Cardiovascular studies during and following simulation and weightlessness.

Two major characteristics of space flight may be simulated to some degree by bed rest and by water immersion. These are inactivity and the lack of the hydrostatic stress on the systematic circulation. Studies prior to the advent of space flight led to the prediction of orthostatic intolerance, muscular weakness, and calcium loss. Prolonged space flight has heightened the interest in these simulations to provide information concerning the temporal cause of the "deconditioning" or disuse syndrome and the value of simulation for testing the effectiveness of remedial measures. This report will eva

Human autonomic responses to actual and simulated weightlessness.

Orthostatic dysfunction occurs after exposure to microgravity, and is not completely understood. The authors developed a device for stimulating carotid baroreceptors to test the hypothesis that exposure to microgravity leads to impairment of arterial baroreflex mechanisms. Data obtained before and after two head-down bedrest studies and before and after brief Space Shuttle missions indicate that baroreceptor-cardiac reflex control is impaired by simulated or actual weightlessness. The authors speculate that arterial baroreflex derangements combine with blood volume reductions and increased ven

Skeletal abnormalities in rats induced by simulated weightlessness.

A hypokinetic model has been developed which attempts to simulate the weightlessness experienced during space flight. Male rats (approximately 130 g) were suspended from the model with a head-down tilt for a two-week period. Total mechanical unloading of the hind limbs and partial unloading of the fore limbs occurred. In comparison to pair-fed control rats, the skeletal alterations in the proximal tibial and humeral metaphyses of suspended rats were determined to be a diminished rate of longitudinal bone growth, a reduced mass of mineralized tissue, and an accumulation of marrow fat. Also, sus

Skeletal response to simulated weightlessness: a comparison of suspension techniques.

The skeletal response to simulated weightlessness was studied in rats subjected to two different methods of suspension. Skeletal unloading of the hind limbs for a two week period was achieved by use of either a back harness or tail traction. In comparison to pair-fed control rats, back-suspended rats failed to gain weight whereas tail-suspended rats exhibited normal weight gain. Quantitative bone histomorphometry revealed marked skeletal abnormalities in the proximal tibial metaphysis of back-suspended rats. Loss of trabecular bone mass in these animals was due to a combination of depressed lo

Postural reactions of circulation and its regulation during simulated weightlessness.

The extention and intensification of space exploration the influence of weightlessness on human organism and the formation of a new level of adaptation. The studies of blood circulation is very important because of frequent occurrence of cardiovascular disorders in the middle age subjects. In connection with extension and intensification of space exploration the influence of weightlessness on human organism and the formation of a new level of adaptation mechanisms acquires a special significance. The data obtained in recently undertaken model experiments and also during space flights indicate

Cardiovascular deconditioning effects of long-term simulated weightlessness in rats.

The aim of our serial work was to investigate the cardiovascular deconditioning effect of long-term simulated weightlessness and to elucidate its mechanisms. Our research goal was established in view of the following three reasons. Firstly, even after several decades of extensive research, there still exist significant gaps in our knowledge concerning microgravity induced cardiovascular effect. Secondly, to guarantee the health and safety of astronauts in the future prolonged missions, it is important to understand the cardiovascular adaptation to long-term weightlessness. Thirdly, the reporte

Orthostatic tests after a 4-day confinement or simulated weightlessness.

Besides microgravity, inactivity is likely to play a role in the cardiovascular deconditioning after space flights and weightlessness simulations. The aim of the study was to compare the effects of a 4-day head-down bed rest (HDBR) (-6 degrees) and a 4-day confinement (C) on cardiovascular responses to orthostatic stress. Eight male subjects underwent head-up tilt (HUT) (+60 degrees) and lower-body negative pressure (LBNP) (-20, -30, -40 and -50 mmHg) before (D-1) and at the end (R1) of each situation. Blood pressure, heart rate variability (HRV) and spontaneous baroreflex slope (SBS) were det

Effects of Long-Term Simulated Weightlessness on Retinal Microcirculation and Visual Electrophysiology.

OBJECTIVE: To investigate the mechanisms of ocular injuries in astronauts due to gravity deficit by examining changes in retinal microcirculation and visual electrophysiology in macaques subjected to simulated weightlessness. METHODS: The head-down recumbency of macaques was used to simulate the movement of blood to the side of the head that occurs without microgravity. Head-down recumbency was performed with the head tilted downwards at a recommended angle of 10°. The macaques in the control group were similarly tethered to the rope but could be held in a normal position. The whole experimen

Reorganization of the mouse oocyte' cytoskeleton after cultivation under simulated weightlessness.

