Emerging Verticillium Wilt of Napa Cabbage in Korea: Morphological, Molecular, and Pathogenic Characterization of Verticillium dahliae and Verticillium longisporum (A1/D1).

Verticillium wilt is an emerging threat to Brassicaceae crops worldwide, yet information on the species diversity and pathogenicity of Verticillium on Napa cabbage (Brassica rapa subsp. pekinensis) in South Korea remains limited. During surveys in 2020 and 2021, patches of wilted Napa cabbage plants were observed in highland cultivation regions, with disease incidence ranging from 1 to 10%. Twenty symptomatic plants were collected, and fifteen fungal isolates were obtained through tissue culture. Morphological observations and multilocus phylogenetic analyses based on four loci (ACT, EF, GPD,

Diversity of endophytic fungi from different Verticillium-wilt-resistant Gossypium hirsutum and evaluation of antifungal activity against Verticillium dahliae in vitro.

Cotton plants were sampled and ranked according to their resistance to Verticillium wilt. In total, 642 endophytic fungi isolates representing 27 genera were recovered from Gossypium hirsutum root, stem, and leaf tissues, but were not uniformly distributed. More endophytic fungi appeared in the leaf (391) compared with the root (140) and stem (111) sections. However, no significant difference in the abundance of isolated endophytes was found among resistant cotton varieties. Alternaria exhibited the highest colonization frequency (7.9%), followed by Acremonium (6.6%) and Penicillium (4.8%). Un

Entomopathogenic potential of Verticillium and Acremonium species (Deuteromycotina: Hyphomycetes).

Hyphomycetes with conidia formed in slimy heads from hyaline mycelium were isolated from a range of insect hosts. Isolation on artificial medium and microscopic examination revealed that these fungi in many cases were not Verticillium lecanii despite a superficial resemblance to this common entomopathogen. The fungi were identified as Verticillium fusisporum, Verticillium psalliotae, Verticillium lamellicola, and species of Acremonium. Isolates of these fungi were bioassayed against the sweet-potato whitefly (Bemisia tabaci) and against the housefly (Musca domestica) to examine their entomopat

GhMAC3e is involved in plant growth and defense response to Verticillium dahliae.

GhMAC3e expression was induced by various stresses and hormones. GhMAC3e may regulate plant growth by influencing auxin distribution, and play important roles in Verticillium wilt resistance via mediating SA signaling. The MOS4-Associated Complex (MAC) is a highly conserved protein complex involved in pre-mRNA splicing and spliceosome assembly, which plays a vital role in plant immunity. It comprises key components such as MOS4, CDC5, and PRL1. MAC3A/B, as U-box E3 ubiquitin ligases, are crucial for various plant processes including development, stress responses, and disease resistance. Howeve

[Infectivity of the entomopathogenic fungus Verticillium lecanii in mice and guinea pigs].

The resistance of plague insects to chemical insecticides as well as the importance of a healthy environment demands an alternative for agricultural plagues. Among others, biological control seems an alternate strategy with fungal entomopathogens playing a relevant role. The hyphomycete Verticillium lecanii is a natural bioregulator of aphids, scales and white-flies that attack different agricultural plantations. Its use in biological control programs must be assessed previously by safety procedures such as its innocuity in mammals and useful animals and plants. The aim of this study was point

VdPRMT1 Is Required for Fungal Growth, Metabolism, and Pathogenicity in Verticillium dahliae.

Protein arginine methyltransferases (PRMTs) are key regulators of diverse cellular processes in eukaryotes, including transcriptional regulation, RNA processing, signal transduction and DNA repair. However, the biological functions of PRMTs in Verticillium dahliae remain largely unexplored. In this study, we identified a PRMT1 homolog in V. dahliae. Targeted deletion of VdPRMT1 resulted in severely impaired hyphal growth, sporulation, stress responses and pathogenicity. Subcellular localization analysis showed that VdPRMT1 is distributed in both the nucleus and cytoplasm of hyphae. Host-induce

Vayg1 is required for microsclerotium formation and melanin production in Verticillium dahliae.

The fungus Verticillium dahliae causes vascular wilt disease on many plant species, including economically important crop and ornamental plants worldwide. It produces darkly pigmented resting structures known as microsclerotia, which are able to survive for up to 14years in soil, and represent one of the defining characteristics of this species. The pigment produced in V. dahliae is dihydroxynaphthalene (DHN)-melanin, a form of melanin common among fungi and named so for the intermediary of this melanin biosynthetic pathway. In this study, we characterized the function of the V. dahliae Vayg1

A novel hexasegmented virus isolated from the phytopathogenic fungus Verticillium nonalfalfae.

In this study, we describe a novel virus, tentatively named Verticillium nonalfalfae virus M (VnaVM), identified in the phytopathogenic fungus Verticillium nonalfalfae isolated from hop (Humulus lupulus L.). The VnaVM genome consists of six double-stranded RNA (dsRNA) segments ranging from 1051 to 2401 bp. DsRNAs 1-3 encode an RNA-dependent RNA polymerase (RdRp), a hypothetical protein and a methyltransferase (MTR), respectively, while dsRNA6 encodes a proline-alanine-serine-rich protein (PASrp). These four proteins share less than 58% amino acid identity with homologs encoded by viruses in th

The Verticillium dahliae Small Cysteine-Rich Protein VdSCP23 Manipulates Host Immunity.

