Vascular Injury After Stenting - Insights of Systemic Mechanisms of Vascular Repair.

BACKGROUND: The role of circulating progenitor cells (CPC) in vascular repair following everolimus-eluting stent (EES) implantation is largely unknown. The aim of the study was to investigate the relationship between temporal variation in CPC levels following EES implantation and the degree of peri-procedural vascular damage, and stent healing, as measured by optical coherence tomography (OCT). METHODS AND RESULTS: CPC populations (CD133+/KDR+/CD45low) included patients with stable coronary artery disease undergoing stent implantation, and were evaluated using a flow cytometry technique both

Complement activation and vascular injury in systemic lupus erythematosus.

The deposition of immune complexes within blood vessel walls results in the potential for complement activation and the release of chemotactic factors, such as fragments of C5 (C5fr). The generation of C5fr results in the intravascular aggregation of neutrophils with subsequent leukostatic occlusion of the pulmonary arterioles. The generation of C5fr may contribute to the pathogenesis of adult respiratory distress syndrome and other diseases. Studies were undertaken to determine the role of circulating complement derived peptides and intravascular neutrophil activation in systemic lupus erythe

Polymer-induced central nervous system complications following vascular procedures: spectrum of iatrogenic injuries and review of outcomes.

Polymer substances are commonly applied as surface coatings on endovascular catheters and vascular devices. Adverse effects related to their use have been reported, although the overall clinical significance and appropriate methods of detection of these complications have been unclear. In this analysis, we systematically reviewed clinical and diagnostic features in 32 patients (age, 36-87years; mean, 59years) in whom intracranial polymer reactions were documented following vascular interventions. Associated neuroradiologic and neuropathologic findings were variable and included cerebral vascul

Endothelial GSDMD underlies LPS-induced systemic vascular injury and lethality.

Endothelial injury destroys endothelial barrier integrity, triggering organ dysfunction and ultimately resulting in sepsis-related death. Considerable attention has been focused on identifying effective targets for inhibiting damage to endothelial cells to treat endotoxemia-induced septic shock. Global gasdermin D (Gsdmd) deletion reportedly prevents death caused by endotoxemia. However, the role of endothelial GSDMD in endothelial injury and lethality in lipopolysaccharide-induced (LPS-induced) endotoxemia and the underlying regulatory mechanisms are unknown. Here, we show that LPS increases

[A case of chronic renal insufficiency due to vascular injury of systemic sclerosis].

A 48-year-old woman with systemic sclerosis (SSc) and rheumatoid arthritis was admitted to our hospital because of renal dysfunction. She had no hypertension at the time of admission and mild hypertension for only one and a half months until the time of admission. After admission, she received angiotensin-converting enzyme-inhibitor, but her renal dysfunction did not improve. She then had thrombotic microangiopathy with thrombocytopenia, and was treated with plasma exchange five times, but her renal dysfunction persisted. The renal biopsy specimens showed an arteriosclerotic lesion with intima

Protection of the Vascular System by Polyethylene Glycol Reduces Secondary Injury Following Spinal Cord Injury in Rats.

BACKGROUND: Polyethylene glycol (PEG) is a hydrophilic polymer, which has been known to have a neuroprotective effect by sealing the ruptured cell membrane, but PEG effects on the vascular systems and its underlying mechanisms remain unclear. Here, we showed the neuroprotective effect of PEG by preventing damage to the vascular system. METHODS: A spinal contusion was made at the T11 segment in male Sprague-Dawley rats. PEG was injected into the subdural space immediately after SCI. Vascular permeability was assessed for 24 h after SCI using intraperitoneally injected Evans blue dye. Junctiona

Systemic Loss of FKBPL Uncovers Diabetes-Dependent Pathways of Myocardial and Vascular Injury.

BACKGROUND: Impaired angiogenesis underpins cardiovascular disease, particularly in people with diabetes; however, molecular mechanisms are still poorly understood. This study aims to decipher the role of an emerging antiangiogenic protein, FKBPL (FK506-binding protein-like), on cardiac structure and function, vascular integrity, and inflammatory signaling in in vivo and in vitro models of diabetes. METHODS: In vivo, FKBPL transgenic mice (fkbpl+/-) were used to examine the metabolic and cardiovascular function and FKBPL-mediated mechanisms in streptozotocin-induced diabetes. In addition, com

Vascular endothelial cell injury in progressive systemic sclerosis and other connective tissue diseases.

