Ultrastructural plasticity in the plant-parasitic nematode, Bursaphelenchus xylophilus.

Phenotypic plasticity is one of the most important strategies used by organisms with low mobility to survive in fluctuating environments. Phenotypic plasticity plays a vital role in nematodes because they have small bodies and lack wings or legs and thus, cannot move far by themselves. Bursaphelenchus xylophilus, the pathogenic nematode species that causes pine wilt disease, experiences fluctuating conditions throughout their life history; i.e., in both the phytophagous and mycetophagous phases. However, whether the functional morphology changes between the life phases of B. xylophilus remains

Nematicidal activity of marcenmicin D2 from Fusarium sp. against Bursaphelenchus xylophilus.

BACKGROUND: Pine wilt disease (PWD) caused by Bursaphelenchus xylophilus is a devastating forest disease worldwide, and there is an urgent need for environmentally friendly biocontrol agents. Cyclic lipopeptides from microbial sources exhibit diverse biological activities, but their nematicidal potential against B. xylophilus remains largely unexplored. RESULTS: A cyclic lipopeptide, marcenmicin D2, was isolated from the endophytic fungus Fusarium sp. SSY-3. Marcenmicin D2 exhibited potent nematicidal activity against B. xylophilus in a concentration- and time-dependent manner, with a median

Volatile Organic Compounds from Wolfiporia hoelen Exhibit Nematicidal Activity Against Bursaphelenchus xylophilus.

Pine wilt disease, caused by the pinewood nematode (PWN) Bursaphelenchus xylophilus, devastates pine forests worldwide. Although the medicinal fungus Wolfiporia hoelen exhibits nematicidal activity against PWN, its specific bioactive constituents remain unidentified. This study investigated the nematicidal activity of volatile organic compounds (VOCs) released from W. hoelen against PWN. Results indicated that VOCs from W. hoelen exhibit significant inhibitory efficacy against second-stage juveniles (J2s) of PWN, achieving 90% mortality after 72 h of exposure. In contrast, ovicidal activity wa

Bioinformatics analysis of the sodium-dependent neurotransmitter transporter family in Bursaphelenchus xylophilus.

Pine wilt disease (PWD), caused by Bursaphelenchus xylophilus, poses a significant threat to global pine forests. The sodium-dependent neurotransmitter transporter (SNF) plays a critical role in the reuptake of neurotransmitters from the synaptic cleft, thereby terminating neural signal transmission and maintaining nervous system homeostasis. This study aimed to comprehensively identify all members of the SNF in B. xylophilus and elucidated their roles in nematode development and nervous system function. Through genome-wide data analysis, we identified 18 SNFs in B. xylophilus and thoroughly c

Physiological Measurements and Transcriptomics Reveal the Fitness Costs of Monochamus saltuarius to Bursaphelenchus xylophilus.

The pine wood nematode (PWN) uses several Monochamus species as vehicles, through a temporary hitchhiking process known as phoresy, enabling it to access new host plant resources. Monochamus saltuarius acts as a new and major vector of the PWN in Northeastern China, showing lower PWN carrying capacity and a shorter transmission cycle compared to established vectors. The apparently altered symbiotic relationship offers an interesting area for researching the costs and adaptions involved in nematode-beetle, a specialized phoresy. We analyzed the response and fitness costs of M. saltuarius throug

Two alcohol dehydrogenases enhance Bursaphelenchus xylophilus resistance in Pinus massonniana via 3-hexenol biosynthesis accumulation.

Green leaf volatiles (GLVs), oxygenated compounds released by plants under biotic and abiotic stresses, play essential roles as signaling molecules in plant immune responses. However, the defense mechanisms mediated by these volatiles remain poorly characterized in conifers, particularly regarding Pinus massoniana against the invasive pine wood nematode (PWN, Bursaphelenchus xylophilus). Here, we systematically integrated transcriptomic and metabolic profiles to compare the defense responses of resistant and susceptible P. massoniana following PWN inoculation. Our results revealed the alpha-li

Development of Metal-Organic Framework-Based Emamectin Benzoate Nanopesticides for Effective Control of Bursaphelenchus xylophilus.

Pine wilt disease is caused by Bursaphelenchus xylophilus invasion and has a great impact on global pine resources. Injection of emamectin benzoate (EB) into pine trunks is an effective way to control B. xylophilus. However, EB has limited aqueous solubility, easily photodissociates, and its long-term use causes resistance problems. In this study, the metal-organic framework material ZIF-8 was prepared, and EB was loaded onto ZIF-8 through physical adsorption, resulting in a pH-responsive EB@ZIF-8 nanopesticide with a high drug loading rate (36.04%). Simulated release experiments of EB@ZIF-8 u

Synergistic antinematodal effects of fluopyram combined with chlorfenapyr and its potential mechanism against Bursaphelenchus xylophilus.

