Robust and efficient protocols for fertilization and early embryo care of Xenopus laevis and Xenopus tropicalis are essential for experimental success, as well as maintenance and propagation of precious animal stocks. The rapid growth of the National Xenopus Resource has required effective implementation and optimization of these protocols. Here, we discuss the procedures used at the National Xenopus Resource, which we found helpful for generation and early upkeep of Xenopus embryos and tadpoles.

Skeletal malformations in captive-bred, adult Xenopus spp., have not previously been reported. Here we describe 10 sexually mature, genetically modified laboratory frogs (6 Xenopus laevis and 4 Xenopus tropicalis) with axial skeletal abnormalities. The young adult frogs were described by veterinary staff as presenting with "hunchbacks," but were otherwise considered to be in good health. All affected frogs were genetically engineered using various techniques: transcription activator-like effector nucleases (TALEN) editing using thyroid hormone receptor α TALEN mRNA, restriction enzyme-mediated

The epithelial Na+ channel (ENaC) is modulated by various extracellular factors, including Na+, organic or inorganic cations, and serine proteases. To identify the effect of the divalent Ni2+ cation on ENaCs, we compared the Na+ permeability and amiloride kinetics of Xenopus ENaCs (xENaCs) and rat ENaCs (rENaCs) heterologously expressed in Xenopus oocytes. We found that the channel cloned from the kidney of the clawed toad Xenopus laevis [wild-type (WT) xENaC] was stimulated by external Ni2+, whereas the divalent cation inhibited the channel cloned from the rat colon (WT rENaC). The kinetics o

The embryos of the African clawed frog, Xenopus laevis, are a powerful substrate for the study of complex fundamental biological and disease mechanisms in neurobiology, physiology, molecular biology, cell biology, and developmental biology. A simple and straightforward technique for generating a large number of developmentally synchronized embryos is in vitro fertilization (IVF). IVF permits simultaneous fertilization of thousands of eggs but requires the death of the parental male, which may not be feasible if the male comes from a stock of precious animals. An alternative to euthanizing a pr

Xenopus laevis and Xenopus tropicalis embryos are widely used in developmental biology, genetics and disease modeling, and can be generated through in vitro fertilization (IVF), fertilizing with either fresh or cryogenically frozen sperm, or by natural matings. This chapter describes protocols developed and used successfully over two decades by the European Xenopus Resource Centre (EXRC) in Portsmouth, UK to consistently and reliably induce ovulation and generate embryos for both Xenopus laevis and Xenopus tropicalis. The following protocols describe the complete workflow for collecting Xenopu

Cryopreservation of Xenopus sperm offers several advantages: it reduces the number of live animals needed and maintained for in vitro fertilization, enables efficient archiving of genetically altered lines, and facilitates easy transfer between laboratories, lowering animal suffering and overall costs. Sperm freezing and in vitro fertilization using frozen sperm are simple procedures that require only standard laboratory equipment. Testes are macerated in Leibovitz L-15 medium, combined with a cryoprotectant made from egg yolk and sucrose and slowly frozen overnight in a polystyrene box at -80

Collecting eggs from adult Xenopus laevis and Xenopus tropicalis to raise healthy embryos and tadpoles is relatively simple but requires careful handling of the frog. Eggs can be fertilized through natural matings or by in vitro fertilization and examined visually. Here we review how eggs are obtained and how to recognize healthy eggs that will develop into high-quality embryos.

