Membranous nephropathy in the UK Biobank.

BACKGROUND: Despite MN being one of the most common causes of nephrotic syndrome worldwide, its biological and environmental determinants are poorly understood in large-part due to it being a rare disease. Making use of the UK Biobank, a unique resource holding a clinical dataset and stored DNA, serum and urine for ~500,000 participants, this study aims to address this gap in understanding. METHODS: The primary outcome was putative MN as defined by ICD-10 codes occurring in the UK Biobank. Univariate relative risk regression modelling was used to assess the associations between the incidence

Epidemiology of Cholesteatoma in the UK Biobank.

OBJECTIVES: To identify factors associated with cholesteatoma in a large UK cohort. Although some risk factors are frequently reported (male sex, history of chronic otitis media), other associations require further evidence (deprivation, smoking). DESIGN AND SETTING: Briefly, 1140 cholesteatoma cases from UK BioBank were compared to 4551 non-cholesteatoma middle ear disease and 493 832 ear disease-free controls. Adjusted odds ratios were calculated for demographic factors including age, sex, ethnicity, deprivation and smoking status with logistic regressions. Odds ratios for overlapping ICD-1

Demographics of lipoprotein(a) and stroke: The UK biobank.

INTRODUCTION: Lipoprotein(a) [Lp(a)] is associated with ischemic stroke, but the strength of association based on demographic differences remains unclear. We aimed to investigate the association between Lp(a)>125 nmol/L and stroke by age, sex, and racial/ethnic subgroups. METHODS: Using data from the UK Biobank, we included 353,309 participants with an Lp(a) measurement without a history of atherosclerotic cardiovascular disease (ASCVD). Stroke was defined as ischemic stroke or hemorrhagic stroke (with subtypes subarachnoid hemorrhage and intracerebral hemorrhage). Cox proportional hazards mo

Genetic stratification of depression in UK Biobank.

Depression is a common and clinically heterogeneous mental health disorder that is frequently comorbid with other diseases and conditions. Stratification of depression may align sub-diagnoses more closely with their underling aetiology and provide more tractable targets for research and effective treatment. In the current study, we investigated whether genetic data could be used to identify subgroups within people with depression using the UK Biobank. Examination of cross-locus correlations were used to test for evidence of subgroups using genetic data from seven other complex traits and disor

BACKGROUND: Antidepressants are the most frequently prescribed medications in psychiatry. Medical records of thousands of individuals provide a valuable opportunity to explore prescribing patterns and identify factors that influence treatment outcomes. AIMS: To investigate antidepressant change patterns in depression and non-depression indications and assess clinical and genetic factors associated with outcomes of antidepressant treatment. METHODS: Using primary care records from the UK Biobank, we examined outcomes including number of antidepressant changes and discontinuation due to either

Optimising fibromyalgia criteria: evidence from the UK Biobank.

OBJECTIVE: To evaluate the performance of the 2016 revisions to the modified fibromyalgia criteria (FM2016) in identifying individuals with self-reported fibromyalgia in a large population sample, and to test whether simplified symptom thresholds and alternative definitions of widespread pain improve case-finding utility. METHODS: We used data from 167,184 UK Biobank participants who completed an "Experience of Pain" online questionnaire. Logistic regression and clinical utility indices (CUI+ and CUI-) were used to assess the performance of the FM2016 criteria and alternative modifications in

Genetic and Phenotypic Features of Schizophrenia in the UK Biobank.

IMPORTANCE: Large-scale biobanks provide important opportunities for mental health research, but selection biases raise questions regarding the comparability of individuals with those in clinical research settings. OBJECTIVE: To compare the genetic liability to psychiatric disorders in individuals with schizophrenia in the UK Biobank with individuals in the Psychiatric Genomics Consortium (PGC) and to compare genetic liability and phenotypic features with participants recruited from clinical settings. DESIGN, SETTING, AND PARTICIPANTS: This cross-sectional study included participants from th

Meaning in life and Parkinson's disease in the UK Biobank.

INTRODUCTION: Meaning in life is an aspect of eudaimonic well-being associated with lower dementia risk. This research examines whether this protective association extends to Parkinson's disease (PD). METHODS: Participants (N = 153,569) from the UK Biobank reported on their meaning in life. Cases of PD were identified through health records. RESULTS: Meaning in life was associated with a 50 % lower likelihood of prevalent PD (OR = 0.68, 95 % CI = 0.59-0.78). Over the 5-year follow-up, meaning was associated with a 35 % lower risk of incident PD (HR = 0.74, 95 % CI = 0.65-0.83), an associatio

Marijuana and Myocardial Infarction in the UK Biobank Cohort.

