Sex-specific differences influence aging and disease onset. Using Caenorhabditis elegans , we previously found that hermaphroditic nematodes activate a robust mitochondrial unfolded protein response (UPR mt ) in the intestines, while female nematodes do not. We hypothesized that the accumulation of vitellogenins in females was suppressing the UPR mt . Loss of vitellogenins via RNAi enhanced the UPR mt in hermaphrodites but had no impact on the UPR mt of females unless a combination of vitellogenins was simultaneously knocked down. Furthermore, combinatorial loss of vitellogenins did not restor

In vertebrates, the provision of nutrients to developing embryos varies widely, ranging from yolk-dependent strategies to highly specialized forms of placental nourishment. Vitellogenins (VTGs) are essential proteins for egg yolk formation in oviparous and lecithotrophic species. In contrast, in eutherian mammals, the loss of VTGs is associated with the evolution of matrotrophy (placentotrophy and lactation), where maternal nutrition via the placenta replaces the need for large yolk reserves during embryonic development. BACKGROUND: Reproduction is a critical process in the insect life cycle, with the ovary serving as the central organ responsible for population maintenance. Successful development of the ovary is dependent on vitellogenin (Vg) transport into oocytes via the vitellogenin receptor (VgR). Exploring the VgR function is crucial for understanding the physiological mechanisms of insect ovarian development. However, the functional role of VgR in Bactrocera dorsalis (Hendel), a notorious agricultural invasive pest with exceptional reproductive plasticity, remains unclear.
RESULTS: Here, we identified

BACKGROUND: Culex quinquefasciatus, a cosmopolitan, domestic, and highly anthropophilic mosquito, is a vector of pathogenic arboviruses such as West Nile virus and Rift Valley virus, as well as lymphatic filariasis. The current knowledge on its reproductive physiology regarding vitellogenin expression in different tissues is still limited.
OBJECTIVES: In this study, we analysed the transcriptional profiles of vitellogenin genes in the fat body and ovaries of C. quinquefasciatus females during the first gonotrophic cycle.
METHODS: C. quinquefasciatus ovaries and/or fat bodies were dissected i

The salmon louse Lepeophtheirus salmonis Krøyer affects a variety of wild salmonoid hosts, but is also an important pest in aquaculture, which is a globally important and rapidly growing industry. Salmon lice have large reproductive outputs, and knowledge of reproductive processes may be crucial for the control of this parasite. Here, we report on the characterisation of 2 vitellogenins (LsVit1 and LsVit2), which are the precursors of salmon-louse egg-yolk glycoprotein. The structure of LsVit1 and LsVit2 was examined and compared to that in other oviparous animals. Phylogenetic analysis of LsV

UNLABELLED: In environmental science, omics-based approaches are widely used for the identification of gene products related to stress response. However, when dealing with non-model species, functional prediction of genes is challenging. Indeed, functional predictions are often obtained by sequence similarity searches and functional data from phylogenetically distant organisms, which can lead to inaccurate predictions due to quite different evolutionary scenarios. In oviparous females, vitellogenin production is vital for embryonic development, ensuring population viability. Its abnormal prese

Ooencyrtus kuvanae is an egg parasitoid species that attacks the egg masses of Dendrolimus spp. and Lymantria spp. in China, which gives it a temporal niche advantage against pest infestations by Dendrolimus spp. and Lymantria spp. Moreover, it has a short life cycle, high offspring outcome, and female-biased population, showing distinctive ovarian development and oviposition behaviors and thus providing an ideal model for reproductivity and regulatory mechanisms. Previous studies have found that the Vitellogenin (Vg) and Vitellogenin Receptor (VgR) genes play important regulatory roles in the

Nutritional effects on trait inheritance are widespread in animals including human famines, representing some of the most severe environmental influences on organismal phenotypes and can be transmitted over generations. However, the chemical nature of the stimuli inducing such memory remains elusive. The nematode Pristionchus pacificus exhibits mouth-form plasticity including predation and responds to a multigenerational Novosphingobium diet with the induction and transgenerational inheritance of the predatory morph. We show that bacteria-derived vitamin B12 is necessary and sufficient for tra

The induction of vitellogenin in oviparous vertebrates has become the gold standard biomarker of exposure to estrogenic chemicals in the environment. This biomarker of estrogen exposure also has been used in arthropods, however, little is known of the factors that regulate the expression of vitellogenin in these organisms. We investigated changes in accumulation of mRNA products of the vitellogenin gene Vtg2 in daphnids (Daphnia magna) exposed to a diverse array of chemicals. We further evaluated the involvement of hormonal factors in the regulation of vitellogenin expression that may be targe

