DNA Vaccines Against Maedi-Visna Virus.

Maedi-visna virus (MVV) is an ovine retrovirus of the Lentivirus genus, responsible for a chronic and progressive disease of sheep with a high prevalence all over the world. Therefore, measures aiming at the control of MVV infection are necessary, and the development of DNA vaccines may be the ideal approach. A DNA vaccine is an antigen-encoding bacterial plasmid designed to mimic infections safely, with ability to generate both humoral and cellular long-lasting immune responses once it is delivered to the host. Here, we describe the development and evaluation of DNA vaccines against ovine maed

Autoimmune reactivity in sheep induced by the visna retrovirus.

Sheep, either naturally or experimentally infected with visna virus, were examined for the production of autoantibodies. Elevated titres of IgM and IgG antiglobulins and rheumatoid factors as well as antibodies to ssDNA, cardiolipin or histones were found in serum of both groups of animals using ELISA techniques. IgA autoantibodies were usually absent or present in low titres. The specificity of the autoantibody reactivities was confirmed by inhibition assays, affinity chromatography, Western blotting and agglutination methods. Antiglobulin titres were slightly elevated in synovial fluids, alt

Maedi-visna virus detection in ovine third eyelids.

Maedi-visna is a systemic disease of sheep caused by a lentivirus, maedi-visna virus (MVV), which mainly affects the lungs and central nervous system but may also affect the mammary glands, joints and other tissues. The aim of the present study was to determine whether the third eyelid was affected in cases of systemic infection. Third eyelid and lung samples from sheep naturally infected with maedi were used. Total DNA was extracted from paraffin-wax-embedded tissues, and a nested polymerase chain reaction (PCR) was performed to amplify MVV proviral DNA. The samples were also tested by in-sit

Investigations of a flock heavily infected with maedi-visna virus.

Following the discovery that a flock of sheep in England was infected with the virus of maedi-virus, several seropositive sheep were brought to the Central Veterinary Laboratory and kept isolated and under observation for up to three years before being further examined at necropsy. Meanwhile, sheep in the infected flock which died or were culled were examined after death for evidence of the actual disease. At necropsy pulmonary disease was a common (although not always the sole) finding, the lesions being mostly chronic pasteurellosis and pulmonary adenomatosis. One of the 45 carcases examined

[Studies on the epizootiology of maedi/visna in sheep (author's transl)].

The transmission of maedi/visna from a severely infected flock to their progeny was studied. At birth, the lambs were divided at random in four groups of approximately forty animals each, which were exposed to the parent flock for zero hours, ten hours, six weeks and one year respectively. Although more than 80 percent of the ewes were affected with maedi, lambs which were separated from the ewes immediately after birth continued to be free from maedi/visna virus infection during an eight years period of observation. In addition, antibodies to the virus were not detected in this group. It is t

Association of Maedi Visna virus with Brucella ovis infection in rams.

Maedi Visna Virus (MVV) is the etiological agent of a systemic disease of sheep, which causes lesions in lungs, the central nervous system, joints, and mammary glands. It has been speculated that the association with Brucella ovis may lead to the venereal shedding of the virus. In this work, samples of epididymis from ten rams positive for MVV and infected experimentally with Brucella ovis, were subjected to liquid-phase PCR, immunohistochemistry (IHC) and in situ PCR tests, aimed at identifying the pathogens in a tissue context. IHC was carried out using a monoclonal antibody raised against p

Cellular immune response in rabbits immunized with purified Visna virus.

Visna virus-sensitized lymphocytes were demonstrated in the peripheral blood of rabbits following immunization with purified, concentrated visna virus. Lymphocytes sensitization was maximal at about 2 weeks and fell to control levels in 5 weeks. A delayed-type cutaneous hypersensitivity to the virus was observed at 2 weeks and lasted for at last 9 weeks. Both reactions increased after a booster injection. Although sera from immunized rabbits had precipitating antibodies against visna virus protein P25, their lymphocytes were not stimulated by P25. However, there was a slight reaction to P25 in

Pathogenesis of visna. I. Sequential virologic, serologic, and pathologic studies.

A total of 56 Icelandic sheep were infected with visna virus by intracerebral injection of strain 1514 and the course of infection was followed for 12 months. Virus was isolated from more than 90 per cent of the animals, primarily from central nervous system and lymphoid tissues. However, titers of free infectious virus were minimal and virus isolation often required the use of tissue explants. All sheep raised serum-neutralizing and complement-fixing antibodies beginning 1 to 3 months after infection. Differences in neutralization titers against the infecting strain (1514) and a reference str

Detection of maedi-visna virus in the kidneys of naturally infected sheep.

Infections with maedi-visna virus (MVV) cause progressive inflammation in different organs, mainly the lung, mammary gland, brain and joints. The aim of the present study was to investigate whether the kidney represents a viral target in natural MVV infection. For this, kidney samples from 13 sheep naturally infected with MVV were examined by histology, polymerase chain reaction (PCR), and immunohistochemistry. The kidneys of nine animals showed membranoproliferative glomerulonephritis and interstitial nephritis. The inflammatory infiltrate consisted of lymphocytes, plasma cells and macrophage

Lentivirus-host interactions: lessons from visna and caprine arthritis-encephalitis viruses.

