Inverse expression of ubiquitin-specific peptidase 19 and caspase 7 correlates with gastric neoplastic transformation.

BACKGROUND: Ubiquitin-specific protease 19 (USP19) is a deubiquitinylase (DUB) that is part of the USP family, the largest group of DUBs in humans. Growing evidence has indicated that USP19 is involved in tumor progression and serves as a new prognostic marker for various malignant disorders. Interestingly, USP19 has been shown to have both promoting and inhibiting effects on the onset and development of different neoplasms, depending on the specific tissue type. DUBs including USP19 affect a variety of cell functions including apoptotic cell death. Herein, caspase 7 acts as a key executioner

Structural insights into the interaction between testis-specific Y-encoded-like protein 5 and ubiquitin-specific protease 7.

The Alternative Lengthening of Telomeres (ALT) mechanism enables telomere maintenance, contributing to the immortality of certain cancer cells. Disrupting the interaction between testis-specific Y-encoded-like protein 5 (TSPYL5) and ubiquitin-specific protease 7 (USP7) has emerged as a promising strategy to target ALT-dependent cancers. While the N-terminal MATH domain of USP7 mediates the protein interaction, the regions of TSPYL5 involved in binding remain unclear. Here, we present a structural analysis of the TSPYL5-USP7 interaction to guide targeted therapeutic strategies. We showed that T

RNF4 and USP7 cooperate in ubiquitin-regulated steps of DNA replication.

DNA replication requires precise regulation achieved through post-translational modifications, including ubiquitination and SUMOylation. These modifications are linked by the SUMO-targeted E3 ubiquitin ligases (STUbLs). Ring finger protein 4 (RNF4), one of only two mammalian STUbLs, participates in double-strand break repair and resolving DNA-protein cross-links. However, its role in DNA replication has been poorly understood. Using CRISPR/Cas9 genetic screens, we discovered an unexpected dependency of RNF4 mutants on ubiquitin specific peptidase 7 (USP7) for survival in TP53-null retinal pigm

Ubiquitin-Specific Proteases (USPs) and Metabolic Disorders.

Ubiquitination and deubiquitination are reversible processes that modify the characteristics of target proteins, including stability, intracellular localization, and enzymatic activity. Ubiquitin-specific proteases (USPs) constitute the largest deubiquitinating enzyme family. To date, accumulating evidence indicates that several USPs positively and negatively affect metabolic diseases. USP22 in pancreatic β-cells, USP2 in adipose tissue macrophages, USP9X, 20, and 33 in myocytes, USP4, 7, 10, and 18 in hepatocytes, and USP2 in hypothalamus improve hyperglycemia, whereas USP19 in adipocytes, US

Targeting USP7 in sarcomas: opportunities and challenges.

Sarcomas are a heterogeneous group of aggressive mesenchymal tumors with limited therapeutic options in the advanced setting. Novel actionable targets and biology-driven therapies are needed to improve patients' outcome. The identification of common pathways engaged by different disease drivers may suggest new therapeutic strategies exploitable across sarcomas. Deregulation of the ubiquitin-proteasome system (UPS) has provided attractive targets for therapeutic intervention also in these malignancies. As sarcoma cells hijack UPS to stabilize aberrant oncogenic signaling and/or inactivate tumor

Targeting the deubiquitinase USP7 for degradation with PROTACs.

Targeting deubiquitinating enzymes (DUBs) has emerged as a promising therapeutic approach in several human cancers and other diseases. DUB inhibitors are exciting pharmacological tools but often exhibit limited cellular potency. Here we report PROTACs based on a ubiquitin-specific protease 7 (USP7) inhibitor scaffold to degrade USP7. By investigating several linker and E3 ligand types, including novel cereblon recruiters, we discovered a highly selective USP7 degrader tool compound that induced apoptosis of USP7-dependent cancer cells. This work represents one of the first DUB degraders and un

Integrating multi-omics data reveals function and therapeutic potential of deubiquitinating enzymes.

