Proteomic comparison of Hypnale hypnale (hump-nosed pit-viper) and Calloselasma rhodostoma (Malayan pit-viper) venoms.

UNLABELLED: Treatment of Hypnale hypnale bites with commercial antivenoms, even those raised against its sister taxon Calloselasma rhodostoma, has never been clinically successful. As these two genera have been separated for 20million years, we tested to see whether significant variations in venom had accumulated during this long period of evolutionary divergence, and thus could be responsible for the failure of antivenom. Proteomic analyses of C. rhodostoma and H. hypnale venom were performed using 1D and 2D PAGE as well as 2D-DIGE. C. rhodostoma venom was diverse containing large amounts of

An enzyme-linked immunoassay for detection of North American pit viper venoms.

We describe a method for immunodetection of North American pit viper venoms in clinical materials. Antibody-enzyme conjugates prepared against venoms of the western diamondback rattlesnake (Crotalus atrox), Mojave rattlesnake (Crotalus scutulatus), and copperhead (Agkistrodon contortrix) detect homologous venoms in concentrations of 0.1-.01 mcg/ml using a double antibody sandwich technique. Venoms of 10 additional species of U.S. pit vipers were detected in concentrations of 10 mcg/ml or less. Venoms of 4 species could not be detected at levels likely to be encountered in clinical situations.

Viper venoms and coumarin-induced prothrombin. A comparison of several one-stage methods employing three different venoms as thromboplastins.

Three venoms obtained from three vipers, namely Echis carinatus, Notechis scutatus scutatus and Oxyuranus scutellatus, have been used as thromboplastin in a one-stage assay of coumarin-induced prothrombin. Regardless of the venom used, prothrombin resulted to be low in coumarin-treated patients. The mean values obtained were 27.2, 33.6, and 24.2%, respectively. These values were comparable to those obtained by means of the classical one-stage method (24.8%). A good correlation was observed among the different methods. However, the levels observed using the Notechis scutatus scutatus venom meth

Viperous fangs: development and evolution of the venom canal.

Fangs are specialised long teeth that contain either a superficial groove (Gila monster, Beaded lizard, some colubrid snakes), along which the venom runs, or an enclosed canal (viperid, elapid and atractaspid), down which the venom flows inside the tooth. The fangs of viperid snakes are the most effective venom-delivery structures among vertebrates and have been the focus of scientific interests for more than 200 years. Despite this interest the questions of how the canal at the centre of the fang forms remains unresolved. Two different hypotheses have been suggested. The mainstream hypothesis

Selectivity screening of cytotoxicity evoked by viper venoms and their toxins after nanofractionation.

Cytotoxicity is a major pathological effect that can occur during snakebite envenoming. To better understand the underlying biochemical and molecular mechanisms behind snake venom-induced cytotoxicity, it is essential to use appropriate in vitro tools for bioassaying cytotoxicity evoked by snake venoms. Identifying the toxins causing cytotoxicity is also important in this regard, particularly in the context of developing more effective snakebite treatments. Cytotoxicity induced by venom toxins can result in local pathologies in snakebite victims, which can result in long-term morbidity, and is

Bite First, Bleed Later: How Philippine Trimeresurus Pit Viper Venoms Hijack Blood Clotting.

The Philippines has a high diversity of venomous snake species, but there is minimal information on their envenomation effects. This is evidenced by the small number of case reports, the poor reporting of envenomation cases, and the absence of specific antivenoms apart from one against the Philippine cobra (Naja philippinensis). This study sought to profile the action of selected Philippine pit viper venoms on blood coagulation and to investigate whether commercially available non-specific antivenoms can provide adequate protection against these venoms. Venom from the pit vipers Trimeresurus f

Comparative proteomics of geographically distinct saw-scaled viper (Echis carinatus) venoms from India.

Snakebite is a socio-economic problem in tropical countries and it is exacerbated by geographical venom variation of snakes. We investigated on venom variation in geographically distinct populations of Echis carinatus from three ecologically distinct regions: Tamil Nadu (ECVTN), Goa (ECVGO), and Rajasthan (ECVRAJ). Venom was fractionated by RP-HPLC, combined with SDS-PAGE, and subjected to tandem mass spectrometry. Toxins were identified, and their relative abundance was estimated. Using NCBI database of Echis genus, we queried the MS/MS spectra, and found 69, 38 and 38 proteins in ECVTN, ECVG

Snake Venom Metalloproteinases and Their Peptide Inhibitors from Myanmar Russell's Viper Venom.

Russell's viper bites are potentially fatal from severe bleeding, renal failure and capillary leakage. Snake venom metalloproteinases (SVMPs) are attributed to these effects. In addition to specific antivenom therapy, endogenous inhibitors from snakes are of interest in studies of new treatment modalities for neutralization of the effect of toxins. Two major snake venom metalloproteinases (SVMPs): RVV-X and Daborhagin were purified from Myanmar Russell's viper venom using a new purification strategy. Using the Next Generation Sequencing (NGS) approach to explore the Myanmar RV venom gland tran

Kinetic study of Russell's viper venom in envenomed patients.

