Contribution of increased mitochondrial free Ca2+ to the mitochondrial permeability transition induced by tert-butylhydroperoxide in rat hepatocytes.

Previously, we showed that the oxidant chemical, tert-butylhydroperoxide (t-BuOOH), induces a mitochondrial permeability transition (MPT) in intact hepatocytes, causing lethal cell injury. Here, we investigated the role of mitochondrial free Ca2+ in t-BuOOH cytotoxicity to 1-day-cultured rat hepatocytes using confocal microscopy of autofluorescence and parameter-indicating fluorophores. t-BuOOH (100 micromol/L) caused an early increase of mitochondrial free Ca2+, as assessed by confocal microscopy of Rhod-2 fluorescence. Increased mitochondrial Ca2+ was followed by onset of the MPT, as evidenc

Protective effect of Paeonia anomala extracts and constituents against tert-butylhydroperoxide-induced oxidative stress in HepG2 cells.

The fruit and root parts of Paeonia anomala L. are used for the treatment of many kinds of disorders in Mongolian traditional medicine. The protective effect of a fruit extract from P. anomala against tert-butylhydroperoxide-induced cell damage was evaluated in human hepatoma HepG2 cells and compared to that of a root extract from P. anomala on the basis of cell viability, generation of intracellular reactive oxygen species, cellular total glutathione concentration, and anti-genotoxicity. The fruit extract of P. anomala showed excellent protection against the oxidative stress when compared to

Endogenous and exogenous nitric oxide enhance the DNA strand scission induced by tert-butylhydroperoxide in PC12 cells via peroxynitrite-dependent and independent mechanisms, respectively.

A short-term exposure to tert-butylhydroperoxide (tB-OOH) promoted a concentration-dependent formation of DNA single-strand breaks in PC12 cells. These events were paralleled by an increase in the cytosolic concentration of Ca2+ that was in part cleared by the mitochondria. Unlike the extent of Ca2+ mobilization and/or mitochondrial Ca2+ clearance, the DNA strand scission evoked by the hydroperoxide was markedly reduced by the nitric oxide (NO) scavenger 2-phenyl-4,4,5,5-tetramethylimidazolin-1-oxyl-3-oxide (PTIO) or by the NO synthase inhibitor N-nitro-L-arginine methylester (L-NAME). Different signalling pathways mediate the opposite effects of endogenous versus exogenous nitric oxide on hydroperoxide toxicity in CHP100 neuroblastoma cells.

The results presented in this study indicate that the toxic response brought about by increasing concentrations of tert-butylhydroperoxide in CHP100 cells was mitigated significantly by exogenously added nitric oxide donors via a cyclic GMP-independent mechanism. In contrast with these results, endogenous nitric oxide generated by the Ca2+-mobilizing agent caffeine was found to increase hydroperoxide toxicity. Under these conditions, nitric oxide was not directly toxic to the cells. Rather, nitric oxide was found to promote the caffeine-mediated release of Ca2+ from ryanodine-sensitive Ca2+ st

Oxidative erythrocyte membrane damage in hereditary spherocytosis.

The occurrence, in Hereditary Spherocytosis, of an oxidative damage to red blood cell membranes was studied by "in vitro" treatment of the erythrocytes with tert-butylhydroperoxide, methylene blue, or phenylhydrazine. Spherocytes were found to be more sensitive than normal erythrocytes to the action of these drugs. Tert-butylhydroperoxide caused a more intense lipid peroxidation as well as more extensive membrane protein alterations, namely spectrin degradation, formation of high molecular weight aggregates, and globin binding to the membrane. Marked spectrin degradation was also induced by me

Variations of adenine nucleotide levels in normal and pathologic human erythrocytes exposed to oxidative stress.

Normal human erythrocytes suspended in isotonic saline at 0.5 haematocrit displayed, after 30 min exposure to 1 mM tert-butylhydroperoxide at 37 degrees C, a marked increase of NADPH, while the concentration of the other adenine nucleotides was almost unchanged. Hereditary Spherocytosis and glucose-6-phosphate dehydrogenase-deficient red blood cells exhibited, under basal conditions, higher levels of most of the nucleotides assayed and significant amounts of hypoxanthine. Treatment with tert-butylhydroperoxide caused, in glucose-6-phosphate dehydrogenase-deficient erythrocytes, a pronounced de

Aurocyanide, an active metabolite of auranofin, exerts cytoprotective effects via ROS-mediated Nrf2 activation in HepG2 cells.

