Multi-drug resistance in nematodes is a serious problem as lately several resistant phenotypes have emerged following the intermittent usage of synthetic nematicides. Contemporary research continues to focus on developing and/or repurposing small molecule inhibitors that are eco-friendly. Here, we describe the repurposing of the indole derivative, 5-iodoindole, as a nematicide for the root-knot nematode, Meloidogyne incognita. 5-Iodoindole effectively killed juveniles and freshly hatched juveniles by inducing multiple vacuole formation. Notably, at higher dosage (50 μg/mL), 5-iodoindole induce

Meloidogyne incognita is a destructive and economically important agricultural pest. Similar to other plant-parasitic nematodes, management of M. incognita relies heavily on chemical controls. As old, broad spectrum, and toxic nematicides leave the market, replacements have entered including fluensulfone, fluazaindolizine, and fluopyram that are plant-parasitic nematode specific in target and less toxic to applicators. However, there is limited research into their modes-of-action and other off-target cellular effects caused by these nematicides in plant-parasitic nematodes. This study aimed to

Sugarcane (Saccharum officinarum L.) is an important economic crop in tropical and subtropical countries and regions. During an investigation of crop nematodes in Guangdong, a new disease caused by a new species, Heterodera parasacchari n. sp., of the subfamily Heteroderinae was found on sugarcane. In this study, the new species is described, and its pathogenicity to sugarcane is confirmed. The new species is characterized by having lemon-shaped cysts with a prominent neck and vulval cone and being ambifenestrate, with a strongly developed underbridge with finger-like projections, developed bu

There is limited research about the impacts of new nematicides, including fluazaindolizine, fluopyram, and fluensulfone, on the plant-parasitic nematode Meloidogyne incognita, despite it being a pervasive agricultural pest. In this study, M. incognita second-stage juveniles were exposed for 24-h to fluensulfone, fluazaindolizine, fluopyram, and oxamyl and total RNA was extracted and sequenced using next-generation sequencing to determine gene expression. The effects of nematicide exposure on cellular detoxification pathways, common differentially expressed (DE) genes, and fatty acid and retino

The root-knot nematodes of the genus Meloidogyne are important and damaging parasites capable of infecting most flowering plants. Within this genus, several species of the Meloidogyne incognita group show evidence of paleopolyploidy in their genomes. We used our software tool POInT, the Polyploidy Orthology Inference Tool, to phylogenetically model the gene losses that followed that polyploidy. These models, and simulations based on them, show that three of these species (M. incognita, M. arenaria and M. javanica) descend from a single common hybridization event that yielded triplicated genome

In our early experiments, the chloroform extract of D. crassirhizoma was demonstrated to contain the highest concentrations of total phloroglucinols among several extract fractions and possessed the most effective nematicidal activity. This study aimed to ascertain the ultrastructural changes in M. incognita after treatment with a D. crassirhizoma chloroform extract at 1 mg·mL⁻¹ for 24 h. It was found that the extract exhibited significant destructive effects on the worm's ultrastructure and caused distinctive damage to body surfaces and internal structures. These results will contribute to a

A number of different plant parasitic nematode species are found associated with sugarcane in South Africa. Of these, the root-knot nematode (Meloidogyne javanica), the lesion nematode (Pratylenchus zeae) and the dagger nematode (Xiphinema elongatum) are potentially the most damaging pests. Identification and enumeration of the number of these nematodes are necessary for providing advice to farmers as well as studying the effects of various treatments in field and glasshouse trials. We report on the development, use, and extent of specificity of three sets of primers, for M. javanica, P. zeae

Root-knot nematodes are among the most destructive plant parasites on Earth. Many aspects of their biology are fascinating, including their diverse reproductive modes, their extreme polyphagy, their unknown origin, and their sophisticated plant manipulation systems. Although genome analyses have illuminated certain points of their biology, recent advances in sequencing technologies have revealed some chromosomal idiosyncrasies. This includes unique telomere systems unparalleled in any other organism and complex holocentromere organisations. Evidence of polyploid hybrid genome structures in som

Infections caused by root-knot nematodes (RKNs) significantly impair vegetable growth and crop yield, posing a severe threat to global food security. Our previous study indicated that fungal-derived 2-furoic acid was a promising lead compound for the exploitation of eco-friendly nematicides. However, the exact molecular mechanism remains poorly understood. In this study, we observed behavioral changes in the nematodes following treatment with 100 μg/mL of 2-furoic acid. Subsequently, transcriptomics and metabolomics were combined to identify changes in differentially expressed genes (DEGs) and

