Experimental infection of Didelphis marsupialis with vesicular stomatitis New Jersey virus.

Although vesicular stomatitis has been present for many years in the Americas, many aspects of its natural history remain undefined. In this study, we challenged five adult Virginia opossums (Didelphis marsupialis) with vesicular stomatitis New Jersey serotype virus (VSNJV). Opossums had no detectable antibodies against VSNJV prior to being inoculated with 10(6.5) median tissue culture infective doses (TCID(50)) of VSNJV by two routes; intraepithelial/subepithelial (IE/SE) inoculation and scarification in the muzzle (SM). Clinical response was monitored daily and animals were tested for viral

Genetic diversity of enzootic isolates of vesicular stomatitis virus New Jersey.

The RNA genomes of 43 vesicular stomatitis virus (VSV) isolates of the New Jersey (NJ) serotype were T1-ribonuclease fingerprinted to compare the extent of genetic diversity of virus from regions of epizootic and enzootic disease activity. Forty of these viruses were obtained from Central America during 1982 to 1985. The other three were older isolates, including a 1970 isolate from Culex nigripalpus mosquitos in Guatemala, a 1960 bovine isolate from Panama, and a 1976 isolate from mosquitos (Mansonia indubitans) in Ecuador. The data indicate that extensive genetic diversity exists among virus

Coinfection with a rhabdovirus: vesicular stomatitis virus of Indiana and New-Jersey serotypes.

Coinfection of cells with vesicular stomatitis virus (VSV) of Indiana and New-Jersey serotypes were performed. Thermosensitive mutants (ts) of VSV Indiana and the wild type strain (+) of New-Jersey were used. Harvests and titrations were made at permissive(PT) and nonpermissive (NPT) temperatures. It was shown that the harvest was mainly composed of one parental-like infectious particles. The dominance of one serotype over the other was shown to be a function of the relative multiplicity of the two viruses; the presence of a thermosensitive lesion imparts a disadvantage to the corresponding se

Potentiation of vesicular stomatitis New Jersey virus infection in mice by mosquito saliva.

Saliva of arthropod vectors can modulate vertebrate host immunological functions in many ways. To investigate if vesicular stomatitis New Jersey virus (VSNJ) infection could be potentiated by arthropod saliva, mice in three different age groups (3 days, 3 weeks, or > 8 months) were exposed to VSNJ-infected mosquitoes or were needle injected with an equivalent dose of VSNJ (titre 1.5-3 logs). Previous studies have demonstrated that VS viruses do not replicate in mice older than 3 weeks of age. Infection was monitored by examining serum for the presence of VSNJ at 2 days postinfection (PI) or fo

Potential for contact and mechanical vector transmission of vesicular stomatitis virus New Jersey in pigs.

OBJECTIVE: To determine whether swine could be infected with vesicular stomatitis virus New Jersey (VSV-NJ) by routes simulating mechanical vector and contact transmission, and to document clinical response and measure source, duration, and extent of associated viral shedding. ANIMALS: 48 approximately 10-kg male pigs. PROCEDURE: 7 inoculation routes were tested at 3 virus concentrations (10(6), 10(4), and 10(2) median tissue culture-infective doses [TCID50]). Inoculation routes included intradermal (snout), topical application of virus to a scarified area (ear or mucous membrane of the lip)

Pathogenesis of experimental vesicular stomatitis virus (New Jersey serotype) infection in the deer mouse (Peromyscus maniculatus).

The pathogenesis of vesicular stomatitis virus (VSV) infection has not been investigated previously in native New World rodents that may have a role in the epidemiology of the disease. In the present study, 45 juvenile and 80 adult deer mice (Peromyscus maniculatus) were inoculated intranasally with VSV New Jersey serotype (VSV-NJ) and examined sequentially over a 7-day period. Virus was detected by means of immunohistochemistry and in situ hybridization in all tissues containing histologic lesions. Viral antigen and mRNA were observed initially in olfactory epithelium neurons, followed by olf

Intracellular formation of two soluble glycoproteins in BHK cells infected with vesicular stomatitis virus serotype New Jersey.

Infection of BHK 21 cells with VSV serotype New Jersey gave rise to three intracellular viral glycoproteins: the membrane-integrated G protein and the two soluble glycoproteins Gs and Gss which lacked the cytoplasmic and transmembrane domains as was deduced from limited chemical cleavage of the glycoproteins by hydroxylamine. Both soluble glycoproteins were completely protected by the microsomal membrane against proteolytic digestion. The soluble glycoproteins were formed in the endoplasmic reticulum because both were fully endo H sensitive after a 5-min pulse with [35S]methionine. Protease in

Nucleotide sequence of a cDNA clone encoding the entire glycoprotein from the New Jersey serotype of vesicular stomatitis virus.

The nucleotide sequence of the mRNA encoding the glycoprotein from the New Jersey serotype of vesicular stomatitis virus (VSV) was determined from a cDNA clone containing the entire coding region. The sequence of 12 5'-terminal noncoding nucleotides present in the mRNA but not in the cDNA clone was determined from a primer extended to the 5' terminus of the mRNA. The mRNA is 1,573 nucleotides long (excluding polyadenylic acid) and encodes a protein of 517 amino acids. Only six nucleotides occur between the translation termination codon and the polyadenylic acid. Short homologies between the un

Pathogenesis of Vesicular Stomatitis New Jersey Virus Infection in Deer Mice ( Peromyscus maniculatus) Transmitted by Black Flies ( Simulium vittatum).

