Аннотация

Monoclonal antibodies (mAbs) represent a powerful targeted therapeutic arsenal for treating diverse diseases, ranging from infectious and chronic conditions to cancer. However, successful development requires overcoming several complex stability and solubility challenges in the final stages. This review examines the discovery, characterization, and therapeutic development of monoclonal antibodies (mAbs), with particular emphasis on inherent stability challenges, including aggregation and degradation, that can compromise mAb efficacy and safety. We demonstrate how novel formulation strategies can address these intrinsic limitations to expand the therapeutic potential of an antibody. Our analysis emphasizes critical aspects of immunogenicity, antibody aggregation and degradation, and advanced characterization methods. We examine liquid, lyophilized, PEGylated, and spray-dried formulations, as well as methods to enhance stability through mutagenesis and chemical modifications. Additionally, we explore how antibody-drug conjugates (ADCs), controlled drug-delivery methods, and nanotechnology-based approaches can enhance bioavailability and therapeutic efficacy. We also cover initial antibody discovery, including high-throughput screening and advanced biophysical characterization techniques that ensure mAb functionality, specificity, and structural integrity. This comprehensive overview of current advances and challenges in biologics should serve as a valuable guide for academic researchers and biopharmaceutical professionals, providing essential knowledge and tools for navigating mAb development and advancing this therapeutics toward clinical success.