BACKGROUND: Root-knot nematodes (Meloidogyne incognita) are one of the main pathogens affecting agricultural production. Conventional fumigants like methyl bromide have been phased out because of environmental risks, while alternatives face challenges of soil toxicity and resistance evolution. This study aims to develop a novel microencapsulated dual-agent system combining allyl isothiocyanate (AITC) and avermectin B2 (AVB2) for sustainable nematode control.
RESULT: Bioassay results showed that AITC and AVB2 had high toxicity against M. incognita with median lethal concentration (LC50) values

Potato cyst nematodes (PCNs), Globodera rostochiensis, are an important group of quarantine plant-parasitic nematodes that pose a serious threat to the potato industry and global food security due to the lack of effective control measures. In this study, intercropping with Houttuynia cordata is shown to effectively reduce PCN populations while increasing potato yield. This effect is associated with the release of a key allelopathic volatile compound, 1-decanol, which exhibits strong nematicidal activity (LC50 = 31.5 μg/mL) markedly exceeding that of the commercial nematicide avermectin (LC50 =

Plant parasitic nematodes (PPNs) are highly destructive and difficult to control, while conventional chemical nematicides are highly toxic and cause serious environmental pollution. Additionally, resistance to existing pesticides is becoming increasingly common. Biological control is the most promising method for the controlling of PPNs. Therefore, the screening of nematicidal microbial resources and the identification of natural products are of great significance and urgency for the environmentally friendly control of PPNs. In this study, the DT10 strain was isolated from wild moss samples an

An unknown root-knot nematode was found at high density on grape roots collected from Yunnan Province. Morphometric traits and measurements, isozyme phenotypes, and molecular analysis clearly differentiated this nematode from previously described root-knot nematodes. This new species is described, illustrated and named Meloidogyne vitis sp. nov. The new species can be distinguished from other Meloidogyne spp. by a unique combination of characters. Females display a prominent neck, an excretory pore is located on the ventral region between 23rd and 25th annule behind lips, an EP/ST ratio of app

Root-knot nematodes (RKNs; Meloidogyne spp.) are among the most widespread and damaging plant-parasitic nematodes known. Although M. incognita is the most prevalent RKN species, other species, including M. enterolobii, M. floridensis, and M. haplanaria, have recently been detected in multiple Georgia, United States, vegetable fields. Producers use chemical nematicides to manage M. incognita populations because most vegetable crops do not have effective RKN host resistance. We evaluated the effects of sublethal doses of nonfumigant nematicides-fluazaindolizine, fluensulfone, fluopyram, and oxam

Ilicic acid, nerolidol, and 9-hydroxynerolidol are major components of the aerial parts of Dittrichia viscosa. These components were selectively isolated in multigram quantities and used as lead compounds to generate diversity in the search for new natural-product-derived pesticides. A total of 29 derivatives of these three molecules-some of which are known natural products-were generated by subjecting these natural products to different transformations. In order to explore potential applications in sustainable biocontrol, some of the compounds generated were evaluated for plant protection pot

Root-knot nematodes (RKN) of the genus Meloidogyne are obligatory plant endoparasites that cause substantial economic losses to agricultural production and impact the global food supply. These plant parasitic nematodes belong to the most widespread and devastating genus worldwide, yet few measures of control are available. The most efficient way to control RKN is deployment of resistance genes in plants. However, current resistance genes that control other Meloidogyne species are mostly inefficient on Meloidogyne enterolobii. Consequently, M. enterolobii was listed as a European Union quaranti

Root knot nematodes are the most devastating root pathogens, causing severe damage and serious economic losses to agriculture worldwide. Octanoic acid has been reported as one of the nematicides, and its mode of action is not fully understood. The main objective of this study was to elucidate the effect of octanoic acid on Meloidogyne incognita by transcriptomic analysis combined with physiological and biochemical assays. In the toxicity assays with octanoic acid, the threshold concentration with nematicidal activity and the maximum concentration to which nematodes could respond were 0.03 μL/m

Arbuscular mycorrhizal fungi (AMF) are generally recognized to induce plant growth and prime plants against soil-borne parasites, such as plant parasitic nematodes. However, the effectiveness of commercial formulates containing AMF has been questioned. Increasing amounts per plant of one commercial AMF-containing formulate, reported in the text as Myco, were used to detect the effects on growth of tomato plants and the resistance induced against root-knot nematodes (RKNs) The doses used per plant (0.5, 1.0, 2.0 g, reported as Myco1, Myco2, Myco3, respectively) were soil-drenched to growing pot
