cell-like characteristics of tumors is an important strategy to inhibit the occurrence and development of tumors. miRNAs are key regulators of the stemness of cancer. Here, we aimed to investigate the role and regulatory mechanism of miR-191-3p in the stemness properties of esophageal cancer cells. METHODS: Esophageal cancer

FOXA1 activates NOLC1 transcription through NOTCH pathway to promote cell stemness in lung adenocarcinoma.

Tumor cell stemness plays a pivotal role in generating functional heterogeneity within tumors and is implicated in essential processes such as drug resistance, metastasis, and cell proliferation. Therefore, creating novel tumor diagnostic techniques and therapeutic plans requires a knowledge of the possible processes that preserve the stem cell-like qualities of cancers. Bioinformatics analysis of NOLC1 expression in lung adenocarcinoma (LUAD) and prediction of its upstream transcription factors and their binding sites were completed. RT-qPCR detection of NOLC1 and FOXA1 expression, colony for

Interleukin-6 is a cofactor for the growth of myeloid cells from human bone marrow aspirates but does not affect the clonogenicity of myeloma cells in vitro.

Several groups have claimed that IL-6 is a growth factor for human myeloma cells in vitro. Bone marrow aspirates from 30 patients at different stages of treatment with VAMP/high dose melphalan, were examined for myeloma colony formation (MY-CFUc) using a clonogenic assay in vitro. Myeloma cells from 16/30 patients produced MY-CFUc in our assay system, which uses heavily irradiated HL60 cells as an underlay in soft agar. These heavily irradiated cells were shown to be essential for the inhibition of granulocyte-macrophage colonies (GM-CFUc). The addition of recombinant human IL-6 (10 ng/plate)

Up-Regulation of FBXO17 Promotes Ferroptosis and Suppresses the Stemness of Prostate Cancer Cells Through Wnt/β-Catenin Pathway.

As a critical component of E3 ubiquitin ligase complexes, FBXO17 exhibits either tumor-promoting or tumor-suppressing in a variety of tumors, but its biological function and molecular mechanism in prostate cancer (PCa) are still unclear. FBXO17 expression in PCa cell lines and normal prostate epithelial cells (RWPE-1) was assessed using Western blot. The malignant biological behavior of the cells was evaluated using EdU staining, Transwell assay, and flow cytometry. The proportion of CD44+CD133+ stem cell subsets was determined via flow cytometry, and the stemness of PCa cells was evaluated us

CircRNA CDR1as promotes hepatoblastoma proliferation and stemness by acting as a miR-7-5p sponge to upregulate KLF4 expression.

Hepatoblastoma (HB) is a malignant embryonal tumor of the liver that consists of heterogenous populations of stem/progenitor cells. Although circular RNAs (circRNAs) play an essential role in tumor development, the effects of circRNA on the proliferation of HB cells, especially cancer stem cells (CSCs), remain unclear. We found that the circRNA, CDR1as, was highly expressed in CSC-enriched populations of HB cell lines. Results from flow cytometric and sphere-forming assays revealed that CDR1as knockdown in HB cell lines decreased the proportion of stem cells. The Cell Counting Kit-8 (CCK-8) as

Assessing tumor heterogeneity and FLOT/FOLFOX-induced alterations in cancer stem cells and cancer immune checkpoint molecules using patient-derived tumor organoids.

BackgroundGastric cancer is a deadly disease with poor prognosis and unfavorable therapeutic outcomes. It is known that a major cause of treatment failure for gastric cancer is related to tumor heterogeneity. However, it is difficult to assess heterogeneity, as the composition of tumors is dynamic and cancer treatments reshape cell diversity and gene/protein expression profiles.ObjectiveWe utilized gastric cancer patient-derived tumor organoids to monitor tumor heterogeneity and the impacts of chemotherapy (FLOT and FOLFOX) on cancer stem cells (LGR5 and CD133) and cancer immune checkpoint mol

Comparative in vitro activity of 4'-deoxy-4'-iododoxorubicin and other anthracyclines in the human tumor clonogenic assay.