Deubiquitinating enzymes (DUBs), ~100 of which are found in human cells, are proteases that remove ubiquitin conjugates from proteins, thereby regulating protein turnover. They are involved in a wide range of cellular activities and are emerging therapeutic targets for cancer and other diseases. Drugs targeting USP1 and USP30 are in clinical development for cancer and kidney disease respectively. However, the majority of substrates and pathways regulated by DUBs remain unknown, impeding efforts to prioritize specific enzymes for research and drug development. To assemble a knowledgebase of DUB

Therapeutic inhibition of USP7 promotes antitumor immune responses.

BACKGROUND: Ubiquitin-specific peptidase 7 (USP7) is a deubiquitinating enzyme that removes ubiquitin from specific protein substrates to modify their degradation rates thereby regulating crucial cellular processes integral to cancer. Conspicuously, overexpression of USP7 is strongly associated with the progression and poor prognosis in various cancers. Therefore, the design of potent and selective USP7 inhibitors poses an attractive therapeutic approach. The mechanism of action of USP7 inhibitors in cancer cells relies on MDM2 depletion and the restoration of p53. METHODS: In this study, we

Disrupting the OTUD4-USP7 deubiquitinase complex to suppress herpesvirus replication: a novel antiviral strategy.

The development of effective and broad-spectrum antiviral therapies remains an unmet need. Current virus-targeted antiviral strategies are often limited by narrow spectrum of activity and the rapid emergence of resistance. As a result, there is increasing interest in alternative approaches that target host cell factors critical for viral replication. One promising strategy is the targeting of deubiquitinases (DUBs), enzymes that regulate key host and viral proteins involved in viral reactivation and replication. In this study, we explore the potential of targeting a DUB complex for antiviral t

Sertraline and Astemizole Enhance the Deubiquitinase Activity of USP7 by Binding to Its Switching Loop Region.

The heterozygous loss-of-function mutations of USP7 lead to the occurrence of Hao-Fountain syndrome, and chemical activators targeting USP7 could potentially serve as a treatment option for the disease. Here, in this study, two drugs Sertraline and Astemizole were identified to act as the agonists of USP7 by binding to its switching loop region. Moreover, although two compounds and USP7's self-activation C-terminal peptide (CTP) share the same binding pocket in the enzyme, joint activation toward full-length USP7 was observed for sertraline/astemizole and the CTP. According to the published da

Antagonistic Ubiquitin Switching by USP7 and RNF40 Orchestrates KDM6A Homeostasis to License Coronavirus Susceptibility.

Lysine demethylase 6A (KDM6A) is a critical epigenetic regulator implicated in development, cancer, and viral infection. Although KDM6A enhances coronavirus entry by modulating viral receptor expression, the mechanisms governing its protein stability remain unknown. Here, we show that ubiquitin-specific protease 7 (USP7) promotes diverse coronavirus infection, including SARS-CoV, SARS-CoV-2, MERS-CoV, and MHV, and represents a broad-spectrum anti-coronavirus target. Genetic and pharmacological inhibition of USP7 attenuates the expression of coronavirus receptors ACE2, DPP4, and Ceacam1, thereb

Ubiquitylation-oxaliplatin-related prognosis signature reveals the landscapes of immune responses, cell communication, and therapeutic sensitivity for colorectal cancer.

Ubiquitylation plays a crucial role in posttranslational modification, and its dysregulation is linked to chemosensitivity. However, its role in oxaliplatin (OXA) resistance in colorectal cancer (CRC) remains unclear. Transcriptomic data from the cancer genome atlas (TCGA) and Gene Expression Omnibus were used to develop a ubiquitylation-OXA resistance-related risk score (URGScore), integrating immunological, mutational, and clinical features. Single-cell RNA sequencing (scRNA-seq) identified OXA-resistant cell populations, and Genomics of Drug Sensitivity in Cancer predicted drug sensitivity.

USP7 attenuates endoplasmic reticulum stress-induced apoptotic cell death through deubiquitination and stabilization of FBXO7.

Parkinson's disease (PD) is a common neurodegenerative disease (NDD) characterized by the loss of dopaminergic neurons in the substantia nigra. Similar to other NDDs, the buildup of toxic protein aggregates in PD leads to progressive neuronal loss, culminating in neurodegeneration. Accumulating evidence indicates that alterations in subcellular organelles, particularly the endoplasmic reticulum (ER), are critically involved in pathological neurodegenerative events in NDDs, including PD. Mutations in the F-box only protein 7 (FBXO7 or PARK15) gene have been found to cause early onset autosomal

MLKL-USP7-UBA52 signaling is indispensable for autophagy in brain through maintaining ubiquitin homeostasis.

