On the existence of a mono-vinyl d-urobilin.

Chromic acid degradation of a d-urobilin, obtained after incubation of bilirubin in fecal bacterial cultures, gave methylvinylmaleimide and methylethylmaleimide. The d-urobilin, molecular weight 588, C(33)H(40)-N(4)O(6), clearly showed the presence of both vinyl and ethyl resonances in the nuclear magnetic resonance spectrum. These results point unambiguously to a urobilin structure with one vinyl and one ethyl beta-substituent.

Microbiome-derived cancer: the catabolism of bilirubin to urobilin in the liver-gut axis and its consequences.

Colorectal cancer (CRC) is the second leading cause of cancer-related deaths globally and is associated with factors, such as obesity, inflammation, and metabolic disorders. Bilirubin, a byproduct of heme degradation, is increasingly recognized as a signaling molecule with antioxidant properties that protect against obesity by reducing oxidative stress, decreasing inflammation, and activating the nuclear receptor PPARα, which enhances fat metabolism and utilization. The gut microbiome converts bilirubin to urobilinogen via bilirubin reductase, which is then rapidly oxidized to urobilin, thereb

An effective urobilin clearance strategy based on paramagnetic beads facilitates microscale proteomic analysis of urine.

Urine provides an ideal source for disease biomarker discovery. High-adhesion contaminants such as urobilin, which are difficult to remove from urine, can severely interfere with urinary proteomic analysis. Here, we aimed to establish a strategy based on single-pot, solid-phase-enhanced sample preparation (SP3) technology to prepare samples for urinary proteomics analysis that almost completely eliminates the impact of urobilin. A systematic evaluation of the effects of two urinary protein precipitation methods, two types of protein lysis buffers, and different ratios of magnetic digestion bea

Stercobilin and Urobilin in Aqueous Media: Existence of Specific H-Aggregates and Nonspecific Higher Aggregates at Different Concentrations.

Fecal matter is considered to be one of the primary sources of water pollution. Understanding the aggregation behavior of the fecal pigments (FPs) could play a critical role in their detection and analysis. This work shows that in aqueous media, the fluorescence of FPs indicates the presence of multiple emitting species, which have been assigned to monomers, lower-order H-aggregates (dimers), and higher-order H-aggregates. Steady-state absorbance, fluorescence and time-resolved fluorescence decay studies conclude that the emission of FPs in aqueous medium indicates H-type of aggregation, even

Bilirubin bioconversion to urobilin in the gut-liver-kidney axis: A biomarker for insulin resistance in the Cardiovascular-Kidney-Metabolic (CKM) Syndrome.

The rising rates of obesity worldwide have increased the incidence of cardiovascular disease (CVD), making it the number one cause of death. Higher plasma bilirubin levels have been shown to prevent metabolic dysfunction and CVD. However, reducing levels leads to deleterious outcomes, possibly due to reduced bilirubin half-life that escalates the production of its catabolized product, urobilinogen, produced by gut bacteria and naturally oxidized to urobilin. Recent findings suggest that the involvement of the microbiome catabolism of bilirubin to urobilin and its absorption via the hepatic por

Suppressing Hepatic UGT1A1 Increases Plasma Bilirubin, Lowers Plasma Urobilin, Reorganizes Kinase Signaling Pathways and Lipid Species and Improves Fatty Liver Disease.

Several population studies have observed lower serum bilirubin levels in patients with non-alcoholic fatty liver disease (NAFLD). Yet, treatments to target this metabolic phenotype have not been explored. Therefore, we designed an N-Acetylgalactosamine (GalNAc) labeled RNAi to target the enzyme that clears bilirubin from the blood, the UGT1A1 glucuronyl enzyme (GNUR). In this study, male C57BL/6J mice were fed a high-fat diet (HFD, 60%) for 30 weeks to induce NAFLD and were treated subcutaneously with GNUR or sham (CTRL) once weekly for six weeks while continuing the HFD. The results show that

l-Stercobilin-HCl and d-Urobilin-HCl. Analysis of Their Chiroptical and Conformational Properties by VCD, ECD, and CPL Experiments and MD and DFT Calculations.

Vibrational circular dichroism (CD) and IR spectra of dichloromethane solutions of l-stercobilin and d-urobilin hydrochlorides have been recorded in the mid-IR region. The spectra are best interpreted by combining molecular dynamics calculations and density functional theory (DFT) calculations within the quantum mechanics/molecular mechanics ONIOM-type framework, and the combined predicted results are better and more informative than the more standard analysis provided by DFT calculations. The same approach also sheds light on the Cotton effect sign inversion of room temperature versus low-tem

Disrupted bilirubin metabolism and gut microbiome dysregulation: A link to cardio-renal-hepatic-metabolic health in obstructive sleep apnea.