Auranofin, an FDA-approved oral gold(I) compound for rheumatoid arthritis, is being repurposed due to its established clinical safety and emerging therapeutic potential. Given its extensive in vivo metabolism, identifying active metabolites and characterizing their biological activities are critical for understanding its therapeutic mechanisms. Here, we evaluated the cytoprotective effects of seven potential auranofin metabolites against tert-butylhydroperoxide-induced oxidative injury in HepG2 cells. Among them, only aurocyanide markedly improved cell viability, as demonstrated by MTT and liv

Synthesis and in Vivo Evaluation of Triphenylphosphonium Conjugated Trimetazidine with Enhanced Cardioprotection and Ability to Restore Mitochondrial Function.

Trimetazidine exhibits great therapeutic potential in cardiovascular diseases and mitochondria-mediated cardioprotection by trimetazidine has been widely reported. In this study, to enhance its cardioprotection, the triphenylphosphonium-based modification of trimetazidine was conducted to deliver it specifically to mitochondria. Fifteen triphenylphosphonium (TPP) conjugated trimetazidine analogs were designed and synthesized. Their protective effects were evaluated in vivo using a tert-butyl hydroperoxide (t-BHP) induced zebrafish injury model. Structure-activity relationship correlations reve

A Standardized Protocol for Inducing Stress-Induced Premature Senescence in Primary Human Melanocytes Using Tert-butyl Hydroperoxide.

Environmental stressors, including UV radiation, air pollution, or cigarette smoke, drive extrinsic skin aging. Exposure to these extrinsic aging factors promotes the accumulation of senescent cells, which contributes to the formation of wrinkles and pigmentation disorders. Melanocytes, which provide primary UV protection, exhibit reduced activity with age. Senescent melanocytes accumulate in aged skin and contribute to its aging appearance; However, there are few models to study melanocyte senescence, and this work aims to develop a reproducible model for further research. In this study, we p

Versatile Butenolide Syntheses via a Structure-Oriented C-H Activation Reaction.

Despite significant advances in forging a single bond via C-H activation in the past decade, one-step construction of biologically or chemically important scaffolds from abundant feedstock chemicals, namely, structure-oriented C-H activation, remains a significant challenge. Since feedstocks often contain a single functional group, multiple C(sp3)-H bond functionalizations are often necessary toward the assembly of densely functionalized scaffolds. As classic pharmacophores and versatile synthetic intermediates, butenolides have received extensive attention from the pharmaceutical industry and

Modeling Heme Peroxidase: Heme Saddling Facilitates Reactions with Hyperperoxides To Form High-Valent FeIV -Oxo Species.

Saddle-shaped hemes have been discovered in the structures of most peroxidases. How such a macrocycle deformation affects the reaction of FeIII hemes with hydrogen peroxide (H2 O2 ) to form high-valent Fe-oxo species remains uncertain. Through examination of the ESI-MS spectra, absorption changes and 1 H NMR chemical shifts, we investigated the reactions of two FeIII porphyrins with different degrees of saddling deformation, namely FeIII (OETPP)ClO4 (1OE ) and FeIII (OMTPP)ClO4 (1OM ), with tert-butyl hydroperoxide (tBuOOH) in CH2 Cl2 at -40 °C, which quickly resulted in O-O bond homolysis fro

Mitochondrial metabolism of a hydroperoxide to free radicals in human endothelial cells: an electron spin resonance spin-trapping investigation.

Oxidative damage to the vascular endothelium may be an important event in the promotion of atherosclerosis. Several lines of evidence suggest that lipid hydroperoxides may be responsible for the induction of such damage. Hydroperoxides cause loss of endothelial cell integrity, increase the permeability of the endothelium to macromolecules, and compromise its ability to control vascular tone via the secretion of vasoactive molecules in response to receptor stimulation. The molecular mechanisms responsible for these effects are, however, poorly understood. In this paper, we describe an e.s.r. sp

Solution-Phase Chemical Synthesis of Modified RNA Dinucleotides.

This article describes a simple, reliable, efficient, and improved solution-phase method for the gram-scale chemical synthesis of RNA dinucleotides such as pAm pA, pAm pG, and pAm pU that utilizes phosphoramidite chemistry. The overall synthetic strategy involves three steps. The first step involves the coupling reaction between 5'-O-MMT protected nucleoside-3'-O-phosphoramidite and a protected nucleoside containing a free 5'-OH group in the presence of tetrazole, followed by the oxidation of phosphite triester using tert-butyl hydroperoxide to give the corresponding protected Nm pN. Next, the

NIS-catalyzed reactions: amidation of acetophenones and oxidative amination of propiophenones.

