Copper Isotopes and Copper to Zinc Ratio as Possible Biomarkers for Thyroid Cancer.

Thyroid cancer is the most common endocrine cancer. There is no systematic screening for such cancer, and the current challenge is to find potential biomarkers to facilitate an early diagnosis. Copper (Cu) and zinc (Zn) are essential micronutrients involved in the proper functioning of the thyroid gland, and changes in their concentrations have been observed in the development of cancer. Previous studies have highlighted the potential 65Cu/63Cu ratio (δ65Cu) to be a cancer biomarker. This study tests its sensitivity on plasma samples (n = 46) of Algerian patients with papillary thyroid carcino

Determination of metal ion transport rate of human ZIP4 using stable zinc isotopes.

The essential microelement zinc is absorbed in the small intestine mainly by the zinc transporter ZIP4, a representative member of the Zrt/Irt-like protein (ZIP) family. ZIP4 is reportedly upregulated in many cancers, making it a promising oncology drug target. To date, there have been no reports on the turnover number of ZIP4, which is a crucial missing piece of information needed to better understand the transport mechanism. In this work, we used a nonradioactive zinc isotope, 70Zn, and inductively coupled plasma mass spectrometry to study human ZIP4 (hZIP4) expressed in Human embryonic kidn

[Studies on the zinc bioavailability using stable isotope techniques].

Zinc is one of essential trace elements in humans. Zinc homeostasis is mainly regulated by the changes of intestinal absorption efficiency and endogenous excretion, namely the regulation of bioavailability. Studies on zinc bioavailability play an important role on a thorough understanding of zinc metabolism and reliable assessment on population zinc status. Stable isotopes are valuable tools for research on mineral bioavailability and metabolism. Zinc stable isotopes can be used for all population as tracers with no exposure to radiation. Since 1980s' many different stable isotope methods have

Plasma and tissue zinc enrichment altered by different sources of zinc using stable isotopes in weaned male Holstein calves facing dietary antagonism.

Trace minerals are an important component of ruminant nutrition. Furthermore, understanding the bioavailability of various trace mineral sources is essential for accurate dietary formulation. The objective of this trial is to determine the effect of a Zn antagonism, elevated S, on the resulting plasma and tissue Zn enrichment, while also determining if differences occur depending on Zn source, Zn glycinate, and Zn methionine hydroxy analog chelate, in Holstein calves. Thirty weaned male Holstein calves were used in a randomized complete block design. Calves were fed a texturized starter varyin

Soil (microbial) disturbance affect the zinc isotope biogeochemistry but has little effect on plant zinc uptake.

Zinc (Zn) is an important micronutrient but can be toxic at elevated concentrations. We conducted an experiment to test the effect of plant growth and soil microbial disturbance on Zn in soil and plants. Pots were prepared with and without maize and in an undisturbed soil, a soil that was disturbed by X-ray sterilization and a soil that was sterilized but reconditioned with the original microbiome. The Zn concentration and isotope fractionation between the soil and the soil pore water increased with time, which is probably due to physical disturbance and fertilization. The presence of maize in

Stage-resolved zinc isotope dynamics in firefighters' hair after firefighting.

Firefighting exposes firefighters to combustion-derived particulate matter enriched in hazardous metals, yet biomarkers capable of integrating repeated fire events and recovery under real exposure conditions remain limited. This study quantified hair metal concentrations and δ66Zn in 53 hair samples collected from 15 Beijing firefighters across five stage-defined periods spanning pre-mission, active firefighting, and recovery. Hair δ66Zn ranged from -0.79‰ to -0.13‰ (mean -0.44‰) and displayed a non-linear incident-recovery trajectory, enriched in the heavier Zn isotopes after repeated mission

Nucleosynthetic isotope anomalies of zinc in meteorites constrain the origin of Earth's volatiles.

