Wolinella recta has been associated with adult periodontitis, but its role in the disease remains uncertain. This report clarifies the distribution of W. recta in periodontally healthy and diseased subjects, and treated patients with recurrent disease. A specific polyclonal rabbit antiserum against W. recta strain 372 was used for indirect immunofluorescence localization of W. recta in dental plaque from untreated and treated patients. Supragingival plaque was collected from 15 periodontally healthy individuals (H), 10 adults with mild gingivitis (G1), 8 with severe gingivitis (G2) and 15 with

The hydrogenase (Hyd) isolated from the cytoplasmic membrane of Wolinella succinogenes consists of three polypeptides (HydA, HydB and HydC) and contains cytochrome b (6.4 mumol/g protein), which was reduced upon the addition of H2. The enzyme catalyzed the reduction of 2,3-dimethyl-1,4-naphthoquinone with H2, in contrast to an earlier preparation which was made up of HydA and HydB only and did not contain cytochrome b (Unden, G., Böcher, R., Knecht, J. & Kröger, A. (1982) FEBS Lett. 145, 230-234). This suggests that HydC is a cytochrome b which serves as a mediator in the electron transfer fro

Global warming is moving more and more into the public consciousness. Besides the commonly mentioned carbon dioxide and methane, nitrous oxide (N2O) is a powerful greenhouse gas in addition to its contribution to depletion of stratospheric ozone. The increasing concern about N2O emission has focused interest on underlying microbial energy-converting processes and organisms harbouring N2O reductase (NosZ), such as denitrifiers and ammonifiers of nitrate and nitrite. Here, the epsilonproteobacterial model organism Wolinella succinogenes is investigated with regard to its capacity to produce and

BACKGROUND AND OBJECTIVES: Helicobacter and Wolinella are gram-negative bacteria belonging to the Helicobacteraceae family. While Helicobacter species are well-known for their role in gastric disorders, emerging evidence suggests their presence in the oral cavity and potential involvement in periodontal diseases.
MATERIALS AND METHODS: Helicobacter and Wolinella species were investigated in 122 saliva and periodontal plaque samples from dogs and cats by DNA extraction, PCR amplification, and 16S rDNA gene identification.
RESULTS: Comparing the periodontitis group and the healthy group, a hig

We describe the first case of gastritis in a male Australian sea lion (Neophoca cinerea) in which members of the family Helicobacteraceae, particularly the genus Wolinella, were detected. The sea lion exhibited clinical signs of gastrointestinal disease, including abdominal pain, lack of appetite, and lethargy. Examination of one ileal and five gastric biopsy specimens collected over a 10-year period revealed persistent fibrosis and/or superficial focal erosion and ulceration of the lamina propria. Spiral-shaped organisms 5 to 12 microm long were observed in two of the gut biopsy specimens. Wh

Clinical isolates of agar-pitting, formate-fumarate-requiring, anaerobic gram-negative bacilli were recharacterized in consideration of the species descriptions of Bacteroides ureolyticus and the newly described B. gracilis, Campylobacter concisus, and Wolinella sp. During an 11-year period, 7.5% (101 of 1,341) of all specimens yielding anaerobes were found to contain an organism in this group. Of the 71 isolates that were available for study, 43 were B. ureolyticus, 23 were B. gracilis, and 5 were in the Wolinella-C. concisus group. The role in infection and patterns of antimicrobial suscepti

Campylobacter rectus, formerly known as Wolinella recta, is an anaerobic Gram-negative bacillus, generally recognized as an agent responsible for severe periodontitis; only two cases of extra-oral infections have been reported. The first case of septicemia with C rectus and Actinomyces odontolyticus is described in a 37-year-old farmer who suffered from severe sacroiliitis. Also presented are a review of C rectus in human pathology, and a brief review of pyogenic sacroiliitis, a rather rare disease.

Monoclonal antibodies were produced against an acid glycine extract of Helicobacter pylori ATCC 43504T. One of these appeared to be specific for H. pylori; it recognized all H. pylori isolates by an indirect immunofluorescence assay (IIF) but it did not cross-react with the other strains tested, including different species of the genera Helicobacter, Campylobacter, and Wolinella. Different strains of members of the families Enterobacteriaceae and Pseudomonadaceae or other gram-negative bacteria tested also gave negative reactions. Indirect immunofluorescence assay of antral biopsy specimens id

DNA of type strain Campylobacter pylori NCTC 11637 and 32 other strains of C. pylori recovered from gastric biopsy specimens was examined by thermal denaturation for its guanine-plus-cytosine (GC) content. The GC content of strain NCTC 11637 was 35.6 mol % (standard deviation (SD) 0.3), and the GC content of the 32 other C. pylori strains ranged from 34.1 to 37.5 mol % (average value 35.2, SD 1.0). A total of 14 type strains of other Campylobacter and Wolinella species were included in this study and the results obtained were compared with those cited in the literature.

