Comparative efficacy of alpha-linolenic acid and gamma-linolenic acid to attenuate valproic acid-induced autism-like features.

The present study was undertaken to elucidate the effect of alpha-linolenic acid (ALA, 18:3, ω-3) and gamma-linolenic acid (GLA, 18:3, ω-6) on experimental autism features induced by early prenatal exposure to valproic acid (VPA) in albino wistar pups. The pups were scrutinized on the accounts of behavioral, biochemical, and inflammatory markers, and the results suggested that the GLA can impart significant protection in comparison to ALA against VPA-induced autism features. When scrutinized histopathologically, the cerebellum of the GLA-treated animals was evident for more marked protection t

Effects of gamma-linolenic and dihomo-gamma-linolenic acids on 7,12-dimethylbenz(alpha)anthracene-induced mammary tumors in rats.

The effects of pure gamma-linolenic acid (GLA; C18:3, n-6) and dihomo-gamma-linolenic acid (DGLA; C20:3, n-6) were investigated in 7,12-dimethylbenz(alpha)anthracene (DMBA) (10 mg/rat)-induced mammary tumors in Sprague-Dawley rats. 0.15 g of GLA, DGLA, or corn oil (CO) were administered (two times per week) by oral intubation, for 12 weeks to rats maintained on a 5% (w/w) CO diet. Tumor incidence, tumor multiplicity, and percent of tumor-bearing rats were highest in the CO group. Tumor multiplicity was significantly reduced in the GLA group (p = 0.015). Feeding of GLA and DGLA resulted in sign

Inhibition of methyl mercury chloride-induced chromosomal damage by gamma-linolenic acid.

The genotoxic potential of methyl mercury chloride (MMC) was investigated in human lymphocyte cultures by tests for chromosomal aberrations and sister chromatid exchange. The protective role of gamma-linolenic acid, a derivative of dietary essential fatty acid, on MMC-induced genetic damage was evident from the reduced frequency of aberrations and sister chromatid exchanges.

Gamma-linolenic acid in borage oil reverses epidermal hyperproliferation in guinea pigs.

As dietary sources of gamma-linolenic acid [GLA; 18:3(n-6)], borage oil (BO; 24-25 g/100 g GLA) and evening primrose oil (PO; 8-10 g/100 g GLA) are efficacious in treating skin disorders. The triglycerol stereospecificity of these oils is distinct, with GLA being concentrated in the sn-2 position of BO and in the sn-3 position of PO. To determine whether the absolute level and/or the triglycerol stereospecificity of GLA in oils affect biological efficacy, epidermal hyperproliferation was induced in guinea pigs by a hydrogenated coconut oil (HCO) diet for 8 wk. Subsequently, guinea pigs were fe

Multiple oral mucoceles treated with gamma-linolenic acid: report of a case.

Multiple mucoceles are an extremely rare condition in the oral cavity. We report a case which responded entirely to systemic gamma-linolenic acid but which recurred several months following discontinuation of therapy. Spontaneous resolution of the condition seems unlikely and it would appear that gamma-linolenic acid could have an effect on minor salivary gland composition and/or viscosity and may be worth prescribing on a trial basis prior to surgical treatment of this rare condition.

gamma-Linolenic acid supplementation can affect cancer cell proliferation via modification of fatty acid composition.

We examined the effect of gamma-linolenic acid (GLA) supplementation on the growth and fatty acid composition of three human tumor cell lines (the neuroblastoma CHP-212, the tubal carcinoma TG, and the colon carcinoma SW-620), in order to evaluate the relationship between GLA-induced tumor cell death and the distribution of fatty acids in tumor cells. At the highest GLA concentrations (10 and 20 micrograms/ml), the DNA synthesis was completely abolished; at 5 micrograms/ml GLA only SW-620 cells did not proliferate, while CHP-212 and TG cells showed a residual [3H]-thymidine incorporation. Fatty acid modification of cultured neuroblastoma cells by gamma linolenic acid relevant to its antitumor effect.

Two human neuroblastoma cell lines, NCG and GOTO, were used to study the cytotoxic effect of gamma linolenic acid (GLA). The cell growth inhibition of these culture cells by GLA was found to be associated with striking membrane fatty acid modification. When culture cells were exposed to 20 micrograms/ml and 60 micrograms/ml GLA for 48 hr, polyenoic acids in cell membrane phospholipids (PC, PE, PI, PS) and triglyceride significantly increased; 1.8-21.0 fold for NCG and 1.04-11.5 fold for GOTO, in association with decreased monoenoic acids. The most remarkable changes were; increase of C18:3, C2

Long-term use of gamma-linolenic acid-enriched fat emulsion (PFE 4501): case report.

