Plastic changes in the peri-infarct cortex are thought to play a key role in post-stroke functional recovery, yet the circuit-level mechanisms underlying such reorganization remain incompletely understood. In this study, we investigated chronic changes in perilesional cortical excitability and synaptic organization using a mouse model of middle cerebral artery occlusion (MCAO). Evoked cortical activity was assessed by voltage-sensitive dye (VSD) imaging following direct cortical stimulation. The ischemic hemisphere exhibited significantly greater response amplitude and broader spatial spread t

The insular cortex (IC) is a pain-related brain region that receives various types of sensory input and processes the emotional aspects of pain. The present study was conducted to investigate spatiotemporal patterns related to neuroplastic changes in the IC after nerve injury using voltage-sensitive dye imaging. The tibial and sural nerves of rats were injured under pentobarbital anesthesia. To observe optical signals in the IC, rats were re-anesthetized with urethane 7days after injury, and a craniectomy was performed to allow for optical imaging. Optical signals of the IC were elicited by pe

We have developed a high pressure optical cell to study large-scale transient neuronal coalitions--"assemblies" using voltage sensitive dye (VSD) fluorescence combined with fast CCD imaging of brain slices under hyperbaric conditions. The new cell has been tested at pressures up to P = 150-200 atm, corresponding to the range over which effects such as "pressure-reversal" of anaesthesia have been described previously. Brain slices were maintained in a flow of artificial cerebrospinal fluid (aCSF) and hyperbaric conditions were controlled to within +/- 0.2 atm using a back-pressure regulator pla

To obtain a more complete description of individual neurons, it is necessary to complement electrical measurements with technologies such as voltage imaging with intracellular dyes, which permit massive parallel recording from many sites on neuronal processes. Utilizing such an approach, we investigate the functional structure of the mitral cell, the principal output neuron in the rat olfactory bulb. These experiments were designed to determine the number, location, and the stability of spike trigger zones, the excitability of terminal dendritic branches, the pattern and nature of spike initia

Dysfunctions of neuronal and network excitability have emerged as common features in disorders associated with intellectual disabilities, autism, and seizure activity, all common clinical manifestations of Rett syndrome (RTT), a neurodevelopmental disorder caused by loss-of-function mutations in the transcriptional regulator methyl-CpG-binding protein 2 (MeCP2). Here, we evaluated the consequences of Mecp2 mutation on hippocampal network excitability, as well as synapse structure and function using a combination of imaging and electrophysiological approaches in acute slices. Imaging the amplit

In early visual cortex different stimulus parameters are represented in overlaid feature maps. Such functioning was extensively explored by the use of drifting gratings characterized by orientation, spatial-temporal frequency, and direction of motion. However surprisingly, the direct cortical visuotopic drift of the gratings' stripy pattern has never been detected simultaneously to these stationary feature maps. It therefore remains to be demonstrated how physical signals of grating motion across the cortex are represented independently of other parametric maps and thus, how multi-dimensional

Slow changes in [Ca(2+)](i) reflect increased neuronal activity. Our study demonstrates that single-trial fast [Ca(2+)](i) imaging (≥200 Hz sampling rate) revealed peaks each of which are associated with single spike discharge recorded by consecutive voltage-sensitive dye (VSD) imaging in enteric neurones and nerve fibres. Fast [Ca(2+)](i) imaging also revealed subthreshold fast excitatory postsynaptic potentials. Nicotine-evoked [Ca(2+)](i) peaks were reduced by -conotoxin and blocked by ruthenium red or tetrodotoxin. Fast [Ca(2+)](i) imaging can be used to directly record single action poten

1. In this paper we describe properties of a video imaging system used to acquire voltage-sensitive dye fluorescence signals from the salamander olfactory bulb. Sources of noise in these signals were evaluated in preparations stained with the potentiometric probe RH-414. These were compared with noise levels in signals obtained from a light-emitting diode array designed to stimulate the experimental conditions with light levels similar to those seen in the salamander bulb recordings. These experiments define a number of determinants of video image quality to standardize optical voltage measure

Multiple-site optical recordings with NK2761, a voltage-sensitive absorption dye, were applied to the embryonic chick olfactory system, and the functional development of olfactory nerve (N.I)-related neural circuits was examined in the forebrain. The stimulation of the N. I elicited neural responses in N.I-olfactory bulb (OB)-forebrain preparations at the embryonic 8-12 day (E8-E12) stages. At the E11 stage, we functionally identified two circuits projecting from the OB to the forebrain. The first circuit passed through the ventral side of the forebrain and spread in the dorso-caudal direction

Despite current progress achieved in the surgical technique of radical prostatectomy, post-operative complications such as erectile dysfunction and urinary incontinence persist at high incidence rates. In this paper, we present a methodology for functional intra-operative localization of the cavernous nerve (CN) network for nerve-sparing radical prostatectomy using near-infrared cyanine voltage-sensitive dye (VSD) imaging, which visualizes membrane potential variations in the CN and its branches (CNB) in real time. As a proof-of-concept experiment, we demonstrate a functioning complex nerve ne

