The association of Yarrowia lipolytica with onychomycosis.

Onychomycosis, a common fungal nail infection, is typically caused by dermatophytes or Candida species, but rare fungal pathogens can also be involved. We describe a 20-year-old woman with persistent nail shedding who was unresponsive to standard antifungal treatments. Microscopic nail examination, fungal culture, and ITS sequencing repeatedly identified the presence of Yarrowia lipolytica (formerly Candida lipolytica), a dimorphic yeast more often associated with systemic bloodstream infections and not previously identified in patients with onychomycosis. Susceptibility testing revealed the r

Yarrowia lipolytica and pollutants: Interactions and applications.

Yarrowia lipolytica is a dimorphic, non-pathogenic, ascomycetous yeast species with distinctive physiological features and biochemical characteristics that are significant in environment-related matters. Strains naturally present in soils, sea water, sediments and waste waters have inherent abilities to degrade hydrocarbons such as alkanes (short and medium chain) and aromatic compounds (biphenyl and dibenzofuran). With the application of slow release fertilizers, design of immobilization techniques and development of microbial consortia, scale-up studies and in situ applications have been pos

Spatial Engineering of Yarrowia lipolytica for the Biosynthesis of Capsanthin.

Capsanthin is a high-value tetraterpenoid widely utilized in the food, cosmetic, and pharmaceutical industries. However, its traditional production via plant extraction and chemical synthesis faces severe sustainability and cost constraints. The oleaginous yeast Yarrowia lipolytica (Y. lipolytica) offers a superior chassis due to its robust mevalonate pathway and abundant lipid droplets for pigment sequestration. In this study, we systematically engineered Y. lipolytica for the de novo biosynthesis of capsanthin. By heterologously co-expressing of Capsicum annuum capsanthin/capsorubin synthase

Multiple-copy integration in the yeast Yarrowia lipolytica.

Using an EcoRI-BglII fragment of the G unit of the rDNA of Y. lipolytica and a set of 11 deletions in the URA3 promoter, we have constructed several plasmids to test gene amplification in the rDNA. These plasmids contain the rDNA fragment for integration, defective versions of the URA3 gene, the XPR2 gene encoding alkaline extracellular protease (AEP) as a reporter gene, and part of the pBR322 plasmid for selection and replication in E. coli. Among these plasmids, one corresponds to a deletion which allows multiple integration into the rDNA (plasmid pINA773). Two other plasmids (pINA767 and pI

Engineering Yarrowia lipolytica for Enhanced Production of Arbutin.

Arbutin, a glycoside, is derived from the leaves of several plants, including wheat, pear, and bearberry plants, and has a significant role in the treatment of melanoma, cystitis, and cough. Here, we aimed to modify Yarrowia lipolytica to produce arbutin. To construct the arbutin synthetic pathway in Y. lipolytica, three genes (chorismate pyruvate-lyase (UbiC), 4-hydroxybenzoate 1-hydroxylase (MNX1), and hydroquinone glucosyltransferase (AS)) were codon-optimized and heterologously expressed. To maximize arbutin production, seven arbutin-biosynthesis molecular targets were overexpressed, and w

Musico-bioremediation of seafood canning wastewater by Yarrowia lipolytica.

Due to the lack of water resources and the harmful effects of wastewater on environment and human health, treatment of wastewater becomes necessary. The present study explored the effect of musical sounds on the biological treatment of seafood canning wastewater by using Yarrowia lipolytica. Our results showed that low frequency (21 Hz to 1356 Hz) and high frequency (21 Hz to 16,214 Hz) musical sounds stimulated the growth of Y. lipolytica and increased the polluant removal efficiency. Such treatment decreased significantly the chemical oxygen demand (COD) and salinity as well as the color of

Reactivation of the alternative oxidase of Yarrowia lipolytica by nucleoside monophosphates.

The study of the effect of nucleoside phosphates on the activity of cyanide-resistant oxidase in the mitochondria and submitochondrial particles of Yarrowia lipolytica showed that adenosine monophosphate (5'-AMP, AMP) did not stimulate the respiration of intact mitochondria. The incubation of mitochondria at room temperature (25 degrees C) for 3-5 h or their treatment with ultrasound, phospholipase A, and the detergent Triton X-100 at a low temperature inactivated the cyanide-resistant alternative oxidase. The inactivated alternative oxidase could be reactivated with AMP. The reactivating effe

De novo biosynthesis of cannabinoids and their analogs in Yarrowia lipolytica.