Material inherited from different nucleosynthesis sources imparts distinct isotopic signatures to meteorites and terrestrial planets. These nucleosynthetic isotope anomalies have been used to constrain the origins of material that formed Earth. However, anomalies have only been identified for elements with high condensation temperatures, leaving the origin of Earth's volatile elements unconstrained. We determined the isotope composition of the moderately volatile element zinc in 18 bulk meteorites and identified nucleosynthetic zinc isotope anomalies. Using a mass-balance model, we find that c

In Vitro Anticancer Activity of the Light Stable Zinc Isotope (64Zn) Compounds.

BACKGROUND/AIM: Today, stable isotopes of zinc are actively used for diagnostic purposes in oncology. However, there is extremely limited data on the attempts to apply stable zinc isotopes in cancer therapy or about the molecular mechanisms of their effects on the biology of tumor cells. Therefore, in this in vitro research, we evaluated the cytotoxic activity of stable zinc isotope (64Zn) enriched compounds against malignant cells and determined the mechanisms of their action. MATERIALS AND METHODS: Malignant and non-malignant cells of different histogenesis were used as objects of the study

Research on zinc electrodeposition for target preparation in medical isotope 67Cu production.

As an ideal dual-functional radionuclide for diagnostics and therapeutics, the clinical translation of 67Cu is constrained by the stringent purity requirements of the 68Zn target material during cyclotron production. To address the issues of impurity interference in traditional electrodeposition processes, this study developed an electrolyte system devoid of extraneous ionic impurities and organic additives. Under the pre-optimized basic parameters (current density 30 mA/cm2, temperature 40 °C, Zn2+ concentration 20 g/L, mass ratio of ZnO to ammonium chloride 1:10), the regulation of pH 1-pH 5

Mechanism of zinc stress on magnesium deficiency in rice plants (Oryza sativa L.): Insights from magnesium isotopes.

Magnesium (Mg) and zinc (Zn) are essential nutrients for plants. Mg deficiency often occurs in rice plants grown in Zn-polluted soil. However, the mechanism for this correlation is unclear. Here, we performed culture experiments on rice plants (Oryza sativa L.) and used Mg isotopes to investigate mechanisms of Zn stress on plant Mg deficiency. Our results show that excess Zn can significantly reduce the uptake of Mg in rice tissues. The root displays positive Δ26Mgplant-nutrient values (δ26Mgplant-δ26Mgnutrient; 1.90 ‰ to 2.06 ‰), which suggests that Mg enters the root cells mainly via Mg-spec

A critical assessment of copper and zinc isotopes as tracers for antifouling paint contamination in coastal sediments.

Antifouling paints (AFPs) are a major source of copper (Cu) and zinc (Zn) to coastal environments yet tracing their contribution to sediment metal contamination remains challenging due to overlapping sources and complex biogeochemical processing. We combined field observations, sediment core analysis, and in situ exposure experiments to evaluate the potential of Cu and Zn stable isotopes as tracers of AFP-derived contamination in a tropical marina (Bracuhy Marina, Brazil). Elemental data from a geochemically constrained sediment core reveal pronounced Cu and Zn enrichment following marina oper

The Use of Stable Zinc Isotope Soil Labeling to Assess the Contribution of Complex Organic Fertilizers to the Zinc Nutrition of Ryegrass.

Manure and sewage sludge are known to add significant amounts of zinc (Zn) and other metals to soils. However, there is a paucity of information on the fate of Zn that derives from complex organic fertilizers in soil-plant systems and the contribution of these fertilizers to the Zn nutrition of crops. To answer these questions, we grew Italian ryegrass in the presence of ZnSO4, sewage sludge, and cattle and poultry manure in an acidic soil from Heitenried, Switzerland, and an alkaline soil from Strickhof, Switzerland, where the isotopically exchangeable Zn had been labeled with 67Zn. This allo

Zinc Stable Isotope Fractionation Mechanisms during Adsorption on and Substitution in Iron (Hydr)oxides.

