Identification of roughskin newt medullary vasotocin target neurons with a fluorescent vasotocin conjugate.

Arginine8 vasotocin (AVT), a neurohypophyseal peptide in nonmammalian vertebrates, plays a key role in the regulation of social behaviors related to reproduction. In male roughskin newts (Taricha granulosa), AVT is an important facilitator of several reproductive behaviors, including courtship clasping of females. Although AVT is known to act in certain brain regions and AVT receptors have been localized to some extent, specific target neurons for this peptide have not been identified in any species. Internalization of a receptor-specific conjugate of AVT and the fluorescent dye Oregon green w

From vasotocin to stress and cognition.

Sex and stress hormones coordinate experience and behaviour with physiological regulations. In the brain the sex hormones act to promote the repertoire of affiliative and reproductive behaviours. Stress hormones target in particular brain circuits underlying emotional arousal and cognition. To exert these actions the hormones operate in concert with neuropeptide secreting systems. Here I will discuss three examples of hormone action on brain and behaviour. First in the song bird manipulation of brain vasotocin promotes acquisition of a stable stereotyped song pattern. Second in mammal's centra

Sleep in the cat: raphe dorsalis and vasotocin.

To determine the relationship between the raphe nuclei and the sleep-inducing pineal peptide arginine vasotocin (AVT), we investigated the hypnogenic effects of intraventricularly administered AVT in adult cats with electrolytic lesions performed either in the raphe dorsalis nucleus (RDN) or the raphe pontis nucleus (RPN). The following results were observed: (a) RDN lesions produced a quantitative decrease in NREM sleep and a transitory increase in REM sleep and narcoleptic-like alterations of REM sleep in the first 2-5 postoperative days. On the eighth postoperative day, normal (prelesion) s

Arginine-vasotocin (AVT)--a pineal hormone in mammals.

Covering more than 30 years of studying the pineal peptide hormones physiology, this study largely discusses the main personal research as well as international literature indirectly or directly supporting the idea that the nonapeptide hormone arginine-vasotocin (AVT) is a pineal hormone in mammals. After a short review of the problem, the data are structured in two chapters: one including all the indirect arguments, i.e. those results demonstrating that the ly synthetic exogenous AVT mimics all the effects attributed to the pineal gland itself or to its releasing hormone the indole melatonin,

Arginine vasotocin is a player in the multifactorial control of spermatogenesis in zebrafish.

This study investigated the role of vasotocin (AVT) in the multifaceted regulation of spermatogenesis in zebrafish, with a focus on its interaction with gonadotropin hormones. Using an ex vivo cultured testis, we explored the interaction of AVT with gonadotropins, LH and FSH, to regulate different stages of germ cell development. In this study, we used recombinant zebrafish FSH and hCG to stimulate the LH-induced response due to the limited availability of zebrafish LH, as it is known to exert similar effects on testicular function. Treatment with AVT enhanced FSH-induced proliferation of earl

Integrated Role of Arginine Vasotocin in the Control of Spermatogenesis in Zebrafish.

Arginine vasotocin (AVT) has recently emerged as a local regulator of testicular function in fish. Using ex vivo culture system, it was demonstrated that AVT directly stimulates androgen-dependent basal spermatogenesis in zebrafish. In the presence of gonadotropins, AVT enhanced FSH-induced development of early phases of spermatogonial proliferation while blocking FSH-mediated spermiogenesis. However, AVT promoted proliferation of LH-induced pre-meiotic and meiotic germ cell populations without affecting the final stages of spermiogenesis. These findings led to the hypothesis that AVT plays a

Mechanism of 1-deamino-arginine vasotocin induced natriuresis in rats.

1-Deamino-arginine vasotocin (1dAVT) induced diuresis and a considerable increase in urinary sodium excretion in female Wistar rats. Sodium fractional excretion rose up to 19.3 ± 1.1%. An increase in urine flow rate after 1dAVT (0.5 nmol/kg body-weight [bw]) injection was accompanied by a significant rise of the solute-free water reabsorption. The 1dAVT-induced natriuresis was as high as natriuresis produced by injection of a maximal dose of furosemide (10mg/kg bw). Roles of arginine vasotocin receptors in the brain and pituitary of submammalian vertebrates.

This chapter reviews the functions of arginine vasotocin (AVT) and its receptors in the central nervous system (CNS) of primarily submammalian vertebrates. The V1a-type receptor, which is widely distributed in the CNS of birds, amphibians, and fish, is one of the most important receptors involved in the expression of social and reproductive behaviors. In mammals, the V1b receptor of arginine vasopressin, an AVT ortholog, is assumed to be involved in aggression, social memory, and stress responses. The distribution of the V1b-type receptor in the brain of submammalian vertebrates has only been

Renal response of roosters with diabetes insipidus to infusions of arginine vasotocin.

In a strain of white Leghorn chicken with symptoms of hereditary diabetes insipidus (DI) and with elevated circulating levels of the antidiuretic hormone arginine vasotocin (AVT), we investigated whether the defect is based on nonfunctional endogenous AVT or on a diminished renal response to AVT. DI chickens responded to hyposmotic infusions with a higher urine flow and lower glomerular filtration rate (GFR) than healthy control birds. Urine osmolality, fractional excretion, and clearance of sodium were lower in DI birds while potassium values were similar in both groups. Hence, the high urine

REM sleep dependent release of vasotocin into cerebrospinal fluid of narcoleptics.

