Bacterial type VI secretion systems (T6SSs) are molecular devices that traverse the bacterial envelope and mediate the translocation of protein effectors directly into prokaryotic or eukaryotic cells, thereby interfering with diverse cellular processes in target organisms. T6SSs involved in interbacterial competition have been widely described and functionally characterized, together with a remarkable arsenal of cognate toxins. In contrast, comparatively less is known about anti-eukaryotic T6SSs. This review summarizes recent advances in the study of T6SSs that target eukaryotic cells, highlig

In Salmonella, the type III and type VI secretion systems (T3SS and T6SS) play critical roles in pathogenicity. The bacterium harbors two distinct T3SSs: T3SS-1, encoded by SPI-1, promotes host cell invasion, whereas T3SS-2 (located within SPI-2) facilitates systemic infection. Both systems translocate effector proteins that disrupt normal host cell functions. Meanwhile, the T6SS enhances competitive fitness in the gut by targeting rival bacteria and supports evasion of the host immune response. Recent evidence indicates that these secretion systems share common regulatory components, includin

Edwardsiella ictaluri is a Gram-negative facultative intracellular fish pathogen causing enteric septicemia of catfish (ESC). While various secretion systems contribute to E. ictaluri virulence, the Type VI secretion system (T6SS) remains poorly understood. In this study, we constructed 13 E. ictaluri T6SS mutants using splicing by overlap extension PCR and characterized them, assessing their uptake and survival in channel catfish (Ictalurus punctatus) peritoneal macrophages, attachment and invasion in channel catfish ovary (CCO) cells, in vitro stress resistance, and virulence and efficacy in

Bacteria that live in the environment have evolved pathways specialized to defend against eukaryotic organisms or other bacteria. In this manuscript, we systematically examined the role of the five type VI secretion systems (T6SSs) of Burkholderia thailandensis (B. thai) in eukaryotic and bacterial cell interactions. Consistent with phylogenetic analyses comparing the distribution of the B. thai T6SSs with well-characterized bacterial and eukaryotic cell-targeting T6SSs, we found that T6SS-5 plays a critical role in the virulence of the organism in a murine melioidosis model, while a strain la

The Enterobacter genus contains 23 species that include common nosocomial pathogens capable of causing a wide variety of infections. We obtained all available Enterobacter genomes and retained 4,805 high-quality genomes after quality control. Genome sequencing analysis of Enterobacter species revealed the presence of type VI secretion systems (T6SS) in these bacteria, but systematic analysis and comparison of these systems among different species are limited. We found that these bacteria code for three distinct types of T6SS, each with a unique set of diverse predicted effectors. Whereas at le

Microbial species often exist in complex communities where they must avoid predation and compete for favorable niches. The type VI secretion system (T6SS) is a contact-dependent bacterial weapon that allows for direct killing of competitors through the translocation of proteinaceous toxins. Vibrio cholerae is a Gram-negative pathogen that can use its T6SS during antagonistic interactions with neighboring prokaryotic and eukaryotic competitors. The T6SS not only promotes V. cholerae's survival during its aquatic and host life cycles, but also influences its evolution by facilitating horizontal

Klebsiella pneumoniae is a nosocomial opportunistic pathogen that can cause pneumonia, liver abscesses, and infections of the bloodstream. The resistance and pathogenicity of K. pneumoniae pose major challenges to clinical practice. However, the ecology and pathogenic mechanisms of K. pneumoniae have not been fully elucidated. Among these mechanisms, the secretion systems encoded by strains of the bacteria confer adaptive advantages depending on the niche occupied. The type VI secretion system (T6SS) is a multi-protein complex that delivers effector proteins to the extracellular environment or

Infections from Campylobacter jejuni pose a serious public health problem and are now considered the leading cause of foodborne bacterial gastroenteritis throughout the world. Sequencing of C. jejuni genomes has previously allowed a number of loci to be identified, which encode virulence factors that aid survival and pathogenicity. Recently, a Type VI secretion system (T6SS) consisting of 13 conserved genes was described in C. jejuni strains and recognised to promote pathogenicity and adaptation to the environment. In this study, we determined the presence of this T6SS in 63 Spanish C. jejuni

The acute diarrheal disease cholera is caused by the marine bacterium Vibrio cholerae. A type VI secretion system (T6SS), which is structurally similar to the bacteriophage cell-puncturing device, has been recently identified in V. cholerae and is used by this organism to confer virulence toward phagocytic eukaryotes, such as J774 murine macrophages and Dictyostelium discoideum. We tested the interbacterial virulence of V. cholerae strain V52, an O37 serogroup with a constitutively active T6SS. V52 was found to be highly virulent toward multiple Gram-negative bacteria, including Escherichia co

The type VI secretion system (T6SS) is a proteinaceous weapon used by many Gram-negative bacteria to deliver toxins into adjacent target cells. Vibrio cholerae, the bacterium responsible for the fatal water-borne cholera disease, uses the T6SS to evade phagocytic eukaryotes, cause intestinal inflammation, and compete against other bacteria with toxins that disrupt lipid membranes, cell walls and actin cytoskeletons. The control of T6SS genes varies among V. cholerae strains and typically includes inputs from external signals and cues, such as quorum sensing and chitin availability. In the foll

