The SH protein of mumps virus is a druggable pentameric viroporin.

Viral infections are on the rise and drugs targeting viral proteins are needed. Viroporins constitute a growing group of virus-encoded transmembrane oligomeric proteins that allow passage of small molecules across the membrane. Despite sparsity in viroporin structures, recent work has revealed diversity in both the number of transmembrane helices and oligomeric states. Here, we provide evidence that the small hydrophobic protein (SH) from mumps virus is a pentameric viroporin. From extensive biophysical data, a HADDOCK model of full-length SH shows its intracellular C-terminal region to form a

The citrus tristeza virus p33 protein functions as a viroporin.

Viroporins are viral proteins that form ion channels in the membranes of the host and, thus, alter the host ion homeostasis to create more favorable environments for the virus. Since the discovery of the ion channel activity of the M2 protein encoded by influenza virus A (species: Alphainfluenzavirus influenzae), many additional viral proteins have also been characterized as viroporins. However, most viroporins known thus far belong to animal viruses, while the discovery of plant virus viroporins has significantly lagged. In this work, we present evidence that the p33 protein, a membrane-assoc

Beyond the Viroporin: The Envelope Protein as a Therapeutic Target in Viral Immunomodulation.

In their comprehensive review, Shekhar et al. (Rev Med Virol. 2025; 35:e70067) provide a valuable synthesis of the viral envelope (E) protein's role in subverting innate immunity and its potential as a pharmacological target. While we commend this timely work, our letter offers a critical perspective on the translational challenges and complexities that warrant deeper consideration. We argue that the context-dependent functionality of the E protein-wherein its immunomodulatory effects vary with infection stage, cell type, and viral load-poses a significant hurdle for therapeutic intervention,

Inhibition of SARS CoV Envelope Protein by Flavonoids and Classical Viroporin Inhibitors.

Severe acute respiratory syndrome coronavirus (SARS-CoV), an enveloped single-stranded positive-sense RNA virus, is a member of the genus Betacoronavirus, family Coronaviridae. The SARS-CoV envelope protein E is a small (∼8.4 kDa) channel-forming membrane protein whose sequence is highly conserved between SARS-CoV and SARS-CoV-2. As a viroporin, it is involved in various aspects of the virus life cycle including assembly, budding, envelope formation, virus release, and inflammasome activation. Here, SARS-CoV E protein was recombinantly expressed in HEK293 cells and channel activity and the eff

Viroporin activity of the JC polyomavirus is regulated by interactions with the adaptor protein complex 3.

Viroporins, which are encoded by a wide range of animal viruses, oligomerize in host cell membranes and form hydrophilic pores that can disrupt a number of physiological properties of the cell. Little is known about the relationship between host cell proteins and viroporin activity. The human JC polyomavirus (JCV) is the causative agent of progressive multifocal leukoencephalopathy. The JCV-encoded agnoprotein, which is essential for viral replication, has been shown to act as a viroporin. Here we demonstrate that the JCV agnoprotein specifically interacts with adaptor protein complex 3 throug

The Small Cysteine-Rich Protein of Potato Mop-Top Virus Exhibits Viroporin Activity.

Potato mop-top virus (PMTV) is the causal agent of potato tuber spraing disease, which causes significant economic losses to potato production worldwide. The 3'-proximal end of PMTV genomic RNA3 encodes an 8 kDa cysteine-rich protein (8K) that is not essential for replication and movement but contributes to virus infection and symptom development. Here, we demonstrate that PMTV 8K forms endomembrane multimers, alters the membrane permeability of Escherichia coli, and possesses potassium and proton conductance activity. In addition, our data reveal that two conserved cysteine residues in the ce

Topology and biological function of enterovirus non-structural protein 2B as a member of the viroporin family.