The biological properties of the ruminant animal lentiviruses, visna and caprine arthritis-encephalitis viruses, closely resemble those of their human counterparts, the human immunodeficiency viruses (HIV). All of these viruses are morphologically identical and are disseminated from host to host in nature during exchange of body fluids. Artificial conditions that favor excess exchange of such fluids precipitate epidemics by these viruses. The strategy of replication of the animal viruses in tissue culture and in vivo are very similar to that of the human virus. Virus replication is highly prod

Clinical and pathological investigation of Maedi-visna outbreak in a farm from Bulgaria.

BACKGROUND: In the autumn of 2024, an outbreak of Maedi-visna (MV) was identified in Bulgaria in the village of Sigmen, Burgas district. The disease is a slowly progressive, persistent, usually subclinical viral infection in sheep, distributed worldwide. Difficulties in early diagnosis are the long period between infection and the dynamics of the antibody titer (seroconversion), the emission of an infectious onset without clinical signs, the lack of vaccines, as well as some ongoing misunderstandings in the epidemiology and the transmission mechanisms, which determine the complex nature of the

Experimental infection of sheep with visna/maedi virus via the conjunctival space.

Experiments were performed to determine whether visna/maedi virus (VMV), a small ruminant lentivirus (SRLV), could infect sheep via ocular tissues. The EV1 strain of VMV was administered into the conjunctival space of uninfected sheep, and the animals monitored for the presence of provirus DNA and anti-VMV antibodies in blood. The results showed that provirus DNA appeared in peripheral blood mononuclear cells of all animals within a few weeks of receiving either 10(6) TCID50 or 10(3) TCID50 of VMV. Of the animals receiving the higher dose of virus via the conjunctival space, two seroconverted

Pulmonary adenomatosis and maedi-visna in Ethiopian central highland sheep: a microscopic study.

Microscopic examination of pneumonic lungs of the Ethiopian highland sheep (n = 35) was made and compared with the pneumonic lungs from ten sheep and 66 goats from the lowlands. Lesions compatible with sheep pulmonary adenomatosis (SPA; 8/35, 22.8%), and maedi-visna (MV; 9/35, 25.7%) were recorded only in sheep from the central highlands. Interstitial pneumonia (43.2%), bronchopneumonia (35.1%), and verminous pneumonia (6.3%) were recorded in both sheep and goats from the high- and the lowlands. SPA was documented for the first time in sheep from Ethiopia in this report. We believe that MV and

[Indurative mastitis in a herd of Dorper sheep caused by an infection with Maedi Visna virus].

This case report describes indurative mastitis in a herd of sheep caused by Maedi Visna virus (MVV) infection. Reduced udder formation after delivery, small, indurated udders and increased losses of lambs were observed in a herd of Dorper sheep. Examination of the mammary gland and milk did not reveal findings characteristic of chronic bacterial mastitis. The protein supply was insufficient which may have contributed to reduced milk yield, but was considered unlikely as cause for the induration of the mammary gland. Nineteen of the 21 mothers were positive for MVV by serology. Mammary gland an

Study of compartmentalization in the visna clinical form of small ruminant lentivirus infection in sheep.

BACKGROUND: A central nervous system (CNS) disease outbreak caused by small ruminant lentiviruses (SRLV) has triggered interest in Spain due to the rapid onset of clinical signs and relevant production losses. In a previous study on this outbreak, the role of LTR in tropism was unclear and env encoded sequences, likely involved in tropism, were not investigated. This study aimed to analyze heterogeneity of SRLV Env regions--TM amino terminal and SU V4, C4 and V5 segments--in order to assess virus compartmentalization in CNS. RESULTS: Eight Visna (neurologically) affected sheep of the outbreak

Ovine aortic smooth muscle cells allow the replication of visna-maedi virus in vitro.

Visna-maedi virus induces in sheep an interstitial lung disease characterised by an accumulation of smooth muscle cells (SMC) or myomatosis. Infection by HIV-1 has been recently associated with disorders of the vessel-derived cells: primary pulmonary hypertension, coronary artery disease and smooth muscle tumors in humans. We hypothesized that, besides their regular targets (i.e. macrophages and lymphocytes), lentiviruses could infect smooth muscle cells. Smooth muscle cell cultures derived from ovine aorta were infected with visna-maedi virus strain K1514. The cultured cells were smooth muscl

Immune reactions and immunopathologic changes induced in ovine fetuses and lambs by maedi-visna virus.

Seventy- to 80-day fetuses of Merino ewes were inoculated intramuscularly in utero and 2-week-old lambs of the same breed intratracheally with 10(6.3) TCID50/0.1 ml of maedi-visna virus strain K1512 isolated in Iceland. While no precipitins appeared in the serum of fetuses, such antibodies were demonstrable in the lambs from postinoculation (PI) day 30. Indirect immunofluorescence (IIF) revealed the presence of antibodies in samples from both fetuses and lambs; the detectability of these antibodies, however, varied even within a given animal during the experiment. The serologic results were in
