Edible Vaccines: A Patent-Driven Exploration of Immunization Technologies.

Vaccines are biological preparations that improve immunity to particular diseases. Particularly for poor developing nations, edible vaccines show significant potential as a financially advantageous, simple to administer, straightforward to store, fail-safe, and socially and culturally acceptable vaccine delivery system. A vaccine incorporates the gene-encoding bacterial or viral disease-causing agent in plants without losing its immunogenic property. Potatoes, tomatoes, rice, soybeans, and bananas are the primary plants for edible vaccines. It activates the systemic and mucosal immunity respon

Plant-based edible vaccines: Can cholera be the case study in Africa?

Vaccines are employed as a sanitary approach that is implemented to lessen the hurdles caused by infectious diseases on the safety of public health. A vaccine is biologically made from inactive components of microbes, to enhance immunity and as a defense mechanism adverse to parasitic, bacterial and viral illnesses. Nonetheless, the mode of production that involves purification is quite costly, more so, to low and middle-income countries, especially in Africa. Conventional oral cholera vaccines, though commercially available, face logistical challenges to be transported and distributed to targ

Plant and probiotic-based edible vaccines for Helicobacter pylori: Research status and application prospects.

Over half of the global population suffers from Helicobacter pylori infection, linked to gastrointestinal disorders and even gastric cancer. As antibiotic resistance increases, so do treatment failure rates, creating an urgent demand for more effective treatments. Plant-derived or probiotic-expressed antigenic proteins elicit immunity via oral delivery, offering a cost-effective anti-colonization strategy. Current edible plants and Lactococcus lactis expression systems leverage genetic engineering to produce recombinant Helicobacter pylori antigen proteins. Oral administration of these vaccine

Stable transformation of Spirulina (Arthrospira) platensis: a promising microalga for production of edible vaccines.

The planktonic blue-green microalga Spirulina (Arthrospira) platensis possesses important features (e.g., high protein and vital lipids contents as well as essential vitamins) and can be consumed by humans and animals. Accordingly, this microalga gained growing attention as a new platform for producing edible-based pharmaceutical proteins. However, there are limited successful strategies for the transformation of S. platensis, in part because of an efficient expression of strong endonucleases in its cytoplasm. In the current work, as a pilot step for the expression of therapeutic proteins, an

Plant Biofactories for Vaccination: Progress in Edible Vaccine Technologies as a Sustainable Approach to Global Immunization.

Globally, the pathogen-driven enteric diseases remain a leading cause of morbidity and mortality, disproportionately affecting populations in low-income regions. While significant strides have been made in vaccine research, challenges such as high production costs, complex manufacturing processes, and inefficient distribution continue to hinder global vaccine accessibility. To overcome these challenges, plant-based edible vaccines have emerged as a promising and innovative alternative, offering advantages such as affordability, ease of administration, and reduced reliance on cold-chain infrast

Development of an edible rabies vaccine in maize using the Vnukovo strain.

The objective of this study was to obtain transgenic maize expressing the rabies virus glycoprotein (G) of the Vnukovo strain and to evaluate its immunogenicity in mice, by the oral route. The ubiquitin maize promoter fused to the whole coding region of the rabies virus G gene, and a constitutive promoter from cauliflowermosaic virus (CaMV)were used. Maize embryogenic callus were transformed with the above construct by biolistics. Regenerated maize plants were recovered and grown in a greenhouse. The presence of the G gene and its product was detected by PCR and western blot, respectively. The

Recent Progress and Challenges in the Development of Edible Vaccines Produced by Genetically Modified Plants.

Biotechnologists have pioneered the idea of an edible vaccination in recent years. Subunit vaccines, such as those used to create edible vaccines, involve the introduction of certain genes into transgenic plants, which are subsequently coaxed into producing the corresponding protein. Bananas, potatoes, legumes, lettuce, soybeans, corn, and rice are all examples of foods that fall under this category. They have a low unit cost, can be stored conveniently, and are simple to administer to patients of varying ages. There is great hope that the use of edible vaccinations, particularly in underdevel

New advances in the production of edible plant vaccines: chloroplast expression of a tetanus vaccine antigen, TetC.

Vaccines are a proven method of controlling disease. However there are issues with the delivery and administration of vaccines. A particular problem is that the majority of vaccines currently used are injected, which can be unsafe if needles are reused in areas where blood-borne diseases are prevalent. Vaccines targeting the mucosal immune system avoid many of the problems associated with injections. One potential form of mucosal vaccine is based on the expression of vaccine antigens in plants. Current research in this area has focused on the expression of immunogens from the plant's nuclear g

Peptide immunotherapy for allergic diseases using a rice-based edible vaccine.

PURPOSE OF REVIEW: Plant pollens are the most common cause of seasonal allergic disease. The number of patients undergoing treatment for allergies to the pollen of Japanese cedar (major antigens: Cry j 1 and Cry j 2) has increased steadily each year. Integration of an effective, safe and inexpensive clinical program would be greatly improved by addressing deficiencies in systemically delivered immunotherapy. RECENT FINDINGS: We have demonstrated that feeding mice transgenic rice seeds accumulating the T-cell epitope peptides of Cry j 1 and Cry j 2 before systemic challenge with total protein

Immunogenicity of norovirus VP1-expressing silkworm (Bombyx mori) pupae as an edible vaccine candidate.