Single-step amination: the N-iodosuccinimide (NIS)-catalyzed amidation of acetophenone derivatives by using tert-butylhydroperoxide (TBHP) as an oxidant is presented. A variety of acetyl derivatives of heterocyclic compounds were easily converted to their corresponding ketoamides under these conditions. A new, NIS-catalyzed amination of propiophenone and its derivatives in the presence of TBHP to furnish the corresponding 2-aminoketone derivatives is the first reported single-step amination of propiophenone derivatives.

13C and 31P NMR studies of glucose and 2-deoxyglucose metabolism in normal and enzyme-deficient human erythrocytes.

The flux of 13C-labeled glucose through the Embden-Meyerhof and pentose phosphate pathways was studied by 13C NMR in intact erythrocytes isolated from normal subjects or from patients suffering of glucose-6-phosphate dehydrogenase (G6PD, EC 1.1.1.49) deficiency. Similar rates of glucose catabolism and similar fluxes of the 13C-label into 2,3-bisphosphoglycerate and lactate were found, under basal conditions, in normal and in G6PD-deficient erythrocytes incubated in the presence of either [1-13C]- or D[6-13C]glucose. [Inactivation of Ras1 in Fission Yeast Aggravates the Oxidative Stress Response Induced by Tert Butyl Hydroperoxide (tBHP)].

Ras proteins are small GTPases and function as molecular switches to regulate cellular homeostasis. Ras-dependent signalling pathways regulate several essential processes such as cell cycle progression, growth, migration, apoptosis, and senescence. The dysregulation of Ras signaling pathway has been linked to several pathological outcomes. A potential role of RAS in regulating the redox signalling pathway has been established that includes the manipulation of ROS levels to provide a redox milieu that might be conducive to carcinogenesis. Access to Polyheterocyclic Compounds through Iron(II)-Mediated Radical Cascade Cyclization Utilizing 2-Ethynylbenzaldehydes and Aryl Isonitriles.

An oxidative radical cascade addition cyclization approach for the synthesis of quinoline-based π-extended polyheterocyclic compounds is reported. Eco-friendly iron catalysis and inexpensive tert-butylhydroperoxide (TBHP) as the oxidant have been utilized in the transformation of various readily available ortho-alkynylated aromatic aldehydes as radical precursors with aryl isonitriles as radical acceptors. Indole and thiophene-based carbaldehydes allow the preparation of quinolines that are π-conjugated with an additional heteroarene moiety in a single sequence by applying the introduced metho

Using blue-light driven iron-catalysis to afford molecular diversity in phosphindole oxide derivatives.

We have developed original blue-light promoted iron-catalyzed reactions of activated alkynes with secondary phosphine oxides in the presence of tert-butylhydroperoxide under argon at room temperature. Depending on the nature of the iron precursors, benzo[b]phosphole oxides were obtained using Fe(NO3)3 in methanol and 9-phenyltribenzo[b,e,g]phosphindole 9-oxide derivatives utilizing Fe(OTf)2 in acetic acid. Control experiments revealed that radical processes are involved, permitting the proposal of a reasonable mechanism.

Apoptosis and the adaptive response in human lymphocytes.

PURPOSE: To determine whether the sensitivity of human lymphocytes for apoptosis induced by either a membrane oxidizing agent or a DNA damaging agent is modified by an adaptive response. MATERIALS AND METHODS: Peripheral blood lymphocytes from normal human donors were exposed to low doses of the DNA damaging agent gamma-radiation, or the membrane oxidizing agent t-butyl hydroperoxide (t-BuOOH), incubated for various times and then tested for their sensitivity to induction of apoptosis by a subsequent exposure to a high dose of either agent. Apoptosis was measured using a fluorescent assay of

Iodo-sulphonylation of 1,6-enynones: a metal-free strategy to synthesize N-substituted succinimides.

An iodine-mediated radical cyclization of 1,6-enynones with sulphonyl hydrazides using tert-butyl hydroperoxide (TBHP) as the oxidant has been developed for the synthesis of iodo-sulphonylated-succinimide derivatives. The notable advantages of the developed method are metal-free conditions, broad functional group tolerance, column chromatography-free purification, high stereoselectivity (E isomer), shorter reaction times, and the cascade construction of three new bonds (C-S, C-I, and C-C). The synthetic application of the iodo-functionality has been extended to the Heck coupling reaction with