To enhance the chemical control of pine wilt disease (PWD) and mitigate resistance arising from the prolonged use of a single-agent pesticides, the nematicides fluopyram (FLU) and chlorfenapyr (CHL) were combined (FLU_CHL) to determine the optimal ratio and assess their effects on pine wood nematodes (PWNs) and the potential synergistic mechanisms. Results revealed that the optimal synergistic mass ratio of FLU to CHL was 1:3, with nematode survival rate, egg hatchability, and population size significantly lower than with FLU or CHL alone. At this ratio, FLU_CHL also induced morphological abno

Inducing the Entry of Third Stage Dispersal Juveniles of Bursaphelenchus xylophilus into Cryptobiosis Through Osmotic Regulation.

Cryptobiosis is a state where organisms lose nearly all their internal water and enter anhydrobiosis under extreme environmental stress. The dispersal third-stage juveniles (pre-dauer juveniles, ) of Bursaphelenchus xylophilus can enter cryptobiosis through dehydration and revive upon rehydration when environmental conditions improve. Osmotic regulation is crucial for their survival in this process. In this study, specimens of B. xylophilus were collected from dead Pinus massoniana due to pine wilt disease in Ningbo, Zhejiang Province, China. Following immersion in 8% potassium chloride (KCl)

Comparative Transcriptome Analysis of Pine Trees Treated with Resistance-Inducing Substances against the Nematode Bursaphelenchus xylophilus.

The pinewood nematode (PWN) Bursaphelenchus xylophilus causes pine wilt disease, which results in substantial economic and environmental losses across pine forests worldwide. Although systemic acquired resistance (SAR) is effective in controlling PWN, the detailed mechanisms underlying the resistance to PWN are unclear. Here, we treated pine samples with two SAR elicitors, acibenzolar-S-methyl (ASM) and methyl salicylic acid (MeSA) and constructed an in vivo transcriptome of PWN-infected pines under SAR conditions. A total of 252 million clean reads were obtained and mapped onto the reference

Chitosan oligosaccharide enhances the tolerance of Pinus koraiensis to Bursaphelenchus xylophilus through the flavonoid biosynthesis pathway.

Chitosan oligosaccharide (COS) is a widely used plant inducer. However, research on its application as an inducer for controlling plant-parasitic nematodes remains limited. In this study, Pinus koraiensis was treated with three concentrations of COS, followed by inoculation with pine wood nematode (PWN, Bursaphelenchus xylophilus). The efficacy of these COS concentrations in enhancing the resistance of P. koraiensis to PWN was evaluated using the relative tolerance index (RTI) and the relative anti-nematode index (RAI). Among the treatments, 0.5 % COS exhibited the optimal induction effect, ef

Detection of Bursaphelenchus Xylophilus, Causal Agent of Pine Wilt Disease on Pinus pinaster in Northwestern Spain.

The pine wood nematode, Bursaphelenchus xylophilus (Steiner & Buhrer) Nickle, a quarantine organism, causes serious damage to pines worldwide. In Europe, it was first detected in Portugal in 1999 (3) and the pathogen was thought to be restricted to this area. However, in 2008, B. xylophilus was isolated from a single tree in the Cáceres Region (Extremadura) of Spain, bordering Portugal (2). The region of Galicia has approximately 383,632 ha of Pinus pinaster Aiton that constitutes more than 40% of the surface of Spain. Since 1999, we have analyzed 5,155 samples to monitor the presence of the p

Identification and Validation of Reference Genes for Gene Expression Analysis in Monochamus saltuarius Under Bursaphelenchus xylophilus Treatment.

A special mutual relationship exists between the pine wood nematode (PWN) Bursaphelenchus xylophilus and its vector beetles of genus Monochamus, which enables PWN to spread, at the same time provides longhorned beetles with more weak hosts. PWN are attracted to the pupal chambers and then carried inside the trachea of beetle adults, which is a necessary part to complete the B. xylophilus infection cycle. The growth and immune responses of the vector beetle will affect this carrying process, however, they were rarely studied in Monochamus saltuarius. Real-time quantitative polymerase chain reac

Genome-Wide Identification and Expression Analysis of m6A Regulators in Bursaphelenchus xylophilus Across Developmental and Stress Conditions.

m6A represents a prevalent epitranscriptomic modification in eukaryotes. The dynamic balance of m6A modification is governed by methyltransferases (writers), demethylases (erasers), and binding proteins (readers). m6A regulators are integral to critical biological processes, including embryonic development, cell differentiation, and stress responses. B. xylophilus, a highly destructive invasive plant-parasitic nematode, has caused considerable ecological and economic damage worldwide. However, the m6A regulatory system in PWNs has not yet been investigated. In this study, we systematically ide