Obstructive Sleep Apnea (OSA) is no longer viewed solely as a sleep related respiratory disorder but as a systemic disease intricately linked to Cardio-Renal-Hepatic-Metabolic (CRHM) axis. Chronic intermittent hypoxia (CIH), oxidative stress, and sympathetic overactivation in OSA lead to endothelial dysfunction, inflammation, and metabolic imbalance. Emerging evidence highlights the gut microbiome as a key mediator in OSA-related metabolic disturbances, where hypoxia-induced dysbiosis reduces microbial diversity and promotes systemic inflammation via endotoxemia and toll-like receptor (TLR) ac

Transforming Toxicity into Therapy: Exploring Bilirubin's Benefits and Its Molecular Role in Cardiac Health and Disease.

Bilirubin, historically recognized solely as a waste product of heme catabolism, has recently gained attention for its potential protective role in the cardiovascular system. Experimental and clinical studies suggest that bilirubin exhibits potent antioxidant, anti-inflammatory, anti-apoptotic, and cytoprotective properties that may protect the heart against oxidative stress, ischemia-reperfusion injury, and the progression of cardiovascular diseases, such as heart failure. As an endogenous hormone, bilirubin activates peroxisome proliferator-activated receptor-α (PPARα), a nuclear receptor th

The human intestinal microflora.

The major host defense mechanisms against bacterial overgrowth in the small bowel are the normal propulsive activity of the bowel itself and gastric acid secretion. Microbial interactions are a major factor in regulating the indigenous bacterial flora. Studies of the bacterial enzymes of the gut suggest that changes in diet may lead to marked changes in the colonic flora. Antibiotics affect the composition of the colonic microflora. The microflora also influence the degradation of mucin, the conversion of urobilin to urobilinogen, of cholesterol to coprostanol, and the production of short chai

Integrative microbiome and metabolome profiles reveal the impacts of periodontitis via oral-gut axis in first-trimester pregnant women.

BACKGROUND: Periodontitis results from host-microbe dysbiosis and the resultant dysregulated immunoinflammatory response. Importantly, it closely links to numerous systemic comorbidities, and perplexingly contributes to adverse pregnancy outcomes (APOs). Currently, there are limited studies on the distal consequences of periodontitis via oral-gut axis in pregnant women. This study investigated the integrative microbiome-metabolome profiles through multi-omics approaches in first-trimester pregnant women and explored the translational potentials. METHODS: We collected samples of subgingival pl

Hematuria in rabbits.

A nine year retrospective study of hematuria in 14 New Zealand White rabbits was conducted to classify possible etiologies of this clinical finding. Physical examination, laboratory tests, radiography and postmortem examination were utilized in most cases to verify the presence of hematuria and to determine its etiology. Uterine adenocarcinoma was diagnosed in two rabbits. Three rabbits had uterine polyps with hemorrhage. Renal infarction with hemorrhage was diagnosed in three rabbits. Urolithiasis with secondary urethral obstruction and hemorrhagic cystitis was identified as the cause of hema

[Changes of the clinico-biochemical indices in the rumirid juice and urine in experimental aflatoxicosis of dairy cows].

The health condition and course of fermentation processes in the rumen were studied in four cows of the Red Spotted breed at the age of four to nine years. The clinico-biochemical indices in the rumen liquor and urine were used. The experimental animals were exposed to a mixture of aflatoxins applied in the dose of 200 mg B1 and 80 mg B2. The toxic action of aflatoxins manifested itself as inappetence, increased temperature, changes in the pulse and respiration rate and reduced activity of the proventriculi. Diarrhoea was observed in two animals. The pH value, total acidity and ammonia level i

Intestinal colonization leading to fecal urobilinoid excretion may play a role in the pathogenesis of neonatal jaundice.

BACKGROUND: Neonatal hyperbilirubinemia remains of concern because of the potential danger for the central nervous system. Because urobilinogen is a nontoxic derivative of bilirubin, the current study was conducted to examine the fecal excretion of urobilinoids and bilirubin in healthy newborns and infants, as well as their intestinal bacteria capable of reducing bilirubin, to assess a possible relation to serum bilirubin levels during the first weeks of life. METHODS: Bilirubin pigments, urobilinoids, and porphyrins were measured in stools of infants during the first week (group A, n = 60) a

Bilirubin Hepatic and Intestinal Transport and Catabolism: Physiology, Pathophysiology, and Benefits.

Bilirubin, a metabolite derived from heme degradation, has traditionally been regarded as a waste product and a marker of liver injury. However, increasing evidence suggests that bilirubin also functions as a hormone, and reduced levels are associated with metabolic dysfunction. Studies have shown a strong association between low circulating bilirubin levels and an increased risk of metabolic disorders and cardiovascular disease. To advance bilirubin-based treatment strategies, it is essential to elucidate the mechanisms underlying bilirubin transport and metabolism. Therefore, we provide an i