Norovirus (NV) gastroenteritis remains a global health challenge, particularly affecting infants and the elderly. Despite its significant clinical impact, no licensed vaccine is currently available. This study explores a novel approach to NV vaccine development using the silkworm-baculovirus expression vector system (silkworm-BEVS), focusing on non-purified VP1-expressing silkworm pupae as a potential edible vaccine candidate. We assessed the immunogenicity of orally administered VP1-expressing silkworm pupae in mice using two delivery formats: crude pupal suspensions administered by oral gava

Expression of protective antigen in transgenic plants: a step towards edible vaccine against anthrax.

Protective antigen (PA) is the most potent molecule for vaccination against anthrax. In the present study, we have successfully integrated protective antigen gene in nuclear genome of tobacco plants by Agrobacterium mediated leaf-disc transformation method. Expression of protective antigen gene was detected by immunoblot analysis using antisera raised against purified PA. A distinct band of approximately 83kDa lighted up in the protein extracted from transformed plants while there was no such band in untransformed plants. The plant expressed PA showed biological activity just like native PA, w

Efficacy of heat-inactivated Mycobacterium bovis vaccine delivered to European badgers (Meles meles) through edible bait.

Badgers (Meles meles) are a major tuberculosis (TB) reservoir in Europe, with the potential to transmit infection to cattle. Here we assessed whether a recently described oral tuberculosis vaccine based on heat-inactivated Mycobacterium bovis (HIMB), delivered as edible baits, can protect badgers from infection. Eight badgers were given individually five baits, each one consisting of a ball of peanut butter, natural peanut and oat flakes including a dose of the vaccine containing 5 × 107 colony-forming units. In parallel, a control group of seven badgers did not receive the vaccine. One month

A Preliminary Study of a Lettuce-Based Edible Vaccine Expressing the Cysteine Proteinase of Fasciola hepatica for Fasciolosis Control in Livestock.

Oral vaccination with edible vaccines is one of the most promising approaches in modern vaccinology. Edible vaccines are an alternative to conventional vaccines, which are typically delivered by injection. Here, freeze-dried transgenic lettuce expressing the cysteine proteinase of the trematode Fasciola hepatica (CPFhW) was used to orally vaccinate cattle and sheep against fasciolosis, which is the most important trematode disease due to the parasite's global distribution, wide spectrum of host species and significant economic losses of farmers. In the study, goals such as reducing the intensi

Leveraging the potential of bacterial lateral gene transfer in boosting the efficacy of an edible probiotic prototype yogurt vaccine for COVID-19.

Administration of coronavirus disease-2019 (COVID-19) vaccines with appropriate booster doses through painful injections under clinical supervision was challenging during the recent COVID-19 pandemic. As an alternative solution, we designed a safer, edible probiotic yogurt vaccine prototype (YoVac) that can be orally consumed by circumventing painful injections and clinical supervision. We hypothesized that YoVac prepared using Lactobacillus carrying an antigen coding gene (donor) can transfer the same to other bacteria (recipients) in the human gut microbiome (hgMb) through lateral gene trans

Constructing and transient expression of a gene cassette containing edible vaccine elements and shigellosis, anthrax and cholera recombinant antigens in tomato.

Shigella dysenteriae causing shigellosis is one of the diseases that threaten the health of human society in the developing countries. In Shigella, IpaD gene is one of the key pathogenic genes causing strong mucosal immune system reactions. Anthrax disease is caused by Bacillus anthracis. PA protective antigen is one of the subunits in anthrax toxin complex responsible for the transfer of other subunits into the cytosol of host cells. The 20 kDa subunit of PA (PA20) has the property of immunogenicity. CTxB or B subunit of Vibrio cholerae toxin (CT) is a non-toxic protein and has the function t

Development of transgenic algae strain expressing CTB-M2e fusion gene an approach towards the development of a universal edible vaccine in algae.

Avian Influenza, the most studied virus, is of high concern due to its zoonotic pandemic potential. In recent years, several influenza vaccines have been used with the broad goal of managing and in certain cases, eliminating the disease. The matrix 2 extracellular domain (M2e), is one of the key targets of the universal influenza vaccine, a liner peptide that is conserved throughout all influenza A subtypes virus. Many recombinant influenza proteins have been expressed in yeast and plants for vaccine development. A remarkable development has been made in the field of biotechnology to explore t

Duckweed-based edible vaccine confers complete protection against avian infectious bronchitis virus by inducing robust mucosal and systemic immunity.

Molecular farming using edible plants offers a cost-effective strategy for vaccine production because plants serve as both bioreactors and oral delivery systems. Owing to its rapid clonal propagation and high protein content, duckweed represents a promising platform for this purpose. Here, we developed an edible duckweed-based vaccine targeting avian infectious bronchitis virus (IBV), a highly contagious respiratory pathogen in poultry, and directly compared its efficacy to that of a commercial live-attenuated H120 vaccine. Transgenic duckweed expressing the IBV antigen peptide EpiC was orally

Expression of a modified Mannheimia haemolytica GS60 outer membrane lipoprotein in transgenic alfalfa for the development of an edible vaccine against bovine pneumonic pasteurellosis.

The GS60 antigen is one of the protective antigens of Mannheimia haemolytica A1. GS60 contains conserved domains belonging to the LppC family of bacterial outer membrane lipoproteins. A high antibody titer to GS60 has been shown to be significantly correlated with resistance to pneumonic pasteurellosis. Calves vaccinated with a commercial vaccine (Presponse) and demonstrating protection against M. haemolytica A1 produced antibodies directed against GS60. Alfalfa was chosen as the platform for an edible vaccine. Agrobacterium tumefaciens was used to mediate the transformation of alfalfa with se
