We used computational tools to study the tymoviral proteome. Past gene sequence comparisons had shown that tymovirus genomes encode five proteins already identified in other viruses; however, our AlphaFold3 modelling found, in addition to their five α-helical and β-sheet structures, five regions of 'intrinsically disordered protein' (IDP). Notably, one of these IDPs is the movement protein (MP), distinguishing it from the structured MPs of other plant viruses. The MP is the largest IDP having some features of structured proteins including genus-specific conserved motifs. All tymovirid methyltr

Japanese Encephalitis (JE) is a mosquito borne arboviral infection caused by Japanese Encephalitis Virus (JEV). It is a major cause of viral encephalitis in Asian countries including India. In the present study, we have used a Tymovirus [i.e. Physalis Mottle Virus (PhMV) coat protein (CP)], which forms virus like particles (VLPs) as a template to display immunodominant epitopes of JEV envelope (E) protein. The immunodominant epitopes of JEV were inserted at the N-terminus of the wild type PhMV CP, and these constructs were cloned and expressed in Escherichia coli. The chimeric proteins were pu

The argonaute (AGO) family proteins play a crucial role in preventing viral invasions through the plant antiviral RNA silencing pathway, with distinct AGO proteins recruited for specific antiviral mechanisms. Our previous study revealed that Nicotiana benthamiana AGO5 (NbAGO5) expression was significantly upregulated in response to bamboo mosaic virus (BaMV) infection. However, the roles of NbAGO5 in antiviral mechanisms remained to be explored. In this research, we examined the antiviral functions of NbAGO5 in the infections of different viruses. It was found that the accumulation of NbAGO5 w

The management of emerging plant viruses presents significant challenges for global agriculture, requiring innovative approaches beyond conventional control strategies. Traditional methods rely on cultural practices, vector management, and breeding for genetic resistance, and these approaches are often time-consuming and may have limited effectiveness against emerging viral strains. Spray-Induced Gene Silencing (SIGS), involving topical application of virus-derived double-stranded RNA (dsRNA) to trigger plant defense mechanisms, offers a promising alternative strategy. However, the application

Recessive resistance, achieved through mutations in host susceptibility genes, offers an effective way for controlling plant viruses. One well-studied gene family involved in such resistance is the eukaryotic translation initiation factor 4E (eIF4E) gene family, which includes eIF4E, eIFiso4E and the atypical novel cap-binding protein (nCBP). Although gene pyramiding of the eIF4E family may provide a promising strategy for broadening virus resistance, it has so far been applied only to a limited set of gene combinations and target viruses. To deepen our understanding of the practicality of eIF

Focusing on the utility of ticks as xenosurveillance sentinels to expose circulating pathogens in Kenyan drylands, host-feeding ticks collected from wild ungulates [buffaloes, elephants, giraffes, hartebeest, impala, rhinoceros (black and white), zebras (Grévy's and plains)], carnivores (leopards, lions, spotted hyenas, wild dogs), as well as regular domestic and Boran cattle were screened for pathogens using metagenomics. A total of 75 host-feeding ticks [Rhipicephalus (97.3%) and Amblyomma (2.7%)] collected from 15 vertebrate taxa were sequenced in 46 pools. Fifty-six pathogenic bacterial sp

Nairoviruses are responsible for numerous diseases that affect both humans and animal. Recent work has implicated the viral ovarian tumor domain (vOTU) as a possible nairovirus virulence factor due to its ability to edit ubiquitin (Ub) bound to cellular proteins and, at least in the case of Crimean-Congo hemorrhagic fever virus (CCHFV), to cleave the Ub-like protein interferon-stimulated gene 15 (ISG15), a protein involved in the regulation of host immunity. The prospective roles of vOTUs in immune evasion have generated several questions concerning whether vOTUs act through a preserved specif

Viruses and virus-like particles (VLPs) have been demonstrated to be robust scaffolds for the construction of nanomaterials. In order to develop new nanoprobes for time-resolved fluoroimmuno assays as well as to investigate the two-dimensional self-assembly of viruses and VLPs, the icosahedral turnip yellow mosaic virus (TYMV) was investigated as a potential building block in our study. TYMV is an icosahedral plant virus with an average diameter of 28 nm that can be isolated inexpensively in gram quantities from turnips or Chinese cabbage. There are 180 coat protein subunits per TYMV capsid. T

Developing new adjuvants and vaccination strategies is of paramount importance to successfully fight against many life-threatening infectious diseases and cancer. Very few adjuvants are currently authorized for human use, and these mainly stimulate a humoral response. However, specific Abs are not sufficient to confer protection against persisting infections or cancer. Therefore, development of adjuvants and immunomodulators able to enhance cell-mediated immune responses represents a major medical need. We recently showed that papaya mosaic virus nanoparticles (PapMV), self-assembled from the
