BACKGROUND: Hydrogen sulfide (H2S) is a key gaseous signaling molecule involved in various physiological and pathological processes. Its abnormal levels are associated with diseases such as cancer and neurodegeneration, necessitating sensitive and selective detection tools. Fluorescent probes based on donor-π-acceptor (D-π-A) structures enable real-time, high-resolution imaging of H2S. However, challenges remain in developing probes with low background signals, rapid responses, and ultralow detection limits for use in complex biological systems.
RESULTS: We developed a D-π-A fluorescent probe (Probe-1) using benzochromene as the donor, 7-hydroxyindanone as the acceptor, and a conjugated double bond as a π-bridge. The acryloyl moiety functions as both the PET-based quencher and the Ester Bond function as H2S recognition unit. Upon H2S exposure, ester bond cleavage occurs, restoring fluorescence at 603 nm within 3 min. Probe-1 exhibits excellent linearity from 0 to 0.1 pM (y = -2566.73 log10x + 1635.69 (R2 = 0.998) with a detection limit of 0.06 pM. It enables efficient H2S imaging in murine tumor models.
SIGNIFICANCE: This study presents a low-background, "turn-on" fluorescent probe with ultrahigh sensitivity for H2S detection. Its rapid response and biological applicability support its use in real-time imaging and disease monitoring. Probe-1 offers a valuable platform for studying H2S-related physiological and pathological processes.