The lumbar cerebrospinal fluid (CSF) collected from 6 narcoleptic patients (three males and three females) aged between 31 and 47 years, as well as from 2 patients having Pickwickian syndromes with hypersomnia attacks (two males), aged 43-51 years, contained relatively high amounts of arginine vasotocin (AVT) only if the CSF was removed after a REM sleep period. The release was not induced by darkness since the same release of AVT occurred when the CSF was removed after a REM sleep period during daylight hours. No detectable AVT levels have been found in the CSF samples of four narcoleptics co

Arginine Vasotocin Directly Regulates Spermatogenesis in Adult Zebrafish (Danio rerio) Testes.

The neuropeptide vasopressin is known for its regulation of osmotic balance in mammals. Arginine vasotocin (AVT) is a non-mammalian homolog of this neuropeptide that is present in fish. Limited information suggested that vasopressin and its homologs may also influence reproductive function. In the present study, we investigated the direct effect of AVT on spermatogenesis, using zebrafish as a model organism. Results demonstrate that AVT and its receptors (avpr1aa, avpr2aa, avpr1ab, avpr2ab, and avpr2l) are expressed in the zebrafish brain and testes. The direct action of AVT on spermatogenesis

Neural distribution of vasotocin receptor mRNA in two species of songbird.

The neurohypophyseal hormones vasopressin and oxytocin are produced and released within the mammalian brain, where they act via multiple receptor subtypes. The neural distributions of these receptors, for example, V1a and oxytocin receptors, have been well described in many mammals. In birds, the distribution of binding sites for the homologous neuropeptides, vasotocin (VT) and mesotocin, has been studied in several species by using synthetic radioligands designed to bind to mammalian receptors. Such binding studies, however, may not reveal the specific distributions of each receptor subtype.

Effects of arginine vasotocin on REM sleep in narcoleptics and in symptomatic hypersomniacs.

Arginine vasotocin (AVT), in doses of 1.5 microgram, administered intranasally to two narcoleptics (a male aged 42 years and a female aged 35 years), as well as to 5 symptomatic hypersomniacs (3 females and 2 males), aged 25-57 years, induced very rapidly in most instances in sleep with REM periods at onset and markedly increased in amount of REM sleep during a fixed period of 180 min of sleep recording. After AVT administration there was a significant shortening of the time of occurrence of the first REM sleep period both in narcoleptics (t test = 0.05 greater than P greater than 0.025) and i

Vasotocin influences mother-offspring associations in a facultatively family-living lizard.

Parent-offspring associations provide the basis for more complex care and breeding systems. One proximate mechanism by which parental behavior is facilitated across animal taxa is via nonapeptide hormones such as arginine vasotocin (AVT). However, existing research largely comes from obligately social species, and whether such mechanisms are conserved in social systems where care is facultative remains unclear. Here, we used a facultatively family living skink, Liopholis whitii, to test the role of AVT in mediating 2 key parental behaviors: parent-offspring associations (measured by proximity

Influence of age on August levels of pineal immunoreactive arginine vasotocin in rats.

The present study was undertaken to evaluate carefully the influence of age on physiological levels of arginine vasotocin-like peptide in rat pineal glands. Glands were collected from male and female rats aged 13, 33, 53, and 73 days on August 4, 12, and 19, 1984. Individual extracts were assayed for arginine vasotocin (AVT) by radioimmunoassay. The results confirm our previous observation that rat pineal AVT immunoactivity (iAVT) increases significantly during August each year; and in this study, each group of rats reached the same peak level of iAVT (700-750 pg/gland) regardless of age or ge

Immunocytochemical localization of somatostatin and vasotocin in the brain of the Pacific hagfish, Eptatretus stouti.

The brain of the Pacific hagfish, Eptatretus stouti, was studied immunocytochemically using antisera against somatostatin (SRIH), arginine vasopressin (AVP), and adrenocorticotropic hormone (ACTH). SRIH-immunoreactive perikarya were distributed bilaterally in the postoptic nucleus and in the hypothalamic nucleus. Although several short, stained fibers were observed in the vicinity of the perikarya, SRIH-immunoreactivity was not found in the neurohypophysis, nor in other parts of the brain. On the other hand, presumed arginine vasotocin (AVT) perikarya were distributed in an arc-shaped region e

Influence of arginine vasotocin on a genetically determined excessive appetite for water in chickens.

Females from two inbred strains of chickens, one normal and one having hereditary diabetes insipidus, were treated with five levels (2.5, 1.0, 0.5, 0.25, and 0.1 units) of Schwartz-Mann (Grade A, 100 units/mg.) arginine vasotocin via ic. injections at 2-hr. intervals. Paired sibs, one normal and one having diabetes insipidus (di), from an F2 population were used in one experiment to compare responses at the level of 2.5 units. Only di females were used for the remaining four levels of treatment. Water intake was determined every two hours and calculated as a percentage of water (ml.)/body wt.