Serum levels of Russell's viper venom in 30 patients bitten by Russell's viper were measured by an ELISA. In the initial serum samples, which were collected immediately after admission to the hospital (0.5-19 hr after the bite), venom was detected in 24 patients (80%), with levels ranging from 3 to 92 ng/ml. These levels correlated well with the patient's clinical signs. At 6-12 hr after antivenom therapy, the venom levels in most of the serum samples (21 of 24, 87%) had decreased to undetectable levels. In the remaining patients, whose serum venom levels could be detected 36-72 hr after thera

Phospholipase A2 diversity and polymorphism in European viper venoms: paradoxical molecular evolution in Viperinae.

We report the diversity and polymorphism of phospholipase A(2) (PLA(2)) transcripts from snakes belonging to nine European viper subspecies. This diversity results in the expression of a combination of six PLA(2) species--ammodytin I1, ammodytin I2, ammodytin L, ammodytoxin, vaspin A and vaspin B--with 19 known isoforms of the first five of these species. Most of the European viper venoms studied contained either a myotoxin or a neurotoxin, and all contained ammodytin I1 and ammodytin I2. There is no evidence that a given pattern of PLA(2) species constitutes a taxonomic criterion, and isoform

Alleviation of viper venom induced platelet apoptosis by crocin (Crocus sativus): implications for thrombocytopenia in viper bites.

Viper envenomations are characterized by prominent local and systemic manifestations including hematological alterations. Snake venom metalloproteinases (SVMPs) and phospholipase A2 (PLA2) plays crucial role in the pathophysiology of hemorrhage by targeting/altering the platelets function which may result in thrombocytopenia. Platelets undergo the classic events of mitochondria-mediated apoptotic pathway due to augmented endogenous reactive oxygen species (ROS) levels. The observed anticoagulant effects during viper envenomations could be due to exacerbated platelet apoptosis and thrombocytope

Immunological Cross-Reactivity and Neutralizing Efficacy of Antivenom Against Five Medically Important Iranian Viper Venoms.

BACKGROUND: Antivenom efficacy relies on the venom antigen composition and the range of antibody cross-reactivity. This research compared the biochemical profiles of five medically relevant Iranian vipers, Gloydius caucasicus, Pseudocerastes persicus, Montivipera raddei, Echis carinatus, and Macrovipera lebetina, and compared the cross-reactivity and neutralisation efficacy of laboratory-prepared monovalent. METHODS: Venom was characterised using SDS-PAGE and reverse-phase HPLC (RP-HPLC). Monovalent antisera were raised in rabbits against each species, and a separate group was immunised with

Venomics of the asp viper Vipera aspis aspis from France.

The asp viper Vipera aspis aspis is a venomous snake found in France, and despite its medical importance, the complete toxin repertoire produced is unknown. Here, we used a venomics approach to decipher the composition of its venom. Transcriptomic analysis revealed 80 venom-annotated sequences grouped into 16 gene families. Among the most represented toxins were snake venom metalloproteases (23%), phospholipases A2 (15%), serine proteases (13%), snake venom metalloprotease inhibitors (13%) and C-type lectins (12%). LC-MS of venoms revealed similar profiles regardless of the method of extractio

Severe cases of green pit viper snake venom poisoning.

Two severe cases of green pit viper bites were reported. The clinical manifestations were severe bleeding and diarrhea. The first patient had acute renal failure complicated from severe diarrhea and hypovolemia. Hematological findings showed hypofibrinogenemia and thrombocytopenia. Renal biopsy performed in the first case showed a picture compatible with that of the recovery phase of acute tubular necrosis. Green pit viper antivenine serum together with symptomatic and supportive treatment was given. Both patients recovered uneventfully.

Coagulant component in Cerastes cerastes (Egyptian sand viper) venom.

A coagulant component has been purified from Cerastes cerastes venom, using gel filtration on a Sephadex G 100 (fine) column followed by chromatography on a Bio-Rex 70 column. This compound had a proteolytic effect and could coagulate human plasma deficient in factor VIII or (VIII + IX) with the formation of a firm clot. It could also clot plasma deficient in factor X, but the clot formed was soft and not complete. The compound had no effect on platelet aggregation and was nontoxic. This compound is believed to be primarily a factor X activator, as it could replace factor VIII in hemophilic pl

Haemostasis-altering effects of Milos viper (Macrovipera schweizeri) venom.

Haemotoxicity is one of the primary symptoms of viperid envenomation, manifesting in cardiovascular and haemostatic disturbances such as hypotension, haemorrhage, and coagulopathy. Bites by the Milos viper (Macrovipera schweizeri) have been reported to induce symptoms affecting the blood system, including fibrinogenolysis, erythrocytopenia, and venom-induced consumption coagulopathy. Consistent with these reports, its venom contains a variety of haemotoxic components and has been observed to exert strong procoagulant activity on human plasma. However, a more comprehensive analysis of the effec