BACKGROUND: The nucleus basalis magnocellularis/Meynert (NBM) has been explored as a new target for deep brain stimulation for neurological disorders. Although anatomical studies suggest the existence of cholinergic topographical projections of the NBM, it is still unknown whether NBM subregions differentially activate the frontal cortex.
OBJECTIVE: To investigate the topography between the NBM and frontal cortex.
METHODS: Electrical stimulation was applied to the anterior and posterior sites of the NBM in rats, and the evoked frontal activity was investigated using voltage-sensitive dye (VS

In a previous study, we applied a multiple-site optical recording technique with a voltage-sensitive dye to the embryonic chick olfactory system and showed that functional synaptic transmission in the olfactory bulb was expressed at embryonic 6-7-day stages. It is known that oscillations, i.e. stereotyped sinusoidal neural activity, appear in the olfactory system of various species. The focus of the present study is to determine whether the oscillation is also generated in the embryonic chick olfactory bulb and, if this is the case, when the oscillation appears and how its profiles change duri

Fast, multiple-site optical recording and video imaging techniques were combined to visualize the olfactory processing stream as it flowed through rat olfactory bulb slices stained with the voltage-sensitive dye RH155. A 464 element photodiode detector array was used to record the voltage-sensitive dye signals. Focal electrical stimulation of the olfactory nerve layer evoked relatively large optical responses in the olfactory nerve and glomerular layers but only small responses within the external plexiform layer. With paired-pulse stimulation, glomerular attenuation was evident in signals rec

Exposure to environmental chemicals such as bisphenol A (BPA) may affect neural circuit function in subtle and spatially distributed ways that are difficult to attribute to a single circuit element. Here, we developed a voltage-sensitive dye (VSD) imaging analysis pipeline for acute male mouse hippocampal slices. This approach measures stimulus-evoked fractional changes in VSD fluorescence (ΔF/F 0) to generate pixel-wise normalized maps, averages response magnitude within anatomically defined regions of interest (ROIs), and visualizes control-versus-compound differences using pixel-wise statis

Previously, we discovered a novel type of depolarization wave in the embryonic chick brain by using a multiple-site optical recording technique with a fast voltage-sensitive dye. This depolarization wave traveled widely over almost all the region of the CNS. This profile has raised the possibility that the depolarization wave plays some global roles in development of the CNS, rather than contributing to a specific neuronal circuit formation. To obtain more information concerning this issue, in the present study, we examined whether the depolarization wave was triggered by various types of peri

In vivo calcium imaging is an incredibly powerful technique that provides simultaneous information on fast neuronal events, such as action potentials and subthreshold synaptic activity, as well as slower events that occur in the glia and surrounding neuropil. Bulk-loading methods that involve multiple injections can be used for single-cell as well as wide-field imaging studies. However, multiple injections result in inhomogeneous loading as well as multiple sites of potential cortical injury. We used convection-enhanced delivery to create smooth, continuous loading of a large area of the corti

BACKGROUND: The antiepileptic drugs carbamazepine and gabapentin are effective in treating neuropathic pain and trigeminal neuralgia. In the present study, to analyze the effects of carbamazepine and gabapentin on neuronal excitation in the spinal trigeminal subnucleus caudalis (Sp5c) in the medulla oblongata, we recorded temporal changes in nociceptive afferent activity in the Sp5c of trigeminal nerve-attached brainstem slices of neonatal rats using a voltage-sensitive dye imaging technique.
RESULTS: Electrical stimulation of the trigeminal nerve rootlet evoked changes in the fluorescence in

Cortical spreading depression (i.e. waves of cellular depolarization, CSD) causes the aura symptoms in classical migraine, and may contribute to delayed cellular damage after an ischemic or traumatic insult to the brain. In the latter cases, secondary neuronal injury may be worsened by some of the cerebral blood flow (CBF) changes that are associated with CSD. Here, we describe a new method for the simultaneous, live imaging of local cellular depolarization and CBF changes (i.e. two variables with well-defined and important biological significance), through a closed cranial window prepared in

1. Video imaging of changes in voltage-sensitive dye (VSD) fluorescence was used to analyze spatial and temporal properties of activity patterns in the in vivo salamander olfactory bulb and primordium piriform cortex after electric stimulation. Distribution of activity among and within the neuronal layers was analyzed after orthodromic stimulation of the whole olfactory nerve (ON), isolated fascicles, or local epithelial sites, and after antidromic stimulation of the medial olfactory tract (OT). 2. Optical signals propagated through the bulbar layers with a sequence that correlates with electr

Hybrid voltage indicators (HVIs) are chemogenetic sensors that combines the superior photophysical properties of organic dyes and the genetic targetability of protein sensors to report transient membrane voltage changes. They exhibit boosted sensitivity in excitable cells such as neurons and cardiomyocytes. However, the voltage signals recorded during long-term imaging are severely diminished or distorted due to phototoxicity and photobleaching issues. To capture stable electrophysiological activities over a long time, we employ cyanine dyes conjugated with a cyclooctatetraene (COT) molecule a