Verticillium wilt caused by Verticillium dahliae is a notorious soil-borne fungal disease and seriously threatens the yield of economic crops worldwide. During host infection, V. dahliae secretes many effectors that manipulate host immunity, among which small cysteine-rich proteins (SCPs) play an important role. However, the exact roles of many SCPs from V. dahliae are unknown and varied. In this study, we show that the small cysteine-rich protein VdSCP23 inhibits cell necrosis in Nicotiana benthamiana leaves, as well as the reactive oxygen species (ROS) burst, electrolyte leakage and the expr

Bioprotection of Olive Trees Against Verticillium Wilt by Pomegranate and Carob Extracts.

Bioprotection through the use of plant extracts is an environmentally friendly strategy in crop protection. Effective control of Verticillium wilt of olive (Olea europaea; VWO), caused by Verticillium dahliae, has proven challenging because of the ineffectiveness of chemicals, which makes it necessary to search for new control tools. Thus, the aim of this study was to evaluate the effect of pomegranate (Punica granatum) and carob (Ceratonia siliqua) extracts on VWO. Extracts derived from pomegranate peels and carob pods and leaves were obtained using ethanol, methanol, or ethyl acetate as a so

Comparative Proteomic Analysis of Gossypium thurberi in Response to Verticillium dahliae Inoculation.

Verticillium wilt is threatening cotton productivity globally. This disease is caused by soil-borne Verticillium dahliae which directly infects cotton roots, and exclusively colonizes and occludes xylem vessels, finally resulting in necrosis, defoliation, and most severely, plant death. For the first time, iTRAQ (isobaric tags for relative and absolute quantification) was applied to screen the differentially expressed proteins of Gossypium thurberi inoculated with V. dahliae. A total of 6533 proteins were identified from the roots of G. thurberi after inoculation with V. dahliae, and 396 showe

Heterologous Expression of the Melon CmVQ23 Positively Regulates Resistance to Verticillium dahliae in Arabidopsis.

Verticillium dahliae is a devastating soil-borne fungal pathogen that causes severe yield losses in melon (Cucumis melo L.) and other crops. Identifying novel resistance genes is crucial for sustainable disease management. In this study, we characterized the function of CmVQ23, a candidate gene previously identified through QTL mapping, in mediating defense against V. dahliae using heterologous expression in Arabidopsis thaliana. Subcellular localization assays revealed that the CmVQ23-eGFP fusion protein predominantly localized to the nucleus, consistent with its predicted role as a co-factor

Ergosterol-targeting fusion antifungal peptide significantly increases the Verticillium wilt resistance of cotton.

Increasing the targeting ability of antifungal proteins towards specific components of fungal cells has the potential to improve their antifungal activity and reduce harmful effects to nontarget cells. To obtain effective disease resistance genes against cotton Verticillium wilt, we constructed several fusion genes, in which binding domains targeting chitin, sphingolipid or ergosterol in the fungal cell wall or cell membrane were individually fused to the antifungal peptide BbAFP1 from entomopathogenic fungus Beauveria bassiana. Transient expression of fusion genes in cotton cotyledons indicat

Fluorescent pseudomonads pursue media-dependent strategies to inhibit growth of pathogenic Verticillium fungi.

Verticillium species represent economically important phytopathogenic fungi with bacteria as natural rhizosphere antagonists. Growth inhibition patterns of Verticillium in different media were compared to saprophytic Aspergillus strains and were significantly more pronounced in various co-cultivations with different Pseudomonas strains. The Brassica napus rhizosphere bacterium Pseudomonas fluorescens DSM8569 is able to inhibit growth of rapeseed (Verticillium longisporum) or tomato (Verticillium dahliae) pathogens without the potential for phenazine or 2,4-diacetylphloroglucinol (DAPG) mycotox

Multi-source UAV remote sensing for cotton Verticillium wilt resistance grading using TPAC-Net.

INTRODUCTION: Cotton Verticillium wilt resistance grading under field conditions remains challenging because disease symptoms are spatially heterogeneous within plots, local pathological cues are unevenly distributed across high-resolution unmanned aerial vehicle (UAV) orthomosaics, and disease-related responses are not expressed with the same sensitivity or spatial consistency across RGB, raw multispectral (MS), and vegetation-index (VI) representations. METHODS: To address these challenges, this study proposes TPAC-Net, a UAV-based Tri-source Pathological Alignment and Coupling Network for

Cotton SNARE complex component GhSYP121 regulates salicylic acid signaling during defense against Verticillium dahliae.

Syntaxin of plant (SYP) plays a crucial role in SNARE-mediated membrane trafficking during endocytic and secretory pathways, contributing to the regulation and execution of plant immunity against pathogens. Verticillium wilt is among the most destructive fungal diseases affecting cotton worldwide. However, information regarding SYP family genes in cotton is scarce. Through genome-wide identification and transcriptome profiling, we identified GhSYP121, a Qa SNARE gene in Gossypium hirsutum. GhSYP121 is notably induced by Verticillium dahliae, the causal agent of Verticillium wilt in cotton, and
