Wolffian duct development.

The Wolffian ducts (WDs) are the progenitors of the epididymis, vas deferens and seminal vesicles. They form initially as nephric ducts that acquire connection to the developing testis as the mesonephros regresses. The development of the WDs is dependent on androgens. Conventionally, the active androgen is believed to be testosterone delivered locally rather than via the systemic circulation. However, recent studies in marsupials show that 5α-reduced steroids are essential and that these can induce virilisation even when they are delivered via the systemic circulation. The development of the W

Regulation of Wolffian duct development.

Wolffian ducts (WDs) are the embryonic structures that form the male internal genitalia. These ducts develop in both the male and female embryo. However, in the female they subsequently regress, whereas in the male they are stabilised by testosterone. The WDs then develop into separate but contiguous organs, the epididymis, vas deferens and seminal vesicles. Recently, considerable progress has been made in identifying genes that are involved in these different stages of development which is described in this review. In addition, WD development in (atypical forms of) cystic fibrosis and interse

Hemihaematometra with persistent undeveloped Wolffian duct.

A case of hemihaematometra in a non-communicating rudimentary horn of the uterus in a 14 year old girl is presented. Exploratory laparotomy revealed a persistent undeveloped Wolffian duct. This is the second case in world literature where a persistent mesonephric duct without any development is reported.

Wnt9b enables androgen action to maintain Wolffian ducts in mice.

The Wolffian duct (WD) is the embryonic primordium that gives rise to the epididymis, vas deferens, and seminal vesicle. The androgen action in the mesenchyme is the predominant driver for fetal WD maintenance, which is essential for male fertility. However, the androgen's capability of promoting WD maintenance was completely lost in the absence of Wnt9b in mice. In this study, we followed up with this interesting phenomenon and revealed cellular and molecular mechanisms whereby Wnt9b facilitates WD maintenance in male embryos. Wnt9b belongs to the WNT family of secreted proteins and is expres

Organ Culture and Whole Mount Immunofluorescence Staining of Mouse Wolffian Ducts.

Tubal morphogenesis is a fundamental requirement for the development of most mammalian organs, including the male reproductive system. The epididymis, an integral part of the male reproductive tract, is responsible for sperm storage, maturation, and transport. The adult epididymis is a highly coiled tube that develops from a simple and straight embryonic precursor known as Wolffian duct (WD). Proper coiling of the epididymis is essential for male fertility, as sperm in the testis are unable to fertilize an oocyte. However, the mechanism responsible for epididymal development and coiling remain

Associated anomalies of mullerian and wolffian duct structures.

Over a ten-year period, we identified 457 patients with renal or uterine anomalies. Only 12 of 55 patients with anomalies of mullerian development had an intravenous pyelogram, which revealed an associated renal anomaly in five. Conversely, only two of 21 women with renal anomalies had a gynecologic evaluation; one of them had a bicornuate uterus. Although the association between anomalies of mullerian and wolffian development has been known for some time, the appropriate studies to demonstrate such an association are not routinely done in clinical practice. All female patients with congenital

Epithelial and mesenchymal fate decisions in Wolffian duct development.

Wolffian ducts (WDs) are the paired embryonic structures that give rise to internal male reproductive tract organs. WDs are initially formed in both sexes but have sex-specific fates during sexual differentiation. Understanding WD differentiation requires insights into the process of fate decisions of epithelial and mesenchymal cells, which are tightly coordinated by endocrine, paracrine, and autocrine signals. In this review, we discuss current advances in understanding the fate-decision process of WD epithelial and mesenchymal lineages from their initial formation at the embryonic stage to p

Wolffian duct differentiation by physiological concentrations of androgen delivered systemically.

In developing mammalian males, conversion of the Wolffian ducts into the epididymides and vasa deferentia depends on androgen secretion by the testes, whereas in females these ducts remain in a vestigial form or regress. However, there is continuing uncertainty whether the androgen needs to be delivered locally, either by diffusion from the adjacent testis or, by secretion into the lumen of the duct, or whether circulating androgens maintain and virilize the Wolffian ducts. To resolve this uncertainty, we transplanted either day 0-2 or day 8-9 post-partum testes beneath the flank skin of three

Crucial Roles of the Mesenchymal Androgen Receptor in Wolffian Duct Development.