Cannabis sativa has long been a cornerstone of both medicinal and cultural practices, with its therapeutic use spanning over 2700 years. Central to its therapeutic effects are cannabinoids, which interact with the endocannabinoid system to influence various physiological processes such as anxiety, pain, and inflammation. Despite its benefits, cannabinoid production faces challenges and scarcity from plant extraction. This work leverages Yarrowia lipolytica as a platform for cannabinoid biosynthesis. By optimizing the precursor supply, engineering biomolecular condensate-like dual prenyltransfe

Engineering a stable high-copy episomal plasmid in Yarrowia lipolytica.

Episomal plasmids are widely used in yeast metabolic engineering for their flexibility and high transformation efficiency. However, their application in Yarrowia lipolytica is often limited by low copy number and insufficient genetic stability. In this study, a robust episomal plasmid that balanced genetic stability and protein expression capacity was developed in Y. lipolytica. The autonomously replicating sequence (ARS) elements were selected, and ARS1 was identified as a favorable element that balanced genetic stability and gene expression capacity. To improve plasmid copy number, different

[Multidimensional metabolic engineering of Yarrowia lipolytica for β-carotene production].

β-carotene is a natural lipid-soluble carotenoid widely used in the food and pharmaceutical industries. Although metabolic engineering efforts for the production of β-carotene via microbial fermentation have achieved certain progress, there remains room for further improvement. Compared with Saccharomyces cerevisiae or Escherichia coli, Yarrowia lipolytica exhibits a pronounced lipid-flux orientation in its core metabolic logic, conferring a natural advantage for the production of lipid-soluble compounds. This study aims to engineer Y. lipolytica strains for efficient β-carotene production. Fi

Engineering Yarrowia lipolytica for production of medium-chain fatty acids.

Lipids are naturally derived products that offer an attractive, renewable alternative to petroleum-based hydrocarbons. While naturally produced long-chain fatty acids can replace some petroleum analogs, medium-chain fatty acid would more closely match the desired physical and chemical properties of currently employed petroleum products. In this study, we engineered Yarrowia lipolytica, an oleaginous yeast that naturally produces lipids at high titers, to produce medium-chain fatty acids. Five different acyl-acyl carrier protein (ACP) thioesterases with specificity for medium-chain acyl-ACP mol

Biosynthesis of the antioxidant γ-glutamyl-cysteine with engineered Yarrowia lipolytica.

The dipeptide γ-glutamylcysteine (γ-GC), the first intermediate of glutathione (GSH) synthesis, is considered as a promising drug to reduce or prevent plethora of age-related disorders such as Alzheimer and Parkinson diseases. The unusual γ-linkage between the two constitutive amino acids, namely cysteine and glutamate, renders its chemical synthesis particularly challenging. Herein, we report on the metabolic engineering of the non-conventional yeast Yarrowia lipolytica for efficient γ-GC synthesis. The yeast was first converted into a γ-GC producer by disruption of gene GSH2 encoding GSH syn

Asphaltenes biodegradation from heavy crude oils by the yeast Yarrowia lipolytica.

Heavy crude oil reserves are characterized by their high viscosity and density, largely due to significant quantities of asphaltenes. The removal of asphaltene precipitates from oil industry installations is crucial, as they can contaminate catalysts and obstruct pipelines. Therefore, this study aimed to bio-transform heavy oil asphaltenes into smaller molecules using the yeast Yarrowia lipolytica, known for its ability to efficiently degrade hydrophobic substrates. For this purpose, asphaltenes were extracted from crude oil samples, and yeast growth was assessed in a mineral medium containing

Biosynthesis of cannabinoid precursor olivetolic acid in genetically engineered Yarrowia lipolytica.

Engineering microbes to produce plant-derived natural products provides an alternate solution to obtain bioactive products. Here we report a systematic approach to sequentially identify the rate-limiting steps and improve the biosynthesis of the cannabinoid precursor olivetolic acid (OLA) in Yarrowia lipolytica. We find that Pseudomonas sp LvaE encoding a short-chain acyl-CoA synthetase can efficiently convert hexanoic acid to hexanoyl-CoA. The co-expression of the acetyl-CoA carboxylase, the pyruvate dehydrogenase bypass, the NADPH-generating malic enzyme, as well as the activation of peroxis
