Transposons are mobile genetic elements found within the genomes of various organisms including bacteria, fungi, plants and animals. Fragments of the transposon Tn1721 were found included in the genome of Xylella fastidiosa strain 9a5c. Regions from such fragments were PCR-amplified using specially designed primers (TNP(1) and TNP(2)). In order to detect insertions of the Tn1721 element, both primers were used and one of them included a region of the transposon (TNP(1)) and the other one had the right repeat and part of the bacterial chromosome (TNP(2)). The PCR products obtained from strain 9

A simple defined solid medium containing citrate and succinate, three amino acids (L-glutamine, L-asparagine, and L-cysteine), hemin chloride, potato starch, gellan gum (GelRite), and mineral salts supported the growth of grape strains of Xylella fastidiosa, the bacterial pathogen that causes Pierce's disease of grape. Isolation efficiency from infected grape plant samples, determined by the number of colony forming units recovered, on the defined medium was slightly less ( approximately 10-fold) or indistinguishable from two standard rich media used for culturing X. fastidiosa, PWG and PD3, r

BACKGROUND: The management of Xylella fastidiosa infections relies on integrated strategies of controlling vector populations and removing inoculum sources. The impact of a subspecies pauca strain of this bacterium (XfDD) was particularly severe in olive in Apulia, Italy, where an entire economy was affected, and a territory was devastated. There is an urgent request for strategies to combat the pathogen in planta, as well as for bioinsecticides targeting its vector insect species, and that these should conform to criteria of environmental acceptance. Therefore, this study analyses the Mechani

Xylella fastidiosa (Xf) is a Gram-negative bacterium responsible for severe diseases in several commercially significant crops, including olive, grapevine, citrus and almond. Its management is particularly challenging due to its transmission via widespread vector insects, its ability to form biofilms, its high genetic diversity and, sometimes, latent symptoms. Current control strategies focus on integrated and preventive approaches, including the use of resistant varieties, agronomic practices, and vector control through chemical and biological methods. Direct control of the bacterium has alwa

The pathogenicity of Xylella fastidiosa is associated with its systematic colonization of the plant xylem, forming bacterial biofilms. Mechanisms of bacterial transport among different xylem vessels, however, are not completely understood yet, but are strongly influenced by the presence of extracellular polymeric substances (EPS), which surrounds the assembly of cells forming the biofilm. In this work, we show quantitative measurements on the elastic properties of the system composed by EPS and bacterial cell. In order to investigate the mechanical properties of this system, force spectroscopy

Strains of Xylella fastidiosa isolated from grape, almond, maple, and oleander were characterized by enterobacterial repetitive intergenic consensus sequence-, repetitive extragenic palindromic element (REP)-, and random amplified polymorphic DNA (RAPD)-PCR; contour-clamped homogeneous electric field (CHEF) gel electrophoresis; plasmid content; and sequencing of the 16S-23S rRNA spacer region. Combining methods gave greater resolution of strain groupings than any single method. Strains isolated from grape with Pierce's disease (PD) from California, Florida, and Georgia showed greater than prev

Xylella fastidiosa subsp. fastidiosa (Xff), the causal agent of Pierce's Disease (PD) in grapevine, is a major threat to global viticulture. However, the mechanisms governing infection establishment, persistence, and recovery within grapevine hosts remain insufficiently characterized. We developed a novel physiologically based infection model that explicitly describes the population dynamics and within-plant spread of Xff in grapevine plants as a function of temperature and host phenology. Grapevines are represented as structured hosts composed of two interacting compartments: permanent woody

Following a request from the European Commission, EFSA was asked to provide urgent technical assistance in the field of plant health as regards the list of host plants of Xylella fastidiosa reported in Appendix В of the recently published EFSA Scientific Opinion on X. fastidiosa. In this technical report the list of host plants is categorised focusing on plant species traded as plants for planting and taking into account available data and information on the trade and cultivation of host plants and on infection with X. fastidiosa. Based on this characterisation, a definition of the host plants

Xylella fastidiosa is a plant-pathogenic bacterium native to the Americas. It has a wide host range and causes significant diseases in economically important crops, including grapevines, citrus, and olive trees. Since 2013, this bacterium has been detected in Europe and other countries of the Mediterranean basin, leading to the identification of several subspecies (multiplex, fastidiosa, and pauca) and sequence types (STs) in various plant species in Italy, France, the Balearic Islands and mainland Spain, Portugal, Israel, and Lebanon. This study focuses on genomic analyses of the subspecies p

The outcome of infection can be seen as a race between the pathogen and the plant immune system. In this review, we use the case study of Xylella fastidiosa (Xf) and explain how the bacterium exploits its slow and fastidious growth to remain below detection thresholds, thereby delaying immune activation. Its self-limiting behavior in the sessile state provides a temporal window for transitioning into a more exploratory lifestyle, enabling systemic colonization of the host. As bacterial populations expand, vessel occlusion and immune overactivation at late stages occur, often propagating beyond