Type VI secretion systems (T6SS) are a class of macromolecular secretion machines that are utilized by a number of bacteria for inter-bacterial competition or to elicit responses in eukaryotic cells. Acinetobacter baumannii is an opportunistic pathogen that causes severe infections in humans. These infections, including pneumonia and bacteremia, are important, as they are often associated with hospitals and medical-settings where they disproportionally affect critically ill patients like those residing in intensive care units. While it is known that A. baumannii genomes carry genes whose predi

The type VI secretion system (T6SS) is a versatile nanomachine that injects effectors into target cells, playing a role in bacterial competition and virulence. While widespread in Gram-negative bacteria, T6SS prevalence varies across species and strains, and its distribution in Escherichia coli remains underexplored despite it being an important intestinal (IPEC) and extraintestinal (ExPEC) pathogen. Here, we examined the prevalence of T6SS subclasses (T6SSi) across 131,610 E. coli genomes, which we annotated for clinical relevance. T6SS genes were identified while focusing on the three subcla

Extraintestinal pathogenic Escherichia coli (ExPEC) is a distinctive group of Escherichia coli strains capable of causing infections outside the gastrointestinal tract of humans and certain animals. ExPEC includes the pathotypes avian pathogenic E. coli (APEC), neonatal meningitis E. coli (NMEC), sepsis-associated E. coli (SEPEC), and uropathogenic E. coli (UPEC). These pathotypes share multiple virulence factors, including the Type VI Secretion System (T6SS), a nanomachinery that has been described as important for E. coli pathogenesis. In this study, we applied genome-wide association analys

Vibrio parahaemolyticus is a significant marine pathogen causing gastroenteritis and wound infections in humans. Its pathogenicity is mediated by key virulence factors, including the type VI secretion system (T6SS). This review comprehensively synthesizes current knowledge on the functional divergence and regulatory networks of T6SSs in V. parahaemolyticus, with emphasis on T6SS1 and T6SS2. T6SS1, enriched in clinical isolates, is activated under high-salt and warm conditions and primarily facilitates antibacterial competition and adhesion to human epithelial cells in strains such as RIMD22106

The type VI secretion system (t6ss) is a recently characterized secretion system which appears to be involved in bacterial pathogenesis as a potential nano-syringe for the translocation of effector proteins into the eukaryotic host cell cytoplasm. Until now no evidence was provided for the presence of t6ss in the genomes of the sequenced representatives of Klebsiella spp., including the human opportunistic pathogen Klebsiella pneumoniae. However, in a previous study by Lawlor et al. (2005), were revealed two insertion mutants in hypothetical proteins of K. pneumoniae with decreased ability to

We characterized the fluorescence intensity distribution within the epithelia and stroma of frozen human cervical tissues at the following excitation-emission wavelength pairs: 440, 525 nm and 365, 460 nm. The intensities at both excitation-emission wavelength pairs are significantly lower in the epithelia of severely dysplastic tissues, relative to that in normal and inflammatory tissues. Furthermore, there are small differences in (1) the epithelial intensity of severe dysplasia and mild dysplasia at 440, 525 nm and (2) the stromal intensity of inflammatory and severely dysplastic tissues at

Bacteria inhabiting the mammalian gut coexist in dense communities where contact-dependent antagonism mechanisms are widespread. The type VI secretion system (T6SS) is an interbacterial toxin delivery pathway prevalent among gut Bacteroidales, yet its function in naturally evolved microbiomes remains poorly defined. Here, we examine the role of the T6SS in Bacteroides within a physiologically relevant gut community derived from wild mice (the WildR microbiome). Using newly developed genetic tools and a strategy for functional replacement of strains within the WildR community, we demonstrate th

Pseudomonas aeruginosa is an opportunistic pathogen and a major driver of morbidity and mortality in people with Cystic Fibrosis (CF). The Type VI secretion system (T6SS) is a molecular nanomachine that translocates effectors across the bacterial membrane into target cells or the extracellular environment enabling intermicrobial interaction. P. aeruginosa encodes three T6SS clusters, the H1-, H2- and H3-T6SS, and numerous orphan islands. Genetic diversity of T6SS-associated effectors in P. aeruginosa has been noted in reference strains but has yet to be explored in clinical isolates. Here, we

The health of mammals depends on a complex interplay with their microbial ecosystems. Compartments exposed to external environments such as the mucosal surfaces of the gastrointestinal tract accommodate the gut microbiota, composed by a wide range of bacteria. The gut microbiome confers benefits to the host, including expansion of metabolic potential and the development of an immune system that can robustly protect from external and internal insults. The cooperation between gut microbiome and host is enabled in part by the formation of partitioned niches that harbor diverse bacterial phyla. Ba

The Burkholderia pseudomallei K96243 genome encodes six type VI secretion systems (T6SSs), but little is known about the role of these systems in the biology of B. pseudomallei. In this study, we purified recombinant Hcp proteins from each T6SS and tested them as vaccine candidates in the BALB/c mouse model of melioidosis. Recombinant Hcp2 protected 80% of mice against a lethal challenge with K96243, while recombinant Hcp1, Hcp3, and Hcp6 protected 50% of mice against challenge. Hcp6 was the only Hcp constitutively produced by B. pseudomallei in vitro; however, it was not exported to the extra
