To determine the pathogenic potential of the vesiculovirus Piry for domestic animals, two ponies, two steers, three sheep, three goats and three pigs were inoculated intradermally in the tongue or, in the case of the pigs, in the snout, heel and coronary band. Inoculated animals were housed in one room and allowed to mingle freely with an equal number of uninoculated contact animals of each species. Clinical signs of infection, consisting of elevated temperature and ulcers at the inoculation sites, were only observed in the ponies, but all inoculated animals developed specific antibody followi

Bats are the natural reservoirs for a variety of emerging and re-emerging viruses. Among them, rabies virus (genus Lyssavirus, family Rhabdoviridae) is one of the first and most emblematic described in these animals. Since its first description, several new bat lyssaviruses have been regularly identified. In addition to lyssaviruses, other bat rhabdoviruses have also been discovered, including members of the genera Vesiculovirus, Ledantevirus and, more recently, Alphanemrhavirus and Tupavirus. However, the family Rhabdoviridae is one of the most abundant and diverse viral families, with 434 of

Chandipura vesiculovirus (CHPV) is a rapidly emerging pathogen responsible for causing acute encephalitis. Due to its widespread occurrence in Asian and African countries, this has become a global threat, and there is an urgent need to design an effective and nonallergenic vaccine against this pathogen. The present study aimed to develop a multi-epitope vaccine using an immunoinformatics approach. The conventional method of vaccine design involves large proteins or whole organism which leads to unnecessary antigenic load with increased chances of allergenic reactions. In addition, the process

The characteristics and ecology of Isfahan virus, a new vesicular stomatitis virus (VSV) serotype, are described. Two strains of the agent were isolated from pools of Phlebotomus papatasi collected in Iran in 1975. Its animal pathogenicity, growth rate, cytopathic effect, and plaque morphology are similar to those of the other VSV serotypes. Electron microscopic examination of the virus demonstrated a bullet shape, the presence of truncated particles and maturation at plasma membranes. Antigenic relationships between Isfahan virus and three other VSV serotypes (Cocal, Piry, and Chandipura) wer

Vesicular stomatitis is an infectious disease that occurs mainly in countries of the Western Hemisphere and affects cattle, swine and horses. The clinical symptoms in cattle and swine are similar to foot-and-mouth disease and include vesicular ulceration of the tongue and mouth. The disease requires a rapid and accurate differential diagnosis, aiming for immediate implementation of control measures. The objective of the present study was to develop and perform validation tests of multiplex RT-qPCR(s) for the detection of RNA from Alagoas vesiculovirus, considering the parameters of sensitivity

Nonlimit chemical cleavage at tryptophan residues of protein labeled at the amino terminus afforded a simple procedure for generating specific fragments and for mapping tryptophan positions. A comparison of the matrix (M) and nucleocapsid (N) proteins of four members of the Vesiculovirus group by this procedure suggests considerable conservation of tryptophan number and location in the four serotypes examined.

Vesiculovirus carajas (CARV) is a pathogen with neuroinvasive potential, yet its impact on neuroinflammation and sickness behavior remains poorly understood. In this study, we investigated the neuropathological and immunological responses to CARV encephalitis in adult BALB/c mice. Mice were intranasally inoculated with either infected or uninfected brain homogenates, and clinical, histopathological, and cytokine profiles were analyzed. CARV antigens were primarily detected in necrotic neurons, with prominent microglial activation near the ventricles and blood vessels. By day 10 post-infection,

Asp-Glu-Ala-Asp (DEAD) box helicase 3 X-linked (DDX3X) plays important regulatory roles in the replication of many viruses. However, the role of DDX3X in rhabdovirus replication has seldomly been investigated. In this study, snakehead vesiculovirus (SHVV), a kind of fish rhabdovirus, was used to study the role of DDX3X in rhabdovirus replication. DDX3X was identified as an interacting partner of SHVV phosphoprotein (P). The expression level of DDX3X was increased at an early stage of SHVV infection and then decreased to a normal level at a later infection stage. Overexpression of DDX3X promote

Phosphoprotein (P), co-factor of the polymerase (large protein, L) of single-stranded negative-sense RNA viruses, is phosphorylated during viral infection and its phosphorylation has been reported to play important roles in viral replication. However, the function of P phosphorylation in viral replication is still far from clear. Snakehead vesiculovirus (SHVV) is a kind of fish rhabdovirus that has caused serious economic losses in snakehead fish culture in China without any effective preventive or therapeutical measures currently. In this study, 4D label-free phosphoproteomics sequencing of S

This study investigates potential epitopes in the glycoprotein and RNA of Chandipura vesiculovirus (CHPV) using MHC Class I (HLA-A0201) and MHC Class II (DRB1_0101) molecules with 3D structures derived from x-ray crystallography. Computationally derived peptides were mapped and subjected to in silico docking, revealing promising targets for CD8+ cytotoxic and CD4+ helper T cells. Key factors analysed include solvent accessible surface area (SASA), Debye-Waller factor (B-factor), and polar bond interactions. Post-docking, removal of N-acetylglucosamine (NAG) increased peptide stability and redu