Almond leaf scorch is a disease caused by Xylella fastidiosa, which is increasingly widespread globally in the main almond cultivation areas. Previously confined to America, in the last decade this disease has been reported in Iran and southern Europe. In this comparative study, the resistance to X. fastidiosa subsp. multiplex of 13 almond cultivars (Mamaei, Non-Pareil, Sefied, Rabie, Ferragnes, Shahrood21, Thompson, Merced, Marcona, Frudeul, Kapariel, Princess, and Tuono) grafted onto seedlings of Talkhe was evaluated in controlled conditions. Plants were artificially inoculated and maintaine

Xylella fastidiosa, which causes Pierce's disease of grapevine and other important plant diseases, is a xylem-limited bacterium that depends on insect vectors for transmission. Although many studies have addressed disease symptom development and transmission of the pathogen by vectors, little is known about the bacterial mechanisms driving these processes. Recently available X. fastidiosa genomic sequences and molecular tools have provided new routes for investigation. Here, we show that a diffusible signal molecule is required for biofilm formation in the vector and for vector transmission to

High rates of homologous recombination (HR) in the bacterial plant pathogen Xylella fastidiosa have been previously detected. This study aimed to determine the extent and explore the ecological significance of HR in the genomes of recombinants experimentally generated by natural transformation and wild-type isolates. Both sets of strains displayed widespread HR and similar average size of recombined fragments consisting of random events (2-10 kb) of inter- and intrasubspecific recombination. A significantly higher proportion and greater lengths (>10 kb, maximum 31.5 kb) of recombined fragments

Copper-based compounds are widely used in agriculture as a chemical strategy to limit the spread of multiple plant diseases; however, the continuous use of this heavy metal has caused environmental damage as well as the development of copper-resistant strains. Thus, it is important to understand how the bacterial phytopathogens evolve to manage with this metal in the field. The MqsRA Toxin-Antitoxin system has been recently described for its function in biofilm formation and copper tolerance in Xylella fastidiosa, a plant-pathogen bacterium responsible for economic damage in several crops worl

Xylella fastidiosa is the causal agent of Pierce's disease in grapevines. The mechanisms of pathogenicity are largely due to occlusion of xylem vessels by aggregation of X. fastidiosa and biofilm formation. Xylella fastidiosa was subjected to xylem fluids with varying chemistries to examine the effects of nutritional components on bacterial growth in vitro. The exposure of X. fastidiosa to xylem fluids collected from different Vitis genotypes resulted in highly significant differences in both planktonic growth and biofilm formation. Planktonic growth of X. fastidiosa in Vitis xylem fluid was c

New defense elicitor peptides have been identified which control Xylella fastidiosa infections in almond. Xylella fastidiosa is a plant pathogenic bacterium that has been introduced in the European Union (EU), threatening the agricultural economy of relevant Mediterranean crops such as almond (Prunus dulcis). Plant defense elicitor peptides would be promising to manage diseases such as almond leaf scorch, but their effect on the host has not been fully studied. In this work, the response of almond plants to the defense elicitor peptide flg22-NH2 was studied in depth using RNA-seq, confirming t

Xylella fastidiosa is a global bacterial phytopathogen that colonizes the xylem vessels, leading to their occlusion and thus to plant death. The expression of the amyloidogenic, prion-like WH1 domain of the replication protein RepA from the Pseudomonas savastanoi plasmid pPS10 severely damages Escherichia coli cells. Here, we characterize the WH1 domains in the Rep proteins from the X. fastidiosa plasmids pXF51 and pXF64. The expression of these Rep-WH1 proteins, as happens with the archetypical RepA-WH1 protein from pPS10, decreased the culturability of E. coli in vitro, promoting dormancy. E

Following a request from the European Commission, EFSA was asked to create and regularly update a database of host plant species of Xylella spp. In 2018, EFSA released a new Xylella spp. host plant database that was now updated with informative data extracted from 76 recent publications retrieved through an extensive literature search. This report is related to the third version of the database published in Zenodo in the EFSA Knowledge Junction community, covering articles selected from: a systematic literature review conducted up of 30 June 2019; Europhyt database up to 15 October 2019; and r

Pierce's disease (PD), caused by Xylella fastidiosa (X. fastidiosa), has challenged grape cultivation in the US since the late nineteenth century and has stimulated extensive exploration of grapevine genetic diversity for sources of resistance. Early observations made during the first outbreaks in California, when the disease was known as "Anaheim disease", revealed marked variety-dependent differences in survival. Some varieties were proposed as potential top-grafts or rootstocks, reflecting the recognition of the heritable nature of resistance and its possible incorporation into newly develo

Xylella fastidiosa causes disease in a number of economically important crops, ornamental plants, and shade trees, including grapevine, citrus, oleander, and sycamore. In pecan, X. fastidiosa causes pecan bacterial leaf scorch (PBLS), which leads to defoliation and reduces nut yield. No economically effective treatments are available for PBLS. In order to improve PBLS management practices, it is necessary to determine the subspecies of X. fastidiosa strains that infect pecan so that potential sources of inoculum may be identified.