Wolf-Hirschhorn syndrome (WHS) is a rare developmental disorder characterized by intellectual disability and various physical malformations including craniofacial, skeletal, and cardiac defects. These phenotypes, as they involve structures that are derived from the cranial neural crest, suggest that WHS may be associated with abnormalities in neural crest cell (NCC) migration. This syndrome is linked with assorted mutations on the short arm of chromosome 4, most notably the microdeletion of a critical genomic region containing several candidate genes. However, the function of these genes durin

After an injury, adult mammals do not regenerate or repair their central nervous system in general, and their spinal cord in particular, leading to life-long disabilities. However, many aquatic species such as zebrafish, salamanders and frogs, as well as some land reptiles (turtles and lizards) have the remarkable ability to regenerate their spinal cords. In the case of anurans such as Xenopus, this is a transient ability that disappears after metamorphosis, making it an interesting model with accessible regenerative and nonregenerative stages within the same species. Here, we will summarize o

Xenopus laevis has a ZZ/ZW type sex determination system, and a PCR-based method was developed in 2007 to distinguish ZZ males from ZW females. As X. laevis can regenerate tail tissue as tadpoles, this method can be used to determine sex from DNA extracted from tadpole tail clips, which enables accurate identification of sex. Male X. laevis reach sexual maturity faster than females, so we routinely select male tadpoles for establishing transgenic lines. It may also be useful to stratify behavioral or drug testing results by biological sex. Here, I present a quick and simple multiplex PCR-based

Aeromonas hydrophila caused severe disease in a group of 50 Xenopus leavis three weeks after being transferred from their laboratory conditions. The first clinical signs observed were marked pallor, petechiae, lethargy, anorexia, and edema. The duration of the disease was about 48 days, during which 4 animals acquired the disease and 10 died. The study of the bacteriology of the skin and skeletal muscles showed at necropsy subcutaneous edema, hemorrhage, and ascitis. Aeromonas hydrophila was isolated in 14 of the 50 animals studied.

Aeromonas hydrophila caused severe disease in a group of Xenopus laevis within 3 weeks of receipt. The primary clinical signs were marked pallor of the skin, petechiation, lethargy, anorexia, and edema. The duration of the outbreak was approximately 45 days during which time 21 frogs became sick and 18 died, despite tetracycline therapy. Pale skin, subcutaneous edema and hemorrhages, ascites, and pale livers were seen at necropsy. Aeromonas hydrophila was isolated from skin, and the same organism was isolated in pure culture from skeletal muscle. Tetracycline treatment via stomach tube was eff

Gynogenesis is a form of parthenogenesis in which sperm is required to activate the egg but contributes no genetic material to the embryo. In Xenopus, gynogenetic embryos can be created by using UV-irradiated sperm in in vitro fertilization and then preventing the formation of the second polar body, so the embryo retains two maternal chromosomes. The method is very simple, relies on standard laboratory equipment, and can be used to create homozygous embryos from heterozygous female carriers without the need to raise an F1 generation for in-crosses.

Xenopus laevis (an anuran amphibian) shows limb regeneration ability between that of urodele amphibians and that of amniotes. Xenopus frogs can initiate limb regeneration but fail to form patterned limbs. Regenerated limbs mainly consist of cone-shaped cartilage without any joints or branches. These pattern defects are thought to be caused by loss of proper expressions of patterning-related genes. This study shows that hyperinnervation surgery resulted in the induction of a branching regenerate. The hyperinnervated blastema allows the identification and functional analysis of the molecules con

Craniofacial birth defects occur in 1 out of every 700 live births, but etiology is rarely known due to limited understanding of craniofacial development. To identify where signaling pathways and tissues act during patterning of the developing face, a 'face transplant' technique has been developed in embryos of the frog Xenopus laevis. A region of presumptive facial tissue (the "Extreme Anterior Domain" (EAD)) is removed from a donor embryo at tailbud stage, and transplanted to a host embryo of the same stage, from which the equivalent region has been removed. This can be used to generate a ch

We have cloned a homolog of Pax-6 in Xenopus laevis. Its deduced amino acid sequence has a 95% overall identity with Pax-6 homologs in other vertebrates. It is expressed early in development in cells fated to form the eye and parts of the forebrain, hindbrain, and spinal cord. It has two phases of expression in the eye. In the early phase, from stage 12.5 to stage 33/34, Xenopus Pax-6 is expressed throughout the developing retina. In the late phase, after stage 33/34, it is excluded from mature cells in the outer half of the retina and from cells in the ciliary marginal zone, remaining only in