Wolffian duct (WD) maintenance and differentiation is predominantly driven by the androgen action, which is mediated by the androgen receptor (AR). It is well established that the mesenchyme indicates the fate and differentiation of epithelial cells. However, in vivo developmental requirement of mesenchymal AR in WD development is still undefined. By designing a mesenchyme-specific Ar knockout (ARcKO), we discovered that the loss of mesenchymal Ar led to the bilateral or unilateral degeneration of caudal WDs and cystic formation at the cranial WDs. Ex vivo culture of ARcKO WDs invariably resul

Development and morphogenesis of the Wolffian/epididymal duct, more twists and turns.

The epididymis serves a critical function of preparing the male germ cells for fertilization. In order for the epididymis to carry out this role it must undergo a highly coordinated succession of molecular and morphogenic events during development. These events begin with the formation of the Wolffian or nephric duct, the embryonic precursor of the male reproductive system, and end with the three-dimensional coiled postnatal epididymis that is comprised of several distinctly functional segments. How the duct changes from a simple straight tube to a highly convoluted structure will be the focus

[Malformations of male internal genitalia originating from the Wolffian duct].

This review includes the description and treatment of the various malformations of the male internal genitalia of Wolffian origin. Congenital epididymal cyst is the most frequent anomaly; it is usually asymptomatic so that surgery is seldom necessary. Agenesis of the deferent is usually discovered during surgery for undescended testis and is frequently associated with a homolateral renal agenesis. Ectopia and duplication are other possible anomalies affecting the deferent. Seminal vesicle cysts is mainly discovered after puberty; pelvic pain and/or urinary symptoms are the main revealing signs

Magnetic Resonance Imaging Evaluation of Wolffian Duct Anomalies - OHVIRA and OSVIRA.

Herlyn-Werner-Wunderlich syndrome in females, also known as obstructed haemivagina and ipsilateral renal agenesis (OHVIRA), and Zinner's syndrome, also known as OSVIRA in males, are two rare congenital syndromes affecting the urogenital tract. OHVIRA is obstructed haemivagina and ipsilateral renal agenesis and OSVIRA abbreviation stands for obstructed seminal vesicle and ipsilateral renal agenesis. For a long, these syndromes were considered different entities, owing to our poor understanding of underlying embryopathogenesis; however, in light of the recent acceptance of the Wolffian origin of

In vivo replacement of damaged bladder urothelium by Wolffian duct epithelial cells.

The bladder's remarkable regenerative capacity had been thought to derive exclusively from its own progenitors. While examining consequences of DNA methyltransferase 1 (Dnmt1) inactivation in mouse embryonic bladder epithelium, we made the surprising discovery that Wolffian duct epithelial cells can support bladder regeneration. Conditional Dnmt1 inactivation in mouse urethral and bladder epithelium triggers widespread apoptosis, depletes basal and intermediate bladder cells, and disrupts uroplakin protein expression. These events coincide with Wolffian duct epithelial cell recruitment into Dn

Intrahepatic paratesticular cyst: unique presentation of vestigial remnants of Wolffian duct.

To report on a case of a progressively enlarging intrahepatic cyst in a 1-year-old boy who also presented with bilateral undescended testes. At surgery, the lesion emerged from the anterior surface of the liver but was unexpectedly found to arise from the epididymis of the right testis, which was located intra-abdominally. The histologic findings were consistent with a paratesticular cyst of vestigial remnants derived from the Wolffian duct. Such an unusual event has not been previously reported among the possible location of a paratesticular cyst, nor has it been described in the differential

Activin A is an endogenous inhibitor of ureteric bud outgrowth from the Wolffian duct.

Development of metanephric kidney begins with ureteric bud outgrowth from the Wolffian duct (WD). GDNF is believed to be a crucial positive signal in the budding process, but the negative regulation of this process remains unclear. Here, we examined the role of activin A, a member of TGF-beta family, in bud formation using an in vitro WD culture system. When cultured with the surrounding mesonephros, WDs formed many ectopic buds in response to GDNF. While the activin signaling pathway is normally active along the non-budding WD (as measured by expression of activin A and phospho-Smad2/3), acti

Paravaginal wolffian duct (mesonephros) adenocarcinoma: a light and electron microscopic study.

This is a report of an adenocarcinoma of mesonephric origin studied by light microscopy, electron microscopy, and immunocytochemistry. Unlike previous reports, our lesion was located paravaginally and not in the leaves of the broad ligament or in the cervix. The light microscopic features are similar to those of previous cases in the literature. Although not specific, the ultrastructural features of the tumor are similar to those of mesonephric structures and different from those of müllerian structures. The diagnosis always should be considered when a tumor occurs at the site where mesonephri