A new gamma-linolenic acid-enriched fat emulsion (PFE 4501) was given for 13 mo as part of a home parenteral nutrition regimen to a 28-yr-old patient with cystic fibrosis. Blood biochemistry, hematological data, and fatty acid profile were followed. No side effects were reported, and pulmonary function tests remained stable. gamma-Linolenic acid appears to be safe for clinical use and may benefit patients with derangements in essential fatty acid metabolism. The potential role of a gamma-linolenic acid-enriched intravenous fat emulsion is discussed.

Gamma-linolenic acid induces apoptosis and lipid peroxidation in human chronic myelogenous leukemia K562 cells.

Various polyunsaturated fatty acids, especially gamma-linolenic acid (GLA), inhibit the growth of a variety of tumor cells. Some evidence indicates that polyunsaturated fatty acid can kill cells by apoptosis. In the current study, we tested the apoptotic effect of GLA on human chronic myelogenous leukemia K562 cells. GLA induced K562 cell death in a dose-dependent manner. Typical apoptotic nuclei were shown by staining of K562 cells with DNA-binding fluorochrome Hoechst 33342, characterized by chromatin condensation and nuclear fragmentation. Flow cytometric analysis also demonstrated that GLA

Microbial cell factories for gamma-linolenic acid biosynthesis: metabolic engineering, downstream processing, and future perspectives.

Gamma-linolenic acid (GLA) is a vital anti-inflammatory polyunsaturated fatty acid, yet its traditional supply from plant seeds such as evening primrose and borage is constrained by low yields and climatic instability. Consequently, microbial cell factories present a sustainable and season-independent alternative for scalable GLA production. This review critically synthesizes recent biotechnological advancements in utilizing oleaginous microorganisms as production platforms. It systematically elucidates the microbial lipid biosynthesis pathways and evaluates strategies to enhance GLA accumulat

In vivo and in vitro effects of gamma-linolenic acid and eicosapentaenoic acid on prostaglandin production and arachidonic acid uptake by human endometrium.

This study examines the effect of a 6 month dietary supplement of either gamma-linolenic acid (GLA) or eicosapentaenoic acid (EPA) on the synthesis of prostaglandins E2 and F2 alpha by the endometrium of women with regular menstrual cycles. Samples of endometrium, obtained pre- and post-treatment, were incubated in vitro for 2 h and the prostaglandins E2 and F2 alpha released into the medium measured by radioimmunoassay. The ability of the tissue to take up 14C-arachidonic acid before and after treatment was also examined. Both GLA and EPA caused a marked decrease in the synthesis of prostagla

The use of gamma linolenic acid, linoleic acid and natural vitamin E for the treatment of multicentric lymphoma in two dogs.

The use of Gamma linolenic acid (GLA) and other essential fatty acid (EFA) metabolites in malignant cancer treatment in vitro and in vivo is briefly reviewed. Treatment of two dogs with multicentric lymphoma with large empirical daily doses of a combination of 40 mg gamma linolenic acid, 350 mg linoleic acid and 10 mg natural vitamin E per capsule resulted, after approximately one week, in slight to marked reduction in size of enlarged peripheral lymph nodes, spleen, skin nodules and tonsils. Both animals showed transient improved habitus and appetite, but deteriorated due to complications, ap

Some effects of the essential fatty acids linoleic acid and alpha-linolenic acid and of their metabolites gamma-linolenic acid, arachidonic acid, eicosapentaenoic acid, docosahexaenoic acid, and of prostaglandins A1 and E1 on the proliferation of human osteogenic sarcoma cells in culture.

Gamma-linolenic acid has been shown to suppress the rate of proliferation of a number of malignant cell lines in culture. To test the proposal that this was a specific prostaglandin 1- or 2-series effect, 379 batches of MG63 human osteogenic sarcoma cells were seeded in Greiner flasks and cultured in media supplemented with a range of unsaturated fatty acids and prostaglandins. The monounsaturated fatty acid oleic acid enhanced the rate of cancer cell proliferation. Failure of short-term gamma-linolenic acid treatment to reduce urinary calcium loss of diabetic rats.

Calcium re-absorption in the kidney is impaired in streptozotocin (STZ) diabetic rats, thereby causing hypercalciuria. Increased calcium loss starts within 1-2 days after induction of diabetes and reaches a plateau after 2 weeks. The excessive calcium excretion was previously shown to be reduced by treatment with gamma-linolenic acid (GLA) or evening primrose oil rich in GLA. However, in these studies, the animals were pre-treated for several weeks before injection of STZ. In the present study we investigated whether GLA can reduce calcium excretion when treatment starts at the same time as in
