Volvariella volvacea Polypeptide Mitigates Alcohol-Induced Liver Injury: A Multi-Omics Study.

This study investigated the hepatoprotective mechanisms of Volvariella volvacea fruiting body polypeptide (VVFP, 1-3 kDa) against acute alcohol-induced liver injury using multi-omics approaches. Male ICR mice pretreated with VVFP (100-400 mg/kg) showed significantly prolonged alcohol tolerance latency (p < 0.05) and accelerated sobriety recovery compared to controls. Integrated transcriptomics and metabolomics revealed VVFP's dual regulatory effects: (1) transcriptional regulation of 36 endoplasmic reticulum stress genes (e.g., ERP57, Derl) through protein processing pathways (KEGG:04141), and

Taste properties and thermal stability during processing of Volvariella volvacea Maillard peptides.

V. volvacea is an easy-to-cultivate short-cycle edible mushroom that is rich in protein and high-quality peptides. The V. volvacea peptides, hydrolyzed by alcalase, showed an umami response value of 15.51 ± 0.11 and a salty response value of 4.64 ± 0.27 after the Maillard reaction at 120 °C via electronic tongue. The fluorescence intensity initially increased and subsequently decreased as reaction temperatures rose, and a substantial formation of browning products was observed beyond 120 °C, resulting in a diminished taste perception. High temperatures enhanced the degradation of Maillard inte

Volvariella volvacea polysaccharides alleviate acute liver injury by modulating Nrf2/NF-κB pathway in a dual manner.

Volvariella volvacea is an edible and medicinal fungus; however, its hepatoprotective effects have not been reported. In this study, we evaluated the hepatoprotective effects of V. volvacea polysaccharides (SMP) by constructing a cellular model of H2O2-induced oxidative damage and an animal model of tetrachloromethane (CCl4)-induced acute liver injury in mice. Moreover, we used ML385, a specific Nrf2 blocker, to explore how SMP might regulate the Nrf2 signaling pathway. SMP is a primarily glucose-based polysaccharide with antioxidant capacities. In vitro analyses revealed that the administrati

Exploring the salty peptides of enzymatically hydrolyzed Volvariella volvacea protein: Structural analysis and taste perception insights.

This study aimed to prepare and screen salty peptides and elucidate the factors triggering their saltiness properties and saltiness-enhancing effects. Electronic tongue results indicated that samples treated with Alcalase (AJ) exhibited the highest saltiness intensity (5.05 ± 0.11) and greater saltiness-enhancing compared to those treated with Flavourzyme (AF), Neutrase (AZ), and Papain (AM). Peptides identified by LC-MS/MS were docked to the TMC4 receptor, resulting in 23 peptides with good docked results. The key amino acid residues (Thr431, Arg433, Phe440, Phe432, and Ser273) were discovere

Comprehensive transcriptome and metabolome analyses reveal dynamic changes in quality and anti-stress strategy of Volvariella volvacea during storage.

Volvariella volvacea is perishable and loses marketability within two days after harvest. Here, we compared V. volvacea stored for 0, 1, 2 days (T0, T1, T2) using transcriptomic and metabolomic analyses. In the three comparisons (T1 vs T0, T2 vs T1, T2 vs T0), 1846, 1361, 3599 differentially expressed genes (DEGs) and 763, 797, 1297 different abundant metabolites (DAMs) were identified, respectively. These DEGs and DAMs dynamically regulated postharvest quality of V. volvacea via activating the pentose phosphate pathway, pentose and glucuronate interconversions, glyoxylate and dicarboxylate me

A bio-nanocomposite hydrogel based on β-cyclodextrin and straw mushroom (Volvariella volvacea) soluble polysaccharide for the delivery of capsaicin and alleviation effect on type II diabetes mellitus.

Type II diabetes mellitus (T2DM) remains a global health challenge, necessitating innovative drug delivery strategies to improve treatment efficacy. Capsaicin (CAP), a bioactive compound with promising antidiabetic potential, is limited by its poor solubility and bioavailability. To address these limitations, a hydrogel nanocomposite system (HGNCs) based on β-cyclodextrin (β-CD) and soluble polysaccharides from Volvariella volvacea (VVPS) was developed for the efficient CAP delivery. We further assessed its remission effect on T2DM. The results showed that CAP was successfully encapsulated wit

Functional domains of a pore-forming cardiotoxic protein, volvatoxin A2.

Volvatoxin A2 (VVA2), a novel pore-forming cardiotoxic protein was isolated from the mushroom Volvariella volvacea. We identified an N-terminal fragment (NTF) (1-127 residues) of VVA2 as a domain for oligomerization by limited tryptic digestion. On preincubation of NTF with VVA2, NTF was found to inhibit VVA2 hemolytic activity by inducing VVA2 oligomerization in the solution in the same manner as liposomes. By site-directed mutagenesis, the amphipathic alpha-helix B of NTF or VVA2 was shown to be indispensable for its biological functions. Interestingly, at a molar ratio of recombinant NTF (r

The Inclusion of Dietary and Medicinal Mushrooms into Translational Oncology: Pros and Cons at the Molecular Level.

Mushrooms are valued for their nutritional qualities and have been used in traditional medicine since the Neolithic era. They exhibit various bioactivities, including antioxidant, hypocholesterolemic, immunomodulatory, and anticancer effects. The anticancer effects arise via direct action on tumor cells and indirect modulation of the immune system; the latter is the predominant mechanism. Numerous studies indicate that various mushroom species are potent immunostimulants because their cell wall polysaccharides and proteoglycans are recognized by intestinal immune cells. This enhances antitumor

Clinical Manifestations of Human Exposure to Fungi.

Biological particles, along with inorganic gaseous and particulate pollutants, constitute an ever-present component of the atmosphere and surfaces. Among these particles are fungal species colonizing almost all ecosystems, including the human body. Although inoffensive to most people, fungi can be responsible for several health problems, such as allergic fungal diseases and fungal infections. Worldwide fungal disease incidence is increasing, with new emerging fungal diseases appearing yearly. Reasons for this increase are the expansion of life expectancy, the number of immunocompromised patien