The natural transmission of vesicular stomatitis New Jersey virus (VSNJV), an arthropod-borne virus, is not completely understood. Rodents may have a role as reservoir or amplifying hosts. In this study, juvenile and nestling deer mice ( Peromyscus maniculatus) were exposed to VSNJV-infected black fly ( Simulium vittatum) bites followed by a second exposure to naive black flies on the nestling mice. Severe neurological signs were observed in some juvenile mice by 6 to 8 days postinoculation (DPI); viremia was not detected in 25 juvenile deer mice following exposure to VSNJV-infected fly bites.

Historical Serological Evidence of Human Exposure to Vesicular Stomatitis Virus (New Jersey and Indiana Serotypes) in Dairy Regions of Costa Rica (1999).

BACKGROUND: Vesicular stomatitis is endemic in southern Mexico, Central America, and northern South America. The causative agent, vesicular stomatitis virus (VSV; Rhabdoviridae: Vesiculovirus), includes two serotypes-New Jersey (VSNJV) and Indiana (VSIV)-both present in Costa Rica. Transmission occurs via direct contact, fomites, and insect vectors. Occupational exposure, particularly in livestock workers, is a known risk, and high human seroprevalence has been reported elsewhere in Central America. Some cases present with a self-limited febrile illness, occasionally with vesicular lesions. No

Vesicular stomatitis New Jersey virus (VSNJV) infects keratinocytes and is restricted to lesion sites and local lymph nodes in the bovine, a natural host.

Inoculation of vesicular stomatitis New Jersey virus (VSNJV) by skin scarification of the coronary-band in cattle, a natural host of VSNJV, resulted in vesicular lesions and 6-8 log(10) TCID(50) increase in skin virus titers over a 72 h period. Virus infection was restricted to the lesion sites and lymph nodes draining those areas but no virus or viral RNA was found in the blood or in 20 other organs and tissues sampled at necropsy. Scarification of flank skin did not result in lesions or a significant increase in viral titer indicating that viral clinical infection is restricted to skin inocu

Serologic survey for selected arboviruses and other potential pathogens in wildlife from Mexico.

During 1988 and 1989, a serologic survey of wildlife was conducted in northeastern Mexico to determine the presence, prevalence, and distribution of arboviruses and other selected disease agents. Eighty mammal specimens were tested. Antibodies to vesicular stomatitis-Indiana, Venezuelan equine encephalitis-Mena II, Rio Grande virus, and vesicular stomatitis-New Jersey were detected predominantly in small mammals. Deer and mouflon (Ovis musimon) had antibodies to bluetongue and epizootic hemorrhagic disease. Two species had serologic evidence of recent exposure to Francisella tularensis. A whit

SEROLOGIC SURVEY FOR SELECTED VIRAL AND BACTERIAL SWINE PATHOGENS IN COLOMBIAN COLLARED PECCARIES ( PECARI TAJACU) AND FERAL PIGS ( SUS SCROFA).

In South America, wild populations of peccaries coexist with domestic and feral pigs, with poorly understood consequences. We captured 58 collared peccaries ( Pecari tajacu) and 15 feral pigs ( Sus scrofa) in locations of Colombia where coexistence of these species is known. Blood samples were tested for antibodies against four viral agents, classical swine fever virus (CSFV), Aujeszky's disease virus (ADV), porcine circovirus (PCV-2), and vesicular stomatitis virus (New Jersey and Indiana subtypes) and two bacterial agents, Brucella spp. and six serovars of Leptospira interrogans. Epizootic vesicular stomatitis in Colorado, 1982: infection in occupational risk groups.

In 1982-1983, an epizootic of vesicular stomatitis occurred in the western United States. Veterinarians, research workers, and regulatory personnel who were exposed to vesicular stomatitis virus were examined for patterns of human infection and prevalence of vesicular stomatitis New Jersey serotype neutralizing antibody. Insight into the mechanism of transmission was sought by comparing activities of antibody-positive and antibody-negative persons. A statistically significant risk factor was a history of infected animals sneezing in the face of serosurvey participants. Elevated odds ratios wer

Detection and serotype-specific differentiation of vesicular stomatitis virus using a multiplex, real-time, reverse transcription-polymerase chain reaction assay.

A multiplex, real-time reverse transcription-polymerase chain reaction (RT-PCR) assay was developed that allowed simultaneous detection and rapid differentiation of vesicular stomatitis virus strains--New Jersey (VSV-NJ) and Indiana 1, 2, and 3 (VSV-IN1-3). This assay involves use of a set of VSV universal primers located in the L gene that amplify VSV-IN1-3 and VSV-NJ using probes that allow differentiation of the major serotypes Indiana and New Jersey. The assay was evaluated using reference VSV, foot-and-mouth disease virus, swine vesicular disease virus, and vesicular exanthema of swine vi

Functional analysis of T lymphocyte subsets in antiviral host defense.

The role of different T cell subsets in antiviral host defense was investigated by treating thymectomized C57BL/6 and CBA/J mice with monoclonal rat anti-Lyt-2 or anti-L3/T4 IgG 2b antibodies 14 and 10 days before infection. This treatment depleted the respective T cell subsets to undetectable levels in peripheral blood when assayed by immunofluorescence. In mice treated with anti-Lyt-2, induction of cytotoxic T cells was reduced to less than 1 to 2% after intravenous infection with Armstrong strain of lymphocytic choriomeningitis virus (LCMV). In addition, no primary swelling of the footpad c