Female germ cells provide the structural basis for the development of a new organism, while the main molecular mechanisms of the impact of weightlessness on the cell remain unknown. The aim of this work was to determine the relative content and distribution of the main proteins of microtubules and microfilaments, to assess the relative RNA content of genes in mouse oocytes after short-term exposure to simulated microgravity, and to determine the potential for embryo development up to the 3-cell stage. Before starting the study, BALB/c mice were divided into two groups. One group received water

Structural damage to the rat eye following long-term simulated weightlessness.

To better perform space missions and develop human spaceflights, the eye health of astronauts is receiving increasing attention from researchers. In this study, we used prolonged tail suspension to simulate microgravity cephalad fluid shift in space to observe intraocular pressure (IOP) changes, retinal structure, and optic nerve damage in rats. We observed significant choroidal thickening and optic nerve demyelination lesions in the rats in each experimental group. At the cellular level, retinal ganglion cells (RGCs) survival was significantly reduced, optic nerve oligodendrocytes were reduce

Influence of Social Isolation During Prolonged Simulated Weightlessness by Hindlimb Unloading.

The hindlimb unloading (HU) model has been used extensively to simulate the cephalad fluid shift and musculoskeletal disuse observed in spaceflight with its application expanding to study immune, cardiovascular and central nervous system responses, among others. Most HU studies are performed with singly housed animals, although social isolation also can substantially impact behavior and physiology, and therefore may confound HU experimental results. Other HU variants that allow for paired housing have been developed although no systematic assessment has been made to understand the effects of s

[The pharmacological correction of hemodynamics during the simulation of the early period of adaptation to weightlessness].

Investigations of volunteered subjects in whom antiorthostatic hypokinesia was modeled by tilting the cranial end of the body by -15 degrees were to answer whether pharmacological countermeasures of the vestibular/autonomous syndrome (motion sickness, sea sickness) alter the orthostatic tolerance and cardiovascular parameters. As was stated, the drugs allow controlled correction of hemodynamic resistance to the head-down tilt. Ephedrine, phencarol, ephedrine combined with scopolamine and pipolphen, and stugeron normalized cerebral circulation. Medicaments were unable to moderate the sympathoad

Osteoblast histogenesis in periodontal ligament and tibial metaphysis during simulated weightlessness.

According to nuclear size, fibroblast-like cells adjacent to bone surfaces in the periodontal ligament (PDL) and tibial primary spongiosa (PS) were classified as less differentiated progenitors and committed osteoprogenitors (A/A'), nonosteogenic cells (B), or preosteoblasts (C/D). The ratio of A/A' to C/D cells reflects osteogenic status of bone lining tissue. When 83-day-old rats were subjected to simulated weightlessness (S-W) for 17 d and examined for changes in osteoblast histogenesis, PDL and PS cell populations increased in A/A' cells (p less than 0.01; less than 0.05) but decreased in

Effects of Simulated Weightlessness on Metabolizing Enzymes and Pharmacokinetics of Folic Acid in SD Rats.

Folic acid (FA) affect human physiology and drug metabolism. Up to now, the effect of microgravity on the pharmacokinetics of FA remains unclear. The pharmacokinetics of FA in Sprague-Dawley (SD) rats are laying a foundation for safe medicine administration of astronauts. Proteins expression of such FA metabolic enzymes as Methyltetrahydrofolate reductase (MTHFR), Cystathionine beta synthase (CBS) and Methionine synthase (MS) in a variety of organs was analyzed with Western-Blot, and mRNA expression was detected by RT-PCR. The plasma concentration-time profile of FA in normal or tail-suspended

[Can a tail-suspension model be applied to simulate the reproduction system under weightlessness?].

A tail-suspension model to simulate weightlessness is widely used for studies on physiological changes not only of the musculoskeletal system but also of the cardiovascular system. In the present study, we applied this model to investigate whether or not it is possible for male spermatogenesis to occur, and for female rats to maintain pregnancy in this model. Two male rats were suspended with the right inguinal canal ligated loosely for 14 days, and female rats were suspended after copulation (the day when spermatozoa were identified on a smear from the female rat, was referred to as Day 0 for

Compliance changes in femoral veins of rabbits after 21 days of simulated weightlessness.

Increased venous compliance in lower limbs may be contributed to postflight orthostatic intolerance; however, direct animal studies to address the changes of venous compliance to microgravity have been rare. The purpose of this study was to determine compliance changes in femoral veins of rabbits after 21 days of head-down rest. Head-down rest -20 dgrees rabbit model was used to simulate weightlessness. 24 healthy male New Zealand Rabbits were randomly divided into 21 days of head-down rest group (HDR), horizontal immobilization group (HIG) and Ctrl group (Ctrl), with 8 in each.