Individuals with genetic elimination of MLKL (mixed lineage kinase domain like pseudokinase) exhibit an increased susceptibility to neurodegenerative diseases like Alzheimer disease (AD). However, the mechanism is not yet fully understood. Here, we observed significant compromise in macroautophagy/autophagy in the brains of mlkl knockout (KO) mice, as evidenced by the downregulation of BECN1/Beclin1 and ULK1 (unc-51 like autophagy activating kinase 1). We identified UBA52 (ubiquitin A-52 residue ribosomal protein fusion product 1) as the binding partner of MLKL under physiological conditions.

Deubiquitinating Enzymes Ubiquitin-Specific Proteases 7 and 10 Regulate TAU Aggregation.

Accumulation of the microtubule-associated protein TAU into inclusions is a hallmark of tauopathies including Alzheimer's disease (AD), potentially driven by impaired protein degradation and dysregulated ubiquitination. To explore the role of deubiquitinating enzymes (DUBs), we performed siRNA knockdown screens targeting 93 murine DUBs in rTg4510 cortical cultures. Knockdown and pharmacological inhibition of the ubiquitin-specific proteases 7 (Usp7) and 10 (Usp10) significantly reduced seeded TAU aggregation without affecting soluble TAU levels. These effects were observed in both cortical and

Toosendanin sensitizes hepatocyte ferroptosis via dual inhibition of Nrf2 O-GlcNAcylation and USP7-driven deubiquitination.

Toosendanin (TSN) is the key bioactive component of Melia toosendan Sieb. et Zucc known for alleviating pain and expelling roundworms, but severe hepatotoxicity limited its further application. O-GlcNAcylation is a dynamic and reversible post-translational modification which has emerged as an important regulatory mechanism in cellular response to liver injury. In this study, we investigated the effect of aberrant O-GlcNAcylation and augmented O-GlcNAc signaling via Thiamet G (TMG) on TSN-induced ferroptosis in HepaRG cells. Evolutionarily conserved regulators of tau identify targets for new therapies.

Tauopathies are neurodegenerative diseases that involve the pathological accumulation of tau proteins; in this family are Alzheimer disease, corticobasal degeneration, and chronic traumatic encephalopathy, among others. Hypothesizing that reducing this accumulation could mitigate pathogenesis, we performed a cross-species genetic screen targeting 6,600 potentially druggable genes in human cells and Drosophila. We found and validated 83 hits in cells and further validated 11 hits in the mouse brain. Three of these hits (USP7, RNF130, and RNF149) converge on the C terminus of Hsc70-interacting p

Study on the mechanism of USP7 promoting endometriosis by regulating DNMT1 deubiquitination level.

Endometriosis is characterized by aberrant epigenetic regulation, and our study reveals that both Ubiquitin Specific Protease 7 (USP7) and DNA methyltransferase 1 (DNMT1) are significantly overexpressed in endometriosis tissues. Functional analyses in ectopic endometrial stromal cells (EESCs) indicate that elevated USP7 levels markedly enhance cellular proliferation, migration, and invasion, whereas silencing DNMT1 mitigates these oncogenic properties. Notably, USP7 and DNMT1 co-localize within the nucleus and interact directly, with USP7 modulating DNMT1 stability by attenuating its ubiquitin

Targeted degradation of USP7 in solid cancer cells reveals distinct effects of deubiquitinase degraders and inhibitors.

Proteolysis-targeting chimeras (PROTACs) co-op the ubiquitin system for targeted protein degradation, creating opportunities to interrogate cellular functions of proteins through "chemical knockdown". However, matched pairs of protein degraders and inhibitors, that possess high specificity and chemical complementarity, for individual components of the ubiquitin system have remained scarce. This includes reagents to modulate activity and abundance of deubiquitinases (DUBs). Here, using an integrated chemical biology approach, we explore cellular functions of the DUB USP7 as a case study by comp