A new halogenated anthracycline analog 4'-deoxy-4'-iododoxorubicin (IODO) was compared with doxorubicin (DOX) and deoxydoxorubicin (DEOX) in the human tumor clonogenic assay (HTCA) using human tumor cell lines. For all cell lines tested, IODO had lower ID50 value and thus greater in vitro potency and cytotoxicity than DOX. DEOX had lower average ID50 values than either IODO or DOX in all cell lines except HEC1A, where DEOX was equal to IODO. Analysis of variance likewise confirmed significantly greater activity for IODO versus DOX in most cell lines tested. Previous in vivo studies demonstrate

Morusin reverses paclitaxel resistance by inhibiting the stem cell-like properties of non-small cell lung cancer in a β-catenin-dependent manner.

Drug resistance is the common factor inducing failure of chemotherapy for non-small cell lung cancer (NSCLC), whereas β-catenin-regulated stem cell-like characteristics exert an important effect on tumor cell resistance. Antitumor activity of morusin is well established; however, its impact on paclitaxel (PTX) resistance of NSCLC is largely unexplored. The present work successfully established PTX-resistant NSCLC cells, and applied morusin as an intervention. We used methylthiazolyldiphenyl-tetrazolium bromide assay for assessing drug resistance level. Apoptosis rate was measured through flow

Human uterine leiomyoma stem/progenitor cells expressing CD34 and CD49b initiate tumors in vivo.

CONTEXT: Uterine leiomyoma is the most common benign tumor in reproductive-age women. Using a dye-exclusion technique, we previously identified a side population of leiomyoma cells exhibiting stem cell characteristics. However, unless mixed with mature myometrial cells, these leiomyoma side population cells did not survive or grow well in vitro or in vivo. OBJECTIVE: The objective of this study was to identify cell surface markers to isolate leiomyoma stem/progenitor cells. DESIGN: Real-time PCR screening was used to identify cell surface markers preferentially expressed in leiomyoma side po

TET3 facilitates bladder cancer progression through targeted modulation of stemness pathways.

Bladder cancer is a major health concern, and understanding its molecular mechanisms is essential for developing effective therapies. TET3, a DNA hydroxymethylase, has been linked to tumor progression, but its role in bladder cancer remains unclear. We analyzed single-cell RNA sequencing data and identified TET3 as a key regulator in tumor-promoting cells using scissor analysis. Functional experiments were performed to evaluate the effects of TET3 knockdown on cell proliferation, migration, and gene expression. In vivo tumor growth assays and histological analyses were conducted to assess the

Solvent fractions of selected Ethiopian medicinal plants used in traditional breast cancer treatment inhibit cancer stem cells in a breast cancer cell line.

BACKGROUND: The incidence and mortality of breast cancer in women is increasing worldwide. Breast cancer contains a subpopulation of cells known as cancer stem cells (CSCs). The CSCs are believed to be responsible for chemotherapeutic resistance and are also involved in tumor initiation, progression, evolution, and metastasis to distant sites. The present study aimed to investigate the anti-CSC potential of selected Ethiopian medicinal plants traditionally used for breast cancer treatment. METHODS: The solvent fractions of three medicinal plants (the ethyl acetate fraction of Vernonia leopold

The impact of inhibiting the Hippo signaling pathway effector molecule YAP1 on in vitro glioblastoma and glioblastoma stem cells.

Glioblastoma, a prevalent and aggressive brain tumor in adults, recurs due to local invasiveness, radiation therapy failure, drug resistance, and cancer stem cell presence. The Hippo signaling pathway, regulating organ size and growth, features YAP as a key effector with oncogenic implications in various cancers, including glioma. This study aimed to explore YAP1 as a therapeutic target in glioblastoma by silencing it using siRNA and evaluating the potential of a new inhibitor, CA3 (CIL56), through advanced in vitro analyses. Investigations were conducted on brain cancer and stem cells, alongs

Targeting TYRO3 attenuates apoptotic cell-driven cancer stemness in triple negative breast cancer.

BACKGROUND: Apoptosis is a prevalent feature of the tumor microenvironment, driven not only by the rapid cellular turnover of high-grade cancers but also by chemotherapeutic agents. Although accumulating apoptotic cells can paradoxically support cancer cell survival and contribute to tumor recurrence, the roles of apoptotic cells in triple-negative breast cancer (TNBC) have not been elucidated. TYRO3, a receptor tyrosine kinase frequently overexpressed in cancers, plays a pivotal role in malignant progression. Here, we demonstrate that aggressive breast cancer cells exploit TYRO3 as a phosphat