A highly sensitive and specific chemiluminescent immunoassay (CLIA) was developed for quantification of vitellogenin (Vg) in five salmonids. The CLIA for salmon Vg was performed using the two-site method, with anti-masu salmon beta'-component as primary antibody and chemiluminescent acridinium-labeled anti-rainbow trout lipovitellin F(ab)'(2) as the second antibody. Using cutthroat trout Vg as the standard, the working range of the CLIA was from 60 pg to 500 ng Vg/ml. Intra- and inter-assay coefficients of variation ranged from 3.04 to 6.67% and 3.23 to 5.86%, respectively. For the various sal

The vitellogenin receptor (VgR), which belongs to the low-density lipoprotein receptors (LDLR) family, regulates the absorption of yolk protein accumulated in developing oocytes during oogenesis. In the present study, the full sequence of Rhipicephalus microplus VgR (RmVgR) and the partial sequence of Rhipicephalus appendiculatus VgR (RaVgR) ORF were determined and cloned. The RmVgR amino acid sequence contains the five highly conserved structural motifs characteristic of LDLR superfamily members, the same overall structure as observed in other species. Phylogenetic analysis separated VgRs in

Involvement of androgens in vitellogenin (VTG) synthesis was investigated using the primary hepatocyte cultures of tilapia, Oreochromis mossambicus. Concentration of VTG in the medium was assessed by enzyme-linked immunosorbent assay. When the hepatocytes of females were treated with testosterone (T), 17 alpha-methyltestosterone (MT) and 5 alpha-dihydrotestosterone (DHT), VTG concentration in the medium slightly increased or maintained. DHT, but not T and MT, increased VTG in the medium of male hepatocyte cultures. However, VTG production in the male hepatocytes, which were previously treated

In teleosts, vitellogenin (Vtg) is a phospholipoglycoprotein synthesized by the liver, released into the blood circulation and incorporated into the oocytes via endocytosis mediated by the Vtg receptor (VTGR) to form the yolk granules. The VTGR is crucial for oocyte growth in egg-laying animals but is also present in non-oviparous vertebrates, such as human. The VTGR belongs to the low-density lipoprotein receptor superfamily (LDLR) and is also named very-low-density lipoprotein receptor (VLDLR). In this study, we identified and phylogenetically positioned the VTGR of a basal teleost, the Euro

In the last years, the hormonal balance is threatened by the interferences of substances with hormone-like action (endocrine disruptor chemicals, EDCs) that may harm animal reproduction. Most EDCs are resistant to environmental degradation and are considered ubiquitous contaminants. EDCs may have synthetic or natural origins. Pesticides used in intensive agriculture contain large amounts of chemicals with estrogenic properties, such as the alkylphenol nonylphenol (NP). Besides, animal feeding operations are important sources of natural estrogen metabolites introduced into the environment throu

Vitellogenin induction has been widely used as a biomarker of endocrine disruption in wildlife, but few studies have investigated its use in wild reptiles living in contaminated habitats. This study examined vitellogenin induction in Morelet's crocodiles (Crocodylus moreletii) from wetlands in northern Belize contaminated with organochlorine (OC) pesticides. Vitellogenin was measured in 381 crocodile plasma samples using a vitellogenin ELISA previously developed for this species. Vitellogenin was detected in nine samples, all from adult females sampled during the breeding season. Males and juv

We describe here an in vitro technique to assess the estrogenic activity of chemicals. This technique is based on rainbow trout hepatocytes incubated in a basic medium free of any additional growth factors or estrogenic chemicals and uses the production of vitellogenin (VTG) as a marker for the estrogenic potency of the compounds tested. The system allows at least some of the metabolic transformations which are undertaken by the liver cells in vivo and could therefore be used for xenobiotic compounds which exhibit estrogenic activities after liver metabolic transformation. BACKGROUND: The western flower thrips Frankliniella occidentalis, a worldwide agricultural pest, has developed resistance to an array of insecticides. Spinosad resistance confers an apparent fitness cost in F. occidentalis. In the present study, we compared the reproductive capacities, ovary development, and the expression of the vitellogenin (Vg) gene in spinosad-susceptible (Ivf03) and -resistant (NIL-R) near isogenetic lines of F. occidentalis in order to clarify the reason for the fitness cost in spinosad resistance.
RESULTS: The NIL-R strain exhibited a 17.9% decrease in fecundity (eggs

Insects can survive in nutrient-poor environments owing to nutritional symbionts that produce vitamins and essential amino acids (EAAs). Nonetheless, how symbionts actively benefit from this nutritional symbiosis remains incompletely understood. Horizontally transferred genes (HTGs) expressed in bacteriocytes can function autonomously or cooperatively with symbionts to biosynthesize EAAs or B vitamins. We previously demonstrated that the horizontally transferred panBC and the symbiont Portiera cooperatively synthesize vitamin B5 (pantothenate), thereby enhancing whitefly fecundity. Beyond supp