Although not universally accepted, retrospective histological, roentgenological and microbiological studies have indicated that cross-infection can occur between infected pulps and deep periodontal pockets. This review provides examples of similarities in the microfloras of these adjacent oral sites, supporting the idea that infection spreads from one site to the other. The organisms most often involved are probably bacteroides, fusobacteria, eubacteria, spirochetes, wolinellas, selenomonas, campylobacter, and peptostreptococci. Important qualities of cross-infecting organisms may be the abili

The in vitro activities of 17 antimicrobial agents were evaluated against 46 clinical isolates of formate/fumarate-requiring anaerobic gram-negative bacilli. Strains of Bacteroides ureolyticus (23) were almost uniformly susceptible to the tested antimicrobials, whereas strains of Bacteroides gracilis (18) showed some striking resistance with penicillin active against only 67%, the cephalosporins active against 67%-89%, and clindamycin active against 67%. Although few strains of Wolinella species/C. concisus (5) were available for testing, these isolates tended to be more resistant than B. ureo

Localised osteomyelitis was diagnosed in 2 dogs and 2 cats. The disease was caused by fight wounds in 3 cases. Radiographic examination demonstrated a circumscribed zone of cortical bone lysis, sequestra and periosteal new bone. Each case was treated surgically by sequestrectomy and debridement. Infection was due mainly to anaerobic bacteria. The pathogenic bacteria isolated from the lesions of dogs were Actinomyces viscosus, Fusobacterium nucleatum and Bacteroides spp, and from the lesions in cats were Clostridium villosum , Peptostreptococcus anaerobius, Wolinella recta and Bacteroides gingi

In this article, we consider, in detail, the second half-cycle of the six-electron nitrite reduction mechanism catalyzed by cytochrome c nitrite reductase. In total, three electrons and four protons must be provided to reach the final product, ammonia, starting from the HNO intermediate. According to our results, the first event in this half-cycle is the reduction of the HNO intermediate, which is accomplished by two PCET reactions. Two isomeric radical intermediates, HNOH(•) and H2NO(•), are formed. Both intermediates are readily transformed into hydroxylamine, most likely through intramolecu

Bacteroides gracilis is a gram-negative anaerobic bacillus which requires formate and fumarate for growth; it has been implicated in periodontal disease and serious infections of the head and neck. In this study, Bacteroides ureolyticus (NTU) medium was tested for its ability to allow the growth of B. gracilis and other formate-fumarate requiring gram-negative anaerobes and to enable the recovery of these organisms from clinical specimens. All reference strains grew on NTU medium with the exception of Wolinella recta and formate-fumarate requiring organisms were isolated from 18 of 20 samples

Many studies indicate a strong association of Actinobacillus actinomycetemcomitans with localized juvenile periodontitis. Species associated with adult periodontitis include Bacteroides forsythus, Porphyromonas gingivalis, Prevotella intermedia, A. actinomycetemcomitans, and Wolinella recta. Capnocytophaga species may be important in pubertal gingivitis. An unnamed spirochete related to Treponema pallidum has been identified in acute necrotizing ulcerative gingivitis lesions. Species isolated from prepubertal periodontitis, peri-implantitis, pericoronitis, and human immunodeficiency virus ging

A total of 57 bacterial strains (26 different species) which may be isolated from subgingival plaque were tested for their in vitro susceptibility to phenoxycthanol, a commonly-used antiseptic and preservative. Ninety-five percent of the strains, including those associated with chronic inflammatory periodontal disease, were susceptible to concentrations of phenoxyethanol used topically (2% w/v). Phenoxyethanol at a concentration of 1% (w/v) was also found that to have a rapid bactericidal effect achieving a 99.9% kill in 5 minutes or less for species such as Bacterodides gingivalis, Fusobacter

Hydrogen sulfide (H2S) is a gaseous gut metabolite with disputed effects on gastrointestinal health. Monitoring H2S concentration in the gut would provide insight into its role in disease, but is complicated by sulfide's reactivity and volatility. Here we develop a transcriptional sulfide biosensor in E. coli. The sensor relies on enzymatic oxidation of sulfide catalyzed by a sulfide:quinone reductase (Sqr) to polysulfides, which bind to the repressor SqrR, triggering unbinding from the promoter and transcription of the reporter. Through promoter engineering and improving soluble SqrR expressi

Chlorhexidine is widely used as a mouth rinse in the prevention and treatment of periodontal diseases and dental caries. The purpose of the present study was to evaluate the in vitro antimicrobial effect of chlorhexidine in a controlled release delivery system. The controlled release dispenser comprised a polymeric inner core matrix containing the medicament with an outer vinyl membrane controlling the drug release. The effect on the following bacteria was studied: Actinobacillus actinomycetemcomitans, Actinomyces viscosus, Streptococcus mutans, Wolinella recta, Bacteroides gingivalis, Bactero