The Zn isotope fingerprint is widely used as a proxy of various environmental geochemical processes, so it is crucial to determine which are the mechanisms responsible for isotopic fractionation. Iron (Fe) (hydr)oxides greatly control the cycling and fate and thus isotope fractionation factors of Zn in terrestrial environments. Here, Zn isotope fractionation and related mechanisms during adsorption on and substitution in three FeOOH polymorphs are explored. Results demonstrate that heavy Zn isotopes are preferentially enriched onto solids, with almost similar isotopic offsets (Δ66/64Znsolid-so

Zinc source differentiation in hydrothermal vent mollusks: Insight from Zn isotope ratios.

Hydrothermal vent represents an extreme environment where metal-enriched fluids are in contact with chemosymbiotic animals. In the present study, Zn isotopic compositions were determined in multiple tissues of three dominant hydrothermal vent mollusks (the mussel Bathymodiolus marisindicus and two gastropods Chrysomallon squamiferum and Gigantopelta aegis) collected from a hydrothermal vent field (Southwest Indian Ridge in the Indian Ocean). We found approximately 1.78‰ differences in the δ66Zn values among the three vent mollusks despite of their similar range of Zn concentrations. The signif

Use of Stable Isotopes to Evaluate Bioefficacy of Provitamin A Carotenoids, Vitamin A Status, and Bioavailability of Iron and Zinc.

The ability of nutrition scientists to measure the status, bioavailability, and bioefficacy of micronutrients is affected by lack of access to the parts of the body through which a nutrient may travel before appearing in accessible body compartments (typically blood or urine). Stable isotope-labeled tracers function as safe, nonradioactive tools to follow micronutrients in a quantitative manner because the absorption, distribution, metabolism, and excretion of the tracer are assumed to be similar to the unlabeled vitamin or mineral. The International Atomic Energy Agency (IAEA) supports resear

Copper isotope fractionation and copper-to-zinc ratios reveal altered metal homeostasis in prostate cancer.

Trace metal homeostasis is increasingly recognized as a reflection of cellular metabolism and redox balance. This study applies high-precision copper isotope and elemental ratio analysis to human serum to investigate how prostate cancer alters systemic metal regulation. Using multi-collector inductively coupled plasma mass spectrometry (MC-ICP-MS) adapted from geochemical applications, we quantified copper isotope fractionation and Cu/Zn ratios across disease stages. Patients with localized or newly metastatic prostate cancer exhibited isotopically heavier serum copper compared to healthy cont

Zinc/Silver-Based Hydrogen Sulfide Trapping as an Alternative to Cadmium for Sulfur Stable Isotope Analysis.

RATIONALE: Hydrogen sulfide (H2S) sulfur stable isotope ratio (δ34S) analyses use cadmium (Cd) or zinc (Zn) solutions; however, Cd is toxic, and Zn does not display an obvious change in color at low molar H2S inputs. This study (i) developed a non-toxic, colorimetric mixed zinc/silver (Zn/Ag)-based method and (ii) evaluated the minimum H2S input required for analysis. METHODS: Gaseous H2S was trapped using the Cd- and Zn/Ag-based methods and converted to solid silver sulfide (Ag2S). δ34S stable isotope analysis was completed using elemental analysis-isotope ratio mass spectrometry (EA-IRMS).

Feasibility of simultaneous dual isotope acquisition for myocardial perfusion imaging with a cadmium zinc telluride camera.

BACKGROUND: We studied the impact of technetium-99m (99mTc) in the thallium-201 (201Tl) energy window (70 keV) to determine if CZT cardiac cameras allow us to perform simultaneous dual-isotope acquisition for myocardial perfusion imaging. METHODS: We included 117 consecutive patients. We injected 0.7 MBq/kg of 201Tl at stress, performed the first scan (image T1), then injected at rest 2 MBq/kg of 99mTc-tetrofosmin and immediately acquired a second scan with reconstruction in the energy window of thallium (image T2). A corrected thallium image was created by the subtraction of 99mTc downscatte