Viroporins are a group of transmembrane proteins with low molecular weight that are encoded by many animal viruses. Generally, viroporins are composed of 50-120 amino acid residues and possess a minimum of one hydrophobic region that interacts with the lipid bilayer and leads to dispersion. Viroporins are involved in destroying the morphology of host cells and disturbing their biological functions to complete the life cycle of the virus. The 2B proteins encoded by enteroviruses, which belong to the family Picornaviridae, can form transmembrane pores by oligomerization, increase the permeabilit

The B169L protein of African swine fever virus functions as a viroporin that activates the calcium-mediated inflammasome.

African swine fever (ASF) is a highly contagious and often fatal viral disease caused by African swine fever virus (ASFV), which poses a significant economic burden on the global pig industry. ASFV infection triggers a robust production of proinflammatory cytokines, leading to severe inflammation that contributes significantly to the high mortality rate associated with ASF. However, the underlying mechanisms remain incompletely understood. Here, we identified the ASFV B169L protein (pB169L) as a viroporin that exerts dual functions in viral replication and proinflammatory responses. We demonst

Enterovirus 2B protein: From viroporin to membrane stress sensor, mechanistic principles of multi-pathway regulation.

Enterovirus 2B is a highly conserved, functionally versatile, nonstructural protein. The classic viroporin model, which restricts its role to ion channel formation, cannot account for its interactions with host factors such as Bcl-2-associated X protein, interleukin enhancer-binding factor 2, and karyopherin subunit alpha 1, nor its functional divergence among enteroviruses. We propose a novel, testable membrane stress sensor hypothesis: the transmembrane helices of 2B sense lipid composition and pH changes, and through conformational dynamics, spatiotemporally coordinate calcium signaling, ce

Beyond independent roles: spatiotemporal coupling between viral fusion proteins and viroporins in viral replication and host inflammation.

Membrane fusion and permeabilization are key membrane remodeling processes hijacked by viruses. Although fusion proteins and viroporins have long been characterized as discrete functional modules, emerging evidence supports the spatiotemporal coordination through shared regulation of ion flux, pH homeostasis, membrane tension, organelle stress, and vesicular trafficking during viral infections. These synchronized membrane events synergistically shape viral replication and host immune responses. This review establishes a fusion-ion-inflammation axis to guide mechanistic exploration and antivira

Patch-clamp studies and cell viability assays suggest a distinct site for viroporin inhibitors on the E protein of SARS-CoV-2.

BACKGROUND: SARS-CoV-2 has caused a worldwide pandemic since December 2019 and the search for pharmaceutical targets against COVID-19 remains an important challenge. Here, we studied the envelope protein E of SARS-CoV and SARS-CoV-2, a highly conserved 75-76 amino acid viroporin that is crucial for virus assembly and release. E protein channels were recombinantly expressed in HEK293 cells, a membrane-directing signal peptide ensured transfer to the plasma membrane. METHODS: Viroporin channel activity of both E proteins was investigated using patch-clamp electrophysiology in combination with a

Comparative Genomics of Viral Genomes and Identification of Three Novel Viroporin-Like Superfamilies.

INTRODUCTION: Viroporins are small multifunctional proteins that modify cellular membranes facilitating processes such as viral nucleic acid entry and the release of virions from infected cells. We are interested in studying the evolutionary relationships among these proteins, in particular their organization into families and superfamilies. METHODS: We applied a variety of computational strategies to perform comparative genomics analyses of 120 viral genomes, using the phylogenetic profile method. This allowed the identification of 12 families, organized into four functionally related groups

Biological Function and Application of Picornaviral 2B Protein: A New Target for Antiviral Drug Development.

Picornaviruses are associated with acute and chronic diseases. The clinical manifestations of infections are often mild, but infections may also lead to respiratory symptoms, gastroenteritis, myocarditis, meningitis, hepatitis, and poliomyelitis, with serious impacts on human health and economic losses in animal husbandry. Thus far, research on picornaviruses has mainly focused on structural proteins such as VP1, whereas the non-structural protein 2B, which plays vital roles in the life cycle of the viruses and exhibits a viroporin or viroporin-like activity, has been overlooked. Viroporins ar

Viroporins: emerging viral infection mechanisms and therapeutic targets.