Snakehead vesiculovirus (SHVV) is a type of rhabdovirus that causes serious economic losses in snakehead fish culture in China. However, no specific antiviral drugs or vaccines are currently available for SHVV infection. In this study, 4D label-free ubiquitome analysis of SHVV-infected cells revealed dozens of ubiquitinated sites on the five SHVV proteins. We focused on investigating the ubiquitination of phosphoprotein (P), a viral polymerase co-factor involved in viral replication. SHVV-P was proved to be ubiquitinated via K63-linked ubiquitination at lysine 264 (K264). Overexpression of wil

Investigating the mechanisms of viral attachment and entry into cells is crucial for understanding viral pathogenesis and developing therapeutic strategies. The aim of this review is to characterize pseudo-typed particles based on the vesicular stomatitis virus (VSV) as a convenient and effective tool for studying viral entry into cells, based on literature data (PubMed, Scopus, and Web of Science), and to determine the prospects for combining this method with genetic and protein-based approaches. VSV, a member of the Rhabdoviridae family, has a remarkable capacity for pseudotyping, which invo

In 1983, 17 virus strains were isolated from mosquitoes collected during an outbreak of western equine encephalitis in Santa Fe Province, Argentina. Strains of western equine encephalitis, Venezuelan equine encephalitis, St. Louis encephalitis, and Antequera viruses were isolated, as were several bunyaviruses of the California and Bunyamwera serogroups and a new vesiculovirus. Complement fixation and neutralization tests were used to identify the California serogroup virus as a subtype of Melao virus, the Bunyamwera serogroup virus as a subtype of both Maguari and Playas viruses, and the vesic

The snakehead vesiculovirus (SHVV) is an emerging and highly pathogenic rhabdovirus that infects a variety of economically important fish species, posing a serious threat to the sustainable development of aquaculture. The phosphoprotein (P) of rhabdoviruses is known to facilitate viral replication and host immune evasion, although the precise mechanisms remain unclear. In this study, the spleen transcriptomic analysis of SHVV-infected fish revealed significantly enriched signaling pathways, and pull-down assays coupled with mass spectrometry identified TANK-binding kinase 1 (TBK1) ranked the f

Chronic hepatitis B virus (HBV) infections cause more than 800,000 deaths per year and currently approved treatments do not cure the disease. Because a hallmark of acute infection resolution is the presence of functional CD8+ T cells to the virus, activation of the immune system with therapeutic vaccines represents a potential approach for treating chronic hepatitis B. In this study, we evaluated the immunogenicity and efficacy of two attenuated vesiculovirus-based platforms expressing HBV Core antigen, the highly attenuated vesicular stomatitis virus (VSV) N4CT1 and a unique vaccine platform

Five different phlebotomus-transmitted viruses have been isolated in Italy. Four of them belong to the Bunyaviridae family (genus Phlebovirus). Sandfly fever Sicilian and Sandfly fever Naples viruses were isolated from sick persons during World War II, Toscana and Arbia viruses have been isolated more recently from Phlebotomus perniciosus and P. perfiliewi. Radi virus is the only phlebotomus-transmitted virus isolated in Italy from P. perfiliewi, which belongs to the Rhabdoviridae family (genus Vesiculovirus). The circulation and the natural cycle of these viruses on the basis of seroepidemiol

BACKGROUND: Chandipura virus (CHPV), a neurotropic, negative-sense RNA virus belonging to the genus Vesiculovirus (family Rhabdoviridae), has emerged as a significant cause of acute encephalitis syndrome (AES) in India, predominantly affecting children. Since the major outbreaks in 2003-2004 and the recent resurgence in 2024, CHPV has re-emerged as a high-fatality, climate-sensitive arboviral threat transmitted primarily by phlebotomine sandflies. The absence of specific antivirals or licensed vaccines underscores the need for comprehensive understanding and preparedness.

Bats carry zoonotic viruses which can be harmful to humans. Zoonotic diseases have caused huge economic losses in the production and trade of animal products and recurring diseases outbreaks and global pandemics. Studies have shown that Rabies and rabies related viruses (Lyssavirus genera, family Rhabdoviridae) are spread to humans by bats. The aim of this article is to assess the global distribution of bat Rhabdoviruses, detection and challenges in Africa. Studies have shown that the prevalence of Rhabdoviruses is high in Africa and Asia. In addition to Rabies virus, other bat Rhabdoviruses w

Vesicular stomatitis is a disease of livestock caused by some members of the Vesiculovirus genus (Family Rhabdoviridae), two of which are called 'vesicular stomatitis virus'. Clinical disease presents as severe vesiculation and/or ulceration of the tongue, oral tissues, feet, and teats, and results in substantial loss of productivity. Except for its appearance in horses, it is clinically indistinguishable from foot-and-mouth disease. Unlike foot-and-mouth disease, it is very infectious for man and can cause a temporarily debilitating disease. Vesicular stomatitis occurs seasonally every year i