The growth inhibition of in vitro vascular endothelial cell cultures by sera from patients with connective tissue diseases was studied. Seven out of 30 sera from patients with progressive systemic sclerosis (PSS) exhibited a cytotoxic effect. The cytotoxic serum factor(s) was not specific to patients with PSS, it was also demonstrated in sera from patients with other connective tissue diseases with vascular symptoms (11 out of 27 sera from patients with a mixed connective tissue disease, MCTD; 5 out of 34 sera from patients with a systemic lupus erythematosus, SLE). The cytotoxic effect was no

Pentraxin 3 as a biomarker of local inflammatory response to vascular injury in systemic lupus erythematosus.

Systemic lupus erythematosus (SLE) is an autoimmune disorder with organ injury related to vasculitis. Inflammation of blood vessels results from auto-immunological activation of endothelial cells. The pentraxin 3 (PTX3), might act as an indicator of vasculitides in many diseases. The aim of this study was to determine whether PTX3 might be useful as a marker of vascular injury in SLE. This study was carried out in a group of 56 SLE women, and in the 28 female volunteers control group. All participants' plasma and serum samples were collected to estimate concentrations (ELISA) of PTX3, soluble

High mobility group motif proteins' role in fibrosis, inflammation, and vascular injury in systemic sclerosis.

Systemic Sclerosis (SSc) is an idiopathic systemic autoimmune disease characterized by progressive cutaneous and systemic fibrosis, severe vasculopathy, and multiple humoral and cellular immunological alterations. The pathogenesis of SSc is highly complex and remains incompletely elucidated. The fibrotic process is a crucial component of SSc and is responsible for organ failure and high mortality. Although an increasing understanding of the fibrotic process has enabled the clinical development of antifibrotic therapeutic agents, these agents have limited clinical efficacy. Recently, the potent

Investigation of therapeutic efficacy of intraperitoneal ketamine administration on the brain vascular system in rats with traumatic brain injury.

Traumatic brain injury (TBI) is a condition characterized by high morbidity and mortality. This study investigated the therapeutic effects of ketamine on TBI through electrophysiological and immunohistochemical methods in a model of 30 rats divided into five equal groups. Following trauma, ketamine, mannitol, and hypertonic saline treatments were administered. Analyses conducted on the 10th day post-trauma revealed that ketamine most effectively reduced the expressions of HMGB1, S100b, RAGE, and EGR-1 across all treatment groups. Furthermore, EEG findings demonstrated a significant decrease in

A Biomimetic Nanoparticle System Intercepts and Degrades Thrombospondin-1 to Restore Vascular Homeostasis After Ischemic Injury.

The thrombospondin-1 (TSP-1)/CD47 signaling axis plays a critical role in endothelial injury during hepatic ischemia-reperfusion (IR). Targeting this pathway represents a promising strategy to prevent liver IR injury; however, existing CD47 antagonists cause severe hematologic side effects due to nonspecific binding to erythrocytes. This study developed TAX2 (CD47-derived peptide with the sequence CEVSQLLKGDAC)-functionalized biodegradable nanoparticles (TAX2-NPs) that specifically capture TSP-1 and block its interaction with CD47 on liver sinusoidal endothelial cells (LSECs). TAX2-NPs are com

Traumatic Brain Injury Impairs Systemic Vascular Function Through Disruption of Inward-Rectifier Potassium Channels.

Trauma can lead to widespread vascular dysfunction, but the underlying mechanisms remain largely unknown. Inward-rectifier potassium channels (Kir2.1) play a critical role in the dynamic regulation of regional perfusion and blood flow. Kir2.1 channel activity requires phosphatidylinositol 4,5-bisphosphate (PIP2), a membrane phospholipid that is degraded by phospholipase A2 (PLA2) in conditions of oxidative stress or inflammation. We hypothesized that PLA2-induced depletion of PIP2 after trauma impairs Kir2.1 channel function. A fluid percussion injury model of traumatic brain injury (TBI) in r