Viroporins, virus-encoded small hydrophobic proteins, are involved in critical steps of viral infections by modulating ion homeostasis, disrupting host membrane integrity, and orchestrating key stages of the virus life cycle-from entry and replication to release. Beyond facilitating viral propagation, these pore-forming proteins exacerbate pathogenesis by disrupting cellular ion homeostasis and triggering proinflammatory responses through NLRP3 inflammasome activation. Their dual role in viral fitness and immunopathology positions viroporins as promising antiviral targets for dual-action thera

Viroporins: Structure, function, and their role in the life cycle of SARS-CoV-2.

Viroporins are indispensable for viral replication. As intracellular ion channels they disturb pH gradients of organelles and allow Ca2+ flux across ER membranes. Viroporins interact with numerous intracellular proteins and pathways and can trigger inflammatory responses. Thus, they are relevant targets in the search for antiviral drugs. Severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) underlies the world-wide pandemic of COVID-19, where an effective therapy is still lacking despite impressive progress in the development of vaccines and vaccination campaigns. Among the 29 proteins

Pathophysiological Consequences of Calcium-Conducting Viroporins.

Eukaryotic cells have evolved a myriad of ion channels, transporters, and pumps to maintain and regulate transmembrane ion gradients. As intracellular parasites, viruses also have evolved ion channel proteins, called viroporins, which disrupt normal ionic homeostasis to promote viral replication and pathogenesis. The first viral ion channel (influenza M2 protein) was confirmed only 23 years ago, and since then studies on M2 and many other viroporins have shown they serve critical functions in virus entry, replication, morphogenesis, and immune evasion. As new candidate viroporins and viroporin

Potential Viroporin Candidates From Pathogenic Viruses Using Bacteria-Based Bioassays.

Viroporins are a family of small hydrophobic proteins found in many enveloped viruses that are capable of ion transport. Building upon the ability to inhibit influenza by blocking its archetypical M2 H+ channel, as a family, viroporins may represent a viable target to curb viral infectivity. To this end, using three bacterial assays we analyzed six small hydrophobic proteins from biomedically important viruses as potential viroporin candidates. Our results indicate that Eastern equine encephalitis virus 6k, West Nile virus MgM, Dengue virus 2k, Dengue virus P1, Variola virus gp170, and Variola

The Rotavirus NSP4 Viroporin Domain is a Calcium-conducting Ion Channel.

Viroporins are small virus-encoded ion channel proteins. Most viroporins are monovalent selective cation channels, with few showing the ability to conduct divalent cations, like calcium (Ca2+). Nevertheless, some viroporins are known to disrupt host cell Ca2+ homeostasis, which is critical for virus replication and pathogenesis. Rotavirus nonstructural protein 4 (NSP4) is an endoplasmic reticulum transmembrane glycoprotein that has a viroporin domain (VPD), and NSP4 viroporin activity elevates cytosolic Ca2+ in mammalian cells. The goal of this study was to demonstrate that the NSP4 VPD forms

Activation and Inhibition of the NLRP3 Inflammasome by RNA Viruses.

Inflammation refers to the response of the immune system to viral, bacterial, and fungal infections, or other foreign particles in the body, which can involve the production of a wide array of soluble inflammatory mediators. It is important for the development of many RNA virus-infected diseases. The primary factors through which the infection becomes inflammation involve inflammasome. Inflammasomes are proteins complex that the activation is responsive to specific pathogens, host cell damage, and other environmental stimuli. Inflammasomes bring about the maturation of various pro-inflammatory

NLRP3 Inflammasome Activation by Viroporins of Animal Viruses.

Viroporins are a group of low-molecular-weight proteins containing about 50-120 amino acid residues, which are encoded by animal viruses. Viroporins are involved in several stages of the viral life cycle, including viral gene replication and assembly, as well as viral particle entry and release. Viroporins also play an important role in the regulation of antiviral innate immune responses, especially in inflammasome formation and activation, to ensure the completion of the viral life cycle.