BACKGROUND: Atrial fibrillation, ventricular tachycardia, acute coronary syndromes, and cardiac arrest have been attributed to marijuana. But the National Academy of Science's 2017 Report, The Health Effects of Cannabis and Cannabinoids, found limited evidence that acute marijuana smoking is positively associated with an increased risk of acute myocardial infarction, and uncovered no evidence to support or refute associations between any chronic effects of marijuana use and increased risk of myocardial infarct (MI). AIMS: We sought to determine the association of marijuana smoking with MI in

Genetic predisposition to tinnitus in the UK Biobank population.

Tinnitus, the phantom perception of noise originating from the inner ear, has been reported by 15% of the world's population, with many patients reporting major deficits to cognition and mood. However, both objective diagnostic tools and targeted therapeutic strategies have yet to be established. To better understand the underlying genes that may preclude tinnitus, we performed a genome-wide association study of the UK Biobank's 49,960 whole exome sequencing participants to identify any loci strongly associated with tinnitus. We identified 17 suggestive single nucleotide polymorphisms (p < 1e-

Education is power: preserving cognition in the UK biobank.

INTRODUCTION: Dementia is a debilitating syndrome characterized by the gradual loss of memory and cognitive function. Although there are currently limited, largely symptomatic treatments for the diseases that can lead to dementia, its onset may be prevented by identifying and modifying relevant life style risk factors. Commonly described modifiable risk factors include diet, physical inactivity, and educational attainment. Importantly, however, to maximize the utility of our understanding of these risk factors, tangible and meaningful changes to policy must also be addressed. OBJECTIVES: Here

Cannabis smoking and glaucoma in the UK Biobank cohort.

BACKGROUND: Cigarette smoking is a well-known risk factor for primary open-angle glaucoma (POAG). Tobacco smoke contains a variety of chemicals, including nicotine, free radicals, and carbon monoxide, all of which can play a role in the development of POAG. Cannabis smoke, like tobacco smoke, contains a comparable variety of carcinogenic and toxic compounds. In the present study, we analyzed UK Biobank data to determine whether smoking cannabis, like cigarettes, might be related to POAG. METHODS: Our analysis included all subjects with glaucoma and cannabis smoking information. Data processin

Methodological considerations in the UK Biobank diabetes‑tinnitus study.

Tinnitus is a common and often debilitating condition that substantially affects quality of life. While diabetes has been suggested as a potential risk factor, the relationship remains poorly understood, particularly in large population-based settings where classification of diabetes type often relies on algorithms rather than confirmatory biomarkers. The recent UK Biobank study by Ji and colleagues addresses this gap by reporting that type 2 diabetes is associated with higher tinnitus prevalence. While the study benefits from a large sample and systematic examination of diabetes-related facto

Retinal vasculometry associations with cognition status in UK Biobank.

INTRODUCTION: Retinal vasculometry (RV) provides a neurovascular biomarker which may relate to cognitive status. However, the presence and form of association remains unclear and unexamined at scale. METHODS: Artificial intelligence-enabled RV measures from 66,350 UK Biobank study participants were related to combined cognition scores. Differences in RV were examined per standard deviation (SD) increase in cognitive score, using multilevel linear regression, adjusted for age, sex, measurement center, ethnicity, and within-person RV clustering. RESULTS: One hundred ten thousand two hundred ei

Telomere length and brain imaging phenotypes in UK Biobank.

Telomeres form protective caps at the ends of chromosomes, and their attrition is a marker of biological aging. Short telomeres are associated with an increased risk of neurological and psychiatric disorders including dementia. The mechanism underlying this risk is unclear, and may involve brain structure and function. However, the relationship between telomere length and neuroimaging markers is poorly characterized. Here we show that leucocyte telomere length (LTL) is associated with multi-modal MRI phenotypes in 31,661 UK Biobank participants. Longer LTL is associated with: i) larger global

Prediagnostic Progressive Supranuclear Palsy - Insights from the UK Biobank.

INTRODUCTION: Prediagnostic features of Parkinson's Disease are well described but prediagnostic Progressive Supranuclear Palsy (PSP) is less understood. The diagnosis of PSP is delayed by an average of three years after symptom onset. Understanding the changes that occur in the prediagnostic period will aid earlier diagnosis, clarify the natural history, and may aid the design of early disease-modifying therapy trials. We set out to identify motor and cognitive markers of prediagnostic PSP, with Parkinson's disease as a comparator condition, in a large prospective cohort. METHODS: Baseline U
